Antioxidant Activities of Ipomoea batatas L. Lam. (Purple Sweet Potato) Extracts Cultured in Korea (국내산 자색고구마(Ipomoea batatas L. Lam.) 추출물의 항산화활성 평가)

  • In this study, the antioxidative effects for the extracts of purple sweet potato (Ipomoea batatas L. Lam.) were investigated. The purple sweet potato was extracted with 70% ethanol and the ethyl acetate fraction was obtained from the extracts. The yields of extract and ethyl acetate fraction were 39.2% and 3.49% per dried powder, respectively. To confirm the antioxidative effects of the extracts, free radical scavenging activities (1,1-diphenyl-2-picrylhydrazyl), total antioxidant capacity by luminol-dependent chemiluminescence assay and the protective effects against reactive oxygen species (ROS) in erythrocytes were measured. Free radical scavenging activities ($FSC_{50}$) of the 70% ethanol extract and ethyl acetate fraction were $90.16{\mu}g/mL$ and $7.69{\mu}g/mL$, respectively. The free radical scavenging activity of ethyl acetate fraction was higher than that of (+)-${\alpha}$-tocopherol ($8.98{\mu}g/mL$). Total antioxidant capacities ($OSC_{50}$) of the 70% ethanol extract and ethyl acetate fraction were $5.75{\mu}g/mL$ and $1.92{\mu}g/mL$, respectively. The capacity of ethyl acetate fraction was similar to L-ascorbic acid, known as a prominent water soluble antioxidant ($1.50{\mu}g/mL$). The cellular protective effects of the ethyl acetate fractions on the $^1O_2$-induced cellular damage of human erythrocytes were increased in a concentration dependent manner ($5{\sim}50{\mu}g/mL$). The ${\tau}_{50}$ value in $5{\mu}g/mL$ was 45.6 min which was higher than that of (+)-${\alpha}$-tocopherol in all concentrations. These results indicate that the ethyl acetate fraction of purple sweet potato (I. batatas) has the excellent antioxidative capacity and could be applicable to anti-aging cosmeceutical ingredients for skin aging inhibition.

Anti-aging Effects of the Extracts from Leaf. Stem, Fruit and Seed of Yew (Taxus cuspidata Sieb) by Solvent Extraction Method (용매추출법에 의한 주목의 잎, 줄기, 과실 추출물의 항 노화 효과)

  • Yew (Taxus cuspidata Sieb.) chose that grow as medicine, food, decorative plant in Korea's Kyong-Gi province surroundings. Extracts of yew extracted leaf of 250g and stems of 300g with 1,3-butylene glycol (l,3-BG), propylene glycol (PG) and water. As results, external appearance of leaf extract of yew was slightly brown clear extract. The pH was 5.3${\pm}$0.5, and specific gravity was 1.012${\pm}$0.05, and refractive index was l.375${\pm}$0.05. Also, appearance of stem's extract was slightly brown clear extract, and the pH was 5.4${\pm}$0.5, and specific gravity was 1.016${\pm}$0.05, and refractive index was 1.358${\pm}$0.05. Oil of yew separated from seeds, and extracted polysaccharide high purity from fruits. As a result, specific gravity of oil was 0.987, and obtained 40.0% of yield. Total polyphenols amount of yew extract is detected 0.563% in leaves, 0.325% in stems, whereas total tannins amount contained 0.054% and 0.037% each in leaves and stems. As effect in cosmetics, the anti-oxidative effect by DPPH method is 75.0% in leaves, and stems was 64.0%. Collagen synthesis rate was shown high activity by 54.16% in stem's extract, 33.18% in leaves' extract. Also, PPE-inhibitory activities were 13.7% and 23.5% each in leaves and stems. Anti-inflammatory effect of yew seed oil displayed superior effect of 41% than control. Polysaccharide's molecular weight that is gotten from fruits was 5${\times}$10$^4$-3${\times}$10$\^$5/ dalton, and got 20.0${\pm}$5% of yield.

Biological activity and analysis of α-glucosidase inhibitor from mulberry (Morus alba L.) wine (오디와인의 생리활성 및 α-glucosidase 저해제의 분석)

  • Wine extracts of four different berry fruits, such as mulberry, blueberry, strawberry, and raspberry, were investigated for antioxidant, anti-tyrosinase, and ${\alpha}$-glucosidase activities by using in vitro assays. Additionally, quantitative changes of ${\alpha}$-glucosidase inhibitor in mulberry wine were determined by HPLC according to mulberry cultivars and fermentation process. Among four berry wines examined, mulberry wine showed the most potent ${\alpha}$-glucosidase inhibitory activity with 69.37% at 0.23 mg/mL, while blueberry and strawberry wines exhibited the strongest inhibition against DPPH radical and tyrosinase activity, respectively. Four compounds were isolated and purified from mulberry wine by a series of isolation procedures, such as solvent fractionation, and Diaion HP-20, ODS-A, and Sephadex LH-20 column chromatographies. Among them, Comp. 4 exerted the strongest ${\alpha}$-glucosidase inhibitory activity ($I_C_{50}=31.57{\mu}M$), and its chemical structure was identified as quercetin by UV and NMR spectral analysis. Finally, the "Daeseongppong" (16.83 ppm) muberry wine had larger amount of quercetin than the "Iksuppong" (14.85 ppm) and "Cheongilppong" (8.92 ppm) mulberry wines, but their contents of three mulberry wines decreased considerably with aging process. These results suggest that mulberry wine containing quercetin acted as ${\alpha}$-glucosidase inhibitor may be useful as a potential functional wine for improving diabetic disorder.

Comparison of Antioxidant and Matrix Metalloproteinases Inhibitory Effects of Sorbus commixta Twig Extracts before and after Fermentation with Lactobacillus pentosus (Lactobacillus pentosus에 의한 발효 전후 마가목 가지 추출물의 항산화 활성 및 Matrix Metalloproteinases 발현 억제 효과)

  • In this study, we investigated antioxidative and matrix metalloproteinases (MMPs) inhibitory effects of Sorbus commixta twig extracts and fermented extracts with Lactobacillus pentosus and analyzed active ingredients. The free radical scavenging activity ($FSC_{50}$) of non-fermented and fermented extracts of S. commixta twig' were 41.04 and $58.2{\mu}g/mL$, respectively. In the $Fe^{3+}-EDTA/H_2O_2$ system, the active oxygen scavenging activity ($OSC_{50}$) was 2.6 and $3.0{\mu}g/mL$, respectively. In the dermal fibroblasts, the intracellular reactive oxygen species (ROS) scavenging activity was 35.3% for non-fermented extract and 40.2% for fermented extracts at the concentration of $10{\mu}g/mL$. Also at the same concentration, the expression of MMPs (MMP-1, -2, -3) by western blot was 68.3, 35.0 and 24.2%, respectively for non-fermented extracts and 84.3, 70.5 and 69.2% for fermented extracts. TLC, HPLC, and LC/ESI-MS/MS were used for measuring the changes in the components of the extract before and after fermentation. As a result, caffeic acid, (-)-epicatechin, isoquercitrin, and quercetin were identified. From the results, S. commixta twig fermented extracts by L. pentosus showed greater ROS scavenging activity and inhibitory effects on MMPs expression than those of using non-fermented extracts. Therefore, it is suggested that S. commixta twig fermented extracts can be used as an anti-aging cosmetic material.

Effect of Water Extract of Cynanchi Wilfordii Radix in RANKL-induced Osteoclast Differentiation (백하수오(白何首烏) 물 추출물의 파골세포 분화에 미치는 영향)

  • Osteoporotic fracture became a serious social problem, which related with mortality and morbidity in old age population. Osteoclast which is responsible for bone resorption is originated from hematopoietic cell line and plays a key role osteoporotic bone loss. Cynanchum wilfordii (Asclepiadaceae) roots have been used in Korean folk medicine for the treatment of diabetes mellitus and aging progression. Also, recent studies have shown that the extract and fractions of Cynanchi Wilfordii Radix have various pharmacological actions including scavenging free radicals, enhancing immunity, reducing high serum cholesterol, and anti-tumor activity. However, the effect of extract of Cynanchi Wilfordii Radix in osteoclast differentiation had not been reported. Thus, we evaluated the effect of Cynanchi Wilfordii Radix on receptor activator of nuclear factor-${\kappa}B$ ligand (RANKL)-induced osteoclast differentiation. Through our study, we found that Cynanchi Wilfordii Radix significantly inhibited osteoclast differentiation induced by RANKL. Cynanchi Wilfordii Radix suppressed the activation of p38 pathway and $NF{\kappa}B$ in bone marrow macrophages (BMMs) treated with RANKL. Also, Cynanchi Wilfordii Radix significantly inhibited the mRNA expression of c-Fos, tartrate-resistant acid phosphatase (TRAP), osteoclast-associated receptor (OSCAR), nuclear factor of activated T cells (NFAT)c1 and cathepsin K in BMMs treated with RANKL. Particularly, Cynanchi Wilfordii Radix inhibited the protein expression of c-fos and NFATc1. Taken together, our results demonstrated that Cynanchi Wilfordii Radix may be useful treatment option of bone-related disease such as osteoporosis leads to fracture of bone and rheumatoid arthritis.

Sprouted Black Rice Oligopeptide Induces Expression of Hyaluronan Synthase in HaCaT Keratinocytes and Improves Skin Elasticity (발아 검은쌀 올리고펩타이드의 각질형성세포에서 Hyaluronan Synthase 발현과 피부 탄력 개선 효과)

  • In this study, the effect of oligopeptide ($Oligosproutin^{(R)}$) from sprouted black rice was evaluated for possible improvement in skin elasticity. We examined the changes in gene expression on oligopeptide-treated HaCaT cells using DNA microarray analysis. As a result, oligopeptide treatment showed a differential expression ratio of more than 2-fold : 745 genes were activated and 1011 genes were repressed. One of the most interesting findings is a 2-fold increase in hyaluronan synthase 2 (HAS 2) gene expression by oligopeptide. We also found that oligopeptide increased cell proliferation, HAS2 mRNA expression and intracellular ROS scavenging activity in HaCaT cells. A human clinical study which oil-in-water emulsion with oligopeptide was topically applied showed significant increase in skin elasticity. These results suggest that the sprouted black rice oligopeptide ($Oligosproutin^{(R)}$) can be effective anti-aging ingredient for cosmetics.

Antioxidative Effects of Skinned Mugwort (Artemisia vulgaris L.) Extracts on UV-Irradiated Hairless Mouse Skin (자외선 조사 무모쥐 피부에 도포한 애엽(Mugwort) 추출물의 항산화 효과)

  • This study investigated the antioxidative effect of mugwort (Artemisia vulgaris L.) extracts in hairless mouse skin from oxidative stress induced by UV-irradiation. After topical application on hairless mouse back with basic skin lotion group (control), ascorbic acid group (AA-0.5%, AA-1.0%, AA-2.0%, and AA-5.0%), and mugwort extract group (ME-0.5%, ME-1.0%, ME-2.0%, and ME-5.0%), the animals were irradiated to increasing doses of UVB (60 $mJ{\sim}100$ mJ) for 4 weeks. Hydrogen peroxide of hairless mouse skin homogenate significantly decreased in 2% (p<0.05) and 5% (p<0.05) of ME and AA groups. Hydroxyl radicals were decreased significantly in both of 2% and 5% ME groups as compared to AA groups (p<0.05). Oxidative stress levels deduced by oxidized protein contents were greatly decreased ($14.6{\sim}18.5%$) in all ME treatment groups, while only at 2% of AA treatment group. Lipid peroxide contents were greatly inhibited in all ME and AA treatment groups (p<0.01). Application of ME significantly increased catalase activity, over 25% in all mugwort and AA groups. Glutathione peroxidase activities were increased up to $20.5%{\sim}32.8%$ in 2.0% and 5% ME groups, whereas it increased in all AA groups. These results suggested that mugwort extract was more effective than that of ascorbic acid in protecting hairless mouse skin from photo-irradiation, and can be used as an potential anti-aging cosmetic ingredients.

Studies on the Sugar Analysis and Biological Activity of Sap from Juglans mandshurica Maxim. (가래나무 수액의 당성분 분석 및 생리활성 연구)

  • Even though the saps have been consumed as beverage in Korea, however a little research has been conducted on the chemical composition. We determined free sugars, mineral in the sap from Juglans mandshurica Maxim. collected from BongHwa region. The contents of glucose, sucrose, and fructose in the sap from J. mandshurica were 0.15, 2.73, and 0.09%, respectively. The prominent minerals in the sap from J. mandshurica were calcium and potassium. The contents of Ca, K, Mg, Fe, and Mn of sap from J. mandshurica were 0.61, 0.57, 0.12, 0.002, and 0.014 ppm, respectively. The sap from J. mandshurica show inhibitory effect on elastase. And real-time RT-PCR showed that sap from J. mandshurica increased mRNA level of AQP3 and HAS2 gene and increased hyaluronic acid production in HaCaT cells. These results indicated that the sap from J. mandshurica can potentially be used for developing cosmetic ingredient for skin moisturizing and anti-aging.

Comprehensive Literature Study on Efficacy of Marine Therapeutic Resources (해양치유자원의 효능관련 기존의 연구문헌 분석)

  • Purpose : Marine therapy is an activity that promotes public health such as constitution improvement, immunity improvement, and anti-aging by utilizing marine therapeutic resources such as seawater, mud, seaweed, salt and sea climate. In Europe developed countries, the marine therapy industry has been developing for centuries, with France, Germany, and Israel leading the way. Currently, it has achieved great industrial achievements and is of great help in improving the human health. The purpose of this study is to investigate how marine therapeutic resources benefit to human health, as well as how to study and utilize their efficacy. We analyzed previous research articles related to the effects of marine therapeutic resources. Methods : The study included a total of 830 published literatures in the last 20 years from the Republic of Korea and other contries. PubMed and Google Scholar were utilized to collect the foreign source while the local scientific publications were accessed through the Korean Education and research Information Service (KERIS) and Korean studies Information Service System (KISS). The keywords used to search foreign literature were "marine therapy", "Thalassotherapy", "seawater", "deep seawater", "saline groundwater", "sand therapy", "mud therapy", "hydrotherapy", "seaweed", "Sun light", "sea salt", "marine animal", and "marine microorganisms" were combined, and for the domestic literature, the keywords were "marine therapy", "marine therapeutic resources", "seawater", and "sand". Results : A total of 830 research papers were found as a result of searching for domestic and international papers related to marine therapeutic resources. The collected documents were classified into 175 seawater resources, 259 marine mineral resources, 41 marine environment, and 355 marine organisms. The efficacy of each marine therapeutic resources was analyzed. By resources type, there were about 213 papers on the efficacy of seaweed, followed by about 175 papers on seawater, 142 on microorganisms, 124 on mud/peat, and sand, salt, minerals and others are appeared in order (Table 1). Conclusion : Korea has the highest marine biodiversity index, excellent tidal flats, four distinct seasons, and various sea environments of the East sea, Yellow sea, South sea and Jeju sea. For this reason, Korea has a much more diverse marine therapeutic resources than other advanced countries in the marine therapy industry. prebiously, we thought that the sea was only valuable as a shipping port and fishery industry. But now, it been shown that the ocean can become a new industrial field which can contribute to human health and well-being by providing healing and therapy to people through the gift of marine resources.

Antioxidative Effects and Component Analysis of Extracts of the Rumex acetosa L. (수영 전초 추출물의 항산화 활성 평가 및 성분 분석)

  • In this study, the antioxidative effects and component analysis for the extracts of Rumex acetosa L. were investigated. All experiments were performed with 50% ethanol extract, ethyl acetate fraction and aglycone fraction obtained from dried R. acetosa L.. Free radical scavenging activities (1,1-diphenyl-2- picrylhydrazyl) size of, in the order of aglycone fraction > ethyl acetate fraction > 50% ethanol extract, aglycone fraction ($45.10{\mu}g/mL$) showed the highest radical scavenging activity. Reactive oxygen species (ROS) scavenging activity (total antioxidant capacity, $OSC_{50}$) on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system was also, in the order of ethyl acetate fraction> aglycone fraction> 50% ethyl acetate fraction, ethyl acetate fraction ($2.68{\mu}g/mL$) was shown a great antioxidant capacity. The total antioxidant capacity of the ethyl acetate fraction was found to be greater than L-ascorbic acid, known as a typical hydrophilic antioxidant ($6.88 {\mu}g/mL$). The cellular protective effects of R. acetosa L. extracts on the $^1O_2$-induced cellular damage of human erythrocytes were exhibited at all concentration-dependent ($1{\sim}25{\mu}g/mL$). Especially, aglycone fraction (${\tau}_{50}$, 104.80 min) in $25{\mu}g/mL$ showed the most protective effect among extracts. Components of the ethyl acetate fraction obtained from R. acetosa L. extracts were analyzed by TLC, HPLC chromatogram, LC/ESI-MS/MS. As a result, the ethyl acetate fraction contained several flavonoids, such as orientin, isoorientin, vitexin, isovitexin. These results indicate that the R. acetosa L. extracts can be used as antioxidants in biological systems, particularly skins exposed to UV radiation by quenching and/or scavenging $^1O_2$ and other ROS. Thus, the extracts of R. acetosa L. could be applicable to new anti-aging cosmeceutical ingredients.
