In this study, the bioactivities of ethanol (EELS) and water extract (WELS) from the leaf of Lythrum salicaria L. were investigated. In the anti-cancer activity, the growths of both human prostate cancer (DU145) and human colonic carcinoma cell (HT29) were inhibited up 60% by adding 10 mg/$m{\ell}$ of EELS. Anti-inflammatory activity of EELS and WELS have been evaluated on lipopolysaccharide (LPS) induced release of nitric oxide (NO) by the macrophage RAW 264.7 cells. EELS and WELS inhibited inflammatory by 57.3 and 46.9% in 10 mg/$m{\ell}$, respectively. In the anti-oxidative activity, $IC_{50}$ of DPPH radical scavenging activity was respectively 60.71 and $92.90\;{\mu}g/m{\ell}$ by EELS and WELS. In the anti-diabetic activity, $IC_{50}$ of ${\alpha}$-amylase inhibitory activity of EELS and WELS were respectively 5,250 and $5,020\;{\mu}g/m{\ell}$. $IC_{50}$ of ${\alpha}$-glucosidase inhibitory activity was 7.96 and $68.41\;{\mu}g/m{\ell}$ by EELS and WELS. In the anti-obesity, $IC_{50}$ of lipase inhibitory activity was 880 and $9,840\;{\mu}g/m{\ell}$ by EELS and WELS. Finally, EELS and WELS exhibited anti-oxidative, anti-inflammatory, anti-diabetic activity and anti-obesity. It suggests that Lythrum salicaria L. could be potentially used as a resource of bioactive materials for health functional foods.
Background: The non-saponin fraction of Korean Red Ginseng has been reported to have many biological activities. However, the effect of this fraction on anti-diabetic activity has not been elucidated in detail. In this study, we investigated the effects of KGC05P0, a non-saponin fraction of Korean Red Ginseng, on anti-diabetic activity in vitro and in vivo. Methods: We measured the inhibition of commercially obtained α-glucosidase and α-amylase activities in vitro and measured the glucose uptake and transport rate in Caco-2 cells. C57BL/6J mice and C57BLKS/Jdb/db (diabetic) mice were fed diets with or without KGC05P0 for eight weeks. To perform the experiments, the groups were divided as follows: normal control (C57BL/6J mice), db/db control (C57BLKS/Jdb/db mice), positive control (inulin 400 mg/kg b.w.), low (KGC05P0 100 mg/kg b.w.), medium (KGC05P0 200 mg/kg b.w.), and high (KGC05P0 400 mg/kg b.w.). Results: KGC05P0 inhibited α-glucosidase and α-amylase activities in vitro, and decreased glucose uptake and transport rate in Caco-2 cells. In addition, KGC05P0 regulated fasting glucose level, glucose tolerance, insulin, HbA1c, carbonyl contents, and proinflammatory cytokines in blood from diabetic mice and significantly reduced urinary glucose excretion levels. Moreover, we found that KGC05P0 regulated glucose production by down-regulation of the PI3K/AKT pathway, which inhibited gluconeogenesis. Conclusion: Our study thereby demonstrated that KGC05P0 exerted anti-diabetic effects through inhibition of glucose absorption and the PI3K/AKT pathway in in vitro and in vivo models of diabetes. Our results suggest that KGC05P0 could be developed as a complementary food to help prevent T2DM and its complications.

This study aimed to investigate the physiological characteristics and anti-obesity effects of Lactobacillus plantarum K10. The ${\alpha}-amylase$ inhibitory activity, ${\alpha}-glucosidase$ inhibitory activity, and lipase inhibitory activity of L. plantarum K10 was $94.66{\pm}4.34%$, $99.78{\pm}0.12%$, and $87.40{\pm}1.41%$, respectively. Moreover, the strain inhibited the adipocyte differentiation of 3T3-L1 cells ($32.61{\pm}8.32%$) at a concentration of $100{\mu}g/mL$. In order to determine its potential for use as a probiotic, we investigated the physiological characteristics of L. plantarum K10. L. plantarum K10 was resistant to gentamycin, kanamycin, streptomycin, ampicillin, ciprofloxacin, tetracycline, vancomycin, and chloramphenicol. It also showed higher Leucine arylamidase, Valine arylamidase, and ${\beta}-galactosidase$ activities. Moreover, it was comparatively tolerant to bile juice and acid, exhibiting resistance to Escherichia coli, Salmonella Typhimurium, Listeria monocytogenes, and Staphylococcus aureus with rates of 90.71%, 11.86%, 14.19%, and 23.08%, respectively. The strain did not produce biogenic amines and showed higher adhesion to HT-29 cells compared to L. rhamnosus GG. As a result of the animal study, L. plantarum K10 showed significantly lower body weight compared to the high-fat diet group. The administration of L. plantarum K10 resulted in a reduction of subcutaneous fat mass and mesenteric fat mass compared to the high-fat diet (HFD) group. L. plantarum K10 also showed improvement in gut permeability compared to the HFD positive control group. These results demonstrate that L. plantarum K10 has potential as a probiotic with anti-obesity effects.
In the current study we assessed a new crystallized compound, 5-(1-hydroxybutyl)-4-methoxy-3-methyl-2H-pyran-2-one (C-HMMP), from the endophytic fungus Colletotrichum acutatum residing in the medicinal plant Angelica sinensis for its in vitro antimicrobial, antibiofilm, antioxidant, antimalarial, and anti-proliferative properties. The promising compound was identified as C-HMMP through antimicrobial-guided fraction. The structure of C-HMMP was unambiguously confirmed by 2D NMR and HIRS spectroscopic analysis. Antimicrobial property testing of C-HMMP showed it to be effective against a variety of pathogenic bacteria and fungi with MICs ranging from 3.9 to 31.25 ㎍/ml. The compound displayed excellent antibiofilm activity against C. albicans, S. aureus, and K. pneumonia. Furthermore, the antimalarial and radical scavenging activities of C-HMMP were clearly dosedependent, with IC50 values of 0.15 and 131.2 ㎍/ml. The anti-proliferative activity of C-HMMP against the HepG-2, HeLa, and MCF-7 cell lines in vitro was investigated by MTT assay, revealing notable anti-proliferative activity with IC50 values of 114.1, 90, and 133.6 ㎍/ml, respectively. Moreover, CHMMP successfully targets topoisomerase I and demonstrated beneficial anti-mutagenicity in the Ames test against the reactive carcinogenic mutagen, 2-aminofluorene (2-AF). Finally, the compound inhibited the activity of α-glucosidase and α-amylase with IC50 values of 144.7 and 118.6 ㎍/ml, respectively. To the best of our knowledge, the identified compound C-HMMP was obtained for the first time from C. acutatum of A. sinensis, and this study demonstrated that C-HMMP has relevant biological significance and could provide better therapeutic targets against disease.
식품으로 사용되는 팥은 생팥이 아닌 증자팥을 사용하며, 고온의 열수로 삶은 후 제조된다. 본 연구에서는 팥을 이용한 고부가가치 식품개발 연구의 일환으로, 생팥의 열수 추출물 및 팥을 삶은 후 증자액을 폐기하고 제조한 증자팥의 열수 추출물을 조제하고 이들의 유용성분 및 항산화, 항균, 항당뇨 및 항혈전 활성을 비교 평가하였다. 생팥 및 증자팥의 추출효율은 각각 16.2% 및 14.8%로 생팥이 1.1배 높았으며, 총 폴리페놀, 총 플라보노이드 및 환원당 함량은 생팥이 증자팥보다 각각 2.5배, 2.1배 및 1.5배 높았다. 생팥의 DPPH 음이온 및 ABTS 양이온 소거능 및 환원력 평가 결과, 증자팥보다 모두 높게 나타나, 팥의 항산화 활성이 고온의 열수 삶기 과정 중 소실됨을 알 수 있었다. 반면, nitrite 소거능은 생팥보다 삶은 증자팥에서 더욱 강하게 나타났다. 항균 활성 평가결과, 생팥의 열수 추출물에서만 그람양성 세균에 대한 활성이 나타났으며, 증자팥의 경우 항균 활성은 $500{\mu}g/disc$ 농도까지 나타나지 않았다. 또한 in-vitro $\alpha$-amylase 및 $\alpha$-glucosidase에 대한 저해활성과 항혈전 활성에 관련된 thrombin, prothrombin, coagulation factors에 대한 저해활성 평가 결과, 생팥의 비가열 추출물과는 달리, 생팥 및 증자팥의 열수 추출물은 전분분해 저해 및 항혈전 활성이 인정되지 않았다. 본 연구결과는 생팥 및 증자팥의 열수 추출물의 항산화, 항당뇨 및 항혈전 활성이 비가열 에탄올 추출물보다 미약하며, 생팥의 유용활성이 증자팥에서는 거의 남아 있지 않음을 제시하고 있으며, 팥을 이용한 고부가가치 식품 제조 시 적합한 삶기 공정 및 팥 증자액의 효율적인 이용에 대한 연구가 필요함을 제시하고 있다.
본 연구에서는 상백피의 소화효소 저해활성과 3T3-L1 전지방세포의 분화 억제능을 기반으로 항비만 효능소재로서의 활용가능성을 평가하였다. 상백피의 에탄올 추출물(MRE)은 ${\alpha}$-amylase와 ${\alpha}$-glucosidase, pancreatic lipase를 활용한 소화효소 저해활성 평가 실험에서 각각 $7.86{\pm}0.36$, $0.12{\pm}0.03$, $7.93{\pm}0.11mg/mL$의 $IC_{50}$ 값을 보이며 우수한 억제 활성을 나타냈다. 또한 3T3-L1 전지방세포를 활용한 세포분화억제효능실험에서 MRE 처리군의 세포내 지방 축적율은 농도 의존적으로 감소되었다. 상백피의 항비만 작용 기전을 구명하기 위하여 adipogenesis 및 lipogenesis와 관련된 유전자 발현양상을 분석한 결과, 상백피 추출물 처리군에서는 생체내 지방대사 조절에 중요한 역할을 하는 FAS와 ACC 뿐 아니라 adipogenesis와 lipogenesis와 관련된 주요 전사요소인 $PPAR{\gamma}$와 $C/EBP{\alpha}$, SREBP-1c의 유전자 발현이 현저하게 억제되었다. qRT-PCR 분석 결과, 상백피 추출물의 anti-adipogenesis 효능은 전사단계에서의 관련 유전자 발현억제에 기인한다고 판단되었다. 본 실험결과 상백피 추출물은 전지방세포의 분화와 세포내 지질합성을 저해하고 비만과 관련 된 소화효소에 대한 저해활성을 나타내었다. 이러한 결과를 기반으로 상백피의 비만 예방 소재로서의 잠재적인 가능성을 확인하였다.
In beer, glutathione works as the main antioxidant compound, which also correlates with the stability of the beer flavor. In addition, high residual sugars in beer contribute to major nonvolatile components, which are reflected in a high caloric content. Therefore, in this study, the Saccharomyces cerevisiae GSH1 gene encoding glutamylcysteine synthetase and the Saccharomycopsis fibuligera ALP1 gene encoding ${\alpha}$-amylase were coexpressed in industrial brewing yeast strain Y31 targeting the ${\alpha}$-acetolactate synthase (AHAS) gene (ILV2) and alcohol dehydrogenase gene (ADH2), resulting in the new recombinant strain TY3. The glutathione content in the fermentation broth of TY3 increased to 43.83 mg/l as compared with 33.34 mg/l in the fermentation broth of Y31. The recombinant strain showed a high ${\alpha}$-amylase activity and utilized more than 46% of the starch as the sole carbon source after 5 days. European Brewery Convention tube fermentation tests comparing the fermentation broths of TY3 and Y31 showed that the flavor stability index for TY3 was 1.3-fold higher, whereas its residual sugar concentration was 76.8% lower. Owing to the interruption of the ILV2 gene and ADH2 gene, the contents of diacetyl and acetaldehyde as off-flavor compounds were reduced by 56.93% and 31.25%, respectively, when compared with the contents in the Y31 fermentation broth. In addition, since no drug-resistant genes were introduced to the new recombinant strain, it should be more suitable for use in the beer industry, owing to its better flavor stability and other beneficial characteristics.
The antioxidant and anti-diabetes effects were evaluated by the ethanol extracts from Astragalus membranaceus root classified by years using through DPPH radical scavenging activity, hydroxyl radical (${\cdot}OH$) scavenging activity, total phenolic and flavonoid contents, reducing power activity, $\alpha$-amylase and $\alpha$-glucosidase inhibitory activities. $IC_{50}$ values for DPPH radical scavenging activity of ethanol extract from 6 years old root ($749.25{\mu}g/mL$) was higher than 1 year ($1452.67{\mu}g/mL$) and 3 years old root ($1095.61{\mu}g/mL$). 6 years old root showed better effects in ${\cdot}OH$ scavenging activity ($IC_{50}$: $10.58{\mu}g/mL$), reducing power, total phenolic contents ($26.13{\pm}0.79\;Tan\;{\mu}g/mg$, $24.03{\pm}0.74\;Cat\;{\mu}g/mg$) $\alpha$-amylase ($33.33{\pm}0.55%$) and $\alpha$-glucosidase inhibitory activity ($49.71{\pm}1.01%$). On the other hand, total flavonoid compound contents were estimated much higher in 1 year old root ($44.93{\pm}1.35\;Que\;{\mu}g/mg$, $70.32{\pm}2.03\;Rut\;{\mu}g/mg$) than others. Based on these results, It was suggested that 6 years old root of A. membranaceus has a potential candidate for functional cosmetic and medicine.
Soybean grit를 이용하여 B. subtilis HA에 의한 고체발효를 통해 얻어진 발효물의 발효 시간에 따른 생리활성물질의 변화 및 항산화능을 평가하였다. 발효시간이 증가함에 따라 혈전분해효소 활성 및 protease 활성은 계속적으로 증가하는 경향을 보였으며, ${\alpha}$-amylase 활성은 발효 5일에서 최대값을 나타내었다. Tyrosine 함량은 발효 5일에서 최대값을 보였으며, 콩 가수분해물중에서 분자량 3,000 Da 이하의 분자량을 갖는 콩 펩타이드 및 갈변화색소는 발효시간이 증가함에 따라서 증가하는 경향을 보였다. Soybean grit 발효물의 총 폴리페놀함량은 주정 추출물이 물 추출물보다 전반적으로 높게 나타났으며 주정 추출물은 발효시간 3일째 18.10 mg/g으로 가장 높은 함량을 나타내었다. 항산화능을 나타내는 DPPH 라디칼 소거능은 발효시간 3일째 가장 높은 값을 보였으며, ABTS 라디칼 소거능은 발효 기간이 증가함에 따라 대조군에 비해 증가되었다.
본 연구에서는 갈근 및 그의 주 이소플라본인 puerarin의 활성을 세포 수준에서 분석하였다. 먼저 갈근에 함유된 이소플라본의 양을 분석한 결과, puerarin이 총 이소플라본의 90 % 차지하였다. 다음으로는 puerarin의 항당뇨 활성을 검정한 결과 먼저 탄수화물 및 지방소화효소 저해활성에 대해서는 거의 미비한 것으로 나타났으나 인슐린 감수성 및 지방세포의 분화의 유도에 대해서는 농도 의존적으로 작용하는 것으로 관찰되었다. 따라서 puerarin은 지방조직내로 포도당의 흡수를 촉진함으로서 항당뇨 효능을 발휘하는 것으로 추정된다.
