In this study, hot water and ethanol, acetone, and hexane extracts of the steamed Deodeok were investigated for use as raw materials for natural cosmetics. Hot water and ethanol, acetone and hexane extracts of steamed Deodeok did not indicate cell toxicity up to 300 ㎍/mL concentration in the MTT assay. The tyrosinase inhibition effects of the ethanol extract were higher than the ascorbic acid. The efficacy of the steamed Deodeok's ethanol extract on the elastase inhibition was similar to that of quercetin at higher concentrations. In conclusion, water, ethanol, acetone and hexane extracts of the steamed Deodeok indicated strong possibility as a whitening and anti-wrinkle functional cosmetic ingredient. Among them, the steamed Deodeok's ethanol extract, which showed excellent activity, is highly available because it has high safety compared to other organic solvent extraction methods used in cosmetics, and has minimal irritation to the skin.
UV exposure induces matrix metalloproteinases (MMPs) and extracellular matrix-degrading enzymes expression. We studied the protective effect of Rubus occidentalis seed against UVB-generated skin photoaging using human fibroblast cells (CCD-986sk). We used an ELISA kit to measure the supernatents of procollagen type I and MMP-1 in CCD-986sk cells after they were exposed to UVB irradiation. The CCD-986sk cells that were used with RC-E/E after the UVB irradiation caused higher levels of type I procollagen and lesser levels of MMP-1 compared with the control group. Furthermore, the RC-E/E treated group showed lesser MMP-1 levels and higher procollagen type I levels than the untreated counterpart. Therefore, it can be concluded that Rubus occidentalis seed can prevent from skin photoaging.
Objectives: This study aims to examine the improvement against facial wrinkles after needle-embedding therapy. Methods: The study participants were 10 women treated with needle-embedding therapy in the Korean Medical Clinic from September, 2011 to August, 2014. The subjects were treated every ten days for twenty days, totally three times, and the result of treatment was evaluated five times, such as pre-treatment, after each treatments, one month and two months later of the last treatment with ARAMO-SG. Results: 1. Depth and range of facial wrinkles, facial skin pore test, and sensitivity test were visibly improved after needle-embedding therapy and follow-up compared pre-treatment (p<0.05). 2. The significant improvements in facial skin texture test was not observed and sustained until the follow-up measurement at 2 months. 3. Any adverse reaction related to needle-embedding therapy did not happen. Conclusions: This study suggests that needle-embedding therapy can improve facial wrinkles. The positive results of this study support the requirement for additional research investigating the efficacy of needle-embedding therapy in women.
Pine bark extract is well-known as a very powerful antioxidant, anti-inflammatory, and antibiotic material. French maritime pine bark extract ($Pycnogenol^{(R)}$) of Horphag Research has monopolized the world market over 30 years. Korean red pine bark extract ($Pinexol^{(R)}$) was first manufactured by the patent technology of NutraPharm in Korea in 2006. Feasibility of topical gel and patch formulations of Pinexol was systematically investigated by evaluating the skin absorption of catechin as a reference compound. In vitro hairless mouse skin absorption of catechin from gel formulation was higher than that from patches. However, significant amount of catechin was also deposited inside the skin from patch formulations, which were dependent on the types of pressure sensitive adhesives. Thus, it seems to be feasible to control the topical delivery of Pinexol by using both gel and patch formulations, and be necessary to conduct further systematic investigation.
Low-molecular weight peptides derived from fish collagen exhibit several bioactivities, including antioxidant, antiwrinkle, antimicrobial, antidiabetic, and antihypertension effects. These peptides are also involved in triglyceride suppression and memory improvement. This study aimed to investigate the optimal processing condition for preparing low-molecular weight peptides from flounder skin, and the properties of the hydrolysate. The optimal processing conditions for peptic hydrolysis were as follows: a ratio of pepsin to dried skin powder of 2% (w/w), pH of 2.0, and a temperature of 50℃. Peptic hydrolysate contains several low-molecular weight peptides below 300 Da. Gly-Pro-Hyp(GPHyp) peptide, a process control index, was detected only in peptic hydrolysate on matrix-assisted laser desorption/ionization-time-of-flight(MALDI-TOF) spectrum. 2,2'-azinobis-(3-3-ethylbenzothiazolline-6- sulfonic acid(ABTS) radical scavenging activity of the peptic hydrolysate was comparable to that of 1 mM ascorbic acid, which was used as a positive control at pH 5.5, whereas collagenase inhibition was five times higher with the peptic hydrolysate than with 1 mM ascorbic acid at pH 7.5. However, the tyrosinase inhibition ability of the peptic hydrolysate was lower than that of arbutin, which was used as a positive control. The antibacterial effect of the peptic hydrolysate against Propionibacterium acne was not observed. These results suggest that the peptic hydrolysate derived from a flounder skin is a promising antiwrinkle agent that can be used in various food and cosmetic products to prevent wrinkles caused by ultraviolet radiations.
To evaluate the protective effect of collagen peptide-coated coffee extract on skin aging, cell viability was measured with a MTT assay using cultured CCD-986sk fibroblasts, and its effect on wrinkles in the skin of hairless mice induced by UVB-irradiation was examined. In addition, its effect on procollagen synthesis and anti-oxidative, and its inhibitory activity against collagenase, elastase, tyrosinase and MMP-1 were analysed. After the 30-minute topical treatment, the animals were exposed to UVB irradiation (60-100 mJ/cm2) for 4 weeks and its intensity increased during the period. Under the experimental conditions set in this study, the skin thickness of hairless mice significantly decreased (11.8-21.3%) compared to the control group. Based on these results, the prolonged oral intake of a collagen peptide mixture with coffee is expected to significantly increase the synthesis of procollagen in dermal fibroblasts, thereby contributing to the alleviation of wrinkling and lowered elasticity due to structural damage to the dermal layer caused by UV. The oral intake of collagen-coated coffee contributes to increasing collagen biosynthesis in a dose-dependent manner and alleviates the symptoms of thickened keratin caused by UV irradiation. However, it did not inhibit the enzymes involved in skin aging, whitening, wrinkle improvement, and antioxidation. Based on the these results, it can be concluded that the intake of collagen peptide-coated coffee extract can be utilized as an alternative material for the prevention or treatment of diseases associated with photoaging.
사람의 피부는 자외선, 오염된 공기, 화학제품 등 환경에 끊임없이 노출된다. 특히 자외선은 피부에 다양한 형태로 영향을 주어 주름, 잔주름, 피부거침, 피부건조와 같은 현상을 발생시켜 피부노화를 유발시킨다. 본 연구에서는 전 세계적으로 향나무과의 일종인 다년생 수목인 노간주나무(Juniperus rigida Sieb.) 추출물의 주름개선 효과에 대하여 검증하였다. 주름개선 효과 측정으로 elastase 저해활성을 측정한 결과 노간주나무 열수 추출물 $1,000{\mu}g/ml$에서 10% 이하의 낮은 저해활성을 나타낸 반면에 노간주나무 에탄올 추출물의 $1,000{\mu}g/ml$에서 68.5%의 저해활성을 나타내었으며, collagenase 저해활성을 측정한 결과 $1,000{\mu}g/ml$에서 열수 및 에탄올 추출물은 44.9%, 97.2%로 에탄올 추출물의 경우 ascorbic acid (99.6%)와 유사한 효과로 높은 저해활성을 나타내었다. 또한 섬유아 세포를 이용한 collagen 생합성량을 측정한 결과 노간주나무 에탄올 추출물 $50{\mu}g/ml$에서 151.52%의 생합성 촉진 효과를 나타내어 노간주나무 에탄올 추출물의 주름개선 효과가 우수하다는 것을 확인할 수 있었다. UVA에 의해 발현이 증가되는 MMP-1에 노간주나무 에탄올 추출물이 미치는 영향을 알아본 결과 노간주나무 에탄올 추출물은 $1,00{\mu}g/ml$에서 67.1%의 MMP-1 발현 저해효과를 나타내었다. 노간주나무 에탄올 추출물의 MMP-1의 단백질 및 mRNA의 발현은 $50{\mu}g/ml$의 농도에서 각각 35%, 39%의 저해율을 나타냄에 따라 노간주나무 에탄올 추출물은 collagen을 분해하는 효소인 MMP-1의 발현을 억제하고 광노화에 의한 주름생성억제 활성을 보이는 것을 확인하였다.
본 실험에서는 안전하고 우수한 효과를 지닌 천연항산화제의 광노화에 의한 피부주름 형성을 억제하는 천연물질을 탐색하기 위한 연구의 일환으로 CTL80의 식이투여와 로션 도포가 광노화를 유발한 동물모델에서 주름 형성에 미치는 영향을 살펴보았다. 광노화 유발 동물모델을 만들기 위해 hairless mice에 10주 동안 UVB 조사를 주 3회 실시하였다. 형태학적 변화를 관찰한 결과 UVB를 조사한 군은 각질이 증가하고 주름 능선의 두께가 굵고 주름이 깊게 형성한 반면, 양성대조군인 비타민 C와 CTL80 처리한 군에서 주름 형성이 감소하였으며, 특히 2% CTL80 로션 도포군이 가장 주름 형성을 억제하였다. 피부조직의 H&E 염색을 통한 조직학적인 변화에서는 UVB 조사로 두꺼워진 각질층과 표피층을 관찰하였으며 진피층의 교원섬유가 감소되었음이 관찰되었다. CTL80은 각질층과 표피층의 증가를 감소시켰으며 진피층의 교원섬유를 증가시켰음을 관찰하였다. 형태학적 변화와 같이 2% CTL80 로션 도포군의 효과가 가장 양호하게 나타났다. 항산화 상태에 미치는 영향을 살펴보기 위해 동물 혈청의 SOD와 GSH-Px의 활성을 측정한 결과, 비타민C와 CTL80처리 군에서 UVB만 조사한 군에 비하여 활성이 유의적으로 높아졌음을 살펴볼 수 있었다. 항산화 활성 또한 CTL80에 농도 의존적이지는 않았으나 식이 투여군에 비하여 로션 도포군에서 높은 활성을 보였다. 교원질 합성을 억제하고 분해를 촉진시키는 MMPs(MMP-1, MMP-3, MMP-9)의 발현을 실시간 정량 PCR로 측정하여 발현 변화를 살펴보았다. 그 결과 식이 투여군과 로션 도포군 모두 UVB 조사군에서는 발현이 현저히 높아졌으나 비타민 C와 CTL80 처리 군에서 유의적으로 낮아졌음을 살펴볼 수 있었다. 특히 CTL80 로션 도포군에서 MMP-9의 발현이 확연히 낮아졌음을 관찰할 수 있었다. 이러한 결과를 통하여 CTL80은 높은 항산화 활성을 지녔으며 이러한 항산화능을 통하여 UVB로 광노화를 유발시킨 동물모델에서 주름 생성 억제에 긍정적인 영향을 미쳤고 주름 억제 효능을 지니는 기능성 소재로써 개발 가능성을 확인하였다.
본 연구에서는 한방 발효주 주박의 피부 생리 기능 활성을 알아보기 위해 주박 추출물 및 분획물을 준비하고 항산화, 미백 및 주름 개선 효과를 조사하였다. 그 중 한방 발효주 주박의 에틸아세테이트 분획물(KSD E4-3)이 가장 우수한 항산화 효과를 나타내었으며 이는 다양한 약재의 발효에 의한 효과로 사료된다. KSD E4-3은 tyrosinase 활성 억제 및 tyrosine을 기질로 melanin이 형성되는 pathway에 관여하는 주요한 인자인 TRP-1과 TRP-2를 저해하는 작용 기전을 통해 피부 색소침착의 주요 원인 물질인 melanin 생합성을 농도 의존적으로 저해하는 것을 확인하였다. 또한 KSD E4-3은 피부의 keratinocyte가 생성, 분비하는 MMP-1, -2, -9의 발현을 억제하여 UV 조사에 의한 피부 광노화에 따른 피부 주름 생성을 억제할 수 있음을 확인하였다. 이상의 결과를 종합하면 한방 발효주 주박에서 추출한 KSD E4-3은 피부 미백 및 주름 개선에 우수한 효과를 나타내고 있으므로 기능성 화장품의 주요한 소재로 이용 가치가 높을 것으로 사료된다.
In vitro 항산화 활성을 갖는 율피 추출물의 향장 소재 특성을 조사하였다. 율피로부터의 다양한 추출물 중 총 페놀성 화합물 함량은 60% 메탄올 추출물 및 에틸아세테이트 분획물이 각각 164.82 및 191.14 mg/g으로 가장 높은 것으로 나타났다. 또한, 상기 두 분획물에 의한 in vitro 항산화 활성(DPPH radical scavenging activity, lipid peroxidation 저해)은 농도 의존적으로 증가한다는 것을 확인하였다. 상기 두 분획물은 UV-B(290-320 nm) 영역에서 강한 UV 흡수도를 보여주었다. 특히, 율피 유래 두 분획물은 $1000{\mu}g/mL$에서 42.88 및 33.02%의 elastase 저해 활성을 나타내어 우수한 주름 개선 특성을 가지는 것을 보여주었다. 아울러, 율피로부터의 60% 메탄올 추출물 및 에틸아세테이트 분획물은 피부 상재균에 대하여서도 항균 활성을 지니는 것으로 나타났다. 이러한 결과들은 율피에서 추출될 수 있는 물질이 향장 산업소재로서 활용될 수 있는 가능성을 제시하는 것으로 판단된다.
