Formation of Hydrogen Peroxide by the Ozonation of Aqueous Humic Acid (수중 부식산의 오존처리시 생성되는 과산화수소의 농도 변화에 대한 연구)

  • The changes in $UV_{254}$ and concentrations of $H_2O_2$ formed by ozonation of aqueous humic acid in ozone/high pH, peroxone process and in the presence of radical scavenger, $HCO_3{^-}$ were investigated. This study confirmed that the formation of $H_2O_2$ by ozonation may undergo different reaction pathways compared to those of $UV_{254}$ reduction in the degradation of the humic acid. The concentration of $H_2O_2$ produced by ozonation was found to be increased with decreasing pH of the sample solution due to the higher stability of ozone molecules at acidic conditions. On the while, $UV_{254}$ reduction was found to be higher at alkaline conditions or larger amount of $H_2O_2$ additions as a radical promoter in which the producing of ${\cdot}OH$, ${\cdot}HO_2$ radicals can be more favorable. From the results, it has been suggested that the formation of $H_2O_2$ by ozonation depends mainly on the direct reactions of ozone with humic acid molecules, while $UV_{254}$ reduction is affected by both the indirect reactions of the radicals and direct reactions of ozone with humic acid.

  • A Japanese larch has been reforested very much in Korea, but it is not used as a wood resources for paper pulp by now. So this study is carried out to utilize the larchwood for paper pulp manufacture through the Asplund pulping process. The experiment on increasing in the brightness of the pulp is made through the addition of $MgSO_4$, $ZnSO_4$, $Al_2(SO_4)_3$, and KI as a cellulose stabilizer in chip treatment with caustic soda which is followed by high-temperature defibration and conventional peroxide bleaching (5% NaOH plus 2% additive salt per wood in cold pretreatment), or in high-consistency (30%) pulp bleaching of hydrogen peroxide and peracetic acid (100% acitve oxygen per lignin) for conventional one. The results obtained are as follows: 1. The solution of 0.5% additive salts had different pH by the sort of bases that was pH 5.7 in $MgSO_4$, liquor, pH 4.9 for $ZnSO_4$, and pH 2.9 for $Al_2(SO_4)_3$, and in the precepitation of bases which ranged to pH 6-13 for $MgSO_4$, pH 5-12 for $ZnSO_4$, and pH 3-10 for $Al_2(SO_4)_3$. 2. The cellulose stabilizer affective in high-consistency peroxide bleaching was KI, $MgSO_4$, and $ZnSO_4$, but has made a little improvement in de lignification and brightness of pulp in comparison with no addition. 3. The higher alkalinity in the chip treatment has made the higher strength and brightness of larchwood Aspiund pulp instead of downing the pulp yield. And the effective compound for cellulose stabilizer in caustic soda pretreatment of chip was $ZnSO_4$, $Al_2(SO_4)_3$ and KI in order for the conventional peroxide bleaching after Asplund pulping. 4. Therefore, the more effective additives for cellulose stabilization in high-temperature defibration of larchwood suppose to be $ZnSO_4$, $Al_2(SO_4)_3$, and KI, while KI and $MgSO_4$ for peroxide bleaching.

A Method for Determination of Nitrogen in Ruminant Feedstuffs and Products

  • A method for the determination of nitrogen in ruminant feedstuffs, products and excreta (e.g. milk and urine) using a spectrophotometer is developed, where samples processed for P determination are also used to determine N. Samples are digested with sulphuric acid and subsequently with hydrogen peroxide in Kjeldahl tubes. Digested solutions along with phenol and buffered alkaline hypochlorite reagents are incubated in a water bath at $37^{\circ}C$ for 30 min and presented in the spectrophotometer. The spectrophotometer set at 625 nm measures the concentration of N of each sample. Nitrogen in 261 of the samples was also determined by the classical Kjeldahl method in order to develop a relationship between N determined by the Kjeldahl method (Y) and the colorimetric method (X). The mean value of Y was as high as that of X (0.92 vs. 0.96; p>0.05). The colorimetric method predicted Kjeldahl N highly significantly (Y=0.985X-0.024, $R^2=0.993$, p<0.001; or more simply Y=0.974X, $R^2=0.993$, p<0.001). An analysis of regression found no difference (p>0.05; both t-test and F-test) between colorimetric (0.96% N) and adjusted (0.96% N) N. In comparison with the Kjeldahl method, the analytical capacity of N by colorimetric method increases greatly, where 200-300 determinations of N are possible in a working day. In addition, the system provides an opportunity to use not only the same digested solution for both N and P determination of a particular sample, but also uses the same spectrophotometer to assay both N and P. Therefore, the system may be attractive in situations where both elements of a sample are to be determined. In conclusion, the speed of N determination, low cost, efficient use of labour, time and reagents, fewer items of equipment, and the reduction of environmental pollution by reducing effluent and toxic elements are the advantages of this method of N determination.

Phelligridin D maintains the function of periodontal ligament cells through autophagy in glucose-induced oxidative stress

  • Purpose: The objective of this study was to investigate whether phelligridin D could reduce glucose-induced oxidative stress, attenuate the resulting inflammatory response, and restore the function of human periodontal ligament cells (HPDLCs). Methods: Primary HPDLCs were isolated from healthy human teeth and cultured. To investigate the effect of phelligridin D on glucose-induced oxidative stress, HPDLCs were treated with phelligridin D, various concentrations of glucose, and glucose oxidase. Glucose-induced oxidative stress, inflammatory molecules, osteoblast differentiation, and mineralization of the HPDLCs were measured by hydrogen peroxide (H2O2) generation, cellular viability, alkaline phosphatase (ALP) activity, alizarin red staining, and western blot analyses. Results: Glucose-induced oxidative stress led to increased production of H2O2, with negative impacts on cellular viability, ALP activity, and calcium deposition in HPDLCs. Furthermore, HPDLCs under glucose-induced oxidative stress showed induction of inflammatory molecules (intercellular adhesion molecule-1, vascular cell adhesion protein-1, tumor necrosis factor-alpha, interleukin-1-beta) and disturbances of osteogenic differentiation (bone morphogenetic protein-2, and -7, runt-related transcription factor-2), cementogenesis (cementum protein-1), and autophagy-related molecules (autophagy related 5, light chain 3 I/II, beclin-1). Phelligridin D restored all these molecules and maintained the function of HPDLCs even under glucose-induced oxidative stress. Conclusions: This study suggests that phelligridin D reduces the inflammation that results from glucose-induced oxidative stress and restores the function of HPDLCs (e.g., osteoblast differentiation) by upregulating autophagy.

N-acetyl cysteine inhibits H2O2-mediated reduction in the mineralization of MC3T3-E1 cells by down-regulating Nrf2/HO-1 pathway

  • There are controversial findings regarding the roles of nuclear factor (erythroid-derived 2)-like 2 (Nrf2)/heme oxygenase-1 (HO-1) pathway on bone metabolism under oxidative stress. We investigated how Nrf2/HO-1 pathway affects osteoblast differentiation of MC3T3-E1 cells in response to hydrogen peroxide (H2O2), N-acetyl cysteine (NAC), or both. Exposing the cells to H2O2 decreased the alkaline phosphatase activity, calcium accumulation, and expression of osteoblast markers, such as osteocalcin and runt-related transcription factor-2. In contrast, H2O2 treatment increased the expression of Nrf2 and HO-1 in the cells. Treatment with hemin, a chemical HO-1 inducer, mimicked the inhibitory effect of H2O2 on osteoblast differentiation by increasing the HO-1 expression and decreasing the osteogenic marker genes. Pretreatment with NAC restored all changes induced by H2O2 to near normal levels in the cells. Collectively, our findings suggest that H2O2-mediated activation of Nrf2/HO-1 pathway negatively regulates the osteoblast differentiation, which is inhibited by NAC.

Identification and Antimicrobial Activity Detection of Lactic Acid Bacteria Isolated from Corn Stover Silage

  • A total of 59 lactic acid bacteria (LAB) strains were isolated from corn stover silage. According to phenotypic and chemotaxonomic characteristics, 16S ribosomal DNA (rDNA) sequences and recA gene polymerase chain reaction amplification, these LAB isolates were identified as five species: Lactobacillus (L.) plantarum subsp. plantarum, Pediococcus pentosaceus, Enterococcus mundtii, Weissella cibaria and Leuconostoc pseudomesenteroides, respectively. Those strains were also screened for antimicrobial activity using a dual-culture agar plate assay. Based on excluding the effects of organic acids and hydrogen peroxide, two L. plantarum subsp. plantarum strains ZZU 203 and 204, which strongly inhibited Salmonella enterica ATCC $43971^T$, Micrococcus luteus ATCC $4698^T$ and Escherichia coli ATCC $11775^T$ were selected for further research on sensitivity of the antimicrobial substance to heat, pH and protease. Cell-free culture supernatants of the two strains exhibited strong heat stability (60 min at $100^{\circ}C$), but the antimicrobial activity was eliminated after treatment at $121^{\circ}C$ for 15 min. The antimicrobial substance remained active under acidic condition (pH 2.0 to 6.0), but became inactive under neutral and alkaline condition (pH 7.0 to 9.0). In addition, the antimicrobial activities of these two strains decreased remarkably after digestion by protease K. These results preliminarily suggest that the desirable antimicrobial activity of strains ZZU 203 and 204 is the result of the production of a bacteriocin-like substance, and these two strains with antimicrobial activity could be used as silage additives to inhibit proliferation of unwanted microorganism during ensiling and preserve nutrients of silage. The nature of the antimicrobial substances is being investigated in our laboratory.

Antigenotoxic Effects of Satureja hortensis L. on Rat Lymphocytes Exposed to Oxidative Stress

  • The protective properties of Satureja hortensis L. on the rat lymphocytes DNA lesions were tested. Lymphocytes were isolated from blood samples taken from healthy rats. DNA breaks and resistance to $H_{2}O_{2}$-induced damage were measured with the comet assay. Rat lymphocytes were incubated in S. hortensis ethanolic extract (SHE) (0.05, 0.1, 0.5, 1.0, and 2.5 mg/mL), essential oil (SHEO)(0.05, 0.1, 0.5, 1.0, and 2.5 ${mu}L/mL$), $H_{2}O_{2}$ (50, 100, and 200 ${\mu}M$), a combination of $H_{2}O_{2}$ (200 mM) with either SHE (1.0, 2.5 mg/mL) or SHEO (1.0, 2.5 ${\mu}L/mL$) at $4^{\circ}C$ for 30 min, and the extent of DNA migration was measured using a single-cell microgel electrophoresis technique under alkaline conditions. Treatment of rat lymphocytes with SHE or SHEO resulted in significant reduction of $H_{2}O_{2}$-induced DNA damage compared to controls. SHE exhibited a significant (P<0.01) inhibitory effect on oxidative DNA damage at 2.5 mg/mL. SHEO (1.0 and 2.5 ${\mu}L/mL$) also showed significant inhibitory effects (P<0.01) on $H_{2}O_{2}$ induced chromosomal damage. In conclusion both the ethanolic extract and the essential oil of the plant reversed the oxidative damage to rat lymphocytes induced by hydrogen peroxide.

  • The electrocatalytic reduction of dioxygen is investigated by cyclic voltammetry and chronoamperometry at glassy carbon electrode and carbon microelectrode coated with a variety of cobalt phenylporphyrins. The n value obtained at carbon microelectrode is slightly different from that determined at glassy carbon electrode. Dioxygen reduction catalyzed by the monormeric porphyrin Co(II)-TPP mainly occurs through the $2e^-$ reduction pathway resulting in the formation of hydrogen peroxide, electrocatalytic process carries out $4e^-$ reduction pathway of dioxygen to $H_2O$ at the electrodes coated with bis-cobalt phenylporphyrins. The electrocatalytic reduction of dioxygen is irreversible and diffusion controlled.

Protective Effects of Ursolic Acid on Osteoblastic Differentiation via Activation of IER3/Nrf2

  • Background: Oxidative stress is a known to be associated with in the pathogenesis of many inflammatory diseases, including periodontitis. Ursolic acid is a pentacyclic triterpenoid with has antimicrobial, antioxidative, and anticancer properties. However, the role of ursolic acid in the regulating of osteogenesis remains undetermined. This study was aimed to elucidate the crucial osteogenic effects of ursolic acid and its ability to inhibit oxidative stress by targeting the immediate early response 3 (IER3)/nuclear factor erythroid 2-related factor 2 (Nrf2) pathway. Methods: Cell proliferation was determined using water-soluble tetrazolium salt assay, cell differentiation was evaluated by alkaline phosphatase (ALP) activity, and formation of calcium nodules was detected using alizarin red S stain. Generation of reactive oxygen species (ROS) was determined using by DCFH-DA fluorescence dye in hydrogen peroxide ($H_2O_2$)-treated MG-63 cells. Expression levels of IER3, Nrf2, and heme oxygenase-1 (HO-1) were analyzed using western blot analysis. Results: Our results showed that ursolic acid up-regulated the proliferation of osteoblasts without any cytotoxic effects, and promoted ALP activity and mineralization. $H_2O_2$-induced ROS generation was found to be significantly inhibited on treatment with ursolic acid. Furthermore, in $H_2O_2$-treated cells, the expression of the early response genes: IER3, Nrf2, and Nrf2-related phase II enzyme (HO-1) was enhanced in the presence of ursolic acid. Conclusion: The key findings of the present study elucidate the protective effects of ursolic acid against oxidative stress conditions in osteoblasts via the IER3/Nrf2 pathway. Thus, ursolic acid may be developed as a preventative and therapeutic agent for mineral homeostasis and inflammatory diseases caused due to oxidative injury.

Antioxidaitve and Differentiation Effects of Artemisia capillaris T. Extract on Hydrogen Peroxide-induced Oxidative Damage of MC3T3-E1 Osteoblast Cells (사철쑥(Artemisia capillaris T.) 추출물의 항산화 활성 및 H2O2로 산화적 스트레스를 유도한 조골세포의 활성과 분화에 미치는 영향)

  • In this study, the antioxidative activity of Artemisia capillaris T. extract on the proliferation and differentiation of MC3T3-E1 cells under $H_2O_2$-induced oxidative stress was investigated in order to determine its protective effect against oxidative stress as well as its availability as an antioxidant material related to treatment of bone diseases. As a result, the total polyphenol content of A. capillaris extract was 90.10 mg/g, whereas the flavonoid content was 4.45 mg/g. A. capillaris extract increased proliferation of MC3T3-E1 cells under $H_2O_2$-induced oxidative stress, and also increased the proliferation of differentiated osteoblast cells under oxidative stress. In addition, two differentiation markers, alkaline phosphatase activity and mineralization level, in A. capillaris extract tended to increase. These results indicate that A. capillaris extract suppresses the damage to osteoblasts caused by oxidative stress, which demonstrates its availability as an antioxidant material for preventing bone diseases.
