  • Isolation and Growth Characteristics of AIkalophilic Bacillus sp. for Removal of Anthraquinone Dye. Kim, Jeong-Mog. School of Environmental Information, Taekyeung College, Kyungsan, 712-850, Korea -Alkalophilic strain degrading and decolorizing anthraquinone dye, Remazol brilliant blue R was isolated from natural system and named as Bacillus sp. ARB!. The optimal temperature and pH of Bacillus sp. ARBI were 35°C and 9.0, respectively. The pH of culture media during the fermentation were changed from 10 and 10.5 of initial values to 9.3 and 9.4 after 40 hrs, respectively. Decolorization efficiency in aerobic shaking culture of Bacillus sp. ARBI was markedly higher than that in standing culture. At the optimal culture condition, decolorization efficiency by the Bacillus sp. ARBl was 93% after 32 hrs batch culture. In the case of batch culture using real dye processing wastewater, dye decolorization efficiency of Bacillus sp. ARBl was 78% after 40 hrs.

  • An Alkalophilic Bacillus circulans that can produce significant amount of cyclodextrin glucanotransferase (CGTase) was newly isolated from soil. The culture filtrate was successively purified by ($NH_4$)$_2$$SO_4$precipitation, DEAE-Sephadex column chromatography, and Sephadex G-100 column chromatography. The enzymatic properties, including molecular weight, optimal pH and temperature, stability, and kinetic parameters, were determined. The cyclodextrin synthesis reaction catalized by the purified CGTase was also studied. The sweet potato and corn starch were found to be the most suitable substrates with 60% conversion to cyclodextrin. The highest conversion was achieved at the CGTase concentration of 900-1,100 units/g of soluble starch. The purified CGTase could also catalize the transglycosylation on stevioside.

  • A $\beta$-Galactosidase producing strain, Alkalophilic Bacillus sp, YS-309, has been isolated from soil sample. The strain was capable of producing large amount of intracellular $\beta$-galactosidase in the alkaline media rather than in the neutral media. The preferable medium composition has been determined to be as follows: 0.5% lactose, 0.5% yeast extract, 0.5% soybean meal, 0.1% KH$_2$PO$_4$, 0.02% MgSO$_4$7$H_2O$ 0,0.6% Na$_2$CO$_3$ (pH 9.9). The enzyme was produced by lactose or IPTG as in-ducer. But both Enzyme synthesis and cellular growth were decreased when lactose was added at the higher concentrations than 1.5% (v/v).

Optimization of the Performance of Microbial Fuel Cells Containing Alkalophilic Bacillus sp.

  • A systematic study of microbial fuel cells comprised of alkalophilic Bacillus sp. B-31 has been carried out under various operating conditions. A significant amount of electricity was generated when redox mediators were used. Among the phenothiazine-type redox dyes tested, azure A was found to be the most effective both in maintaining a high cell voltage and for the long-term operation. The maximum efficiency was and for the long-term operation. The maximum efficiency was obtained at ca. $50^{\circ}C$ giving an open circuit voltage of 0.7V. A small change in temperature did not significantly affect the cell performance, but a rapid decrease in performance was observed below $20^{\circ}C$ and above $70^{\circ}C$. It was noticeable that fuel cell efficiency and discharge pattern depended strongly on the carbon source used in the initial culture medium. Regardless of the initial carbon sources, only glucose and trehalose were utilized as substrates. Galactose, however, was not substantially utilized except when galactose was used in the initial medium. Glucose, in particular, showed $87\%$ coulombic efficiency, which was the highest value ever reported, when Bacillus sp. was cultured in a maltose-containing medium. This study demonstrates that highly efficient microbial fuel cells can be constructed with alkalophilic microorganisms by fine-tuning the operating conditions and by carefully selecting carbon sources in the initial culture medium.

  • Alkalophilic sp. YC-335 isolated from soil was capable of producing large amount of cyclodextrin glycosyltransferase (CGTase) in culture broth. This enzyme was successively purified 52.9 folds with 17.8 yield by ethanol precipitation, DEAE-Toyopearl column chromatography and Sephadex G-100 column chromatography. The purified enzyme have a molecular weight of approximately 75,000 estimated by SDS polyaerylamide gel electrophoresis. The optimum pH and temperature for the enzyme activity were 6.0 and 5$0^{\circ}C$, respectively. The enzyme stable between pH 6 and 10, and up to 5$0^{\circ}C$. The thermostability of the enzyme was increased up to 6$0^{\circ}C$ by the addition of 15mM CaCl$_2$.

  • An intracellular invertase was purified to homogeneity from the cell extract of an alkalophilic and thermophilic Bacillus sp. TA-11, which was classified as a new species belonging to Bacillus cereus based on chemotaxanomic and phylogenetic analyses. The purified enzyme with a recovery of 26.6% was determined to be a monomeric protein with a molecular weight of 23 kDa by SDS-PAGE and 26 kDa by gel filtration. The maximum enzyme activity was observed at pH 7.0 and $50^{\circ}C$, and the purified enzyme was stable at the pH range of 5.0 to 8.0 and below $60^{\circ}C$. $K_m$ and $V_{max}$ values of the enzyme for sucrose were 370 mM and 3.0 ${\mu}M$ per min, respectively. The enzyme activity was significantly inhibited by bivalent metal ions ($Hg^{2+}$, $Cd^{2+}$ and $Cu^{2+}$) and sugars (glucose and fructose).

  • Regulation of invertase biosynthesis was studied in thermophilic and alkalophilic Bacillus sp. TA-11. Biosynthesis of the invertase was effectively induced in the presence of 10 mM sucrose for 180 min. Glucose repressed the invertase induction by sucrose and as late as addition time of glucose, the invertase formation was increased, indicating that glucose repression was occurred by inducer exclusion. Catabolite repression was reduced a little by the addition of cAMP for 180 min of induction.

  • The study was performed to investigated the excellent microbial anticaries substance which is more effective than the chlorohexidine in the dental caries treatment. For the screening of alkaliphilic microorganism, more than 1200 bacterial strains were isolated from sea soil sample. A typ-ical strain which produced the most excellent antimicrobial substance was selected. The strain was identified novel alkalophilic Bacillus sp. through the results of morphological, biochemical and chemotaxonomical characteristics and 16S rDNA sequencing and designated as Bacillus alkalophilshaggy JY-827.

  • A bacterium producing alkaline cellulase was isolated from soil, leaf mold and compost, and was identified as alkalophilic Bacillus sp. HSH-810 by morphological, cultural and biochemical determination. The optimum cul-ture condition of Bacillus sp. HSH-810 for the growth and alkaline cellulase production was $30^{\circ}C$ and pH 10.0. The maximum alkaline cellulase production was obtained when 1.0%(w/v) CMC, 0.5%(w/v) peptone, 0.02%(w/v) $CaCl_2$ and 0.02(w/v) $CoCl_2$ were used as carbon source, nitrogen source and mineral source, respectively. The optimum pH and temperature of the enzyme activity were pH 10.5 and $50^{\circ}C$, respectively. This enzyme was fairly stable in the pH range of 6.0-13.0 and at $50^{\circ}C$. For the effect of surfactants, the activity of alkaline cellulase was stable in the presence of sodium-$\alpha$-olefin sulfonate (AOS), sodium dodecyl sulfonate (SDS), Tween 20 and Tween 80, but inhibited by the presence of 0.1 linear alkyl-benzene sulfonate (LAS) sig-nificantly.

  • Alkalophilic bacteria isolated from compost were selected, identified and tested for production of alkaline protease. The bacterium was tentatively assigned to Bacillus sp. based on the morphological, cultural and biochemical characteristics. The optimum pH of growth was 10 Galactose and Sodium glutamate enhanced the production of alkaline protease. The protease was most active at pH 11.0 and $60^{\circ}C$ and stable in the range of pH 5-11 and temp. $30^{\circ}-55^{\circ}C$. The protease was stabilized by the presence of calcium salts.
