Inhibitory Effects of Ginsenoside Rb1 on Atopic Dermatitis-Like Skin Lesions in Mice

  • Allergies are immediate hypersensitive responses to antigens and interleukin (IL)-4 is involved in the initiation and development of allergic responses. $Rb_1$ has been known to have a variety of biological activities including anti-inflammatory activity, but the effect of $Rb_1$ on allergic responses is not known yet. The present study was undertaken to examine whether $Rb_1$ has an inhibitory effect on allergic response in mouse model. In allergic mouse model, our results showed that topical application of $Rb_1$ on atopic dermatitis (AD)-like skin lesions improved skin condition and inhibited starching behaviors. In addition, $Rb_1$ application not only suppressed mRNA expression of IL-4 and IL-10, but also prevented the nuclear factor of activated T cells 1 transcription. Moreover, $Rb_1$ application suppressed IL-4's secretion. Taken together, these results suggest that $Rb_1$ has a potent inhibitory effect in AD-related T cell cytokine production and may be a candidate for therapeutic agent in allergy.

The Use of Complementary and Alternative Medicine in Parents of Children and Adolescents with Allergic Diseases: Community-based Survey (지역사회 기반 소아 청소년 알레르기 환자의 보완대체요법 이용 실태조사)

  • Objectives The purpose of this study is to see the prevalence and the patterns of the use of complementary and alternative medicine (CAM) in children and adolescent patients with allergies. Methods We analyzed data on 547 children and adolescents (age from 0-18 years old) chosen from all regions throughout the country with allergic diseases, including atopic dermatitis, allergic rhinitis, asthma and allergic urticaria. We used multiple logistic regression modeling to predict CAM use based on predictor variables. Results The previous 12-months prevalence of CAM usage in overall was 70.7% (atopic dermatitis, 78.1%; allergic rhinitis, 52.9%; asthma, 70.3%; allergic urticaria, 86.3%). Central and southern regions displayed significantly lower rate of using CAM compare to the northern region, and CAM was less likely to be used for the allergic rhinitis patients than the atopic dermatitis patients. The most commonly used CAM type was natural products (62.2%). Top five of the most frequently used CAM modalities were softener water, vitamin, red ginseng, wood bathing and aloe oil. One of the main reasons for trying CAM was from the 'hope for a more effective outcomes in additional to the conventional medicine' (43.9%). The subjective effectiveness of CAM was found to be excellent in 74.0% of the patients, and 70.3% of the parents were willing to recommend CAM therapies to the others. Conclusions CAM is used widely to treat allergic diseases in children and adolescents in Korea. Korean medical doctors should actively discuss the use of CAM with the patients and provide information on the effectiveness and safety of CAM as guide in making choice for usage of CAM.

  • Atopic dermatitis is estimated to affect 15-20% of the childhood population and there id considerable evidence that the prevalence is increasing. But it is frequently under diagnosed and inappropriately treated yet. Atopic dermatitis can have a large social;. emotional and financial effect on the child and their family. Atopic dermatitis also commonly predated the development of asthma and allergic rhinitis. Therefore early diagnosis and proper treatment are the key for control the atopic dermatitis itself and modify the future repiratory allergies. This review will cover the new diagnostic criteria and treatment briefly.

  • To identify the allergy problem of a 36-year old swimming instructor, who experiences heavy itching and rashes whenever she comes in contact with pool water. Patch tests were performed with European standard series and materials from the work floor. A positive patch test to aluminum chloride and flocculant was observed. Occupational dermatitis is, based on a contact allergy to aluminum chloride in the flocculant.

Clinical Differences between Baekho-tang and Baekhogainsam-tang through Skin Disease Case (피부 질환 증례 연구를 통한 백호탕(白虎湯)과 백호가인삼탕(白虎加人參湯)의 비교)

  • Objective : The purpose of this paper is to analyze the difference between Baekho-tang(白虎湯) and Baekhogainsam-tang (白虎加人蔘湯). Methods : We compared the effect of Baekho-tang on atopic dermatitis and the effect of Baekhogainsam-tang on allergic contact dermatitis. We focused on the level of thirst. Results : After the Baekho-tang treatment, the SCORAD of atopic dermatitis patient decreased $63.8{\rightarrow}4.3$ and VAS of pruritus on atopic dermatitis patient were improved $8{\rightarrow}2$ respectively. Meanwhile, after the Baekhogainsam-tang treatment, the VAS of pruritus on allergic contact dermatitis patient was improved $7{\rightarrow}2$ and the thirst of the patient improved. Conclusions : There is a difference between Baekho-tang and Baekhogainsam-tang at the level of thirst. Baekhogainsam-tang continues to thirst hard enough to find water.

Suppressing Effects of Ethanol Extract from Dioscoreae Rhizoma on atope Dermatitis (부채마 에탄올 추출물의 아토피 피부염 억제 효과)

  • This study was conducted to investigate the effect of 70% ethanol extract (DR) on the mast cell-mediated allergic contact dermatitis induced by dinitrofluorobenzene in BALB / c mice, which affects the cell activity by antigen in RBL-2H3 mast cells Respectively. The ethanol extracts of RBL-2H3 cells activated by DNP-HSA and anti-DNP IgE antibodies inhibited the secretion of ${\beta}$-hexosaminidase, histamine, and IL-4 and $TNF-{\alpha}$ Production was suppressed. In the DNFB-induced contact allergic dermatitis animal model, treatment with ethanol extract reduced ear swelling and inhibited serum histamine and IL-4 secretion, and DR treatment effectively prevented mast cell infiltration in dermatitis-induced areas. As a result, the ethanol extract may be used as a therapeutic agent for mast cell-mediated allergic diseases such as atopic dermatitis.

  • Objective : This study was performed to investigate the effects of Sophorae Radix and Coptidis Rhizoma-iontophoresis in allergic contact dermatitis. Methods : Contact hypersensitivity assay, video microscope, melanin-erythema levels, pH levels, hydration levels, WBC count, RBC count, neutrophil ratio, lymphocyte ratio and total IgE levels were measured. Results : At contact hypersensitivity assay, the right ear swellings in Sophorae Radix and Coptidis Rhizoma-iontophoresis group were decreased compared with control group, but have no statistical significance. At observation of skin morphologic change, many papules were seen in control group and keratins were seen in Sophorae Radix-iontophoresis group. At melanin levels, Sophorae Radix and Coptidis Rhizoma-iontophoresis groups showed no significant difference compared with control group. At pH levels, Sophorae Radix-iontophoresis group showed significant decreased with control group. At hydration levels, Sophorae Radix and Coptidis Rhizoma-iontophoreses groups showed to significant difference compared with control group. WBC count, RBC count and neutrophil ratio were significantly increased in Sophorae Radix and Coptidis Rhizoma-iontophoresis group compared with control group. Lymphocyte ratio and total IgE levels were significantly decreased in Sophorae Radix and Coptidis Rhizoma-iontophoresis groups compared with control group. At morphology of skin, inflammation was decreased and the thickness of epidermis was well preserved in Sophorae Radix and Coptidis Rhizoma-iontophoresis group. Conclusion : Sophorae Radix and Coptidis Rhizoma-iontophoresis had some anti-allergy, anti-inflammatory effects on allergic contact dermatitis, but had no effects on melanin-erythema levels, pH levels and hydration levels of skin.

The Anti-Inflammatory Effects of WooBangHaeGiTang on the Allergic Contact dermatitis (우방해기탕(牛蒡解肌湯)의 알러지성 접촉피부염(接觸皮膚炎)에 대한 소염효과(消炎效果))

  • Objective : This study was performed to investigate the Anti-Inflammatory Effects of WooBangHaeGiTang on the Allergic Contact dermatitis. Method : Contact hypersensitivity assay, video microscope, WBC count, RBU count, neutrophil ratio, lymphpcyte ratio and GOT & GPT were measurd. Result : 1. At Contact hypersensitivity Aasay, at 24 hours after Control group & Sample group showed appreciably more ear swelling than Normal group, at 48 hours after Control group showed more ear swelling than Normal group & Sample group, at 72 hours after Control group & Sample group showed more ear swelling than Normal group 2. Regarding the number of WBC, at 24 or 48 hours after There are no significant difference among Normal group, Control group. Sample group, at 72 hours after Sample group showed more WBC than Control group. 3. Regarding the number of RBC, at 24 hours after Control group showed more RBC than Normal group, at 48 hours after Control group & Sample group showed more RBC than Normal group. at 72 hours after Control group showed more RBC than Normal group. 4. Regarding the percentage of neutrophil in WBC, at 24 & 48 hours after Control group & Sample group showed higher percentage than Normal group. 5. Regarding the percentage of Lymphocyte in WBC, at 24 hours after Sample group showed lower percentage than Normal group, at 48 hours after Sample group & Control group showed lower percentage than Normal group. 6. Regarding the GOT & GPT in blood, the GOT & GPT are in Normal scale, so there are no Hepatotoxicity in Normal group, Control group, Sample group. Conclusions : According to above experiments, WooBangHaeGiTang have some anti allergy effects on allergic contact dermatitis.

Effect of Picrorrhiza Rhizoma on Dinitrofluorobenzene-induced Contact Dermatitis (Type I allergy)

  • The effect of Picrorrhiza Rhizoma (PR) aqueous extracts were evaluated on 2,4-dinitrofluorobenzene (DNFB)-induced contact dermatitis, type I allergic model. Contact dermatitis was induced by sensitization with dinitrophenyl-derivatized ovalbumin (DNP-OVA) and DNFB challenge as antigen. Three different concentrations of PR extracts (300,150 and 75mg/kg) were orally administered to DNP-OVA sensitization mice once a day for 7 days with reference materials; dexamethasone (15mg/kg, intraperitoneal treatment). End of 7 days oral administration of PR extracts or intraperitoneal treatment of dexamethasone, the changes on the edematous changes and scratching behavior were measured. Immediate after DNFB challenge on ear or paw of DNP-OVA sensitized mice, increases of ear and paw thicknesses and weights were detected with anterior ear skin (dermis to epidermis) thickness and paw scratching behavior increases. However, these DNFB-induced increases on ear and paw thicknesses, weights and scratching behaviors were decreased by treatment of all three different dosages of PR extracts and dexamethasone, respectively. In addition, the increases of anterior skin thicknesses were also dramatically inhibited by treatment of all three different dosages of PR extracts and dexamethasone at histopathological observations. The results obtained in this study suggest that oral treatment of PR extracts also has relatively favorable effects on allergic dermatitis.
