급성 위염 동물 모델에서 감국(甘菊) 추출물과 아마인유(亞麻仁油) 혼합물의 위 점막 보호 효과 (Protective Effects of Chrysanthemi Indici Flos Extract and Flaxseed Oil Mixture on HCl/ethanol-induced Acute Gastric Lesion Mice)

  • Objectives : The objective of this study was to investigate the protective effect of Flaxseed oil and Chrysanthemi Indici Flos 50% ethanol extract in an HCl/ethanol induced acute gastritis model. Methods : ICR mice were divided into 6 groups; normal mice (Nor), gastritic mice with distilled water (Veh), gastritic mice with 10 mg/kg sucralfate (SC), gastritic mice with 16 g/㎏ Flaxseed oil (FO), gastritic mice with FO + 50 mg/kg Chrysanthemi Indici Flos (FCL), and gastritic mice with FO + 100 mg/kg Chrysanthemi Indici Flos (FCH). Then, mice were orally administered with 150 mM HCl/60% ethanol and caused acute gastritis. After 1 hr, mice were sacrificed, and blood and stomach tissue were collected. Results : Administration of FCL and FCH to mice prior to the induction of gastritis was found to reduce gastric injury. reactive oxygen species (ROS) and peroxy nitrite ($ONOO^-$) levels of stomach tissues were significantly decreased in FO, FCL, and FCH compared to Veh group. As results of stomach protein analyses, FCL and FCH effectively reduce inflammatory-related factors such as inducible nitric oxide synthase (iNOS), interleukin-6 (IL-6), and interleukin 1 beta ($IL-1{\beta}$) in gastric lesion mice. In addition, nuclear factor kappa B p65 ($NF-{\kappa}B$ p65) and phosphorylation inhibitor of nuclear factor kappa $B{\alpha}(p-I{\kappa}B{\alpha})$ were down-regulated in FCL and FCH administrated gastric lesion mice. Conclusions : These results suggest that FCL and FCH has an inhibitory effect against gastric injury. Therefore, FCL and FCH has the potential to be used as a natural therapeutic drug.

케나프 장대 품종의 부위별 에탄올 추출물의 항혈전 및 항산화 활성 (Evaluation of Antithrombosis and Antioxidant Activities of the Ethanol Extract of Different Parts of Hibiscus cannabinus L. cv. 'Jangdae')

  • 세계 4대 섬유작물중의 하나로 알려진 케나프(Hibiscus cannabinus L.)는 탄소 저감용 및 친환경 생활소재용으로 각광받고 있으며, 2013년 국내 채종 가능한 '장대' 품종이 확립되면서 전국적으로 재배되고 있다. 그러나, 케나프의 부위별 생리활성, 특히 '장대'의 부위별 생리활성 평가는 미미한 실정이다. 본 연구에서는 케나프 '장대'의 종실(seed), 종자 꼬투리(seedpod), 잎(leaf), 줄기(stem), 뿌리(root) 에탄올 추출물을 조제하여 이의 항혈전 및 항산화 활성을 평가하였다. 그 결과 종실 추출물에서 강력한 혈액응고인자 저해활성을, 종자 꼬투리 추출물에서 프로트롬빈 저해 활성을, 줄기 추출물에서 트롬빈 저해활성을 확인하였다. 또한 종자 꼬투리 추출물에서는 항혈전 활성과 연관된 DPPH 음이온 소거능, ABTS 양이온 소거능, 환원력 및 nitrite 소거능이 여타의 부위 추출물보다 유의적으로 강력한 활성을 나타냄을 확인하였다. 본 연구결과는 국내 케나프 친환경 섬유산업의 부산물로 얻어지는 종자 꼬투리, 뿌리 및 줄기를 이용한 고부가가치 생물소재 개발이 가능함을 제시하고 있다.

Rutin alleviated lipopolysaccharide-induced damage in goat rumen epithelial cells

  • Objective: Rutin, also called vitamin P, is a flavonoids from plants. Previous studies have indicated that rutin can alleviate the injury of tissues and cells by inhibiting oxidative stress and ameliorating inflammation. There is no report on the protective effects of rutin on goat rumen epithelial cells (GRECs) at present. Hence, we investigated whether rutin can alleviate lipopolysaccharide (LPS)-induced damage in GRECs. Methods: GRECs were cultured in basal medium or basal medium containing 1 ㎍/mL LPS, or 1 ㎍/mL LPS and 20 ㎍/mL rutin. Six replicates were performed for each group. After 3-h culture, the GRECs were harvested to detect the relevant parameters. Results: Rutin significantly enhanced the cell activity (p<0.05) and transepithelial electrical resistance (TEER) (p<0.01) and significantly reduced the apoptosis rate (p<0.05) of LPS-induced GRECs. Rutin significantly increased superoxide dismutase, glutathione peroxidase, and catalase activity (p<0.01) and significantly decreased lactate dehydrogenase activity and reactive oxygen species and malondialdehyde (MDA) levels in LPS-induced GRECs (p<0.01). The mRNA and protein levels of interleukin 6 (IL-6), IL-1β, and C-X-C motif chemokine ligand 8 (CXCL8) and the mRNA level of tumor necrosis factor-α (TNF-α) and chemokine C-C motif ligand 5 (CCL5) were significantly increased in LPS-induced GRECs (p<0.05 or p<0.01), while rutin supplementation significantly decreased the mRNA and protein levels of IL-6, TNF-α, and CXCL8 in LPS-induced GRECs (p<0.05 or p<0.01). The mRNA level of toll-like receptor 2 (TLR2), and the mRNA and protein levels of TLR4 and nuclear factor κB (NF-κB) was significantly improved in LPS-induced GRECs (p<0.05 or p<0.01), whereas rutin supplementation could significantly reduce the mRNA and protein levels of TLR4 (p<0.05 or p<0.01). INS-1 췌장 베타 세포에서 ferulic acid의 당독성 개선 효과 (Ferulic Acid Protects INS-1 Pancreatic β Cells Against High Glucose-Induced Apoptosi)

  • 제 2형 당뇨병에서 나타나는 인슐린 분비 감소는 베타세포의 자가사멸에 의한 베타세포질량의 급격한 감소로 인한 것으로 보고되고 있으며, 베타세포의 자가사멸을 촉진하는 요인으로 고혈당에 의한 당독성 및 활성산소종들의 증강에 의한 산화스트레스 등이다. Ferulic acid는 항산화, 항염, 항암 등 다양한 생리활성을 나타내며, 본 연구에서는 고혈당으로 유도된 세포 당독성 개선 효과와 그 기전을 INS-1 췌장 베타세포에서 규명하고자 하였다. Ferulic acid는 고농도 포도당 처리된 INS-1 췌장 베타 세포에서 세포 생존율을 증가시키고, 지질과산화물, 세포 내 ROS 및 NO 수준을 감소시켰다. 세포사멸 관련 인자의 유전자 발현결과 pro-세포자가사멸 인자인 bax, cytochrome c, caspase-3 및 caspase-9의 단백질 발현을 유의적으로 감소시켰고, anti-세포자가사멸 인자인 bcl-2 발현을 증가시켰다. Ferulic acid는 annexin V/I propidium iodide 분석을 통하여 고농도 포도당으로 유도된 세포 사멸을 감소시키고, INS-1 췌장 베타세포에서의 인슐린 분비능을 증가시키는 것으로 사료된다. 따라서ferulic acid는 고농도 포도당으로 손상된 INS-1 췌장 베타세포의 보호효과를 나타낸다.

Lipopolysaccharide로 활성화된 RAW 264.7 세포에서 적하수오(Polygonum multiflorum) 추출물의 항염증 효과 검증 (Anti-inflammatory effect of Polygonum multiflorum extraction in activated RAW 264.7 cells with lipopolysaccharide)

  • 적하수오 추출물의 추출용매별 항염증 작용을 알아보기 위해 LPS로 자극된 RAW 264.7 세포를 이용하여 실험을 진행 한 결과 열수 추출물 및 70% 에탄올 추출물 모두 $100{\mu}g/mL$에서 60%이상의 NO 생성 억제 율을 보이는 것을 확인 하였다. 또한 사이토 카인들에 대하여 적하수오 추출물은 TNF-${\alpha}$, IL-$1{\beta}$, IL-6, $PGE_2$ 생성량은 농도 의존적으로 감소하였고, 이는 적하수오 추출물이 염증성 사이토 카인의 발현을 억제 하여 염증 발생을 억제 할 수 있다고 할 수 있다. PMW는 $100{\mu}g/mL$의 농도에서 iNOS 단백질 발현량이 89%, COX-2는 54%의 효과를 나타내었으며, PME는 $100{\mu}g/mL$의 농도에서 iNOS 91%, COX-2는 57%의 단백질 발현 저해효과가 있음을 확인 할 결과로 보아 적하수오는 대식세포에서 NO, TNF-${\alpha}$, IL-$1{\beta}$, IL-6, PGE2 iNOS, COX-2 발현을 억제 하며, 특히 적하수오 70% 에탄올 추출물의 경우 저 농도에서부터 효과를 나타내어 우수한 항염증 소재임을 확인 할 수 있었다.

회화나무 꽃, 열매, 가지 추출물의 피부개선 효과에 관한 연구 (Study on the Effects of the Extracts from Sophora Japonica L. Flowers, Fruits and Branches on Improvements in Skin Condition)

  • 본 연구에서는 Sophora japonica L. (S. japonica)학명의 콩과 식물 낙엽교목인 회화나무의 꽃, 열매, 가지 에탄올 추출물의 총 폴리페놀 함량 및 l,1-diphenyl-2-picryl-hydrazyl (DPPH) 라디칼 소거능 측정, 티로시나제 활성 저해력 측정 및 porcine pancreatic elastase (PPE) 저해력 측정을 통하여 기능성 화장품 소재로서의 활용 가능성을 확인하였다. 그 결과 총 폴리페놀 함량은 평균 각각 261.37 mg/g, 187.49 mg/g, 81.26 mg/g이며, DPPH 라디칼 소거능을 조사한 결과 50~1,000 mg/L 농도에서 회화나무 꽃 추출물은$17.68{\pm}1.59{\sim}51.40{\pm}1.04%$, 열매 추출물은 $27.48{\pm}0.22{\sim}50.89{\pm}0.13%$, 가지 추출물은 $30.79{\pm}0.55{\sim}45.17{\pm}0.83%$ 범위의 소거 활성을 나타내었다. 또한 같은 농도에서 티로시나제 저해능과 PPE 저해능을 실험한 결과 회화나무 꽃 추출물은 $0.27{\pm}0.12{\sim}11.38{\pm}0.57%$, $3.70{\pm}1.23{\sim}7.28{\pm}1.01%$ 열매 추출물은 $0.27{\pm}0.02{\sim}0.82{\pm}0.27%$, $3.06{\pm}2.13{\sim}13.03{\pm}2.99%$, 가지 추출물은 $0.09{\pm}0.16{\sim}0.55{\pm}0.27%$, $6.00{\pm}0.96{\sim}9.71{\pm}0.44%$ 범위로 티로시나제 저해능은 꽃 추출물이 높게 나타났다. 본 연구에서 회화나무의 피부개선에 미치는 효능, 효과를 살펴본 결과 꽃, 열매, 가지 추출물이 각 부위별로 효과가 상이하게 나타남을 알 수 있었다.

  • Effects of Sotosaja hwan on Blood Glucose, Hyperlipidemia, Polyol Pathway and Antioxidative Mechanism in ob/ob Mouse Diabetes is a disease in which the body does not produce or properly use insulin. Etiological studies of diabetes and its complications showed that oxidative stress might play a major role. Therefore, many efforts have been tried to regulate free oxygen radicals for treating diabetes and its complications. Sotosaja-hwan has been known to be effective for the antiaging and composed of four crude herbs. In male ob/ob mouse in severe obesity, hyperinsulinemia and hyperlipidemia, which are features of NIDDM, the hyperglycemic activites and mechanisms of Sotosaja-hwan were examined. Mice were grouped and treated for 5 weeks as follows. Both the lean (C57/BL6J black mice) and diabetic (ob/ob mice) control groups received standard chow. The experimental groups were fed with a diet of chow supplemented with 30 and 90 mg Sotosaja-hwan per 1 kg of body weight for 14 days. The effects of Sotosaja-hwan extract on the ob/ob mice were observed by measuring the serum levels of glucose, insulin, lipid components, and the kidney levels of superoxide anion radical $({\cdot}O_2)$, MDA+HAE, GSH/GSSG ratio, and also the enzyme activities involved in polyol pathway. Sotosaja-hwan lowered the levels of serum glucose and insulin in a dose dependent manner. Total cholesterol, triglyceride and free fatty acid levels were decreased, while the HDL-cholesterol level was increased, in Sotosaja-hwan treated groups. Renal aldose reductase and sorbitol dehydrogenase activities were increased in the ob/ob mice, whereas those were inhibited in the Sotosaja-hwan-administered groups. Sotosaja-hwan inhibited the generation of ${\cdot}O_2$ in the kidney. Finally, MDA+HAE levels was increased and GSH/GSSG ratio was decreased in the ob/ob mice, whereas those were improved in the Sotosaja-hwan-administered groups. Sotosaja-hwan showed the antidiabetic and anti hyperlipidemic activities by regulating the activities of polyol pathway enzymes, scavenging reactive oxygen species and reducing the MDA+HAE levels in the ob/ob mice.

Effects of Woo-Gui-Um on A${\beta}$ Toxicity and Memory Dysfunction in Mice

  • Objectives : Alzheimer's disease (AD) is characterized by neuronal loss and extracellular senile plaque. Moreover, the cellular actions of ${\beta}$-amyloid (A${\beta}$ play a causative role in the pathogenesis of AD. This study was designed to determine whether Woo-Gui-Um, a commonly used Korean herbal medicine, has the ability to protect cortical and hippocampal neurons against A${\beta}_{25-35}$ neurotoxicity Methods : In the present study, the authors investigated the preventative effects of the water extract of Woo-Gui-Um in a mouse model of AD. Memory impairment was induced by intraventricularly (i.c.v.) injecting A${\beta}_{25-35}$ peptides into mice. Woo-Gui-Um extract was then administered orally (p.o.) for 14 days. In addition, A${\beta}_{25-35}$ toxicity on the hippocampus was assessed immunohistochemically, by staining for Tau, MAP2, TUNEL, and Bax, and by performing an in vitro study in PC12 cells. Results : Woo-Gui-Um extract had an effect to improve learning ability and memory score in the water maze task. Woo-Gui-Um extract had significant neuroprotective effects in vivo against oxidative damage and apoptotic cell death of hippocampal neurons caused by i.c.v. A${\beta}_{25-35}$. In addition, Woo-Gui-Um extract was found to have a protective effect on A${\beta}_{25-35}$-induced apoptosis, and to promote neurite outgrowth of nerve growth factor (NGF)-differentiated PC12 cells. Conclusions : These results suggest that Woo-Gui-Um extract reduces memory impairment and Alzheimer's dementia via an anti-apoptotic effect and by regulating Tau and MAP2 in the hippocampus.

자외선B 노출로 인해 손상된 피부세포에 대한 돌배나무잎 추출물의 보호효과 (Protective Effects of Pyrus pyrifolia NAKAI Leaf Extracts on UVB-induced Toxicity in Human Dermal Fibroblasts)

  • 피부 손상은 주로 자외선, 열, 담배 등과 같은 환경적 요인으로부터 초래되는데, 이는 활성산소종의 과생성으로 인한 피부노화와 연관이 있는 것으로 알려져 있다. 돌배(Pyrus pyrifolia NAKAI)는 전 세계적으로 많이 소비되는 과일로써 항암, 항산화, 항염증효과가 알려져 있다. 본 연구에서는 돌배나무잎 추출물(Pyrus pyrifolia leaf extract, PPE)의 ultraviolet B (UVB)스트레스에 대한 피부 섬유아세포 보호효과를 검증하였다. Lactate dehydrogenase assay와 DCF-DA를 이용한 정성분석 실험은 PPE가 인간의 섬유아세포에서 UVB 스트레스에 의해 유발된 세포독성 및 과생성된 활성산소종을 농도 의존적으로 억제할 뿐만 아니라, 미토콘드리아 기능저하, 막전위 저하, 그리고 세포사멸과정의 핵심 인자인 caspase-3 활성도 유의하게 억제함을 보여주었다. 결론적으로, PPE는 UVB스트레스에 의해 과생성된 활성산소종을 억제시켰으며, 이로 인해 생기는 피부세포 사멸을 효과적으로 저해함을 확인하였다.

Effects of Tributyrin on Intestinal Energy Status, Antioxidative Capacity and Immune Response to Lipopolysaccharide Challenge in Broilers

  • This study was carried out to investigate the effects of tributyrin (TB) on the growth performance, pro-inflammatory cytokines, intestinal morphology, energy status, disaccharidase activity, and antioxidative capacity of broilers challenged with lipopolysaccharide (LPS). A total of 160 one-day-old Cobb broilers were allocated to 1 of 4 treatments, with 4 replicated pens per treatment and 10 birds per pen. The experiment consisted of a $2{\times}2$ factorial arrangements of treatments with TB supplementation (0 or 500 mg/kg) and LPS challenge (0 or $500{\mu}g/kg$ body weight [BW]). On days 22, 24, and 26 of the trial, broilers received an intraperitoneal administration of $500{\mu}g/kg$ BW LPS or saline. Dietary TB showed no effect on growth performance. However, LPS challenge decreased the average daily gain of broilers from day 22 to day 26 of the trial. Dietary TB supplementation inhibited the increase of interleukin-$1{\beta}$ (in the jejunum and ileum), interleukin-6 (in the duodenum and jejunum), and prostaglandin $E_2$ (in the duodenum) of LPS-challenged broilers. Similar inhibitory effects of TB in the activities of total nitric oxide synthase (in the ileum) and inducible nitric oxide synthase (in the jejunum) were also observed in birds challenged with LPS. Additionally, TB supplementation mitigated the decrease of ileal adenosine triphosphate, adenosine diphosphate and total adenine nucleotide and the reduction of jejunal catalase activity induced by LPS. Taken together, these results suggest that the TB supplementation was able to reduce the release of pro-inflammatory cytokines and improve the energy status and anti-oxidative capacity in the small intestine of LPS-challenged broilers.
