선이질풀 추출물의 멜라닌 생성 억제 효과 (The Anti-melanogenic Effect of Geranium krameri Extract)

  • 본 연구에서 국립원예특작과학원 인삼특작부 추출물은행에서 분양받은 에탄올 추출물 시료(시료번호: 4)인 선이질풀(Geranium krameri)에 대한 다양한 생리활성을 조사하여 기능성소재 응용가능성을 검토하였다. 선이질풀 추출물은 멜라노마 세포에 대하여 낮은 세포독성을 나타냈다. 세포독성이 거의 없는 농도에서 선이질풀 추출물 처리 시 산화방지 활성($IC_{50}$, $8.72{\mu}g/mL$) 및 B. subtilis, E. coli, C. albicans에 대한 항균활성이 우수하게 나타났으며 타이로시네이즈 활성저해($IC_{50}$, $456.86{\mu}g/mL$) 및 멜라닌 함량 저하($IC_{50}$, $50.35{\mu}g/mL$)를 보여주었다. 또한, B16F10 세포에서 선이질풀 추출물 농도 의존적으로 타이로시네이즈 발현이 억제되었으며, 이는 선이질풀 추출물이 직접적인 타이로시네이즈 활성저해 및 세포 내 타이로시네이즈 발현을 억제시킴으로서 멜라닌 합성을 저해하는 것으로 판단된다. 이와 같은 결과로 미루어볼 때 선이질풀 추출물은 피부미백 소재 등 피부개선 효과를 지닌 기능성 화장품에 활용하기 위한 매우 효과적인 재료가 될 수 있다고 판단된다.

참느릅나무에서 추출된 catechin 유도체 화합물의 멜라닌 생성 억제 효과 (Tyrosinase Inhibition-mediated Anti-melanogenic Effects by Catechin Derivatives Extracted from Ulmus parvifolia)

  • 자외선으로 야기되는 피부조직의 손상에 대한 방어기전으로 멜라닌세포는 멜라닌 색소를 생산한다. 이러한 멜라닌 생성에는 tyrosinase 효소가 중추적인 역할을 수행한다. 그러나 과다한 멜라닌 생성은 피부조직에 주근깨와 검버섯 등의 과다 색소침착 병변을 일으킨다. 따라서 tyrosinase 발현이나 활성을 조절함으로써, 과도한 멜라닌 생성을 억제하는 물질을 발굴하는 것은 임상적인 의미가 있다. Catechin은 tyrosinase 발현을 저해함으로써 멜라닌 생성을 억제하는 화합물로 알려져 있다. 본 연구에서는 B16F10 마우스 흑색종 세포주와 버섯에서 추출된 tyrosinase를 사용하여, 참느릅나무의 줄기 껍질에서 추출 및 동정된 catechin 유도체 화합물(C5A, C7A, C7G, 및 C7X)의 멜라닌 생성 억제 효능을 검증하였다. 먼저, B16F10 세포주에서, 세포독성을 나타내지 않는 농도의 catechin 유도체 화합물 처리는, 모두α-MSH로 촉진된 멜라닌 생합성을 억제하는 효능을 보였다. Catechin 유도체 화합물의 이러한 효능은 멜라닌 생합성 저해제로 잘 알려진 kojic acid와 비슷한 수준이였다. 또한, C5A와 C7A는 버섯에서 추출된 tyrosinase의 효소 활성을 통계적으로 유의하게 억제하였다. 또한, 컴퓨터 분석을 통해 C5A와 C7A는 kojic acid와 유사한 수준으로 tyrosinase의 active site에 결합할 것으로 예측되었다. 이에 더하여, 4종류의 catechin 유도체 화합물 모두가 α-MSH 처리된 B16F10 세포주에서 tyrosinase 단백질 발현을 감소시켰다. 본 연구결과를 종합하면, 참느릅나무에서 추출/동정된 catechin 유도체 화합물은 멜라닌세포의 멜라닌 생성을 효과적으로 억제하였다. 따라서, 본 연구결과는 이러한 catechin 유도체 화합물이 피부의 과다 색소침착 질환 제어를 위한 새로운 조절 물질로 응용될 수 있음을 제시한다.

Lipoteichoic Acid Isolated from Lactobacillus plantarum Inhibits Melanogenesis in B16F10 Mouse Melanoma Cells

  • Lipoteichoic acid (LTA) is a major component of the cell wall of Gram-positive bacteria. Its effects on living organisms are different from those of lipopolysaccharide (LPS) found in Gram-negative bacteria. LTA contributes to immune regulatory effects including anti-aging. In this study, we showed that LTA isolated from Lactobacillus plantarum (pLTA) inhibited melanogenesis in B16F10 mouse melanoma cells. pLTA reduced the cellular activity of tyrosinase and the expression of tyrosinase family members in a dose-dependent manner. The expression of microphthalmia- associated transcription factor (MITF), a key factor in the synthesis of melanin, was also decreased by pLTA. Further, we showed that pLTA activated melanogenesis signaling, such as extracellular signal-regulated kinase (ERK) and phosphatidylinositol 3-kinse (PI3K)/AKT. In addition, the expression of heterogeneous nuclear ribonucleoprotein A1 (hnRNP A1) and HuR, which are important RNA-binding proteins (RBPs), was reduced. pLTA likely degrades MITF via regulation of melanogenic signaling and RNA stability of melanogenic proteins, resulting in the reduction of melanin. Thus, our data suggest that pLTA has therapeutic potential for treating hyperpigmentation disorders and can also be used as a cosmetic whitening agent.

매생이 추출물의 멜라닌생성 억제효과 (Inhibitory Effect on the Melanogenesis of Capsosiphon fulvescens)

  • The green marine algae, Capsosiphon fulvescens is one of the important economic seaweeds cultured in Korea. In this study, we investigated the effects of Capsosiphon fulvescens on melanogenesis using B16 cells. Our results showed that Capsosiphon fulvescens significantly inhibits melanin synthesis and it reduces the activity of tyrosinase, the rate-limiting melanogenic enzyme. Western Blot analysis using anti-tyrosinase antibody revealed that Capsosiphon fulvescens ($10\~40\;{\mu}g/ml$ decreased tyrosinase protein levels. Cell proliferation was dose-dependently inhibited by 10, 20 and 40 ${\mu}g/ml$ Cap­sosiphon fulvescens, without cytotoxicity and morphological change. These results suggest that the depigmenting effect of Capsosiphon fulvescens is correlated with the suppression of tyrosinase activity and protein level, which are key enzymes for melanogenesis.

감태 효소 추출물 및 폴리페놀 추출물의 생리활성에 관한 연구 (Biological Potential of Enzymatic and Polyphenol Extracts from Ecklonia cava)

  • 본 연구에서는 감태 효소 추출물과 그것의 폴리페놀 추출물의 화장품 원료로서의 효능을 알아보기 위하여 항산화, 항당화, 미백, 항염 효과와 관련된 실험을 실시하였다. 감태 효소 추출물과 폴리페놀 추출물은 강력한 라디컬 소거능을 가지고 있으며 BSA/Glucose 시스템에서 최종당화생성물의 형성을 저해하는 항당화 활성과 타이로시네이즈 저해를 통한 우수한 미백력을 가지고 있음을 확인하였다. 또한 두 추출물 모두 세포 내에서 $PGE_2$와 NO 생성 저해를 통한 항염 효과를 나타내었다. 이러한 결과를 종합해 볼 때, 감태 효소 추출물과 그 폴리페놀 추출물은 화장품 원료로서의 응용 가능성이 있을 것으로 사료된다.

Inhibitors of Tyrosinase and Melanogenesis from Galla rhois

  • Previously, a 50% aqueous methanol extract of Galla rhois was shown to be the most potent tyrosinase inhibition activity with an {TEX}$IC_{50}${/TEX}(the concentration causing 50% inhibition of tyrosinase activity) of 0.2mg/ml of 205 crude drug extracts. To isolate tyrosinase inhibitors, the methanol extract was evaporated to a small volume in vacuo, and then partitioned stepwise with benzene and ethyl acetate(EtOAc). the EtOAc fraction was solubilized in 10% MeOH solution, and then fractionated successively by Diaion HP-20 and Sephadex LH-20 column chromatography, and preparative HPLC. Three phenolic compounds were isolated, and characterized as gallic acid(GA), methyl gallate(MG) and 1,2,3,4,6-penta-O-galloyl-$\beta$-D-glucose(PGG) by UV, IR, {TEX}${1}^H${/TEX}-&{TEX}${13}^C${/TEX}-NMR, and FAB-MS spectroscopy, PGG({TEX}$IC_{50}${/TEX}=50$\mu\textrm{g}$/ml) showed a considerable inhibitory effect against mushroom tyrosinase, while GA({TEX}$IC_{50}${/TEX}=1.6mg/ml) and MG({TEX}$IC_{50}${/TEX}=234$\mu\textrm{g}$/ml) did not show an appreciable effect. Meanwhile, MG inhibited greatly melanogenesis in a murine melanocyte cell line, Mel-Ab. MG and PGG showed typical noncompetitive inhibition patterns against mushroom tyrosinase. These results suggest that PGG and MG may be potentially useful as either anti-browning or anti-melanogenic agents in foods and cosmetics.

짚신나물 물 추출물의 항산화 활성 및 미백효과에 관한 연구 (Antioxidant and Whitening Effects of Agrimonia pilosa Ledeb Water Extract)

  • This study was performed to assess the antioxidant activities and whitening effects of Agrimonia pilosa Ledeb on melanin synthesis. The whitening effects of Agrimonia pilosa Ledeb water extracts were examined by in vitro mushroom tyrosinase assay and B16BL6 melanoma cells. We assessed inhibitory effect of Agrimonia pilosa Ledeb water extract on expression of melanogenic enzyme proteins including tyrosinase, tyrosinase-related protein 1 (TRP-1) and tyrosinase-related protein 2 (TRP-2) in B16BL6 cells. Inhibitory effect of Agrimonia pilosa Ledeb onto free radical generation was determined by measuring DPPH and hydroxyl radical scavenging activitie. Our results indicated that Agrimonia pilosa Ledeb water extract effectively inhibited free radical generation. In DPPH and hydroxy radical scavenging activity, Agrimonia pilosa Ledeb water extract had a potent anti-oxidant activity in a dose-dependent manner. They significantly inhibited tyrosinase activity in vitro and in B16BL6 melanoma cells. Also, Agrimonia pilosa Ledeb suppressed the expression of tyrosinase in B16BL6 melanoma cells. These results show that Agrimonia pilosa Ledeb inhibited melanin production on the melanogenesis. The underlying mechanism of Agrimonia pilosa Ledeb on whitening activity may be due to the inhibition of tyrosinase activity. We suggest that Agrimonia pilosa Ledeb may be useful as new natural active ingredients for antioxidant and whitening cosmetics.

성숙도에 따른 복분자 열매의 미백 활성 비교 (Comparison of whitening effect of Rubus coreanus fruit according to maturity)

  • 본 연구에서는 복분자 열매의 성숙도에 따른 미백 활성의 차이를 확인하기 위해 B16F10 cell에서 melanin 생성량을 측정하고, 기능성분인 ellagic acid의 함량 변화를 확인하였다. 또한 ellagic acid의 함량에 따른 tyrosinase 저해 활성도 측정하였다. B16F10 cell에서 MRF 및 IRF 추출물은 200 ㎍/mL 농도까지 세포 독성을 나타내지 않았고, Melanin 생성량은MRF 추출물에 비해 IRF 추출물에서 더 좋은 억제 활성을 보여주었다. 또한 복분자 성숙도에 따른 ellagic acid의 함량 변화를 확인한 결과, MRF 추출물에 비해 IRF 추출물에서 ellagic acid의 함량이 높게 측정되었고, tyrosinase 저해 활성 평가에서도 MRF 추출물에 비해 IRF 추출물에서 더 좋은 억제 활성을 보였다. 결과를 종합해보면, MRF 추출물에 비해 IRF 추출물에서 ellagic acid의 함량이 더 높았으며, 이로 인해 tyrosinase의 저해 및 melanin 생성 억제 또한 IRF 추출물에서 더 좋은 것으로 판단된다. 따라서, 복분자 미성숙과는 천연 미백 소재로써 우수한 기능성 화장품 원료가 될 것이라 생각한다.

B16/F10 흑색종양세포에서 삼내자 메탄올 추출물의 멜라닌 생성에 미치는 억제효과 (Inhibitory Effects of Methanol Extract of Kaempferia galanga on melanogenesis in B16/F10 Melanoma Cells)

  • Objective: Recently the demands for the effective and safe depigmentative and anti-aging agents of the skin have increased due to the medical, pharmaceutical and cosmetic reasons. The purpose of this study is to investigate the MKG(Methanol Extract of Kaempferia galanga) and their dermal bioactivity properties related to cosmeceuticals such as depigmentation. Methods: We assessed inhibitory effects of MKG on melanin production in B16/F10 melanoma cells, on mushroom tyrosinase activity, effects of MKG on the expression tyrosinase, TRP-1, TRP-2, GSK-$3{\beta}$, CREB, MITF in B16/F10 melanoma cells without cytotoxicity range. Cell viability was measured by MTT assay and tyrosinase activity was assessed using by DOPA staining, western-blot analysis. We measured inhibition of melanin synthesis and tyrosinase activity by down-regulation of melanogenic enzyme expressions in ${\alpha}$-MSH induced melanogenesis B16/F10 melanoma cells. Results: MKG inhibited tyrosinase-activity, total melanin contents and dendrite out-growth. MKG inhibited melanogenesis by down-regulation of tyorsinase, TRP-1, TRP-2, CREB, and MITF in B16/F10 cells. The treatment with MKG at the 12.5, $25{\mu}g/ml$ level significantly inhibited the melanin synthesis induced ${\alpha}$-MSH in B16/F10 melanoma cells compared with untreated control. Conclusion: These results suggest that MKG inhibit melanin biosynthesis which is involved in hyper-pigmentation. So MKG is considered to be used as a whitening components reducing cytotoxicity.

Inhibitory Effect of Ceylon Black Tea Extract on the Melanogenesis in 𝛼-MSH Stimulated B16F10 Melanoma Cells

  • The desire to be light skinned is universal among women. Asia has a long history of using skincare formulations as whitening agents. There is an imperative need to develop novel cosmetics from herbal sources due to several unpleasant side effects and high costs. As a result, this study aims to investigate the effect of Ceylon black tea extracts on melanogenesis. Five different Ceylon black tea extracts were prepared and examined for total phenolic content (TPC), total flavonoid content (TFC), DPPH radical scavenging activity, and tyrosinase inhibitory activity. Furthermore, B16F10 melanoma cells were treated with these extracts and tested for cytotoxicity and protein suppression levels. According to the results of this study, the highest TPCs were obtained from ethanol and acetone extractions (240.303 ± 1.389 ㎍/g and 240.202 ± 4.700 ㎍/g, respectively), whereas the highest TFC was obtained from acetone extraction (57.484 ± 0.413 ㎍/g). Ceylon black tea extracted with ethanol exhibited the highest inhibitory activity on tyrosinase with an IC50 value of 0.277 ± 0.017 mg/mL and the highest DPPH radical scavenging activity with an EC50 value of 0.009 ± 0.000 mg/mL. Furthermore, western blot results revealed that tyrosinase, TRP-1, TRP-2, and MITF protein expression levels were dose-dependently suppressed, indicating the applicability of Ceylon black tea extract as a novel melanogenesis inhibitor.
