손바닥선인장(Opuntia humifusa) 줄기 추출물의 항당뇨 효과 (Anti-diabetic Effect of Opuntia humifusa Stem Extract)

  • Opuntia humifusa known as the Eastern prickly pear have been used as a treatment of burns, diarrhea, asthma, rheumatism, gonorrhea, and diabetes in alternative medicine. O. humifusa is widely cultivated in the middle and southern provinces of Korea and distributed in North America. The aim of this study is to investigate anti-diabetic effect of O. humifusa stem (OHS) water or 80% MeOH extract using 3T3-L1 adipocytes and db/db mice animal models. OHS 80% MeOH extract at a dose of $250{\mu}g/ml$ significantly increased the glucose uptake and lipid accumulation compared with the control in 3T3-L1 adipocytes. Blood glucose, plasma total cholesterol and triglyceride levels were significantly reduced by oral treatment of OHS 80% MeOH extract (200 mg/kg BW) for 6 weeks in db/db mice. Also, the oral treatment of OHS 80% MeOH extract slightly changed the plasma insulin and insulin resistance levels in db/db mice, but were no significance in comparison to control. Glucose transporter(GLUT)4 expressions of adipose tissue and muscle were significantly increased more than that in the control. Therefore, these results suggest that OHS 80% MeOH extract inhibits the blood glucose level through regulation of lipid profile, insulin resistance, and GLUT4 expression in db/db mice and its diabetic effect is effective more than water extract.

  • SA51, a medium potency inhibitor of protein tyrosine phosphatase 1B (PTP1B), was identified to be a potent inhibitor of $I{\kappa}B$ kinase ${\beta}$ ($IKK{\beta}$). Consistent with this, SA51 prevented lipopolysaccharide (LPS)-induced breakdown of $I{\kappa}B{\alpha}$ in macrophages. The effects of SA51 in mice were compared with those of structurally related compounds, SA18 and SA32, which were previously reported as inhibitors of both enzymes - less potent against $IKK{\beta}$ but more potent against PTP1B compared to SA51. SA51 improved glucose tolerance and lipid parameters in mice, consistent with the results reported for $IKK{\beta}^{+/-}$ mice. In contrast, SA18 and SA32 showed anti-obesity effects without anti-diabetic effects. Collectively, the effects of SA51 could be due largely to the inhibition of $IKK{\beta}$, whereas SA18 and SA32 may be more likely to inhibit PTP1B, consistent with their relative in vitro inhibitory effects.

Modulation of Pro-inflammatory and Anti-inflammatory Cytokines in the Fat by an Aloe Gel-based Formula, QDMC, Is Correlated with Altered Gut Microbiota

  • Abnormal inflammatory responses are closely associated with intestinal microbial dysbiosis. Oral administration of Qmatrix-diabetes-mellitus complex (QDMC), an Aloe gel-based formula, has been reported to improve inflammation in type 2 diabetic mice; however, the role of the gut microbiota in ameliorating efficacy of QDMC remains unclear. We investigated the effect of QDMC on the gut microbiota in a type 2 diabetic aged mouse model that was administered a high-fat diet. Proinflammatory (TNF-α and IL-6) and anti-inflammatory (IL-4 and IL-10) cytokine levels in the fat were normalized via oral administration of QDMC, and relative abundances of Bacteroides, Butyricimonas, Ruminococcus, and Mucispirillum were simultaneously significantly increased. The abundance of these bacteria was correlated to the expression levels of cytokines. Our findings suggest that the immunomodulatory activity of QDMC is partly mediated by the altered gut microbiota composition.

정향 추출물의 항산화.항혈소판 응집효과 및 혈전 용해능 탐색 (Screening of Antioxidative, Anti-platelet Aggregation and Anti-thrombotic Effects of Clove Extracts)

  • Clove has been frequently used as anti-diabetic, anti-microbial, anti-inflammatory, anesthetic drug and remedies for stomachache by coldness. In this study, the antioxidant activity of extract from Clove was studied in vitro methods by measuring the antioxidant activity by TEAC, measuring the scavenging effects on reactive oxygen species (ROS) [superoxide anion, hydroxyl radical] and on reactive nitrogen species (RNS) [nitric oxide and peroxynitrite] as well as measuring the inhibitory effect on $Cu^{2+}$-induced human LDL oxidation. Anti-platelet aggregation and anti-thrombotic effects of Clove extracts were studied ex vivo methods by mesuring the inhibitory effect on thrombin induced platelet aggregation and the fibrinolytic activity. The Clove extracts were found to have a potent scavenging activity, as well as an inhibitory effect on LDL oxidation in vitro. Moreover Clove extracts were exhibited remarkable inhibitory effect on platelet aggregation and fibrinolytic activity. In conclusion, the Clove extracts have anti-oxidative and anti-atherosclerotic effects in vitro and ex vivo system, which can be used for developing pharmaceutical drug against oxidative stress and atherosclerosis.

Streptozotocin 유도 흰쥐에서 참당귀(Angelica gigas Naki) 추출물의 당뇨 개선 효과 (Hypoglycemic Effect of Angelica gigas Naki Extract in Streptozotocin-induced Diabetic Rats)

  • This study examined the possible hypoglycemic effects Angelica gigas Naki extracts in streptozotocin-induced diabetic rats(STZ+50%, STZ+100% EtOH and STZ+water). The studies showed that administration of the Angelica gigas Naki extract decreased high blood glucose levels(more than 300 mg/$d{\ell}$) to a normal level(104 mg/$d{\ell}$) in the STZ+50% EtOH group. Liver glucokinase levels were significantly increased in STZ+50% EtOH and STZ+100% EtOH groups compared to the STZ group. Moreover, the liver acetyl CoA carboxylase level was significantly increased in STZ+50% EtOH, STZ+100% EtOH and STZ+water groups compared to the STZ group. These results suggest that the Angelica gigas Naki extract in the STZ+50% EtOH group exerted an ameliorable effect and can be used as an anti-diabetic substance, either as a dietary supplements or as a new drug.

Sargassum coreanum extract alleviates hyperglycemia and improves insulin resistance in db/db diabetic mice

  • BACKGROUND/OBJECTIVES: The goal of this study was to examine the effect of Sargassum coreanum extract (SCE) on blood glucose concentration and insulin resistance in C57BL-KsJ-db/db mice. MATERIALS/METHODS: For 6 weeks, male C57BL/KsJ-db/db mice were administrated SCE (0.5%, w/w), and rosiglitazone (0.005%, w/w). RESULTS: A supplement of the SCE for 6 weeks induced a significant reduction in blood glucose and glycosylated hemoglobin concentrations, and it improved hyperinsulinemia compared to the diabetic control db/db mice. The glucokinase activity in the hepatic glucose metabolism increased in the SCE-supplemented db/db mice, while phosphoenolpyruvate carboxykinase and glucose-6-phosphatase activities in the SCE-supplemented db/db mice were significantly lower than those in the diabetic control db/db mice. The homeostatic index of insulin resistance was lower in the SCE-supplemented db/db mice than in the diabetic control db/db mice. CONCLUSIONS: These results suggest that a supplement of the SCE lowers the blood glucose concentration by altering the hepatic glucose metabolic enzyme activities and improves insulin resistance.

용규(龍葵)가 streptozotocin으로 유발된 백서(白鼠)의 실험적 당뇨에 미치는 영향 (Anti-daibetic Effect of Solani nigri Herba on Diabetic Rat Induced by Streptozotocin)

  • The objective of the study was to observe the anti-diabetic effect of Solani nigri Herba(SH). The following experimental results were derived from the measurement of the levels of glucose, GOT and GPT in the serum and weight from a group of experimental rats by way of the injection of streptozotocin as well as the oral administration of Solani nigri Herba water extracts. The glucose level in the serum significantly decreased in the Solani nigri Herba-dosed group(SH2,12.6 mg/100 g rat weight) on the 10th day. The GOT level of SHI(4.2 mg/100 g Solani nigri Herba) and SH2 in the serum slightly decreased, but no significance to compare with control group. The GPT level of SH1 and SH2 in the serum slightly decreased, but no significance to compare with control group. The weight of rat of SH1 and SH2 in the serum slightly increased, but no significance to compare with control group. In conclusion, Solani nigri Herba appeared to have significance inhibitory effects on the rise of blood sugar level in the experimentally induced diabetics.

보리순 분말이 당뇨쥐의 혈당조절에 미치는 효과 (Effect of Young Barley Leaf Powder on Glucose Control in the Diabetic Rats)

  • This study examined the anti-diabetic effects of young barley leaf powder in rats with streptozotocin (STZ)-induced diabetes. Male Sprague-Dawley rats were divided into the non-diabetic (N) and diabetic groups, and fed the following for four weeks. The diabetic groups were further subdivided into three experimental groups: a diabetic control group (STZ), a diabetic group fed 5% barley leaf powder (STZ-BL), and a diabetic group fed 10% barley leaf powder (STZ-BH). Food and water intakes were higher in the diabetic groups than in the N group. Body weight gain was higher in the STZ-BL and STZ-BH groups compared with the STZ group, but there were no significant changes in body weight gain between the diabetic groups. The serum glucose and fructosamine levels were lower in the STZ-BL and STZ-BH groups than in the STZ group. The levels of serum insulin were higher in the STZ-BL and STZ-BH groups than in the STZ group. Serum ALT, AST and ALP activities decreased in the STZ-BL and STZ-BH groups compared with the STZ group, but there was no difference. These results indicate that dietary supplementation of barley leaf powder can attenuate clinical symptoms of diabetes in rats with STZ-induced diabetes.

Hypoglycemic effect of standardized Chrysanthemum zawadskii ethanol extract in high-fat diet/streptozotocin-induced diabetic mice and rats

  • Chrysanthemum zawadskii (CZ) is a perennial herb belonging to the Asteraceae family. CZ is used medicinally to treat inflammatory and uterine diseases in Asia. CZ was extracted with 50% ethanol and CZ extract (CZE; at 125, 250, and 500 mg/kg body weight) was administered orally every day for 5 or 6 weeks to investigate the anti-diabetic effects in streptozotocin (STZ)-induced rats and STZ + high-fat diet (HFD)-fed mice. CZE significantly decreased fasting blood glucose levels in STZ- and STZ + HFD-induced diabetic models. In addition, glucose tolerance and insulin tolerance were improved in the STZ + HFD + CZE group by increasing insulin levels and decreasing hemoglobin A1c (HbA1c) levels in serum. Furthermore, CZE supplements decreased components of the serum lipid profile such as triglyceride, total cholesterol, and low-density lipoprotein cholesterol levels. These results suggest that CZE may be a potential candidate for controlling hyperglycemia.
