Anti-angiogenesis and anti-cell adhesion effects were investigated with different dose of Camellia japonica leaf (CJL) extract for applying anti-cancer, anti-metastasis and anti-obesity. Cytotoxicity on HUVECs was very low at 200 ug/mL of CJL-extract. Anti-angiogenic ratio at increasing dose of 1.5 ug/mL, 3.0 ug/mL, 15 ug/mL and 30 ug/mL was showed 30.7%, 38.5%, 53.8%, and 70.0%, respectively. Also, anti-cell adhesion effect at concentration of $50{\mu}g{/well},\;100{\mu}g{/well}\;and\;200{\mu}g{/well}$ was expressed on E-selectin by 46.7%, 66.7%, and 86.76%, on VCAM-1, 23.0%, 61.5%, and 84.6%, and on ICAM-1, 11%, 55.5%, and 88.8%, respectively. For inquiring anti-angiogenesis mechanism, when western blot was performed with different dose of CJL extract, signal molecules of VEGFR-2, $\beta$-catenin and PI3-K were suppressed. As the signal transduction from VEGFR-2, $\beta$-catenin and PI3-K to NF-${\kappa}$B was interupted, angiogenesis could not be occurred causing not activated NF-kB. C. japonica leaf (CJL) is a useful herb for developing therapeutics of angiogenesis related diseases such as cancer, metastasis, rheumathioid arthritis and obesity.
Objectives This experimental study was designed to investigate the effects of Mulberry leaves contained herbal mixture (MLHM) on body weight, serum lipid level and adipocyte differentiation in high fat diet-fed obese mice. Methods Four-week old mice (wild-type C57/BL6) were used for all experiments. Cells were incubated with MLHM at the indicated concentration (0.04-4mg/ml) for 24h, and growth rate was assessed by MTT ((3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay. 3T3-L1 preadipocytes were incubated in DMEM for 2 days with the indicated concentrations of MLHM, and on Day 6, the cells were fixed and the cellular lipid contents were assessed by Oil-Red-O staining. The expression of peroxisome proliferator-activated receptor ${\gamma}$ (PPAR ${\gamma}$) and cytidine-cytidine-adenosine-adenosine-thymine (CCAAT)/enhancer-binding proteins ${\alpha}$ (C/EBP ${\alpha}$) as adipocyte-specific proteins were determined by real time RT-PCR and western blotting. In addition, body weight gain and serum lipid levels were measured in the mice with obesity induced by the high fat-diet for four weeks. Results Though MLHM did not show toxicity even at the concentration of 4mg/ml, MLHM significantly inhibited the differentiation of 3T3-L1 preadipocites in a dose-dependent manner. Also, MLHM significantly reduced the expressions of PPAR ${\gamma}$ and C/EBP ${\alpha}$ in a dose-dependent manner. Furthermore, MLHM significantly reduced body weight gain and LDL-cholesterol contents in high fat diet-fed obese mice. Conclusions These results demonstrate that MLHM exerts anti-obesity effect in 3T3-L1 cells and mice with obesity by high-fat diet.
Objectives : The objective of this study was to investigate the effect of Myrrh 80% ethanol extract on adipocyte differentiation and adipogenesis in 3T3-L1 cell. Methods : Myrrh was prepared by extracting with 80% ethanol. Cell viability was assessed by MTT assay using 3T3-L1 cells. Anti-obesity activity was measured in lipid droplets and triglyceride (TG) accumulation in 3T3-L1 cells. We also analyzed the expression of C/EBPβ, C/EBPα, PPARγ, SREBP1c, and aP2 by reverse transcriptase polymerase chain reaction (RT-PCR). In addition, we observed the production of fatty acid, acetyl-CoA carboxylase and Oil-red O stainingResults : No cytotoxicity from Myrrh 80% ethanol extracts was observed at the concentration of 1, 10, 100 (㎍/㎖) in 3T3-L1 cells. Treatment with Myrrh significantly suppressed the terminal differentiation of 3T3-L1 in a dose-dependent manner, as confirmed by a decrease in triglyceride and Fatty acid and Acetyl-CoA carboxylase. Also, Myrrh exhibited potential adipogenesis inhibition and downregulated the expression of pro-adipogenic transcription factors, such as sterol regulatory element binding protein-1c (SREBP-1c), peroxisome proliferator-activated receptor-γ (PPARγ), CCAAT/enhancer binding proteins α (C/EBPα) and C/EBPβ, and adipocyte expressed genes, such as adipocyte fatty acid binding protein (aP2) and Fas. In addition, lipid accumulation determined by Oil-red O staining showed that Myrrh extract had inhibitory effects on lipid accumulation in 3T3-L1 cells. Conclusions : These results suggest that Myrrh suppresses obesity factors in 3T3-L1 cells. Myrrh may be a useful medical herbs for attenuating metabolic diseases such as obesity.
Objectives : The aim of this study is to investigate Daesiho-tang(Dachaihu-tang) water extracts (DSTE) have potent anti-obesity activities in a high-fat diet-induced obesity mouse model. Methods : In this study, we designed three groups (normal diet group, high-fat diet group, high-fat diet plus DSTE group for 7-weeks oral administration). Results : Increases in body weight were inhibited by 7-weeks oral administration of DSTE at a 500 mg/kg concentration in this animal model. Results from blood lipid analysis showed that the levels of triglyceride, total cholesterol and LDL-cholesterol were significantly lowered by DSTE administration, also HDL-cholesterol was increased more than high-fat diet-induced obese mouse. To understand the underlying mechanism at the molecular level, the effects of DSTE were examined on the expression of the genes involved in lipogenesis by real-time PCR. In epididymal fat and liver of DSTE-treated mice, the mRNA level of lipogenic genes such as sterol regulatory element binding protein 1 and fatty acid synthase were decreased, which was well correlated with the reduction of the tissues weight. Conclusions : These results suggest that DSTE may have great potential as a novel anti-obesity agent.

We evaluated the effect of anti-obesitic activity of MYE (mulberry leaf + Yacon tuber) extracted from Morus alba as muberry leaf and Smallanthus sonchifolia as yacon. 1%, 3%, or 5% of MYE were treated to Sprague-Dawley rats exposed to a high-fat diet. MYE treated rats were suppressed weight during four weeks, and they lost weight significantly after six weeks. Common blood chemistry panels related to liver function revealed significant improvement in the MYE-treated groups. The expression of leptin as indicators for obesity was decreased in perirenal fat. Such results indicate that MYE could be a promising candidate for the improvement of obesity. In addition, MYE effected on deceased glucose metabolism, reducing the activities of glucose-6-phosphate dehydrogenase (G-6-PDH) and glucokinase related to glycogen synthesis. The fatty liver was observed in high-fat diet-treated rats, resulting from increased number of adipose cells and Ito cells. However, this pathologic change was significantly improved by administration of MYE. MYE have significant effects on antioxdative function and glycometabolism against high fat diet. Thereby, it seems that MYE prevent fatty liver by high-fat diet. Thus it is suggested that MYE would be worth being developed as an biofunctional food to prevent undesirable effects caused by obesity.

Objectives This study was performed to evaluate anti-obesity effects of Crataegus pinnatifida (CP) on high-fat-diet induced obese mice. Methods The experimental animals were divided into four groups: normal diet (NOR) group, high fat diet (HFD) group, HFD+Xenical (XEN) group, and HFD+CP (CP) group. NOR group was fed a normal diet and the other three groups were fed high fat diet during the experiment. After the first two weeks of diet, XEN group and CP group were administered with XEN or CP for seven weeks, respectively. After that, we measured body weight, liver weight, fat weight, food intake, and serum concentrations of lipids and liver enzymes. Also the liver, intestine, fat tissue was removed to estimate the obesity-related mRNA expressions and the stool sample was collected to analyze the gut microbiota. Results We found that body weight, fat weight, and triglyceride level were decreased significantly in CP group compared to HFD group. Also CP significantly suppressed gene expressions associated with lipogenesis and inflammation, and increased gene expressions of browning of white adipose tissue and mitochondrial biogenesis. Moreover, it shifted the microbial diversity closer to that of NOR group and increased Firmicutes/Bacteriodetes ratio. Conclusions These results suggest that CP decrease body weight, fat weight and serum triglyceride. Also it inhibit inflammation and adipogenesis, altering gut microbial diversity and abundance. In conclusion, CP could be used as a therapeutic drug for obesity via gut microbiota modulation.
Objectives: To expand and provide information on the efficacy of herbal medicines, anti-obesity effects were evaluated. In many studies, plant-derived components with anti-obesity efficacies have been investigated for their potential inhibitory effects on 3T3-L1 preadipocyte cells. The purpose of this study was to investigate the anti-obesity effects of 65 herbal medicine in 3T3-L1 preadipocyte cells. Methods: Preferentially, 3T3-L1 cells were treated with 65 herbal medicines (500 ㎍/mL) during differentiation for 8 days. Next, 3T3-L1 cells were treated with selected herbal medicines at concentrations ranging from 50 to 200 ㎍/mL during differentiation for 8 days. The accumulation of lipid droplets was determined by Oil Red O staining. The expressions of genes related to adipogenesis were measured by reverse transcription polymerase chain reaction and Western blot analyses. Results: Among the 65 kinds of herbal medicines, 13 herbal medicines that been shown to be effective against the accumulation of lipid droplets were selected. Finally, selected Banhasasim-tang and Samhwangsasim-tang showed inhibitory activity on adipocyte differentiation at 3T3-L1 preadipocytes without affecting cell toxicity. In addition, Banhasasim-tang and Samhwangsasim-tang significantly reduced the expression levels of several adipocyte marker genes including peroxisome proliferator activated receptor-γ and CCAAT/enhancer binding protein-α. Conclusions : These results suggest that the ability of Banhasasim-tang and Samhwangsasimtang has inhibited overall adipogenesis and lipid accumulation in the 3T3-L1 cells. Banhasasim-tang and Samhwangsasim-tang may be a promising medicine for the treatment of obesity and related metabolic disorders.
Boehmeria nivea (L.) Gaud., a flowering plant, has been widely cultivated in Asian countries including Korea. It has been reported that B. nivea exhibits health beneficial effects for the prevention of inflammation, oxidative stress, and virus-related diseases. In this study, we evaluated the inhibitory effect of B. nivea on adipocyte differentiation and angiogenesis. DPPH radical scavenging activities of 70% ethanol extract of B. nivea (EBN) and water extract of B. nivea (WBN) were $90.8{\pm}1.1%$ and $20{\pm}6.9%$, respectively. EBN was also effective in the reduction of adipocyte differentiation in 3T3-L1 cells. We next examined the transcriptional activity of peroxisome proliferator-activated receptor gamma ($PPAR-{\gamma}$), a pivotal target for anti-obesity. We found that treatment with rosiglitazone induced the transactivation of $PPAR-{\gamma}$. Under the same condition, $800{\mu}g/mL$ EBN reduced the transactivation of $PPAR-{\gamma}$ in rosiglitazone-induced cells. These results demonstrate that EBN-inhibited adipocyte differentiation was accompanied by $PPAR-{\gamma}$ inhibition. The study also tested whether EBN exhibits an anti-angiogenic effect by inhibiting tube formation in HUVECs. We found that EBN effectively inhibits tube formation, suggesting that EBN exhibited an anti-angiogenic effect. Taken together, B. nivea can be used as a functional food for the prevention of obesity and angiogenesis-related diseases including cancer.
Objectives : In this study, we investigated the biological activities such as anti-obesity using Samwondan ethanol extract (SWD). SWD is a complex with Salicornia herbacea Linnaeus, Saururus chinensis Baill and Houttuynia cordata Thunberg as the main raw material. Methods : The SWD was extracted 80% ethanol. 3T3-L1 preadipocytes were induced adipogenesis by differentiation media with SWD at 1 μg/mL, 10 μg/mL, and 100 μg/mL. Effect of SWD performed using MTT assay, oil red O staining (observation by microscope), and reverse transcription polymerase chain reaction. Also we measured production of triglyceride (TG), fatty acid, and acetyl-CoA carboxylase (ACC).Results : Non-cytotoxicity was in all test group from range of 1 μg/mL to 100 μg/mL on pre-adipocyte. The droplet and production of lipid were decreased significantly by the SWD. And TG was decreased by approximately 89%, 85% and 82%, upon the SWD treatment at concentration of 1 μg/mL, 10 μg/mL, and 100 μg/mL. Moreover, the SWD showed inhibitory effects on the expression of the C/EBP (CCAAT/enhaner binding protein)β, C/EBPα, and peroxisome proliferator-activated receptorγgenes in adipocytes. The SWD at 100 μg/mL concentration showed inhibitory effect on fatty acid production by 79%. Also ACC production were decreased dose-dependently.Conclusions : From the results above, we concluded that the SWD indicated significantly anti-obesity effects.

The purpose of this study was to investigate the effect of skate skin collagen peptide (SCP) according to its molecular weight (<1,000 Da or >1,000 Da) which was divided using the ultrafilatration method. The 200 mg/kg collagen peptide was administrated to obesity-induced db/db mice for 8 weeks. As the results, in collagen peptide-treated groups, body weight gain was decreased, the plasma and hepatic concentration of reactive oxygen species decreased and oxidative stress was alleviated. In addition, SCP-treated group showed the significant reduction of the protein expressions of nuclear transcription factors($NF-{\kappa}B$), enzymes(COX2, iNOS), and inflammatory cytokine(IL-6) in hepatic tissue, compared with those of the obese control group. There was a slight difference depending on the molecular weight of collagen peptide, but overall it was not significant. Therefore, SCP effectively inhibited the obesity-induced inflammatory response through attenuation of oxidative stress in the liver.
