녹화 처리기간에 따른 황기, 석결명, 술패랭이꽃 및 질경이 새싹채소의 항산화 효과 변화 (Changes of Antioxidant Effects According to Greening Period of Astragalus membranaceus var. membranaceus, Senna occidentalis, Dianthus longicalyx, and Plantago asiatica Sprout Vegetables)

  • 새로운 항산화 기능성 새싹채소를 개발하고, 항산화효과가 우수한 재배 조건을 구명하기 위하여 황기, 석결명, 술패랭이꽃, 질경이 새싹채소의 재배 중 녹화기간에 따른 항산화효과 변화를 분석하였다. 연구의 결과, 종에 따라 광합성이 항산화능에 미치는 영향이 각기 다르게 나타났다. 황기 새싹채소는 총 폴리페놀 함량은 녹화 0일, 총 플라보노이드 함량은 녹화 3일째에 가장 우수하였으며, DPPH radical 소거능은 녹화 0일, ABTS radical 소거능은 녹화 1일, $Fe^{2+}$ chelating효과는 녹화 0일, 지질과산화 억제활성은 녹화 3일째에 가장 우수하였다. 석결명 새싹채소는 2일 녹화했을 때 항산화물질 함량이 높고 radical 소거능이 우수하였으며, $Fe^{2+}$ chelating효과는 녹화 0일, 지질과산화 억제활성은 녹화 1일째에 가장 우수하였다. 술패랭이 새싹채소는 녹화 3일 째에 항산화물질 함량이 높고, $Fe^{2+}$ chelating 효과가 우수하였으며, radical 소거능과 지질과산화 억제 활성은 녹화시키지 않았을 때 가장 우수하였다. 질경이 새싹채소의 항산화물질 함량 및 radical 소거능은 녹화 2일째에 가장 우수하였으며, $Fe^{2+}$ chelating 효과는 녹화 0일, 지질과산화 억제활성은 녹화 3일째에 가장 우수하였다. 연구의 결과, 식물 종에 목표로 하는 항산화 효과에 따라 광합성이 미치는 영향이 다르므로, 식물 종과 원하는 항산화 효과에 따라 재배 방법을 달리해야 할 것으로 생각되었다.

안태(安胎)에 활용되는 산약(山藥)의 신경세포주에 대한 안전성 및 항산화효과에 대한 연구 (Non-toxic and Anti-oxydative effect of Dioscoreae Rhizoma on PC12 Cell)

  • Purpose : This study examined the non-toxic and the anti-oxidative effect of Dioscoreae Rhizoma on PC12 cells. Sanyak(Dioscoreae Rhizoma; chinese yam, shan yao) is well-known for its curing power for kidney, lung, spleen. Tonifies and augments the spleen and stomach. Tonifies the lung gi and augments the kidney yin. Tonifies the kidneys and also stabilizes and binds. it also binds the essence and treats spermatorrhea, frequent urination, and vaginal discharge. We are therefore interested in whether Dioscoreae Rhizoma is capable of causing abnormal apoptosis processes, and whether this condition can be rectified through Dioscoreae Rhizoma herb treatment. Methods : We used aqueous extract to treat PC12 cells with different concentrations treated with a water or a MeOH extract of Dioscoreae Rhizoma (0, x10, x20, x40, x80). The MTT (3, (4, 5-dimethyl-thiazol) 2, 5-diphenyl-tetraxolium bromide) reduction assay was employed to quantify the differences in cell activity and viability. The Bax expression level was monitored using western-blotting techniques. The patterns of the changes in expression were scanned and analyzed. Results : Bax and GSK-3${\beta}$ promotes cell death and down-regulated during the development of the PC12 cells. This is indicated that Dioscoreae Rhizoma is capable of inducing apoptosis in PC12 cells. The induced cell death and significantly inhibited by Dioscoreae Rhizoma, which can be explained by the increase in the inhibition of Bax and GSK-3${\beta}$ expression. It was also shown that Dioscoreae Rhizoma inhibits the release of $H_2O_2$ and prevents lipid peroxidation. Furthermore, the accumulation of wild type Bax protein significantly downregulated in a dose-dependent manner upon treatment with Dioscoreae Rhizoma. Conclusion : In conclusion, Dioscoreae Rhizoma can induce apoptosis via a Bax-dependent pathway or GSK-3${\beta}$ dependent pathway in PC12 cells into anti-oxidant and protective effect.

B16F1세포에서 항산화 활성 및 멜라닌 합성에 대한 유백피 에탄올 추출물의 효능 (Effect of Ulmus macrocapa Ethanolic Extracts on Anti-oxidant Activity and Melanin Synthesis in B16F1 Cells)

  • 멜라닌은 superoxide, hydroxyl radical, singlet oxygen, hydrogen peroxide와 같은 활성산소를 생성하는 자외선으로부터 피부를 보호하는 핵심적인 역할을 한다. 그러나 지질, 단백질, DNA 산화를 야기시키는 활성산소는 주근깨와 기미의 원인으로 알려진 melanin 과잉생산을 유도한다. 한약재중에서 본 연구에 사용된 유백피는 주요 성분으로 flavonoids를 함유하는 것으로 보고되었다. 본 연구의 목적은 B16F1에서의 유백피 에탄올 추출물의 미백효과 및 항산화 효과를 조사한 것이다. UMEE는 $3.12{\mu}g/ml$ 이하의 농도에서 세포독성을 보여주지 않았다. 항산화 실험에서 UMEE는 높은 환원력과 DPPH 소거효과를 보여주었다. 더욱이 UMEE는 지질과산화 억제효과가 있는 것으로 관찰되었다. UMEE는 in vitro에서 tyrosinase활성에 대한 억제효과는 없었다. 그러나 UMEE는 melanogenesis에서 중요한 효소인 tyrosinase 및 tyrosinase related protein-2 (TRP-2)의 발현을 감소시키는 것으로 나타났다. 이러한 결과들은 UMEE가 항산화 활성뿐 만 아니라 tyrosinase 및 TRP-2의 발현억제를 통한 미백효과를 나타내어, 피부에 미백효과를 줄 수 있는 기능적인 잠재성을 가지고 있다는 것을 암시한다.

Assessment of In vitro Antioxidant, Antidiabetic and Cytotoxic Activities of Sphaeranthus africanus Extracts

  • Sphaeranthus africanus is commonly used as a traditional remedy for sore throats and pain treatment in Vietnam. The aerial parts have been studied for its anti-inflammatory and anti-proliferative properties. However, the antioxidant and antidiabetic potential of the plant has not been explored. In this work, hydrophilic extracts of the plant's aerial parts were prepared in order to investigate its antioxidant and anti-diabetic properties. Also, the cytotoxicity of the root was evaluated and compared to that of the aerial parts. All of the extracts inhibited lipid peroxidation with IC50 values ranging from 2.05 to 3.56 ㎍/mL, indicating substantial antioxidant activity. At an IC50 value of 4.80 ㎍/mL, the 50% ethanol extract exhibited the most potent inhibition of α-glucosidase. The cytotoxic activity of root extracts is 2 to 5-fold less than that of the aerial parts. Nevertheless, dichloromethane and ethyl acetate extracts of the root demonstrated a selective effect on leukemia cells, with no harm towards the normal HEK-293 cell line. This work provides a scientific support for the antioxidant and antidiabetic activity of the plant. Hence, it may find a promising material for the development of novel antioxidant and antidiabetic agents. More research can be conducted on the phytochemistry and anticancer activities of the plant's root.

Aspergillus niger 효소에 의한 길경 사포닌(플라티코딘)의 전환 및 항산화 활성 비교 (The Transformation of Saponin Platycodi Radix by Aspergillus niger and Anti-oxidation Evaluation of the Transformed Metabolites)

  • 본 연구에서는 길경을 추출, 농축, 정제하여 crude platycodin을 얻은 후, 식품 미생물을 이용하여 길경의 배당체인 platycodin의 당 사슬 부분을 일부 가수분해하여 전환할 수 있는 방법을 모색하였다. 그리고 platycodin의 전환 전과 전환 후의 세포주를 이용한 세포 독성, 항산화 활성 및 항산화 효소 활성에 대해 비교하여 보았다. Chinese Hamster lung fibroblast인 V79-4 세포 독성실험 결과, 전환 전에 비교하여 전환 후에 더 나은 세포 생존률을 보였다. 후에 진행된 DPPH 자유기 소거능을 측정실험과 lipid peroxidation 억제능을 알아보기 위하여 malondialdehyde(MDA) 양을 측정한 결과 전환 후에서 더 높은 항산화 활성이 나타나는 결과를 보였다. 따라서, 식품이나 생약 소재 배당체의 구조를 식품 미생물을 통해 안전하게 전환시키면 그에 따라 독성, 활성 등이 변화해 새로운 성질을 가진 유도체를 만들어 낼 수 있고, 이러한 전환체는 상대적으로 높은 생리활성을 가지는 것을 알 수 있었다. 따라서 이상의 결과를 종합하여 보면, 식품이나 생약 소재 배당체의 구조를 식품 미생물을 통해 안전하게 비당체로 전환시키면 그에 따라 독성, 활성 등이 변화해 새로운 성질을 가진 유도체를 만들어 낼 수 있다. 본 연구에서 살펴본 platycodin의 전환 전과 전환 후의 항산화 생리활성은 대부분이 전환 후의 platycodin 활성이 높게 나타났으며, 이는 전환체가 새로운 식품 소재로서의 가능성을 시사한다고 판단된다.

설련의 항산화능와 사람 섬유아세포에서 UVA에 의한 MMP-1발현 저해효과 (Anti-Oxidative and Inhibitory Effect of Saussurea involucrata on MMP-1 in UVA-irradiated Human Dermal Fibroblast)

  • 노화와 연관하여 설련의 항산화 효과, in vitro MMP 활성 저해능, 자외선 조사에 의해 유도된 MMP-1 발현에 대한 영향을 사람섬유아세포를 이용하여 확인하였다. 설련의 항산화 효과를 알아보기 위하여 DPPH radical과 superoxide anion radical 소거효과를 측정하였다. 그 결과 DPPH radical 소거능의 $IC_{50}$ 값은 $3.89{\mu}g/mL$이고, xanthine/xanthine oxidase에 의한 superoxide anion radical 제거능의 $IC_{50}$ 값은 $67.29{\mu}g/mL$이었다. 설련 $1000{\mu}g/mL$에서 93.27%의 지질과산화 저해효과를 나타내었다. MMP-1의 효소활성 저해 효과는 농도 의존적으로 활성을 억제하였으며, $IC_{50}$ 값은 $97.18{\mu}g/mL$이다. 또한 자외선 조사에 의해 사람 섬유아세포에서 발현되는 MMP-1에 대해 단백질의 양적인 변화는 42.86% 감소되었으며, 설련에 의해 농도 의존적으로 MMP-1 mRNA의 발현량도 감소되었다. 이러한 실험결과를 통하여 설련은 항산화 효과뿐만 아니라 자외선 조사에 의해 유도되는 MMP-1 단백질 발현과 mRNA 유전자 수준에서의 조절이 가능함을 확인하였다. 결론적으로 설련은 항산화와 자외선으로부터 생성되는 MMP-1의 발현을 저해함으로써 노화와 따른 피부를 보호하는 항노화 소재로서의 응용가능성을 확인하였다.

BJS-Mix001이 Indomethacin 유발 랫트 위점막 손상에 미치는 영향 (Protective Effects of BJS-mix001, in Indomethacin induced Gastric Damages in Rats)

  • The object of this study was to observe the gatro protective effects of BJS-mix001, a mixed herbal formula consisted of 4 herbal drugs Pinelliae Rhizoma : Coptidis Rhizoma : Massa Medicata Fermentata : Ostreae Testa = 1 : 1 : 1 : 1 (g/g) mixtures, which were main component of oriental medicine for treating various digestive diseases, in indomethacin induced gastric damages in rats. Three different dosages of BJS-mix001 (200, 100 and 50 mg/kg) were once orally administered 30 min before indomethacin treatment. Six hrs after indomethacin treatment, changes on the gross lesion scores, fundic histopathology, MPO activity and anti oxidant activities were observed. The results were compared with omeprazole, antioxidant and proton pump inhibitor 10 mg/kg and DA-9601, a standardized extract of the herb Artemisia asiatica 100 mg/kg treated group, respectively. As results of all three different dosages of BJS-mix001 in the indomethacin induced gastric damaged rats, significant decreased gastric damages were detected as compared with the indomethacin treated control rats. BJS-mix001 also strengthened the antioxidative defense systems - decreased the level of lipid peroxidation and catalase activity but increased the level of superoxide dismutase and glutathione contents. BJS-mix001 showed similar gastro protective effects as compared with equal dosage of DA-9601, and BJS-mix001 50 mg/kg showed slighter effects as compared with omeprazole 10 mg/kg, in the present study. The results obtained in this study suggest that BJS-mix001 showed favorable effects in the indomethacin induced gastric damages mediated by strengthening of the antioxidative defense systems.

Amelioration of 1,2 Dimethylhydrazine (DMH) Induced Colon Oxidative Stress, Inflammation and Tumor Promotion Response by Tannic Acid in Wistar Rats

  • Colon cancer is the third most common malignant neoplasm in the world and it remains an important cause of death, especially in western countries. The toxic environmental pollutant, 1, 2-dimethylhydrazine (DMH), is also a colon-specific carcinogen. Tannic acid (TA) is reported to be effective against various types of chemically induced toxicity and also carcinogenesis. In the present study, we evaluated the chemopreventive efficacy of TA against DMH induced colon toxicity in a rat model. Efficacy of TA against the colon toxicity was evaluated in terms of biochemical estimation of antioxidant enzyme activities, lipid peroxidation, histopathological changes and expression of early molecular markers of inflammation and tumor promotion. DMH treatment induced oxidative stress enzymes (p<0.001) and an early inflammatory and tumor promotion response in the colons of Wistar rats. TA treatment prevented deteriorative effects induced by DMH through a protective mechanism that involved reduction of oxidative stress as well as COX-2, i-NOS, PCNA protein expression levels and TNF-${\alpha}$ (p<0.001) release. It could be concluded from our results that TA markedly protects against chemically induced colon toxicity and acts plausibly by virtue of its antioxidant, anti-inflammatory and antiproliferative activities.

Quercetin Prevents Hydrogen Peroxide-induced Necrotic and Apoptotic Cell Death in Human Colonic Epithelial Cells

  • Quercetin is one of the most distributed flavonoids in the plant kingdom and occurs naturally in a wide range of fruits and vegetables. This study was undertaken to determine whether quercetin exerts beneficial effect against necrotic and apoptotic cell death induced by hydrogen peroxide ($H_2O2$) in intestinal cells using the human-derived cultured T84 colonic epithelial cell line. Necrotic cell death was induced by exposing cells to 0.5 mM $H_2O_2$ for 2 h and apoptosis was induced by incubating cells in normal culture medium for 18 h following exposure of cells to 0.5 mM $H_2O2$ for 2 h. Cell viability was evaluated by the trypan blue exclusion assay and apoptosis was assessed by Hoechst 33258 staining and flow cytometry. $H_2O_2$ induced necrotic cell death in a time and dose-dependent fashion. Both necrotic and apoptotic cell deaths were not prevented by the antioxidants N,N'-diphenyl-p-phenylenediamine(DPPD) and Trolox, whereas both cell deaths induced by the organic hydroperoxide t-butylhydroperoxide (tBHP) were prevented by DPPD, suggesting that $H_2O_2$ induces cell death through a lipid peroxidation-independent mechanism. $H_2O2$-induced necrotic death was prevented by deferoxamine and 3-aminobenzamide, while the apoptotic cell death was not affected by these agents. Quercetin prevented both necrotic and apoptotic cell deaths induced by $H_2O_2$ in a dose-dependent manner. $H_2O_2$ caused activation of poly (ADP-ribose) polmerase (PARP), which was inhibited by deferoxamine, 3-aminobenzamide, and quercetin, but not DPPD. These results indicate that quercetin inhibits both necroticand apoptotic deaths of T84 cells. The anti-necrotic effect of quercetin may be attributed to its iron chelator activity rather than a direct $H_2O_2$ scavenging capacity and antioxidant. The present study suggests that quercetin may play a therapeutic role in the treatment of human gastrointestinal diseases mediated by oxidants.

Plant Extracts and Plant-Derived Compounds: Promising Players in Countermeasure Strategy Against Radiological Exposure: A Review

  • Radiation exposure leads to several pathophysiological conditions, including oxidative damage, inflammation and fibrosis, thereby affecting the survival of organisms. This review explores the radiation countermeasure properties of fourteen (14) plant extracts or plant-derived compounds against these cellular manifestations. It was aimed at evaluating the possible role of plants or its constituents in radiation countermeasure strategy. All the 14 plant extracts or compounds derived from it and considered in this review have shown some radioprotection in different in vivo, ex-vivo and or in vitro models of radiological injury. However, few have demonstrated advantages over the others. C. majus possessing antioxidant, anti-inflammatory and immunomodulatory effects appears to be promising in radioprotection. Its crude extracts as well as various alkaloids and flavonoids derived from it, have shown to enhance survival rate in irradiated mice. Similarly, curcumin with its antioxidant and the ability to ameliorate late effect of radiation exposure, combined with improvement in survival in experimental animal following irradiation, makes it another probable candidate against radiological injury. Furthermore, the extracts of P. hexandrum and P. kurroa in combine treatment regime, M. piperita, E. officinalis, A. sinensis, nutmeg, genistein and ginsan warrants further studies on their radioprotective potentials. However, one that has received a lot of attention is the dietary flaxseed. The scavenging ability against radiation-induced free radicals, prevention of radiation-induced lipid peroxidation, reduction in radiation cachexia, level of inflammatory cytokines and fibrosis, are some of the remarkable characteristics of flaxseed in animal models of radiation injury. While countering the harmful effects of radiation exposure, it has shown its ability to enhance survival rate in experimental animals. Further, flaxseed has been tested and found to be equally effective when administered before or after irradiation, and against low doses (${\leq}5Gy$) to the whole body or high doses (12-13.5 Gy) to the whole thorax. This is particularly relevant since apart from the possibility of using it in pre-conditioning regime in radiotherapy, it could also be used during nuclear plant leakage/accidents and radiological terrorism, which are not pre-determined scenarios. However, considering the infancy of the field of plant-based radioprotectors, all the above-mentioned plant extracts/plant-derived compounds deserves further stringent study in different models of radiation injury.
