Bacterial community structure in the Juam Reservoir was analysed using fluorescent in situ hybridization (FISH) technique from April 2005 to January 2006. Total bacterial numbers varied in the range of 1.58 ${\sim}\;2.73{\times}\;10^6\;cells\;mL^{-1}$ proportional to the concentration of chi-a and had the minimal value in January. The ratios of ${\alpha}\;{\cdot}\;{\beta}\;{\cdot}\;{\gamma}$-subclass proteobacteria and Cytophaga-Flavobacterium (CF) group to total bacteria ranged from 45.1% to 77.5%, and the ratios of ${\alpha}\;{\cdot}\;{\beta}\;{\cdot}\;{\gamma}$-subclasses to total bacteria in June and September with the concentration of chi-a measured were lower than those ratios in April and January. It suggests that enriched growth of Microcystis aeruginosa may inhibit the metabolic activlty of ${\alpha}\;{\cdot}\;{\beta}\;{\cdot}\;{\gamma}$-subclass proteobacteria. However, the ratio of CF group bacteria represented no large change depending on algal bloom. In terms of nitrifying bacteria, the numbers of ammonia-oxidizing bacteria ranged from 9.9 ${\times}\;10^4\;to\;25.5\;{\times}10^4\;cells\;mL^{-1}$ with sharp fluctuation whereas those of nitrite-oxidizing bacteria varied in 8.7${\sim}9.8{\times}10^4\;cells\;mL^{-1}$ without noticeable change except the maximal value of $20.3{\times}10^4\;cells\;mL^{-1}$ in January maybe due to the high DO.

The purpose of this study was to validate the EFDC model for the weir pool of Gangjeong-Goryong Weir located in Nakdong River, and evaluate the effect of flow control and phosphate loading reduction on the water quality and algae biomass by group (Diatom, Green, Cyanobacteria). As a result of model validation using 2018 experimental data,the time series of water level and vertical distribution of water temperature, DO, organic matter, nitrogen, and phosphorus time series were properly simulated. Seasonal fluctuations of algae biomass by group were adequately reproduced, but the deviations between measured and simulated values were significant in some periods. As a result of scenario simulations to control the water level and flow rate, the thermal stratification was resolved as the water level was lowered and the flow rate increased. The flow velocity at which the water temperature stratification was resolved was about 0.1 m/s, which is consistent with the previous study results of Baekje Weir in Geum River. Simulations of the 2Q flow scenario showed that Chl-a decreased by 8.7% and the cell density of diatom and green algae declined. The cell density of cyanobacteria increased, however, because the high concentrations of cyanobacteria in the upstream boundary conditions directly affected downstream due to increased flow velocity. In the scenario simulation of reducing the influent phosphate load concentration (average 0.056 mg/L) to 50%, Chl-a decreased by 13.6%.The results suggest that the upstream algae concentration and phosphorus load reduction should be considered simultaneously with hydraulic control to prevent algal overgrowth of Gangjeong-Goryong Weir.

The effect of various SNPs (silver nanoparticles) on the growth of Microcystis aeruginosa was investigated in laboratory and field experiment. Four SNPs, namely JS47N, JS47N-K2, JS47N/3-1 and JS47N/3-2 were used to this study. The Ag size, concentration and color of these solutions were about $20{\sim}40nm$, $200mg\;L^-1$ and brown, respectively. At 0.01 and $0.1mg\;L^-1$, SNPs inhibited the growth of unicellular M. aeruginosa by 99.4% and 99.9%, respectively. However, SNPs of $1mg\;L^-1$ inhibited the growth of colonial M. aeruginosa by 98.5%, whereas the other three concentrations (0.001, 0.01 and $0.1mg\;L^-1$) had little inhibitory effect. In experimental enclosures from eutrophic lake, cyanobacteria including M. aeruginosa were found to be more sensitive to the SNPs than green algae and diatoms. In conclusion, our study indicates that SNPs has a selective cyanocidal potential when used to M. aeruginosa. We believe that future studies need to test on various other organisms, and determine minimum concentration for field application.

Microalgae are a promising resource in energy and food production as they are cost-effective for biomass production and accumulate valuable biological resources. In this study, CO2 assimilation, biomass, and lipid production of 4 microalgal species (Chlorella vulgaris, Mychonastes homosphaera, Coelastrella sp., and Coelastrella vacuolata) were characterized at different CO2 concentrations ranging from 1% to 9%. Microscopic observation indicated that C. vulgaris was the smallest, followed by M. homosphaera, C. vacuolata, and Coelastrella sp. in order of size. C. vulgaris grew and consumed CO2 more rapidly than any other species. C. vulgaris exhibited a linear increase in CO2 assimilation (up to 9.62 mmol·day-1·l-1) as initial biomass increased, while the others did not (up to about 3 mmol·day-1·l-1). C. vulgaris, Coelastrella sp., and C. vacuolata showed a linear increase in the specific CO2 assimilation rate with CO2 concentration, whereas M. homosphaera did not. Moreover, C. vulgaris had a greater CO2 assimilation rate compared to those of the other species (14.6 vs. ≤ 11.9 mmol·day-1·l-1). Nile-red lipid analysis showed that lipid production per volume increased linearly with CO2 concentration in all species. However, C. vulgaris increased lipid production to 18 mg·l-1, compared to the 12 mg·l-1 produced by the other species. Thus, C. vulgaris exhibited higher biomass and lipid production rates with greater CO2 assimilation capacity than any other species.

Research on the potential biological activity of red alga Symphyocladia spp. has been limited to Symphyocladia latiuscula, which is widely used as a food ingredient in Korea. Here, we examined the biological activity of another species, Symphyocladia linearis, which is found in Korea and was reported as a new species in 2013. The aim of this study was to evaluate the antioxidant, anti-inflammatory, and antibacterial properties of a 70% ethanol extract of S. linearis. Antioxidant activity, which was evaluated using radical scavenging assays, revealed half maximal inhibitory concentration values for 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH) and 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) of 34.57 and 11.70 ㎍/mL algal extract, respectively. Anti-inflammatory activity of the S. linearis ethanolic extract was evaluated using RAW 264.7 cells by measuring the inhibition of lipopolysaccharide-induced nitric oxide (NO) and prostaglandin E2 (PGE2) production. The potential cytotoxicity of NO and PGE2 was first examined, confirming no toxicity at concentrations ranging from 10-100 ㎍/mL. NO production was inhibited 61.1% and 78.0% at 50 and 100 ㎍/mL S. linearis extract, respectively; and PGE2 production was inhibited 69.1%, 83.2%, and 94.8% at 25, 50, and 100 ㎍/mL S. linearis extract, respectively. Thus, the S. linearis extract showed very strong efficacy against PGE2 production. The cellular production of reactive oxygen species, measured using 2',7'-dichlorofluorescin diacetate fluorescence, was inhibited 48.8% by the addition of 100 ㎍/mL S. linearis extract. Antibacterial activity was evaluated using the disc diffusion method and minimum inhibitory concentration (MIC). S. linearis was effective only against gram-positive bacteria, exhibiting antibacterial activity against Staphylococcus aureus with a MIC of 256 ㎍/mL extract and against Bacillus cereus with a MIC of 1,024 ㎍/mL extract. Based on these results, we infer that a 70% ethanolic extract of S. linearis possesses strong anti-inflammatory properties, and therefore has the potential to be used in the prevention and treatment of inflammatory and immune diseases.
Daechung Reservoir has been suffering from severe cyanobacterial blooming periodically due to the water pollutants from the watershed, especially nutrients from nonpoint sources. As a countermeasure, an artificial wetland was constructed to mitigate the pollutant load from the watershed by utilizing the vegetation. We investigated the water quality of the influent and outflow of the wetland during years 2014~2020 to evaluate the performance of pollutant removal through the wetland. Major pollutants (e.g. BOD, COD, SS, T-N, and T-P) were largely reduced during the retention in the wetland while nutrients removal was more efficient than that of organic matters. Pollutant removal efficiency for different inflow concentrations was also investigated to estimate the wetland's capability as a way of managing nonpoint sources. The efficiency of water treatment was significantly higher when inflow concentrations were above 75th percentile for all pollutant, implying the wetland can be applied to the pre-treatment of high pollution load including initial rainfall runoff. Furthermore, the yearly variation of removal efficiency for seven years was analyzed to better understand long-term trends in water treatment of the wetland. The annual treatment efficiency of T-P was very high in the early stages of vegetation growth with high concentration of inflow water. However, it was confirmed that the concentration of inflow water decreased, vegetation stabilized, and the treatment efficiency gradually decreased as the soil was saturated. The findings of the study suggest that artificial wetlands can be an effective method for controlling harmful algal blooms by alleviating pollutant load from the tributaries of Daechung Reservoir.
In this study, we estimated the concentration of chlorophyll-a (Chl-a) using hyperspectral water surface reflectance in an inland weir (Baekjae weir) and estuarine reservoir (Namyang Reservoir) for monitoring the occurrence of algae in freshwater in South Korea. The hyperspectral reflectance was measured by aircraft in Baekjae Weir (BJW) from 2016 to 2017, and a drone in Namyang Reservoir (NYR) from 2020 to 2021. The 30 reflectance bands (BJW: 400-530, 620-680, 710-730, 760-790 nm, NYR: 400-430, 655-680, 740-800 nm) that were highly related to Chl-a concentration were selected using permutation importance. Artificial neural network based Chl-a estimation model was developed using the selected reflectance in both water bodies. And the performance of the model was evaluated with the coefficient of determination (R2), the root mean square error (RMSE), and the mean absolute error (MAE). The performance evaluation results of the Chl-a estimation model for each watershed was R2: 0.63, 0.82, RMSE: 9.67, 6.99, and MAE: 11.25, 8.48, respectively. The developed Chl-a model of this study may be used as foundation tool for the optimal management of freshwater algal blooms in the future.
The effect of ammonia on survival and growth of the larval rockfish, Sebastes schlegeli was examined by a static renewal bioassay method. The tolerance of larvae to ammonia toxicity was more sensitive at the early larvae, but increased with the development of larvae. In 14 day-old-larvae after parturition, the mortality with treatment of each concentration of ammonia was 5% at control group and 0.0112 $NH_3$mg/l, whereas it was increased up to 27.5% at exposure group of 0.1230 $NH_3$mg/l with higher concentration. Regression equation between ammonium concentration(X) and mortality(Y) was followed; Y=0.516+3.482 X($r^2=0.4737$, P<0.01). The NOEC(no-observable-effect concentration) and LOEC (lowest-observable-effect concentration) to mortality compared to control group were 0.100 $NH_3mg/l$ and 0.1230 $NH_3$mg/l, respectively and chronic value(ChV) which is the geometric mean of the NOEC and LOEC was 0.1110 $NH_3$mg/l. Body length after 7-days exposure in control group, 0.0112 $NH_3$mg/l and 0.1230 $NH_3$mg/l were 7.8325mm, 7.700mm and 7.05mm, respectively. The NOEC, LOEC and chronic value(ChV) were 0.0335 $NH_3$mg/l, 0.0558 $NH_3$mg/l and 0.0432 $NH_3$mg/l, respectively.
The objective of this study was to analyze eutrophication characteristics, empirical model analysis, and variation of water quality according to monsoon intensity in Hapcheon Reservoir for 16 years from 2002 to 2017. Long-term annual water quality analysis showed that Hapcheon Reservoir was in a meso-nutrition to eutrophic condition, and the eutrophic state intensified after the summer monsoon. Annual rainfall volume (high vs. low rainfall) and the seasonal intensity in each year were the key factors that regulate the long-term water quality variation provided that there is no significant change of the point- and non-point source in the watershed. Dry years and wet years showed significant differences in the concentrations of TP, TN, BOD, and conductivity, indicating that precipitation had the most direct influence on nutrients and organic matter dynamics. Nutrient indicators (TP, TN), organic pollution indicators (BOD, COD), total suspended solids, and chlorophyll-a (Chl-a), which was an estimator of primary productivity, had significant positive relations (p<0.05) with precipitation. The Chl-a concentration, which is an indicator of green algae, was highly correlated with TP, TN, and BOD, which differed from other lakes that showed the lower Chl-a concentration when nutrients increased excessively. Empirical model analysis of log-transformed TN, TP, and Chl-a indicated that the Chl-a concentration was linearly regulated by phosphorus concentration, but not by nitrogen concentration. Spatial regression analysis of the riverine, transition, and lacustrine zones of $log_{10}TN$, $log_{10}TP$, and $log_{10}CHL$ showed that TN and Chl-a had significant relations (p<0.005) while TN and Chl-a had p > 0.05, indicating that phosphorus had a key role in the algal growth. Moreover, the higher correlation of both $log_{10}TP$ and $log_{10}TN$ to $log_{10}CHL$ in the riverine zone than the lacustrine zone indicated that there was little impact of inorganic suspended solids on the light limitation in the riverine zone.

Harmful algal blooms (HABs) are a serious problem for public health and fisheries industries, thus there exists a need to investigate the possible ways for effective control of HABs. In the present study, we investigated the algicidal effects of a newly developed GreenTD against the HABs (Chattonella marina, Heterosigma akashiwo, Cochlodinium polykriokides, and Heterocapsa circularisquama) and non-HABs (Chaetoceros simplex, Skeletonema sp. and Tetraselmis sp.), which is focused on the different population density and concentration gradients of algicidal substances. The time series viability of target alga was assessed based on the activity of Chl. a photosynthetic efficiency in terms of $F_v/F_m$, and in vivo fluorescence (FSU). Effective control of Raphidophyta, C. marina and H. akashiwo was achieved at a GreenTD concentration of $0.5{\mu}gL^{-1}$ and $0.2{\mu}gL^{-1}$, respectively, and regrowth of both the species was not observed even after 14 days. The inhibitory ratio of the dinoflagellate, C. polykriokides was more than 80% at $0.2{\mu}gL^{-1}$ of GreenTD. H. circularisquama was constantly affected in the presence of $0.2{\mu}gL^{-1}$ of GreenTD in the high- and low-population density experimental groups. On the other hand, diatoms, C. simplex, and Skeletonema sp. were not significantly affected even in the presence of $0.2{\mu}gL^{-1}$ of GreenTD and exhibited re-growth activity with the passage of incubation time. In particular, green alga Tetraselmis sp. remained unaffected even in the presence of the highest concentration of GreenTD ($1.0{\mu}gL^{-1}$), implying that non-HABs were not greatly influenced by the algicidal substances. As a result, the algicidal activity of GreenTD on the harmful and nonharmful algae was as follows: raphidophyte>dinoflagellates>diatoms>green alga. Consequently, our results indicate that inoculation of GreenTD substances into natural blooms at a threshold concentration ($0.2{\mu}gL^{-1}$) can maximize the algicidal activity against HABs species. If we consider the dilution and diffusion rate in the field application, it is hypothesized that GreenTD will demonstrate economic efficiency, thus leading to effective control against the target HABs in the closed bay.

제 19 조 (관할 법원)
