Advances in Biodegradable Polymers for Drug Delivery Systems

  • The recent development of biodegradable polymers for drug delivery system (DDS) has been investigated. The biodegradable polymers for DDS are mainly discussed in two categories: one category is natural biodegradable polymers such as polysaccharides, modified celluloses, poly(${\alpha}$-amino acid)s, modified proteins, and microbial biodegradable polymers; the other is synthetic biodegradable polymers such as poly(ester)s, poly(ortho ester)s, poly(phosphazene)s, poly(anhydride)s, poly(alkyl cyanoacrylate)s, and multiblock copolymers. The bioconjugate polymeric drug delivery systems have been also proposed for the design of biocompatible polymeric controlled drug delivery.

A Trial for Utilizing Flounder Skin Gelatin as an Emulsifier through Enzymatic Modification (가자미피 젤라틴의 효소적 수식에 의한 유화제의 시제)

  • In order to effectively utilize the by-products of sea-food, the utilization of enzyme-modified flounder(Limanda aspera) skin gelatin as an emulsifier was investigated. In the experiment, the gelatin was extracted from the flounder skin with the heat-treatment at $60^{\circ}C$ and in pH 5.0 for 3 hrs with four volumes of distilled water and emulsifiers were enzymatically modified L-leucine alkyl esters$(L-leucine-OC_n$ : n= 2, 4, 6, 8 and 10) to the gelatin$(EMFSG-C_2,\;EMFSG-C_4,\;EMFSG-C_6,\;EMFSG-C_8,\;EMFSG-C_{10})$ for improving the functional properties such as emulsifying activity, emulsifying viscosity, whippability, electric conductivity, critical micelle concentration and interface tension, etc. Also, the functional properties of the L-leucine alkyl ester modified gelatins were compared with those of Tween-60 as reference. Molecular weights of the enzymatically modified flounder skin gelatin(EMFSG) were 20.5kDa. in $EMFSG-C_2.\;19.5 kDa.\;in\;EMFSG-C_4\;and\;16.5kDa.\;in\;EMFSG-C_6,\;EMFSG-C_8$ and $EMFSG-C_{10}$. respectively. Emulsifying activity and emulsifying viscosity in the modified gelatins were risen with increase of carbon number of the introduced L-leucine alkyl esters. Among the modified gelatins, $EMFSG-C_6$ exhibited the highest emulsifying stability and foaming stability, whereas $EMFSG-C_8$ showed the highest whippability. The electric conductivities of the all $EMFSG-C_n$ were linearly risen to critical micelle concentration(CMC) , therefore $EMFSG-C_{10}$ exhibited the lowest CMC value and interface tension, and dense particles in the microscopic observation. In conclusion, the best quality in functional properties was assured on $EMFSG-C_{10}$.

  • The effects of chemical structure of alkyl groups of norbornene carboxylic alkyl esters(methyl, octyl, 4-chlorobenzyl) and endo/exo ratios of norbornene monomers on activity of palladium catalyst and polymerization behavior were investigated. Norbornene ester monomers were synthesized from the reaction of 5-norborene-2-carboxylic acid and various alcohols. Polymerization catalyst, di-${\mu}$-chloro-bis(-methoxybicyclo[2,2,1]-hept-2-ene)palladium(II) (DCBMP), was synthesized according to the literature procedure and silver hexafluoroantimonate ($AgSbF_6$) was used as a conjugate anion source. Gel permeation chromatography (GPC), thermogravimetric analysis (TGA), differential scanning calorimetry (DSC) were the principal techniques for polymer characterization and $^1H$ NMR spectroscopy was used for chemical structures determination of monomers and polymers. For all of the norbonene alkyl esters GPC data showed that when the amounts of endo isomers exceeded those of exo isomers decreased molecular weight polymers were obtained probably due to the decreased catalyst activity. Polymerizations were conducted by varying the monomer/catalyst mole ratios (100:1, 200:1, 300:1). When 300:1 monomer/catalyst ratio was employed it was possible to synthesize high molecular weight ($M_n=27500g/mol$), film forming polymer from exo-norbornene carboxylic acid octyl ester.

Comparative Enantiomer Separation on Chiral Stationary Phases Derived from Chiral Crown Ether by HPLC (고성능 액체 크로마토그래피에서 키랄 크라운 에테르로부터 유도된 키랄 고정상을 이용한 광학분리의 비교)

  • Comparative liquid chromatographic enantiomer separation of ${\alpha}$-amino acids, their esters and primary amino compounds was performed using two chiral stationary phases (CSPs) prepared by covalently bonding (+)-(18-crown-6)-2,3,11,12-tetracarboxylic acid (18-C-6-TA) of the same chiral selector. In general, the separation factors and resolution factors for these analytes on CSP 1 were greater than on CSP 2, while these capacity factors on CSP 2 were quite greater than on CSP 1. Except for leucine methyl ester and phenylalanine methyl ester, the elution orders of all analytes including ${\alpha}$-amino ${\alpha}$-alkyl acids and phenylglycine alkyl esters on CSP 1 are identical to those on CSP 2. This study showed that different connecting structures for these two CSPs might influence their ability to resolve the analytes depending on their structures related to the chiral recognition mechanism.

Structure and Properties of Polynorbornene Derivatives: Poly(norbornene dicarboxylic acid dialkyl ester)s and Poly(norbornene dimethyl dicarboxylate)s

  • Poly(norbornene dimethyl dicarboxylate)s, (PNDMD)s, were prepared by addition polymerization with palladium(II) catalyst from pure exo-monomers, and their structure and properties were compared with those of poly(norbornene dicarboxylic acid dialkyl ester)s, (PNDADA)s. Both polymer series exhibited good solubility in general organic solvents and excellent thermal stability up to $330^{\circ}C$. Wide-angle X-ray scattering (WAXS) study indicated the presence of nano-scale layer-like order in amorphous PNDADAs, while PNDMDs showed random amorphous structure. The glass transition temperatures and dielectric constants of solid polymers were found to decrease as the alkyl side-chain length increases for both polymer series. However, PNDMDs showed lower glass transition temperatures and higher dielectric constants, as compared with those of PNDADAs containing the same alkyl substituents. This difference, caused by the higher side-group mobility of PNDMDs, may be closely related to the nano-scale order in amorphous PNDADAs and its absence in PNDMDs.

Preparation of Alkyl and Aryl-thionocarbamate and their Anthelmintic action (Alkyl 급 Aryl thionocarbamate 류의 합성과 그 구충작용에 관한 연구)

  • Alkyl-phenylthionocarbamates already have been investigated as anthelmintics. Especially ethyl-phenylthionocarbamate is noted for its anthelmintic action. Author studied methods for synthesizing Alkyl-phenylthionocarbamates, Alkyl-p-phenetyl thionocarbamates, Alkyl-cyclohexylthionocarbamates, 5 kinds for each. For alkyl radical $-CH_{3}$, $-C_{2}H_{5}$, -n-propyl, -isobuthyl, -n-buthyl, being chosen. Compounds VIII, X, XIII, XIV, and XV are new compounds. At the sametime author prepared 4 kinds of Arylthionocarbamates, among which XVII, and XIX being new compounds. (Table I) The anthelmintic action of these thionocarbamates has been examined through kymographic records indicating the action of neuromuscular preparation of Eisenia foetida Savigny in the various concentration fo above compounds: -Trendelenburg method. Santonine as the criterion of the estimation. (Table. II) 1. Compounds I-X could be prepared by method A or method B. method A in sealed tube brought on better yield and purity. 2. Compounds XI-XV couldn't be prepared by method A but by method B. 3. Arylthionocarbamates XVI, XVII, XVIII, and XIX couldn't be prepared by method A or method B but by method C. 4. Analyzed N. contained in all compounds. Analyzed C.H. and N. contained in unknown compounds, VIII, X, XIII, XIV, XV, XVII, and XIX. 5. Examination of anthelmintic action has been done under the advice of Prof. J.S.Oh medical college, Seoul Nat. University. 6. Compounds VI-X showed little action. 7. Compounds XVI-XIX showed almost no action. 8. Compounds XI-XV showed stronger action than compounds I-V, which was reported already as a usable anthelmintics. 9. Compounds XI, XII, XIII and XIV showed strongest actions, stronger than santonine as recorded on kymographion of the Trendelenburg method. Provided that, the toxity of these compounds found mild enough for human system, these compounds will serve as anthelmintics of greater powers than compounds I-V. 10. In Alkylthionocarbamates, R'NH-radical more influence on the anthelmintic action than ester radical(-OR).

  • Methoxy polyoxyethylene dodecanoates, kinds of nonionic surfactants, could be obtained from addition of ethylene oxide (5, 7, 9, and 12mol) with fatty acid methyl ester utilizing solid catalyst, metal oxide. Because ethylene oxide (EO) couldn't react directly in acid or alkali catalyst with dodecanoic acid methyl ester (DME) that had no active hydrogen, the reaction to add EO was carried out using active solid catalyst. By using IR, HPLC and $^1H$ NMR analysis, structural confirmation of methyl polyoxy ethylene dodecanate showed high yield ranging from 93 to 97%. EO unit mol number of reacted products was 5.2, 7.1, 9.2 and 12.1 mol respectively. Also, EO adduct distrobution of ethoxylated methyl laurate (MPD) had normal distribution curve like polyoxyethylene alkyl ether (AE).

A Study on Processing and Physical Properties of Isoprene Rubber Involving Norbornene Dialkyl Ester (Norbornene Dialkyl Ester가 첨가된 Isoprene Rubber의 가공성 및 물성에 관한 연구)

  • In this study, we applied six different norbornene dialkyl esters as a plasticizer to an isoprene rubber (IR) and evaluated replaceability of DEHP which is an endocrine disruptor. IR test sheets were prepared by blending IR, norbornene dialkyl ester, vulcanizing agent, etc. and processing properties of the IR were evaluated by measuring Toque, scorch time, cure time and mooney viscosity. Mechanical properties of IR test sheet including hardness, tensile strength, 100% modulus and elongation were also measured and the physical properties of norbornene dialkyl ester applied as a plasticizer were compared to those using DEHN. Both the maximum and minimum toque for the norbornene dialkyl ester as a plasticizer were similar to those of using DEHP. In addition, the scorch and cure time of the former were slightly longer than those of the latter. The mooney viscosity for the case of DEHN was slightly lower than that of the latter. DEHN was also superior to DEHP in terms of processing. The hardness and thermal properties of all IR test sheets were measured to be similar to each other. The linear alkyl chain of norbornene compounds also exhibited good tensile characteristics.

  • The synthetic route and physical properties for three-ring derivatives, trans-2,3,2',3'-tetrafluoro-4-alkyl-4'-(4-alkyl-cyclohexyl)-biphenyl and 4-alkyl-cyclohexane carboxylic acid 2,3,2',3',4'-pentafluoro-biphenyl-4-yl ester are presented. They exhibit broad range nematic phases exceeding 30K and low viscosity of nematic liquid crystal mixtures containing these materials. Their viscosity is compared with that of homologues with a lateral isothiocyanate group and show large negative dielectric anisotropies. The threshold voltage $V_{th}$ was low enough to be operated at a driving voltage of 5V. Our novel LC compounds are suitable materials for the improvement of dielectric anisotropy of vertical align liquid crystal displays (VA-LDCs).

The Synthesis of 1,2-Benzothiazine-3-carboxamidylhydantoin Derivatives and their Antiinflammatory and Analgesic Activities

  • A number of 4-hydroxy-2H (or alkyl)-N-(3-aralkyl-2-thio-1-hydantoinyl)-1, 2-benzothiazine3-carboxamide 1, 1-dioxides were synthesized through the reaction of 4-hydroxy-2H (or alkyl)1, 2-benzothiazine-3-carboxylic methyl ester 1, 1-dioxide and 1-amino-2-thio-3-aralkyl-4-imidazolones in xylene. The compounds synthesized were screened for antinflammatory effect on carrageenin-induced edema in rat and for analgesic effect on acetic acid-induced Writhing syndrome in mice. Most compounds were inhibots of carrageenin-induced rat foot edema and some showed significant antinflammatory activity comparable to that of indomethacin and significant analgesic activity comparable to that of indomethacin and aspirin.
