Evolution of E. coli Phytase for Increased Thermostability Guided by Rational Parameters

  • Phytases are enzymes that can hydrolyze phytate and its salts into inositol and phosphoric acid, and have been utilized to increase the availability of nutrients in animal feed and mitigate environmental pollution. However, the enzymes' low thermostability has limited their application during the feed palletization process. In this study, a combination of B-value calculation and protein surface engineering was applied to rationally evolve the heat stability of Escherichia coli phytase. After systematic alignment and mining for homologs of the original phytase from the histidine acid phosphatase family, the two models 1DKL and 1DKQ were chosen and used to identify the B-values and spatial distribution of key amino acid residues. Consequently, thirteen potential amino acid mutation sites were obtained and categorized into six domains to construct mutant libraries. After five rounds of iterative mutation screening, the thermophilic phytase mutant P56214 was finally yielded. Compared with the wild-type, the residual enzyme activity of the mutant increased from 20% to 75% after incubation at $90^{\circ}C$ for 5 min. Compared with traditional methods, the rational engineering approach used in this study reduces the screening workload and provides a reference for future applications of phytases as green catalysts.

  • This review article addresses the role of safety professionals in the diffusion strategies for predictive analytics for safety performance. The article explores the models, definitions, roles, and relationships of safety professionals in knowledge application, access, management, and leadership in safety analytics. The article addresses challenges safety professionals face when integrating safety analytics in organizational settings in four operations areas: application, technology, management, and strategy. A review of existing conventional safety data sources (safety data, internal data, external data, and context data) is briefly summarized as a baseline. For each of these data sources, the article points out how emerging analytic data sources (such as Industry 4.0 and the Internet of Things) broaden and challenge the scope of work and operational roles throughout an organization. In doing so, the article defines four perspectives on the integration of predictive analytics into organizational safety practice: the programmatic perspective, the technological perspective, the sociocultural perspective, and knowledge-organization perspective. The article posits a four-level, organizational knowledge-skills-abilities matrix for analytics integration, indicating key organizational capacities needed for each area. The work shows the benefits of organizational alignment, clear stakeholder categorization, and the ability to predict future safety performance.

The Dynamics of Social Media Marketing: Unraveling the Impact on Customer Purchase Intentions through Engagement and Trust

  • Purpose: The primary aim of this study is to explore how social media marketing influences customer purchase intention, focusing on the roles of customer engagement and trust as mediators. Research Design, Data, and Methodology: The investigation utilized an online survey conducted in Chinese on the Questionnaire Star platform. It targeted male and female consumers aged 18 and above who purchase products online, yielding 1107 valid responses from across major Chinese cities. Results: Analysis reveals that social media marketing significantly affects customer purchase intention in a positive manner. It also enhances customer engagement and trust, which serve as crucial mediating variables linking social media marketing to purchase intention. The study found that engagement and trust facilitate brand identification and alignment, thereby directly boosting purchase intention. Conclusion: The findings offer essential insights for businesses aiming to improve their social media marketing impact on consumer behavior. It highlights the importance of fostering user engagement and trust, as well as creating a favorable brand image. These elements are key to influencing consumer purchase decisions within social media contexts. The study advises businesses to engage deeply with consumers and build trust, providing practical recommendations for navigating the evolving social media environment towards sustainable growth.

Publication Trends in the Pelvic Parameter Related Literature between 1992 and 2022 : A Bibliometric Review

  • Objective : This study aimed to conduct a bibliometric analysis on pelvic parameter related research over the last 30 years, analyzing trends, hotspots, and influential works within this field. Methods : A comprehensive Web of Science database search was performed. The search yielded 3249 results, focusing on articles and reviews published from 1992 to 2022 in English. Data was analyzed using CiteSpace and VOSviewer for keyword, authorship, and citation burst analysis, co-citation analysis, and clustering. Results : The number of publications and citations related to pelvic parameters has increased exponentially over the last 30 years. The USA leads in publication count with 1003 articles. Top publishing journals include the European Spine Journal, Spine, and Journal of Neurosurgery: Spine, with significant contributions by Schwab, Lafage V, and Protoptaltis. The most influential articles were identified using centrality and sigma values, indicating their role as key articles within the field. Research hotspots included spinal deformity, total hip arthroplasty, and sagittal alignment. Conclusion : Interest in pelvic parameter related research has grown significantly over the last three decades, indicating its relevance in modern orthopedics. The most influential works within this field have contributed to our understanding of spinal deformity, pelvic incidence, and their relation to total hip arthroplasty. This study provides a comprehensive overview of the trends and influential research in the field of pelvic parameters.

전문직 간 교육을 위한 학교 간 협동 사례: 중앙대학교 의과대학과 성신여자대학교 간호대학 (Interprofessional Education Collaboration between Chung Ang Medical School and Sungshin Nursing School)

  • Interprofessional collaboration is crucial for patient-centered care and safety. Since healthcare students will be part of interprofessional teams in the future, they need to understand the unique contributions of various healthcare professions to patient care and develop skills in collaboration, communication, leadership, and mutual respect. In response to this need, healthcare faculties have adopted interprofessional education as an innovative teaching method. However, traditional health education has typically taken place within individual schools, resulting in a limited understanding of other professional roles and identities. In our study, we introduced an interprofessional education model involving two different colleges. A total of 152 undergraduate students, comprising 101 medical students from Chung Ang University and 51 nursing students from Sungshin Women's University, participated in the program. A one-day interprofessional education program was conducted to promote collaboration between medical and nursing students. The program included team building and communication games, scenario-based simulations, such as a "room of errors," and tabletop exercises. Key factors for successful interprofessional education include carefully planned scheduling, leadership, and commitment from participating colleges, faculty support and training, the use of diverse teaching methods and technology, and alignment regarding educational directions among the faculty. We believe that this model may provide valuable insights for healthcare institutions aiming to develop and implement interprofessional curricula.

Antioxidant, anti-inflammatory, and cytotoxic properties of fruiting bodies and their mycelia as sources of Cordyceps

  • Mushrooms play crucial roles as reservoirs of naturally occurring bioactive compounds. Among these, Cordyceps militaris is significant because of its well-established reputation for organoleptic excellence and positive health effects, which have led to its widespread commercialization. In contrast, the key properties of Paecilomyces variotii, an ectomycorrhizal symbiont, has received limited attention. In alignment with current research trends, the study of the mycelia and culture media of these mushrooms hold promise in identifying potential sources of valuable bioactive compounds. In the present study, we investigated C. militaris and P. variotii for their phenolic acids and sterols, assessing antioxidant capacity, anti-inflammatory effects, and anti-proliferative activity. Interestingly, P. variotii mycelia exhibited higher concentrations of ergosterol and phenolic compounds, with comparable levels observed in the fruiting bodies, along with superior antioxidant activity compared to that of C. militaris. In contrast, C. militaris mycelia demonstrated anti-inflammatory effects (which were absent in P. variotii mycelia) and cytotoxicity comparable to, and at times exceeding, that of its fruiting bodies (in contrast to P. variotii). In addition, the species analyzed in this study displayed variations in growth rates and mycelial production, which merit consideration for potential future applications and further study.

Streamlining ERP Deployment in Nepal's Oil and Gas Industry: A Case Analysis

  • Oil and gas industry is a unique sector with complex activities, long supply chains and strict rules for the business. It is important to use enterprise resource planning (ERP) systems to address these challenges as it helps in simplifying operations, improving efficiency and facilitating evidence-based decision making. Nonetheless, successful integration of ERP systems in this industry involves careful planning, customization and alignment with specific business processes including regulatory requirements. Several critical factors, such as strong change management, support of top managers and training that works have been identified in the study. Amongst the hurdles are employee resistance towards the changes, data migration complications and integration with existing systems. Nonetheless, NOCL's ERP implementation resulted in significant improvements in operating efficiency, better data visibility and compliance management. It also led to a decrease in financial reporting timeframes, more accurate inventory tracking and improved decision-making capabilities. The study provides useful insights on how to optimize oil and gas sector ERP implementations; key among them is practical advice including strengthening change management strategies, prioritizing data security and collaborating with ERP vendors. The research highlights the importance of tailoring ERP solutions to specific industry needs as well as emphasizes the strategic role of ongoing monitoring/feedback for future benefits sustainability.

Isolation and characterization of a novel gossypol-degrading bacteria Bacillus subtilis strain Rumen Bacillus Subtilis

  • Objective: The aim of the study was to isolate gossypol-degrading bacteria and to assess its potential for gossypol degradation. Methods: Rumen liquid was collected from fistulated cows grazing the experimental pasture. Approximately 1 mL of the rumen liquid was spread onto basal medium plates containing 2 g/L gossypol as the only source of carbon and was then cultured at $39^{\circ}C$ to isolate gossypol-degrading bacteria. The isolated colonies were cultured for 6 h and then their size and shape observed by microscope and scanning electron microscope. The 16S rRNA gene of isolated colonies was sequenced and aligned using National Center for Biotechnology Information-Basic Local Alignment Search Tool. The various fermentation conditions, initial pH, incubation temperature, inoculum level and fermentationperiod were analyzed in cottonseed meal (CSM). The crude protein (CP), total gossypol (TG), and free gossypol (FG) were determined in CSM after fermentation with isolated strain at $39^{\circ}C$ for 72 h. Results: Screening results showed that a single bacterial isolate, named Rumen Bacillus Subtilis (RBS), could use gossypol as a carbon source. The bacterium was identified by 16S rDNA sequencing as being 98% homologous to the sequence of Bacillus subtilis strain GH38. The optimum fermentation conditions were found to be 72 h, $39^{\circ}C$, pH 6.5, moisture 50%, inoculum level $10^7cell/g$. In the optimum fermentation conditions, the FG and TG content in fermented CSM decreased 78.86% and 49% relative to the control. The content of CP and the essential amino acids of the fermented CSM increased respectively, compared with the control. Conclusion: The isolation of a gossypol-degrading bacterium from the cow rumen is of great importance for gossypol biodegradation and may be a valuable potential source for gossypol-degradation of CSM.

양호한 Finishing을 위한 이론적 근거 및 기준 (Rationale and criteria for excellent finishing)

  • 교정 치료 후 이상적인 교합과 양호한 심미적 결과를 얻기 위해서 일반적으로 교정 장치 제거하기 4개월에서 7개월 전에 detailing을 시행한다. 이와 같은 과정을 finishing이라고 하며, 이 과정은 최종적인 양호한 결과를 얻는데 있어서 필수적이다. 일부 교정의는 finishing을 위한 특별한 이론적 근거 및 기준 없이 이 과정을 교정 치료의 마지막 단계에서 시행하면 되는 것으로 여기고 있다. 그러나, 최종적인 양호한 교정 치료 결과를 얻기 위해서는finishing 과정이 총 치료 계획의 부분으로 치료의 초기 단계에서부터 고려되어야만 하며, 교정 장치 제거를 위한 이론적 근거 및 기준에 근거한 finishing을 위한 checklist가 필요하다. 이 checklist는 크게 네 부류로 분류할 수 있다. 즉, 교합 요인, 심미적 요인, 치주적 요인과 습관적 요인으로 분류할 수 있다. 교합 요인에서 점검해야할 요소로는 alignment, marginal ridge discrepancy, interproximal contact, anterior inclination, posterior inclination, overjet, overbite, arch form과 functional occlusion 등이 있다. 심미적 요인에는 gingival form, crown form, crown width과 crown length 등이 있다. 치주적 요인에는 root angulation, bone level 등이 있다. 습관적 요인에는 mouth breathing, tongue position at rest, tongue thrust habit, lip biting, nail biting과 finger sucking 등이 있다.

관성항법장치 초기정렬시간 단축을 위한 링레이저 자이로 lock-in오차 보상방법의 수치해석적인 분석 (Numerical Research on the Lock-in Compensation Method of a Ring Laser Gyroscope for Reducing INS Alignment Time)

  • 링레이저 자이로에는 입력각속도가 작은 영역에서 비선형적인 출력현상인 lock-in이 발생하는데 이를 제거하기 위하여 레이저 공진기에 정현파 각진동을 인가하는 방법이 주로 적용된다. 그러나, 그 방법을 적용하는 경우에도 각진동 회귀점에서 lock-in에 의한 오차가 남아있게 되는데, 이들 오차에 의하여 링레이저 자이로의 일반적인 오차특성인 랜덤웍이 발생된다. 이 lock-in에 의한 오차를 제거하기 위한 많은 연구결과 중의 한 방법으로써 lock-in오차 보상방법은 공진기 각진동 회귀점을 통과하기 전과 후의 맥놀이신호 주기를 비교하여 오차를 추정하고 보상하는 방법이다. 본 연구에서는 자이로 모델링 및 수치해석적인 방법으로, 이 lock-in오차 보상방법의 이론적인 적용 가능성을 분석하고, 현재 가능 할 것으로 판단되는 맥놀이 신호주기 측정 분해능을 감안하여 이 방법의 적용 효과를 분석하였다. 그 결과 lock-in오차 보상방법에 의하여 랜덤웍이 약 1/2~1/3로 감소될 수 있음을 알 수 있었다. 그러므로 이 방법은 항법장치의 정렬시간을 획기적으로 단축시킬 수 있는 방법이 될 것으로 기대된다.
