Activation of Heme Oxygenase-1 by Mangiferin in Human Retinal Pigment Epithelial Cells Contributes to Blocking Oxidative Damage

  • Mangiferin is a kind of natural xanthone glycosides and is known to have various pharmacological activities. However, since the beneficial efficacy of this compound has not been reported in retinal pigment epithelial (RPE) cells, this study aimed to evaluate whether mangiferin could protect human RPE ARPE-19 cells from oxidative injury mimicked by hydrogen peroxide (H2O2). The results showed that mangiferin attenuated H2O2-induced cell viability reduction and DNA damage, while inhibiting reactive oxygen species (ROS) production and preserving diminished glutathione (GSH). Mangiferin also antagonized H2O2-induced inhibition of the expression and activity of antioxidant enzymes such as manganese superoxide dismutase and GSH peroxidase, which was associated with inhibition of mitochondrial ROS production. In addition, mangiferin protected ARPE-19 cells from H2O2-induced apoptosis by increasing the Bcl-2/Bax ratio, decreasing caspase-3 activation, and blocking poly(ADP-ribose) polymerase cleavage. Moreover, mangiferin suppressed the release of cytochrome c into the cytosol, which was achieved by interfering with mitochondrial membrane disruption. Furthermore, mangiferin increased the expression and activity of heme oxygenase-1 (HO-1) and nuclear factor-erythroid-2 related factor 2 (Nrf2). However, the inhibition of ROS production, cytoprotective and anti-apoptotic effects of mangiferin were significantly attenuated by the HO-1 inhibitor, indicating that mangiferin promoted Nrf2-mediated HO-1 activity to prevent ARPE-19 cells from oxidative injury. The results of this study suggest that mangiferin, as an Nrf2 activator, has potent ROS scavenging activity and may have the potential to protect oxidative stress-mediated ocular diseases.

가미생맥산(加味生脈散) 및 개별약재의 항산화 및 항염증 효능에 대한 비교 연구 (Anti-Oxidative and Anti-inflammatory Effect of Combined Extract and Individual Extract of GamiSaengmaeksan)

  • Objectives : The aim of this study is to investigate the various effects of individual or combined extract of GamiSaengmaeksan (GSS) on cell viability, anti-inflammatory and antioxidant activityMethods : In order to evaluate cytotoxicity, MTT assay was performed. We investigated the levels of proinflammatory cytokines such as tumor necrosis factor (TNF)-α and interleukin (IL)-6 and interleukin (IL)-1β, and nitric oxide(NO) in LPS-induced RAW 264.7 cells to check the effects on anti-inflammatory activity. The level of NO production in RAW 264.7 cells was measured by using Griess reagent. The levels of cytokines and ROS were measured by Luminex and Flow cytometry, respectively.Results : At concentration of 200 ㎍/㎖ GSS, cytotoxicity was observed in RAW 264.7 cells. However, at concentration less than 100 ㎍/㎖ of both combine and individual GSS, cytotoxicity was not observed in Raw 264.7 cells. However, the level of ROS in RAW 264.7 cells were decreased at both extract of 100 ㎍/㎖ GSS. Also, the level of NO in RAW 264.7 cells were decreased from extraction of concentration of 100 ug/ml in GSS and individual-extraction of Liriopis Tuber, White Ginseng and Glycyrrhizae Radix. In addition, productions of pro-inflammatory cytokines (TNF-α) in LPS-induced RAW 264.7 cells were decreased from extraction of concentration of 10 and 100 (㎍/㎖) in GSS and individual-extraction of Liriopis Tuber.Conclusions : It is concluded that combined extract of GSS appears to be more effective in anti-oxidation and anti-inflammatory effect than those in individual-extraction of GSS. These results may be developed as a raw material for new therapeutics to ease the symptoms related with inflammatory and oxidative stress.

팽이버섯 추출물의 항산화 및 항염증 활성 (Antioxidant and Anti-inflammatory Effect of Extracts from Flammulina velutipes (Curtis) Singer)

  • 본 연구는 팽이버섯으로부터 열수 및 에탄올 추출물의 항산화와 항염증 활성을 탐색하였다. 라디칼 소거능을 측정한 결과, DPPH 라디칼 소거능은 팽이버섯 열수 추출물의 경우 천연 항산화제인 비타민 C보다 활성이 우수하지는 않지만 다른 종류의 버섯 추출물보다 우수한 효능을 확인하였고 추후 분리, 정제를 통하여 유효 물질을 발굴하여 새로운 소재를 찾아내는 연구가 필요할 것으로 판단된다. ABTS 라디칼 소거능과 FRAP을 이용한 총항산화 효능에 관한 결과에서도 팽이버섯 추출물이 항산화 효과를 나타내는 것을 확인하였고, FTC법을 이용한 지질과산화 억제 효능을 살펴본 결과도 마찬가지로 열수 추출물이 에탄올 추출물보다 항산화 활성이 우수한 것을 확인하였다. 한편, 세포 독성을 살펴보기 위하여 항산화력이 우수한 팽이버섯 열수 추출물을 이용하여 정상 신경세포에 MTT assay를 수행한 결과, 세포의 생존율은 1.0 mg/mL의 이하 농도까지는 생존에 영향을 미치지 않는 것을 확인하였고 신경세포 보호효능 실험에서는 1.0 mM $H_2O_2$로 유도시킨 세포사에 대해 농도의존적인 신경세포 보호 효과가 있는 것을 확인하였으며, 마크로파지 세포에서도 독성에 대하여 신경세포와 유사한 결과를 확인하였다. NO의 저해효능도 농도 의존적으로 감소하였고 추가적으로 세포의 유전학적 분석인 western blot으로 iNOS와 COX-2를 수행하였을 때 LPS군에 비해 팽이버섯 열수 추출물군에서 iNOS 단백질 발현량이 감소하는 것을 확인하였다. 하지만 좀 더 명확한 결과를 위하여 차후 효소활성 및 동물연구 등이 추가적으로 필요할 것으로 판단된다.

HaCaT세포에서 TNF-α에 의해 유도되는 MMP-1에 대한 Gallic Aicd의 저해 효과 (Inhibitory Effect of Gallic Aicd on TNF-α-induced matrix metalloproteinase-1 (MMP-1) in HaCaT Cells)

  • 본 연구는 항주름 소재를 개발하기 위해서 목단피로부터 gallic acid (GA)를 분리하여 항산화능을 측정하였고, HaCaT 세포에서 세포독성을 측정하였다. 또한 HaCaT 세포에서 tumor necrosis factor alpha (TNF-${\alpha}$)에 의해 유도되는 matrix metalloproteinase-1 (MMP-1) mRNA 발현, protein 발현, 분비에 대한 GA의 영향을 관찰하였다. 결과로써 GA는 30 ${\mu}g/mL$$IC_{50}$과 함께 항산화능을 나타내었고, 그것의 항산화능은 합성항산화제인 butylated hydroxyanisol (BHA)보다 높았다. GA는 HaCaT 세포에서 고농도인 200 ${\mu}g/mL$ 처리시 약한 세포독성을 나타내었다. 또한 HaCaT세포에서 TNF-${\alpha}$ (10 ng/mL)의 처리에 의해 증가된 MMP-1의 mRNA 발현, protein 발현, 분비는 GA의 처리에 의해 농도 의존적으로 유의적인 감소를 나타내었다(p<0.05). 그러므로 GA는 항산화 효과와 TNF-${\alpha}$로부터 유도되는 MMP-1의 발현을 저해함으로써 피부 주름을 개선할 수 있는 주름개선제로서의 활용가능성을 확인하였다.

위중전침의 항산화 효과에 대한 실험적 연구 (A study on the anti-oxidative effect of electroacupuncture at Wijung (BL40) in rats)

  • Objectives : The purpose of this study is to observe the anti-oxidative effects of electroacupuncture at Wijung (BL40) in rats. Methods : The author performed several experimental items including measurements of body weight, liver index, levels of albumin, total bilirubin, LDL-cholesterol, LDH, GOT and GPT in blood serum, and levels of SOD, glutathione, catalase, No and MDA (malondialdehyde) in liver, histological analysis of liver. Results : 1. In the BL40-EA group, liver index was decreased significantly compared to those of the control group, the holder group and the sham-EA group. 2. In the BL40-EA group, the albumin level was increased significantly compared to those of the control and holder group. LDL-cholesterol and GOT level were increased significantly compared to those of the control group and the holder group. 3. In the BL40-EA group, the SOD activity & the Catalase activity were decreased significantly compared to those of the control group and the holder group. 4. In the BL40-EA group, the density of liver tissue was more similarly maintained to the normal group compared to those of the control group and the holder group. Conclusion : These results suggest that electroacupuncture at BL40 has an antioxidant effect in human, and hereafter continuous study & clinical application is needed.

Effect of Astaxanthin on Anti-Inflammatory and Anti-Oxidative Effects of Astaxanthin Treatment for Atopic Dermatitis-induced Mice

  • Background: This study sought to determine whether the antioxidant effects of astaxanthin (AST) could have an anti-inflammatory effect to reduce inflammation caused by atopic dermatitis (AD). Methods: Using a mouse model of AD induced by phtalic acid (PA), the levels of inflammation, inflammatory agents, and evidence of antioxidant activity were examined in PA treated mice (n = 3), PA-AST treated mice (n = 3), and a control group of mice (n = 3). This included measurements of ear thickness, levels of mast cells, IgE, inflammatory cytokine, malondialdehyde (MDA), hydrogen peroxide, HO-1, and GPx-1. Results: AST treatment significantly prevented inflammation as measured by ear thickness (p < 0.05), mast cell count (p < 0.001), and IgE concentration in the blood (p < 0.001). Levels of TNF-α (p < 0.001), IL-1β (p < 0.001), IL-6 (p < 0.001), and MDA (p < 0.05) were also significantly lower. In addition, GSH levels increased significantly (p < 0.001), and the level of hydrogen peroxide significantly reduced (p < 0.01). The expression of HO-1, GPx-1 increased. Conclusion: In this small experimental study, AST acted on inflammatory mechanisms that induced AD, through anti-inflammatory and antioxidant mechanisms, and is a candidate of interest in the clinical treatment of AD.

추출 방법에 따른 자소엽 추출물의 항산화 효과 비교 (Comparison of Anti-Oxidative Activities of Perilla frutescens Extracts by Extraction Methods)

  • 자소엽(perilla frutescens)은 꿀풀과(Labiatae)에 속하며 널리 알려져 있는 약용 식물이다. 본 연구에서는 자소엽을 물, 열수, 초음파 추출 방법으로 추출하여 항산화 효과를 비교하고 가장 효과적인 추출방법을 제시하고자 한다. 물, 열수, 초음파 처리를 통해 각각의 자소엽 추출물을 제조 하였고, DPPH 라디칼 소거능 및 총 페놀 함량을 통해 항산화 효과를 검증하고, 인간 간세포인 HepG2에 대한 세포 독성효과와 hydrogen peroxide ($H_2O_2$)로 유도된 산화적 스트레스로부터 간세포 보호효과를 3-[4,5-dimethylthiazole-2-yl]-2,5-diphenyltetrazolium bromide (MTT) assay로 확인하였다. 자소엽 초음파 추출물은 $5000{\mu}g/mL$농도에서 69.07%의 DPPH 라디칼 소거능을 나타내며 물, 열수 추출물과 비교하여 우수한 항산화 효과를 나타내었다. 또한 총 페놀 함량 측정 결과 $51.60{\pm}1.06mg\;GAE/g$ extract 로서 물, 열수 추출물 보다 높은 총 페놀 함량을 확인하였다. 그러나 산화적 스트레스에 의한 세포 보호효과는 미비하였다. 본 연구를 통해 추출 방법에 따른 항산화 효과의 차이를 확인하였으며, 우수한 항산화 효과를 나타낸 자소엽 초음파 추출물을 이용하여 추가적인 연구가 필요 할 것으로 사료된다.

국내 자생 자원식물들의 항산화 활성 검색 (Screening of Natural Herb Resources for Anti-oxidative Effects in Korea)

  • 자생식물의 항산화 활성 탐색을 위하여 140종의 식물을 80% ethanol로 추출하여 활성을 확인하였다. 그 결과 며느리배꼽이 가장 높은 항산화 활성을 나타내었고(VitaminE, ORACPE=1.0), 쉬땅나무, 찔레나무, 좁쌀풀, 갈참나무, 신갈나무 그리고 싸리의 순으로 항산화활성이 높은 것으로 조사되었다(ORACPE ${\geq}$ 1.50). 또한 80% 에탄올 추출물의 용매별(n-hexane, $CH_2Cl_2$, EtOAc, n-BuOH, water) 분획층 ORAC 결과에서도 며느리배꼽과 쉬땅나무가 각각 $1.13{\pm}0.002$, $1.10{\pm}0.004$, $3.67{\pm}0.232$, $1.62{\pm}0.049$, $1.05{\pm}0.003$; $1.02{\pm}0.007$, $v1.00{\pm}0.016$, $2.94{\pm}0.130$, $1.80{\pm}0.019$ 그리고 $1.04{\pm}0.012$로 높은 값을 나타내었다. 그 중 항산화 활성이 가장 높은 EtOAc 분획층의 유효성분 규명 등의 추가 연구가 필요하다.

Antioxidant Activities and Inhibition Effects on Oxidative DNA Damage of Callus Derived from Abeliophyllum distichum Nakai

  • Abeliophyllum distichum is Korea Endemic Plants and its genetic resources found from Korea only. Bioactivities of A. distichum such as antioxidant, anti-cancer, and anti-inflammatory studies have been proved through many researches. Whereas, there are no studies on the biological activity of its callus extracts. In this study, we investigated the antioxidant activities of callus extracts derived from A. distichum and its inhibitory effect on oxidative DNA damage. The antioxidant activities were assessed using radical scavenging assays with DPPH, ABTS, and reducing power assay and the inhibitory effects on oxidative DNA damage were measured using ${\varphi}-174$ RF I plasmid DNA cleavage assay. In addition, callus extracts derived from A. distichum showed high antioxidant acitivties and no cytotoxicity in NIH/3T3. Also, it has significantly suppressed expression of ${\gamma}$-H2AX and p53 protein and mRNA levels in NIH/3T3 cells exposed to oxidative stress. Therefore, the callus extracts derived from A. distichum has potential antioxidant activity that can provide protective effects against the oxidative DNA damage caused by free radicals. This study suggest that it is valuable as cosmetics and medicine for antioxidant and cancer preventive materials.

Anti-oxidative and Antibacterial Constituents from Sedum hybridum

  • Phytochemical studies on the whole extract of Sedum hybridum L., a Mongolian medicinal plant, has been undertaken to isolate active principles responsible for its anti-oxidative and antibacterial activities. Eighteen known compounds, i.e. (1) quercetin, (2) kaempferol, (3) herbacetin-8-O-${\beta}$-D-xylopyranoside, (4) myricetin, (5) gossypetin-8-O-${\beta}$-D-xylopyranoside, (6) gallic acid, (7) 2,4,6-tri-O-galloyl-D-glucopyranose, (8) 6-O-galloylarbutin, (9) myricetin-3-O-${\alpha}$-L-arabinofuranoside, (10) quercetin-3-O-${\alpha}$-L-arabinofuranoside, (11) caffeic acid, (12) ethylgallate, (13) (-) epigallocatechin-3-O-gallate, (14) palmitic acid, (15) stearic acid, (16) stearic acid ethyl ether, (17) ${\beta}$-sitosterol and (18) ${\beta}$-sitosteryl-O-${\beta}$-D-glucopyranose have been isolated and their molecular structures identified by spectroscopic analysis. Thirteen substances including seven flavonol components (1, 2, 3, 4, 5, 9 and 10), five gallic acid derivatives (6, 7, 8, 12 and 13) and caffeic acid (11) exhibited significant, dose-dependent, DPPH radical scavenging activity. Galloyl esters 12 and 13 were revealed to be main active principles for the antibacterial property of the extract of Sedum hybridum L.
