TOXICOLOGICAL STUDY ON TRADITIONAL KOREAN HERBAL DRUGS (V)

  • Water extracts of 21 traditional Korean herbal drugs were prepared, and a dose range of 100 mg/kg to 400 mg/kg was administered orally into mice once a day for five days. Changes of serum enzyme activities of glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), blood urea nitrogen (BUN), alkaline phosphatase, body weight changes and histo pathological examination of various organs were investigated. Water extract of Ephedra Herba caused severe body weight loss at a dose of 100 mg/kg and death from a dose level of 200 mg/kg by oral administration. Angelica koreanae Radix and Anthrisci Radix showed a slight body weight loss and damages to liver and kidney.

Exmination of Anti-Obesity Effect of Regional Special Natural Products of Anthrisci radix, Psoraleae semen, Siegesbeckiae herba and Corni fructus (지역 특산 천연산물 전호, 파고지, 희첨 및 산수유의 항비만효과 규명)

  • 4 kinds of Regional Special Natural Products (RSNPs), such as Anthrisci radix, Psoraleae semen, Siegesbeckiae herba and Corni fructus were examined to verify for anti-obesity effect. $PPAR\gamma$ (peroxisome proliferator-activated receptor $\gamma$) from 3T3-L1 cell concerning adipocyte differentiation was suppressed by different concentraton of 4 RSNPs with western blot, when treated RSNPs' extract and MDI (IBMX, Dexamethasone, Insulin) at the same time. Also, SREBP-1 (Sterol regulatory element binding protein) controlling lipogenesis and $PPAR\gamma$ expression levels were reduced by these 4 RSNPs' extract, when these chemicals after differentiation of 3T3-L1 cell. And lipid droplets were reduced by 7.5%, 14.4%, 18.3% and 30% at different concentration of Anthrisci radix from Oil Red O staining. Also, it was reduced by 2%, 4.9%, 9.3% and 38% at different concentration of Psoraleae semen. For Siegesbeckiae herba, it was inhibited by 1.4%, 6.4%, 16.4% and 30.1%, respectively. And Corni fructus was also showed by 0.9%, 6.3%, 13.7% and 33% at same concentration of Siegesbeckiae herba. These 4 kinds of RSNPs were expected for a useful material for anti-obesity materials.

Anti-Cell Adhesion Effect of Animal Cell with Regional Special Natural Products of Anthrisci radix, Psoraleae semen, Siegesbeckiae herba and Corni fructus (지역 특산 천연산물 전호, 파고지, 희첨 및 산수유의 동물세포간의 부착 억제효과)

  • For investigation of anti-angiogenesis mechanism of Anthrisci radix, Psoraleae semen, Siegesbeckiae herba and Corni fructus, anti-cell adhesion experiment was performed. The adhesion of U937 cells to IL-$1\beta$-stimulated HUVECs was completely suppressed by 276% at 0.2 mg/L of Anthrisci radix, 220% at 5 mg/L of Psoraleae semen, 158% at 10 mg/L of Siegesbeckiae herba and 132% at 20 mg/L of Corni fructus, respectively. And the adhesion of PMA-chemical stimulated U937 cells to HUVECs, it was inhibited 139% at 0.2 mg/L of Anthrisci radix, 442% at 5 mg/L of Psoraleae semen, 720% at 10 mg/L of Siegesbeckiae herba and 664% at 20 mg/L of Corni fructus. Also, the adhesion of chemical stimulated U937 cells to IL-$1\beta$/chemical stimullated HUVECs, it was inhibited by 286% at 0.2 mg/L of Anthrisci radix, 146% at 5 mg/L of Psoraleae semen, 436% at 10 mg/L of Siegesbeckiae herba and 297% at 20 mg/L of Corni fructus, respectively. It would be a useful substance for anti-cell adhesion based on anti-angiogenesis for anti-obesity and anti-cancer.

  • Sporadic diarrhea occurring predominantly in infants and young children is a significant illness of worldwide importance. Rotaviruses are the etiologic agents for 47% of the cases of infantile diarrhea in Seoul, Korea. This research was undertaken to investigate the inhibitory effect of traditional herbal medicines on rotatvirus infection. Among tested 50 kinds of herbal medicines, Coptidis Rhizoma was best on inhibitory activity of rotavirus infection, followed by Astragali Radix and Anthrisci Radix. The active component of Coptidis Rhizoma was berberine.

Identification keys of Jeonho(Qianhu) and Asam(Eshen) (백화전호(白花前胡)와 자화전호(紫花前胡), 아삼(峨參)의 감별기준(鑑別基準))

  • Objectives : Peucedani Radix ('Jeonho' in Korean) is the root of Peucedanum praeruptorum or Angelica decursiva. However, the root of Anthriscus sylvestris has usually been distributed as Jeonho. This study was performed to determine the discriminative criteria of Jeonho, focused on distribution in Korean markets. Methods : We have determined identification keys to discriminate each Jeonho samples, through observation of external morphology of original plants, and the research of external and internal morphological features of dried Jeonho herbs. Results : Because of identical to the Korean plant name 'Jeonho', Anthrisci Radix had been came into use as a substitute of Jeonho in Korea during the time of Japanese colonial rule. The original plants of Jeonho and its substitute are discriminated with shape of leaf, leaf margin and color of stem. External morphological features of the medicinal herbs of Jeonho are different in the color of cross-sections, pellucid dot, white powder. Internal morphological points, such as fiber bundle of xylem, seconadary mudullary ray and ray of xylem were also used as discriminative criteria for Jeonho. Further details(e.g. identificaion keys) are in the article. Conclusions : We think that these discriminative criteria will be meaningful in identifying the substitutes and adulterants of Jeonho.
