Effects of Sleep on Balance Control and Reaction Time to Visual Stimuli (수면이 균형조절과 시각적 자극 반응시간에 미치는 영향)

  • Objectives: To find evidence that sleep is necessary for normal brain function, thus indicating that declines in both sleep quality and quantity are related to worse performance of many daily tasks and deteriorated physical functions. The present study investigates the relationships of balance control and reaction time with sleep quality. Methods: 58 healthy (male 20, female 38) volunteers with informed consent participated in this study. The Self-reported Pittsburgh Sleep Quality Index (PSQI) was used to evaluate sleep quality and relevant factors, and the subjects were divided into groups A (PSQI < 5) and B ($PSQI{\geq}5$) based on this index. Static balance control and reaction time to visual stimuli were conducted to assess their relationship with sleep quality. Results: Group B exhibited excessive daytime sleepiness significantly more often compared to group A. Static balance control did not markedly change relative to sleep quality, but reaction time and error to visual stimuli were significantly increased in group B compared to group A. Conclusion: These findings indicate that a decline in sleep quality can result in delayed reactions, as well as decreased accuracy of these reactions. They also suggest that low sleep quality may be associated with changes in physical functions, including balance control through reduced selective attention.

Active Component of Fatsia japonica Enhances the Transduction Efficiency of Tat-SOD Fusion Protein both In Vitro and In Vivo

  • It has been reported that Tat-SOD can be directly transduced into mammalian cells and skin and acts as a potential therapeutic protein in various diseases. To isolate the compound that can enhance the transduction efficiency of Tat-SOD, we screened a number of natural products. 3-O-[$\beta$-D-Glucopyranosyl(1$\rightarrow$4)-$\alpha$-L-arabinopyranosyll-hederagenin (OGAH) was identified as an active component of Fatsia japonica and is known as triterpenoid glycosides (hederagenin saponins). OGAH enhanced the transduction efficiencies of Tat-SOD into HeLa cells and mice skin. The enzymatic activities in the presence of OGAH were markedly increased in vitro and in vivo when compared with the controls. Although the mechanism is not fully understood, we suggest that OGAH, the active component of Fatsia japonica, might change the conformation of the membrane structure and it may be useful as an ingredient in anti-aging cosmetics or as a stimulator of therapeutic proteins that can be used in various disorders related to reactive oxygen species (ROS).

Isolation and Identification of Marine Bacteria with High Removal Efficiencies for Nitrogen-Phosphate In Gwangyang bay (광양만 해역에서의 고효율 질소-인 제거 해양박테리아 탐색 및 분리)

  • 371 strains of marine bacteria were isolated from Gwangyang bay in Korea. The dominant species were identified as Pseudomonas aeruginosa, Aeromonas hydrophila, P. fluorescens, P. paucimobilis, Chryseomonas luteola and P. vescularis. To screen marine bacteria capable of removing nutrients and organics, marine bacteria was inoculated in 10 mL of marine broth 2216 (DIFCO) with $NH_3-N$ (100 mg/L), ${NO_3}^{-}-N$ (100 mg/L), and ${PO_4}^{-3}-P$ (10 mg/L) with 1.0% (v/v), and incubated for 12 h. Results from the screening test, showed that the removal efficiencies for $COD_{Cr}$, ammonia niterogen, nitrate nitrogen, and phosphate were over 25% for 16 strains, 15% for 9 strains, 50% for 63 strains, and 15% for 80 strains, respectively. Aeromonas hydrophila, Chryseomonas indologenes, Pseudomonas diminuta, Vibrio parahaemolyticus were selected for nutrients removal experiments. For the batch test, 4 species of marine bacteria were inoculated in modified marine broth containing with nutrients($COD_{Cr}$ 250 mg/L, $NH_3-N$ 40 mg/L, ${NO_3}^{-}-N$ 40 mg/L, ${PO_4}^{3-}-P$ 10 mg/L, respectively), incubated for 10 hr and the removal efficiencies were measured.

Transcriptome Analysis of Human HaCaT Keratinicytes by Ginsenosides Rb1 and Rg1 (진세노사이드 Rb1과 Rg1에 의한 HaCaT 피부각질세포의 전사체 분석)

  • This study examined the efficacy and the mechanism of action of biological response modifiers, ginsenosides Rb1 and Rg1 isolated from Panax ginseng C.A. Meyer on human keratinocytes HaCaT cell lines. A non-significant cytotoxic response was obtained in the HaCaT cell lines on treatment with various concentrations of ginsenosides Rb1 and Rg1 for different time durations. Furthermore, the global changes in the mRNA profile of HaCaT cells were investigated using DNA microarrays after stimulation with the ginsenosides Rb1 and Rg1. Ginsenosides Rb1 and Rg1 strongly increased FGF2 in HaCaT cells, and were found to be a candidate gene for antioxidant activity and elasticity. Other key candidate genes for antioxidant activity, such as FANCD2, LEPR, and FAS, also show enhanced regulation in HaCaT cells treated with ginsenoside Rb1. This study will be useful for understanding the regulatory genes involved in skin elasticity and signal transduction pathway stimulated by the ginsenoside Rb1. This paper currently focuses on the key factors regulating the interaction of anti-aging principles and skin elasticity.

Expression of Mycosporine-like Amino Acids Biosynthetic Genes in the Chlamydomonas sp. Exposed to Radiofrequency (Radiofrequency에 노출된 Chlamydomonas sp.의 mycosporine-like amino acids 생합성 유전자 발현)

  • Mycosporine-like amino acids (MAAs) are UV-absorbing substances, and diverse marine organisms have the evolved the capacity to diminished the direct and indirect damaging effects of environmental ultraviolet radiation by synthesis and accumulation of MAAs. In this study, we manufactured a radiofrequency (RF) generation device and applied to microalgal culture. $0.35{\pm}0.05$ mHz of RF was supplied to culture vessel for Chlamydomonas sp. and samples were harvested at the designated time intervals (1, 0.5, 1 and 2 hr). MAAs biosynthetic genes, dehydroquinate synthase homolog (DHQS-like) and nonribosomal peptide synthetase homolog (NRPS-like), were cloned from Chlamydomonas sp. and their gene expressions under the RF exposure were analyzed using qRT-PCR. DHQS-like and NRPS-like gene expressions of Chlamydomonas sp. exposed to RF were increased 1.46 and 1.19 fold at 1 hr, respectively. These results means that DHQS-like and NRPS-like genes can be good biomarker candidates for diagnosis of MAAs biosynthesis in the Chlamydomonas sp.

Skate cartilage extracts containing chondroitin sulfate ameliorates hyperlipidemia-induced inflammation and oxidative stress in high cholesterol diet-fed LDL receptor knockout mice in comparison with shark chondroitin sulfate

  • BACKGROUND/OBJECTIVES: In this study, we investigated the beneficial effects of skate cartilage extracts containing chondroitin sulfate (SCS) on hyperlipidemia-induced inflammation and oxidative stress in high cholesterol diet (HCD)-fed mice in comparison with the effects of shark cartilage-derived chondroitin sulfate (CS). MATERIALS/METHODS: Low-density lipoprotein receptor knockout (LDLR-KO) mice were fed HCD with an oral administration of CS (50 and 100 mg/kg BW/day), SCS (100 and 200 mg/kg BW/day), or water, respectively, for ten weeks. RESULTS: The administration of CS or SCS reduced the levels of serum triglyceride (TG), total cholesterol (TC), and LDL cholesterol and elevated the levels of high-density lipoprotein cholesterol, compared with those of the control group (P < 0.05). Furthermore, CS or SCS significantly attenuated inflammation by reducing the serum levels of interleukin (IL)-1β and hepatic protein expression levels of nuclear factor kappa B, inducible nitric oxide synthase, cyclooxygenase-2, and IL-1beta (P < 0.05). In particular, the serum level of tumor necrosis factor-alpha was reduced only in the 100 mg/kg BW/day of SCS-fed group, whereas the IL-6 level was reduced in the 100 and 200 mg/kg BW/day of SCS-fed groups (P < 0.05). In addition, lipid peroxidation and nitric oxide production were attenuated in the livers of the CS and SCS groups mediated by the upregulation of hepatic proteins of antioxidant enzymes, such as superoxide dismutase, catalase, and glutathione peroxidase (P < 0.05). CONCLUSIONS: These results suggest that the biological effects of SCS, similar to those of CS, are attributed to improved lipid profiles as well as suppressed inflammation and oxidative stress induced by the intake of HCD.

Research Trends for Nanotoxicity Using Soil Nematode Caenorhabditis elegans (토양선충 Caenorhabditis elegans를 이용한 나노독성 연구동향)

  • Caenorhabditis elegans, a free-living nematode mainly found in the soil pore water, roles the critical function in trophic levels, energy flow, and decomposition in soil ecosystem. C. elegans is commonly used species to test soil toxicity. Recently, they are employed broadly as a test organism in nanotoxicology. In this study, a review of the toxicity of nanomaterials for C. elegans was presented based on SCI (E) papers. The nanotoxicity studies using C. elegans have been reported in 20 instances including the mechanism of toxicity. Most studies used K-medium, S-medium, and NGM (Nematode Growth Medium) plate as an exposure medium to test toxicity of nanoparticles. The effects observed include anti aging, phototoxicity, genotoxicity, and dermal effects on C. elegans exposed to nanoparticles. We found that the toxic mechanisms were related with various aspects such as lifespan abnormality, oxidative stress, distribution of particles on inter-organisms, and stress-related gene analysis. C. elegans has advantage to test toxicity of nanoparticles due to various cellular activities, full genome information, and easy observation of transparent body. C. elegans was considered to be a good test species to evaluate the nanotoxicity.

Electrospray-assisted Preparation of Polymer Microparticles Containing Water-insoluble Bioactive Compounds (전기방사법을 이용한 유용성 생리활성성분을 포함한 고분자 마이크로입자 제조)

  • An anti-aging compound ethyl (4-(2,3-dihydro-1H-indene-5-carboxyamido) benzoate) rapidly crystallizes in emulsion systems, and a flavonoid 3,5,7-trihydroxy-4'-methoxy-8-prenylflavone bearing a whitening function causes coloration of cosmetic compounds when mixed with metal oxides. In this study, an electrospray method was used to encapsulate water-insoluble bioactive compounds in polymeric microparticles. Poly (methyl methacrylate) and polycaprolactone were used to encapsulate ethyl (4-(2,3-dihydro-1H-indene-5-carboxyamido) benzoate) and 3,5,7-trihydroxy-4'-methoxy-8-prenylflavone, respectively. It was found that polymer concentration, the structure of electrospray nozzle, and compatibility between polymers and bioactive compounds were important factors in the preparation of the particles. Polycaprolactone particles encapsulating 3,5,7-trihydroxy-4'-methoxy-8-prenylflavone was effective in preventing coloration of a cosmetic compound when mixed with metal oxides.

Sprouted Black Rice Oligopeptide Induces Expression of Hyaluronan Synthase in HaCaT Keratinocytes and Improves Skin Elasticity (발아 검은쌀 올리고펩타이드의 각질형성세포에서 Hyaluronan Synthase 발현과 피부 탄력 개선 효과)

  • In this study, the effect of oligopeptide ($Oligosproutin^{(R)}$) from sprouted black rice was evaluated for possible improvement in skin elasticity. We examined the changes in gene expression on oligopeptide-treated HaCaT cells using DNA microarray analysis. As a result, oligopeptide treatment showed a differential expression ratio of more than 2-fold : 745 genes were activated and 1011 genes were repressed. One of the most interesting findings is a 2-fold increase in hyaluronan synthase 2 (HAS 2) gene expression by oligopeptide. We also found that oligopeptide increased cell proliferation, HAS2 mRNA expression and intracellular ROS scavenging activity in HaCaT cells. A human clinical study which oil-in-water emulsion with oligopeptide was topically applied showed significant increase in skin elasticity. These results suggest that the sprouted black rice oligopeptide ($Oligosproutin^{(R)}$) can be effective anti-aging ingredient for cosmetics.

Study on the Antioxidant Activity of Geranium nepalense subsp. thunbergii Extract (이질풀 추출물의 항산화 효능에 관한 연구)

  • Antioxidant activity of Geranium nepalense subsp. thunbergii extract was evaluated by DPPH free radical scavenging assay. Geranium nepalense subsp. Thunbergii extract contains tannin, (-)epicatechin, kaempferitin, kaepferol -7-rhamnoside, brevifolin, corilagin, pyrogallol, ellagitannin, geraniin, gallic acid, succinic acid, quercetin, protocatechuic acid, etc. Geranium nepalense subsp. Thunbergii showed excellent antioxidant activity compared to positive control, quercetin. Geranium nepalense subsp. thunbergii extract showed a 98.33 % inhibition of DPPH radical at a concentration of $50\;{\mu}g/mL$. Quercetin showed a 78.05 % inhibition of DPPH radical at the same concentration. To investigate reactive oxygen species (ROS) scavenging activity, Geranium nepalense subsp. thunbergii extract was treated to human keratinocytes (HaCaT). $IC_{50}$ value of Geranium nepalense subsp. thunbergii extract was $43.22\;{\mu}g/mL$ and $IC_{50}$ value of quercetin was $102.35\;{\mu}g/mL$. Geranium nepalense subsp. thunbergii extract showed excellent antioxidant activity. Skin irritation test and cytotoxicity test suggested that Geranium nepalense subsp. thunbergii extract is a safe antioxidant ingredient for cosmetics.
