산초 추출물의 S. mutans 항균활성 및 항산화 효능 연구 (A Study on the Anti-microbial Effect on S. mutans and Anti-oxidant Effect of Zanthoxylum pericarpium Extract)

  • Objectives : The Zanthoxylum pericarpium has been used as oriental spicy seasoning and a medicinal plant from old times. This study was performed to determine the anti-oxidant efficacy of Zanthoxylum pericarpium extract and the anti-microbial effects. Methods : We got Zanthoxylum pericarpium extract using PSE (pressurized solvent extraction) method. The anti-microbial effect of Zanthoxylum pericarpium extract was assessed on Streptococcus mutans(S. mutans) and anti-oxidant effect of the extract was assessed by measuring DPPH radical scavenging activity and SOD like activity. Results : 1. Zanthoxylum pericarpium extract had high anti-microbial activity on S. mutans. 2. DPPH radical scavenging activity significantly increased in the Zanthoxylum pericarpium extract. 3. SOD like activity also significantly increased in the Zanthoxylum pericarpium extract. Conclusions : The PSE extract from Zanthoxylum pericarpium has good anti-microbial and anti-oxidant effects in a concentration-dependent manner.

해조 다당류 추출물의 항균성 및 항염증 기능성평가 (The Functional Effects of Anti-microbial Activity and Anti-inflammatory of Seaweed polysaccharide Extracts)

  • 본 연구는 해조 다당류 추출물을 화장품 원료로 사용할 수 있는지 평가하고자 하였다. 항균 효과를 평가하기 위해 해조 다당류 추출물로 부터 Staphylococcus aureus (S. aureus) 균주에 대한 항균활성을 측정하였다. 또한, RAW 264.7 세포에서 항염증 효과가 확인되었으며, 해조 다당류 추출물을 Sprague-Dawley (SD) 흰쥐의 등쪽 피부에 적용하여 단회투여독성을 평가하였다. 그 결과, 해조 다당류 추출물은 피부섬유 아세포에서 최대 $1,000{\mu}g/mL$ 농도에서 세포 독성을 나타내지 않았다. 또한, 1 % 해조 다당류 추출물로 처리되었을 때, $1.52{\pm}0.34cm$로 생육저해환이 형성되어 S. aureus 균주에 대한 항균 활성을 확인하였다. 해조 다당류 추출물을 RAW 264.7 세포에 처리하면 농도 의존적으로 산화 질소 (NO)의 생성이 감소하고 인터루킨 1 베타 ($IL1{\beta}$), 종양 괴사 인자 알파 ($TNF{\alpha}$)와 같은 염증 관련 사이토카인의 생성이 증가하였으며, 프로스타글란딘 E2 (PGE2)가 감소 하였다. 해조 다당류 추출물을 여러 농도로 흰쥐에 투여했을 때 증상은 14 일 이상 변하지 않았으며 췌장이나 장기 무게에는 변화가 없었다. 결론적으로 우리는 해조 다당류 추출물이 세포 독성을 나타내지 않았으며 항균 및 항염증 효과가 있음을 발견하였다. 따라서, 화장품 원료로 사용하기에 적합하다고 사료된다.

  • Objectives: The purpose of this study was to investigate the in vitro anti-inflammatory, anti-pruritic and antimicrobial effects of the three herbal prescription (EST, SCT, WDT), which has been traditionally used for treating leukorrhea induced by various infections in the female genital tract. Methods: In this experiment, the anti-inflammatory effects were evaluated by Nitric oxide (NO), $Interlukine-1{\beta}$ ($IL-1{\beta}$), Interlukine-2 (IL-2), Interlukine-6 (IL-6), Tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$), Prostaglandin $E_2$ ($PGE_2$), Leukotriene $B_4$ ($LTB_4$) production amount and Inducible nitric oxide synthase (iNOS), Nuclear factor kappa B ($NF-{\kappa}B$), Cyclooxygenase-2 (COX-2) gene expression levels in RAW264.7 cells. And the anti-pruritic effects were evaluated by Histamine, Acetylcholine (ACh), Acetylcholinesterase (AChE), Substance P production amount in Mast cell/9 (MC/9) and Pheochromocytoma 12 (PC12) cells. The anti-microbial effect was measured by inhibition zone diameter on Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, Candida albicans and Aspergillus niger. Results: As a result of measuring anti-inflammatory efficacy, $IL-1{\beta}$, IL-2, IL-6, $TNF-{\alpha}$, $PGE_2$, and $LTB_4$ production amounts were significantly reduced in the EST, SCT, WDT extraction groups compared with the control group, and significantly decreased the amount of $NF-{\kappa}B$, iNOS, and COX-2 gene expression and the amount of Phospho-Inhibitor kappa B alpha ($p-I{\kappa}B-{\alpha}$)/Inhibitor kappa B alpha ($I{\kappa}B-{\alpha}$) and $NF-{\kappa}B$ p65 protein expression. In addition, As a result of measuring the anti-pruritic effect, the amounts of histamine, ACh and Substance P were significantly decreased, and AChE production was slightly decreased, but it's significance did not appear. Finally the anti-microbial effects of EST, SCT, WDT extraction groups against Pseudomonas aeruginosa, Candida albicans and Aspergillus niger was inhibited, however the growth of Escherichia coli and Staphylococcus aureus was not inhibited. Conclusions: These data suggest that EST, SCT, WDT can be used to treat patients with leukorrhea.

  • 본 연구에서는 4급 암모늄에 속하는 didecyldimethylammonium chloride (DDAC)와 유카추출물 (Yucca extract)의 합제인 아쿠아 팜세이프$^{(R)}$의 어류 질병 병원체에 대한 살균효과를 확인하기 위하여 농림수산검역검사본부의 소독제 효력시험 중 세균 및 바이러스 소독제 효력시험에 따라 수행하였다. 소독제와 병원체를 증류수, 경수 그리고 유기물 조건에서 반응시킨 후, 중화액을 이용하여 중화시킨 다음, 중화된 용액을 배지 및 주화세포에서 접종시켜 세균의 성장 및 세포변성효과를 통해 소독제의 효력배수를 결정하였다. 본 연구의 결과, 아쿠아 팜세이프$^{(R)}$는 식중독을 일으키는 공시균주 살모넬라균 (S. typhimurim)에 비하여, 어병세균이 30~40배 이상 높은 희석농도에서 소독효과를 나타내었으며, VHSV에 대하여서는 2,000~3,600배의 유효한 효력배수를 나타내었다. 따라서, 향후, 야외적용시험을 통해 실제 적용에 따른 효과를 확인할 필요가 있을 것으로 사료된다.

  • A new colloidal silver (Ag) system is present in which a fine colloidal Ag is in situ deposited onto functionalized porous poly (ethylene glycol dimethacrylate) (poly (EGDMA)) microspheres. The effectiveness of Ag deposition was investigated considering the surface characteristics of poly (EGDMA) microspheres. The result reported in this study illustrates that the control of surface area and surface functionality (in this study, a hydroxyl group) of poly (EGDMA) microspheres is an important factor that determines practically the degree of deposition of colloidal Ag. The x-ray analysis showed that Ag nanoparticles were dispersed evenly inner and outer surfaces and had a face center cubic (fee) phase. In the preservative efficacy test, the Ag-containing poly (EGDMA) microspheres had a powerful anti-bacterial activity, showing a high potential for a new preservative in cosmetic industry.

Efficacy of Sodium Hypochlorite and Acidified Sodium Chlorite in Preventing Browning and Microbial Growth on Fresh-Cut Produce

  • The use of suitable sanitizers can increase the quality of fresh-cut produce and reduce the risk of foodborne illnesses. The objective of this study was to compare the washing effects of 100 mg/L sodium hypochlorite (SH) and 500 mg/L acidified sodium chlorite (ASC) on the prevention of enzymatic browning and the growth of microbial populations, including aerobic plate counts, E. coli, and coliforms, throughout storage at $4^{\circ}C$ and $10^{\circ}C$. Fresh-cut zucchini, cucumbers, green bell peppers, and root vegetables such as potatoes, sweet potatoes, carrots, and radishes were used. Compared to SH washing, ASC washing significantly (p<0.05) reduced microbial contamination on the fresh-cut produce and prevented browning of fresh-cut potatoes and sweet potatoes during storage. More effective inhibition of aerobic plate counts and coliforms growth was observed on fresh-cut produce treated with ASC during storage at $10^{\circ}C$. Polyphenol oxidase (PPO) activity of fresh-cut potatoes and sweet potatoes was more effectively inhibited after washing with ASC. The use of 500 mg/L ASC can provide effective antimicrobial and anti-browning treatments of fresh-cut produce, including processed root vegetables.

Santalum album Linn wood and its oil: An aromatic Unani traditional medicine with versatile pharmacological activities

  • Santalum album Linn. [Family: Santalaceae] is commonly known as white sandalwood, sandal safaid and safed chandan. It is one of the most valuable trees and second costliest wood in the world. Sandalwood and its oil is extensively used in the Unani and other traditional systems of medicine as it has blood purifier, anti-inflammatory, analgesic, exhilarant, cardiotonic, antiseptic, nervine tonic and expectorant properties. It is used in skin, cardiac, liver, gastrointestinal, respiratory, integument and urogenital disorders. These uses are supported and proven by many in vitro or in vivo studies. The proven pharmacological activities of S. album are antimicrobial, anti-oxidant, anti-inflammatory, antimutagenic and anti-fatigue. The research has proven that sandal oil or its constituents have anti-microbial activity. Sandalwood oil showed skin cancer preventive effect in mice and its constituent alpha santalol showed the anticancer property. The methanolic extract of wood was confirmed for antioxidant, free radical scavenging, analgesic and anti-inflammatory activities. ${\alpha}$ and ${\beta}$ santalols present in sandal oil showed sedative effects. Sandalwood tea had a significant effect on heart muscles of frog and showed increased myocardial contractility. Its oil showed significant changes in hepatic xenobiotic metabolizing enzymes. Sandalwood oil and its major constituents showed less acute oral and dermal toxicity in laboratory animals. Hence, the aforementioned studies justify the uses of sandalwood and its oil mentioned in the classical Unani literature. However, further clinical trials are suggested to confirm its efficacy and safety in humans.

홍화잇꽃의 여드름피부 개선효과 및 세안용 생약식물소재 응용 (Efficacy of Safflower on the Acne Skin and Its Application for Facial Cleansing Biomedical Material)

  • 홍화는 예로부터 '사람의 건강에 도움이 된다'고 해서 '잇꽃'으로 불려져 왔으며 동의보감에서는 꽃잎이 홍색을 나타내기 때문에 홍화(紅花)로 소개되어 있다. 예로부터 홍화는 꽃잎을 압착후 정제된 색소를 이용하여 옷감에 천연색소를 입히는 염료와 연지의 재료로 사용되기도 하였다. 현대에 와서는 홍화의 주성분인 폴리페놀 화합물(Polyphenol cocktail) 성분이 건강식품업계에서 항노화 및 항산화물질로 알려지면서 관심의 대상이 되고 있다. 금번연구에서는 홍화의 주성분인 폴리페놀 화합물이 청소년기 혹은 장년기에서도 고민거리가 되고 있는 여드름균의 생성을 억제시킬 수 있는 항균, 항염효능이 있음을 DDT(Disk Diffusion Test) assay, MTT assay, NF-${\kappa}$B Luciferase activity inhibition assay의 in vitro법으로 확인하고 더불어 인체안면에 대한 임상시험에 사용할 세안화장품 제형의 화장비누 시제품을 제조하여 여드름피부의 개선 효과가 있음을 시험하고자 한다. 또한 이번 연구는 여드름용 화장비누 개발에 진일보하여 약용화장품 산업의 발전에 기여할 것임을 확신한다.

Eruca sativa 에칠아세테이트와 부탄올 분획물의 미백 및 항균효과를 이용한 화장품 응용연구 (Cosmetic Application Using Skin Whitening and Anti-microbial effects of Ethyl Acetate and n-Butanol Fractions from Eruca sativa)

  • Eruca sativa (E. sativa)는 루꼴라라고 불리우고 이탈리아 요리에 많이 사용되는 식물로 Brassicaceae 속에 속하는 다년생 식물이다. E. sativa를 70% 에탄올로 환류추출 및 농축하고 (ES), n-hexane (EHex), ethyl acetate (EEA), chloroform (ECHCl3), n-butyl alcohol (EBuOH) 및 water (EDW) 용매 극성별로 분획하여 미백 효능을 규명하고 화장품 소재로서의 응용성을 연구하였다. ES, EHex, EEA, ECHCl3, EBuOH 및 EDW의 mushroom tyrosinase 활성 저해를 평가한 결과 EEA가 가장 유의하게 저해하였으며, B16F10 멜라노마 세포 내에서의 멜라닌 생성을 확인한 결과 ES, EEA 및 EBuOH를 처리하였을 때 멜라닌 생성이 유의하게 저해되었으며, 특히 EEA 분획물에서 가장 효과가 뛰어났다. 피부염과 여드름을 일으키는 황색포도상구균, 여드름 원인균을 포함한 8종의 세균, 진균에 대해 ES, EHex, EEA, ECHCl3, EBuOH 및 EDW의 항균력과 방부력 시험을 평가한 결과 EEA는 8종의 균 모두 효과적으로 항균력을 나타냈다. ES를 0.05, 0.1%를 함유하여 제조한 토너와 에멀션은 기존 처방에서 방부력을 증가시켜 천연보존제 역할을 할 수 있을 것으로 판단된다. ES를 토너, 에멀션 제형에 0.05, 0.1%로 첨가하여 일광, -20, 4, 25, 55 ℃ 조건에서 4 개월동안 안정성을 평가한 결과 pH 및 점도에 대한 영향은 크게 없었으나 대조군 및 ES를 첨가한 에멀션 55 ℃에서 분리가 일어나는 것을 확인하였다. 또한, 피부침투시험을 통해 EEA는 6 hr에서 0.058%가 침투되어 임상적인 효능을 예측할 수 있었다. 천연소재 E. sativa는 미백 기능성화장품 소재 및 천연 항균 보존제로 활용 가치가 있을 것으로 사료된다.

Propolis 추출물의 병원성 미생물에 대한 항균 효과 (Antibacterial Effects of Propolis Extracts on Pathogenic Bacteria)

  • The optimal concentration of ethanol to separate a high quantity of propolis was $60\%$ but that for the best flavonoids extraction was $80\%$ We compared the yields of propolis from different countries. In this study we used $60\%$ ethanol concentration as a standard. The yield of propolis was proportional to the contents of flavonoids. Namely, Polish propolis which showed the highest yield with $56\%$ by the extraction with $60\%$ ethanol revealed also the highest flavonoids content with $3.49\%$ among all the samples tested The major constituents of propolis differed from country to country. It has been suggested that the different geographical origin influenced the efficacy and the constituents of propolis. Antibacterial activity of ethanol extracted propolis from different countries was tested against 6 microbial strains of type cultures including Gram-positive (Staphylococcus aureus, Streptococcus uberis, Streptococcus agalactiae) and Gram- negative bacteria (Klebsiella pneumoniae, Proteus vulgaris and E coli) in vitro. Propolis extract showed anti-microbial activity against all the tested bacterial strains. In addition, propolis was sensitive to E coli which was resistant to broad spectrum antibiotics like ampicillin. These results showed that propolis may substituted for commercial antibiotics. The efficiency of anti-microbial activity of the propolis was slightly higher in $80\%$ than $97\%$ ethanol extract.
