Anti-Inflammatory Herbal Extracts and Their Drug Discovery Perspective in Atopic Dermatitis

  • Atopic dermatitis (AD) is an allergic disorder characterized by skin inflammation. It is well known that the activation of various inflammatory cells and the generation of inflammatory molecules are closely linked to the development of AD. There is accumulating evidence demonstrating the beneficial effects of herbal extracts (HEs) on the regulation of inflammatory response in both in vitro and in vivo studies of AD. This review summarizes the anti-atopic effects of HEs and its associated underlying mechanisms, with a brief introduction of in vitro and in vivo experiment models of AD based on previous and recent studies. Thus, this review confirms the utility of HEs for AD therapy.

Effect of Juglandis Semen Aqua-acupuncture and Acupuncture on the Allergic Response (호도약침이 알레르기에 미치는 영향)

  • Experimental studies were done to research the dinical effects of Juglandis Semen aqua-acupuncture and acupuncture(BL 13) on the anti-allergic response. anaphylaxis provoked by the compound 48/80, delayed type hypersensitivity response to picryl chloride and SASC and inflammation response to egg albumin. The following results have been obtained: Juglandis Semen aqua-acupuncture and acupuncture(BL13) group were increased the survival rate in compound48/80 induced systemic anaphylactic reaction. Picryl chloride induced contact dermatitis and delayed type hypersensitivity in SRBC challenged mouse were significantly decreased in Juglandis Semen aqua-acupuncture and acupuncture (BL13) group. Inflammation response - WBC, CRP and Nitric Oxide in egg albumin induced allergic rat were significantly decreased in Juglandis Semen aqua-acupuncture and acupuncture(BL13) group. According to the above results, Semen aqua-acupuncture and acupuncture(BL13) both depress the allergy reaction.

Phototherapy in Allergic Rhinitis: From In Vitro Studies to Clinical Trials

  • Phototherapy has a profound immunosuppressive effect and is widely used for the treatment of inflammatory skin diseases. Since allergic rhinitis (AR) shares several common pathologic features with atopic dermatitis, intranasal phototherapy is a novel approach for treating AR. Phototherapy reduces the antigen presenting capacity of dendritic cells, induces apoptosis of immune cells, and inhibits synthesis and release of pro-inflammatory mediators from inflammatory cells. The effectiveness of phototherapy for AR has been extensively researched: from in vitro studies to several clinical trials. Data indicates the potential of intranasal phototherapy as an alternative treatment for AR as well as other inflammatory mucosal diseases. In this review, we introduce the in vitro, in vivo studies, and clinical trials which demonstrated the effect of phototherapy on AR.

Two Cases of Allergic Contact Dermatitis to $Betadine^{(R)}$ ($Betadine^{(R)}$에 의한 알레르기성 접촉피부염 2례)

  • Antiseptics are substances that kill or prevent the growth of microorganisms when applied to living tissue. They must be effective against microoranisms but must also retain their activity in presence of body fluids without being harmful locally or systemically. Among many antiseptics. $Betadine^{(R)}$ has been widely used because of its low toxicity and high germicidal efficacy. We reported 2 cases of allergic contact dermatitis to $Betadine^{(R)}$ in surgical patients. They had eczematous eruption along the $Betadine^{(R)}$ applying sites. Patch tests with $Betadine^{(R)}$ confirmed the diagnosis.

  • Objectives: Epichlorohydrin is a material that has been predicted to have high volatility and strong toxicity and is used normally in working area. Therefore, the hazardous and dangerous level and the foreign management system about epichlorohydrin should be invested at home and abroad and through hazard assessment for occupational environment measurement and exposure status of industrial workers in domestic working area. Methods: To assess risk and to suggest Development and Adoption to prevent health damage of workers owing to the epichlorohydrin exposure, the hazardousness and dangerousness of epichlorohydrin and its practical examples and regulation level for domestic and abroad health impairment are researched on the base of various references. Results: The epichlorohydrin caused skin and mucus membrane irritation, respiratory paralysis, kidney and live damage under the influence of acute toxicity and in animal study, it was confirmed as a doubtful carcinogenic substance to trigger reducement of sperm number and reproduction ability, abnormal spermatogenesis, mutagen, increase of forestomach epithelium and occurrence of papilloma and so on, as well as it induced stimulus asthma and allergic contact dermatitis for exposure workers. Conclusions: Epichlorohydrin was found to occur allergic contact dermatitis, carcinogenesis doubt and reproduction toxicity and was verified as a material which would be established reinforcement of management level to care health of handlers, such as denotement dangerousness of skin absorption.

Geumeunyoungji-tang Inhibits Serum IgE Level in Mouse Dermatitis Induced by 2,4-dinitrofluorobenzene

  • Lonicerae Flos containing formulation, Geumeumyoungji-tang (GYT), is an herbal prescription prepared using 5 different herbal drugs, namely, Lonicerae Flos, Ganoderma, Lactucae Herba, Xanthii Fructus, and Smilacis Rhizoma. This study was focused on the investigation of the pharmacological effects of GYT on allergic reactions. As the first step of the study, GYT was administered BALB/c mice which were sensitized by 2,4-dinitrofluorobenzene (DNFB). As the result GYT ameliorated dermatitis provoked by DNFB. The serum IgE level of the DNFB sensitized-mouse was significantly decreased when GYT was administered. In order to confirm the moderating effect of this prescription on allergic reaction, GYT was pretreated to human mast cells (HMC-1) before they were stimulated by phorbol-12-myristate 13-acetate plus calcium ionophore A23187 (PMACI). GYT suppressed secretion of inflammatory cytokines, interleukin (IL)-6 and IL-8, from HMC-1. Additionally, pretreatment of GYT showed regulating effect on COX-2 expression. Collectively, these findings provide insights into the pharmacological actions of GYT as a potential agent for treatment of allergic dermatitis.

The Clinical Importance of MAST CLA Test of Allergic Diseases (알레르기 질환 환자의 MAST CLA 검사의 임상적 의의)

  • Background and Objectives : The diagnosis of allergic diseases in current oriental medicine is in the state of depending mainly on two factors: some distinctive symptoms and scientific 'opinions', MAST CLA test is also believed to be helpful in Oriental Medicine on making definite diagnosis of allergic diseases and identifying causative antigen. This study will address the clinical importance of MAST CLA test, the residential type of allergic patients and its distribution status, etc. Methods : From March of 2000 through September of 2003, tests were made in Oriental Medicine Hospital of Semyung University on sex, age, types of residence, allergic diseases and MAST CLA system for the patients who showed allergic symptoms and had been diagnosed so in other hospitals. Results : 1. Sex: Among 91 subjects, 38 of them were men, 53 of them were women. 2. Age group : 15.3% of them were in their 40s; 31.8% in 30s; 15.3% in 20s; 17.5% in 10s. 3. Residental type : A.P.T(51 of them, 56%), plain type of house(33 of them, 36.2%) and villa(7 of them, 7.6%). 4. Discases : Among the diseases, allergic dermatitis was most common(69 of them. 75.8%); allergic rhinitis(17 of them, 18.6%); and chronic and acute urticaria(4 of them, 4.3%) in order. 5. Among 91 subjects, 39 of them showed positive reaction to more than one type of antigens in MAST CLA test: 23 of them were men(60.5%); 16 of them. women(30.1%). 6. Among 91 subjects. 53 of them(58.2%) showed positive reaction to antibodies in MAST CLA test: 22 of them were men; 31 of them, women. 7. Antigen distribution order among 39 subjects who showed positive reaction in MAST CLA test: Mite-farinae, Mite-pterony, Housedust in men and women subjects alike.

The Effects of BGG on Various Immunological Factors Related to Pathogenesis of Allergic Dermatitis in NC/Nga Mice Induced by Der-f (보음거풍지양탕(補陰祛風止痒湯)이 아토피 피부염을 유발(誘發)한 NC/Nga 생쥐의 면역(免疫) 조절작용(調節作用)에 미치는 영향(影響))

  • To evaluate the therapeutic effects of BGG on atopic dermatitis, we investigated the composition of immune cells of lymph node, PBMC and skin of Dermatophagoides farinae-induced NC/Nga mice. The levels of immunoglobulins in serum were analyzed at the protein level and the amount of pathologic cytokines were investigated using CD3/CD28 stimulated splenocytes. The results are summarized below; 1. BGG showed no cytotoxic effect up to $200\;{\mu}g/m{\ell}$ on mLFC in vitro. 2. BGG showed no hepatotoxicity in vivo based on the levels of ALT and AST. 3. Atopic dermatitis was improved through naked eye examination. BGG reduced the skin clinical index from 2.9 to 1.3 (p<0.01). 4. H&E and toluidine blue staining of tissue biopsies revealed that BGG inhibited the infiltration of lymphocytes and mast cells to skin. 5. BGG reduced the number of CD19 positive B cells in PBMCs by 16% (p<0.01), whereas cells were increased by 26% (p<0.05) in lymph nodes. 6. BGG reduced the numbers of B220+/CD23+ cells by 15% (p<0.01) and 33% in PBMCs and lymph node, respectively. 7. BGG reduced the numbers of B220+/IgE+ cells in PBMCs and lymph node by 21% and 33% (p<0.01), respectively. 8. BGG suppressed the levels of IgE (13%, p<0.001) as well as IgM (34%, p<0.001), IgG2a (40%, p<0.001) and IgG2b (26%, p<0.05). 9. BGG reduced the levels of IL-4 and IFN-$\gamma$ by 7% (p<0.05) and 13% (p<0.001) in anti-CD3 and anti-CD28-activated splenocytes, respectively. 10. BGG considerably inhibited the production of TNF-$\alpha$ and IL-6 by 42% (p<0.01) and 15% in the serum, respectively. Based on the results above, we concluded that BGG has therapeutic effects on atopic dermatitis by regulating the differentiation of B cells and isotype switching of IgE. Further investigations on the molecular mechanisms of BGG on atopic dermatitis are anticipated.

A Review on Effects of Probiotics in Atopic Dermatitis (아토피 피부염에서 프로바이오틱스 치료 유효성에 관한 문헌적 고찰)

  • Objective : The purpose of this study is to investigate the use of probiotics for cure of atopic dermatitis and suggest for the better method of studies in order to seek more effective treatment. Methods : We searched from Korean Traditional Knowledge Portal(KTKP), Reserach Information Sharing Service(RISS), National Discovery for Science Leader(NDSL) and PubMed. We selected among the papers published from 2005 to 2019 using terms related to 'Atopic dermatitic & Allergy diseases & Probiotics'. Results : 41 papers were selected from the database worldwidely. In these collected 41 related theses, there were 14 experimental studies(34.1%), 16 clinical studies( 39.0%), 11 review studies (26.9%). 23 papers(76.7%) insisted that the effects is but 9 papers(30%) said not effective. Conclusion : The effects of probiotics in atopic dermatitis is investigated positively, however, we have to consider the methods to approach the result including the sort, amount of probiotics, how to culture, how to eat and so on.
