Effect of Anti-oxidant Activity and the Skin Whitening Action on Plantago asiatica L. Root Extract (차전초 뿌리 추출물이 항산화 활성 및 피부미백 작용에 미치는 영향)

  • To investigate the effect of Plantago asiatica L. Root extract on skin care, we measured anti-oxidant activity and whitening action. As a result of measuring DPPH radical scavenging activity to examine independent anti-oxidation of PRE, there was slight scavenging activity. Fluorescent materials DHE, DCF-DA and DHR were each used to measure superoxide anion, hydrogen peroxide, and hydroperoxide created in RAW 264.7 cells, all concentrations were found to dependently prevent ROS production. Tyrosinase activation was found to be blocked dose-dependant. Melanin production was also prevented dose-dependant, but the effects were slight. Therefore, it is expected to be used effectively in development of functional cosmetic materials.

  • This study aims to investigate the biological activities related to hair loss of short anti-oxidant peptides (DK peptides) $5{\alpha}$-reductase inhibition and anti-oxidation. The series of DK peptides were generated amphipathic helical properties using leucines, lysines and tryptophan residues. Cell viability and free radical scavenging activities were performed using 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl-tetrazolium bromide (MTT) and 2, 2-diphenyl-1-picrylhydrazyl (DPPH) assay, respectively. The DK peptides were investigated for inhibitory activity against $5{\alpha}$-reductase. Antioxidant activities were determined by means of, 2, 2-diphenyl-1-picrylhydrazyl (DPPH) assays. All peptides could inhibit $5{\alpha}$-reductase in lipopolysaccharide-stimulated macrophage. In conclusion, DK peptides was suggested as the most attractive ingredients for improving hair loss, because of the high inhibitory against $5{\alpha}$-reductase inhibition.

Effects of Phenylpropanoid Compounds on Melanin Production in B16 Melanoma Cells (B16 Melanoma 세포에서 Phenylprlopanoid 화합물이 Melanin 생성에 미치는 영향)

  • To investigate the relationship between structure and biological activity of phenylpropanoids, we measured effects of phenylpropanoids on anti-oxidant and whitening activity, In DPPH radical scavenging activity, caffeic acid analogues showed the significant anti-oxidant activity. Although phenylpropanoids did not inhibit purified-tyrosinase activity, they significantly inhibited tyrosinase activity and melanin production in MSH-stimulated B16 melanoma cells. However, phenylpropanoids did not affect tyrosinase expression in MSH-stimulated B16 melanoma cells, which suggest that inhibition of MSH-induced melanin production was due to tyrosinase inhibition mediated via other signal pathways but not expression of tyrosinase. Phenylpropanoids also significantly inhibited both hyaluronidase and elastase activity, suggesting that phenylpropanoids may be used as whitening, hydration and anti-wrinkling agents. Hydroxyl residue of aromatic ring in phenylpropanoids plays an important role in anti-oxidant and whitening activity.

Anti-oxidant Activity and Whitening Activity of Bamboo Extracts (대나무 추출액의 항산화 작용과 미백작용)

  • To investigate the possibility of development as a whitening agent using bamboo extracts (Phyllostachys nigra var. henonis), we measured DPPH assay, NBT/XO assay, intracellular ROS scavenging assay, tyrosinase assay and MSH-induced melanin production in B-16 cells. Bamboo extracts had dose-dependently anti-oxidant activity in DPPH, NBT/XO and intracellular ROS assay. Bamboo extracts appear to inhibit xanthine oxidase directly. Bamboo extracts inhibited not only purified tyrosinase activity but also inhibited tyrosinase activity and melanin production in B16 melanoma cells stimulated by $1{\mu}M\;{\alpha}$-MSH. Anti-oxidant activity and cytotoxicity of ethyl acetate fraction was more potent than those of water fraction, whereas whitening effect of water fraction was stronger than ethyl acetate fraction. Therefore, these results suggest that water fraction of bamboo extracts may be useful for the development as whitening agents reducing cytotoxicity.

Anti-Oxidant and Hepato-protective Activities of the Stems of Acanthopanax senticosus

  • The anti-oxidant activities of Acanthopanax senticosus stems were investigated. The n-BuOH fraction of A. senticosus stems exhibited a significant decrease in serum transaminase activities elevated by hepatic damage induced by CCl$_4$-intoxication in rats. The n-BuOH fraction inhibited the sGPT activities by 65.79%. The n-BuOH fraction showed the increase in the anti-oxidant enzymes such as hepatic cytoso1ic superoxide dismutase, catalase and glutathione peroxidase activities by 30.31, 19.82 and 155%, respectively, in CCl$_4$-intoxicated rats. These results suggest that the stems of A. senticosus possess not only the hepatoprotective, but also the anti-oxidant activities in rats.

Anti-oxidant, Anti-inflammatory and Anti-cancer Effect of Methanol Extract of Pogostemon cablin (광곽향 메탄올 추출물의 항산화, 항염증 및 암세포 증식 억제 효과)

  • In the present study, the substance that show anti-proliferation of cancer cells as well as anti-oxidant and anti-inflammatory effect was searched. As a results, the methanol extract of Pogostemon cablin (P. cablin), is a well-known herb for traditional medicine in Korea and China for treating the digestive disorders, less of appetite, vomiting and diarrhea, inhibited the growth of various cancer cells such as A549, HepG2, MCF7 and HT29 cells. Cytotoxic effect of methanol extraction of P. cablin was excellent in A549 cells. P. cablin extract induced cell cycle arrest at G1 phase of A549 in a dose dependent manner. And it induced phosphorylation of p38 and Cdc25A and reduced expression of Cdc25A, Cdks, Cyclins and phospho-Retinoblastoma (Rb) proteins. Therefore, P. cablin extract seems to act through the p38 - Cdc25A - Cdk - Cyclin - Rb pathway in A549 cells. In addition, P. cablin extract showed anti-oxidant effect by DPPH free radical scavenging assay and anti-inflammation effect by inhibition of lipopolysaccharide (LPS)-induced nitric oxide (NO) in RAW 264.7 cells in a dose-dependent manner. Taken together, these results suggest that P. cablin may be used as not only candidate materials for anti-cancer, anti-inflammatory and anti-oxidant, moreover, it would be possible utilized in various health functional food materials.

Anti-oxidant and Anti-microbial Activities of Herb-combined Remedies used in Traditional Korean Medicine for Treating Breast Cancer (유암 처방에 사용되어온 한약재 복합 처방전의 항산화 및 항균활성에 관한 연구)

  • Sipyukmiryuki-eum (SYMYKE), Danjacheongpi-tang (DJCPT), Jipae-san Ⅰ (JPS Ⅰ), Jipae-san Ⅱ (JPS Ⅱ), and Chungganhaeul-tang (CGHUT) are representative herb-combined remedies used in traditional Korean medicine for treating breast cancer patients, as mentioned in "Dongeuibogam." In this study, we investigated the total phenolic contents (TPCs) and the anti-oxidant and anti-microbial activities of hot water and 70% ethanol extracts of these herbal prescriptions. Among the five herb-combined remedies, the extraction yields of the hot water extracts and 70% ethanol extracts were the highest in JPS Ⅱ (34.30%) and DJCPT (30.50%), respectively. The TPCs of the hot water extracts from the herb medicines were rich in the order of JPS Ⅰ < CGHUT < JPS Ⅱ < SYMYKE < DJCPT. In addition, the 70% ethanol extracts from the herb medicines were rich in the order of JPS Ⅱ < JPS Ⅰ < CGHUT < SYMYKE < DJCPT. Among them, DJCPT and SYMYKE displayed a strong anti-oxidant capability, which was determined using ferric-reducing anti-oxidant power and scavenging of 2,2'-diphenyl-1-picrylhydrazyl and 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonate) cationic radical activity assays. In addition, anti-microbial activities against Staphylococcus aureus and Escherichia coli were stronger in the 70% ethanol extracts than in the hot water extracts. Together, these findings reveal a positive relationship between TPCs and their anti-oxidant activities.

Comparison of Anti-Oxidative and Cox-2 Promoter Activities of Lepidoptera Extracts (Lepidoptera 추출물에 의한 항산화 및 Cox-2 프로모터 활성 비교)

  • Lepidoptera (butterflies) extracts, traditionally employed as medicines, have various biological activities. Five species of Lepidoptera (Papilio maackii, Papilio xuthus, Pieris rapae, Eurema hecabe, and Sasakia charonda) were extracted with distilled water (DW), dimethyl sulfoxide (DMSO), ethanol (EtOH), and methanol (MeOH). Each extract was analyzed for anti-oxidant and anti-inflammatory activities using the 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay method, the ferric reducing ability of plasma (FRAP) test, and a cyclooxygenase-2 (COX-2) promoter assay. The results suggest that Lepidoptera extracts have valuable anti-oxidant and anti-inflammatory properties, supporting the idea that the extracts may serve as a food biomaterial(s) preventing oxidative processes and inflammatory damage.

Verification of anti-oxidative activity of Aruncus dioicus, a native plant of Ulleungdo (울릉도 자생식물 삼나물(Aruncusdioicus)의 항산화 활성 검증)

  • The extracts of Aruncus dioicus were investigated for anti-oxidant and biological activities in order to verify its potential as functional ingredients of cosmetic products. The ethyl acetate (EtOAc) extract of A. dioicus showed excellent scavenging activity using DPPH and ABTS anti-oxidant analysis at the $1,000{\mu}g/mL$ concentration. While tyrosinase inhibitory effect was measure for whitening assay and showed 59.2%, the suppression levels of elastase and collagenase were 56% and 90% respectively for anti-wrinkle activity. Therefore, it is plausible to conclude that A. dioicus extract, especially EtOAc fraction which has outstanding whitening, anti-oxidant, and anti-wrinkle activities, could be used as a new functional materials for cosmetics.

Anti-oxidant, anti-inflammatory and Whitening effect of Benincasa hispida seed extract (동과자 추출물의 항산화, 항염 및 미백 효능)

  • To investigate the natural cosmetic ingredients of Benincasa hispida seed extract on skin care, we measured anti-oxidant and anti-inflammatory, and whitening effect. DPPH free radical scavenging activity was increased in a dose-dependent manner. The total phenolic content was higher in methanol extract (22.42±0.002 mgGAE/g) than water extract (9.77±0.002 mgGAE/g). MTT assay was demonstrated that the seed extract did not have a cytotoxic effect in RAW264.7 and B16F10 cell lines. We also examined to find out the inhibitory activity on NO production and secretion of TNF-α cytokine in LPS-induced RAW264.7 cells. In B16F10 melanoma cells, the seed extract significantly suppressed α-MSH induced melanin synthesis. Furthermore, westernblot analysis revealed that methanol extract dramatically downregulated the expression level of MITF, TRP-1 and TRP-2. Taken together, the B. hispida seed extract posses anti-oxidant, anti-inflammatory and skin whitening activities, which might provided its functional efficacy in cosmetic materials.
