허혈(虛血)/재관류(再貫流) 뇌(腦) 조직(組織)에서 녹용(鹿茸) 약침액(藥鍼液)의 항산화작용(抗酸化作用)에 대한 연구(硏究) (Effects of Cervus elaphus extract solution for Aqua-acupuncture on Antioxidation in Rat's Brain induced by ischemia and reflow)

  • Cervus elaphus, being known to reinforce Kidney, have tested to study the effects concerning damages of brain tissue induced by lipid peroxidation. In vitro, the level of lipid peroxide in brain tissue was decreased proportinally according to dose by Cervus elaphus extract solution for aqua-acupuncture (CESAA). It was much more decreased, when lipid peroxidation was induced with Fe(II). And, it was seen proportinally decrease according to the dose of CESAA on xanthine oxidase activities and type conversion ratio. However, I can not find special changes about aldehyde oxidase activities. And, I had observed the effects of CESAA on damages of rat's brain following ischemia and reflow. Before ischemia was caused, CESAA was applied 0.2 ml per 250 g through femoral vein in ischemia and reflow group and normal sailine was applied in normal group. Ischemia was caused by cervical artery's clamp for 30 min and reflowed by clamp remove after 15 min. It was increased on the content of lipid peroxidation, activies and type conversion ratio of xanthine oxidase following ischemia and reflow. However, they were decreased when CESAA was pre-appllied. However, it could not seen special changes on aldehyde oxidase activities, either. In conclusion. CESAA recovers the damage of brain due to ischemia and reflow by decreasing the lipid peroxidation through decreasing of xanthine oxidase activies and type conversion ratio.

  • Jwagyuyeum and Woogyuyeum, being known to reinforce Kidney-yin and -yang, were tested for the effects of on free radical generating enzyme and lipid peroxidation in senile rat brain. In vitro, levels of lipid peroxide in tissues of brain were proportionally decreased to concentration of extracts prepared from Jwagyuyeum and Woogyuyeum. They were much more decreased, when lipid peroxidation was induced with ferrous iron ($Fe^{-2}$). In vivo, after both berbs were administered to the rat. levels of lipid peroxide in brain were decreased. especially it was much more decreased using Jwagyuyeum. Also, enzyme activities of xanthine oxidase and aldehyde oxidase in brain were decreased. The ratio of type conversion of the brain xanthine oxidase was lowered in both, especially Jwagyuyeum was much more done. These results suggest that Jwagyuyeum and Woogyuyeum decrease the activities of free radical generating enzymes such as xanthine oxidase and aldehyde oxidase which form lipid peroxide, Consequently both herbs might delay aging.

백하수오(白何首烏)의 항산화활성(抗酸化活性)과 amino acid의 분포(分布)에 관한 실험적(實驗的) 연구(硏究) (Study on the Antioxidative Effects and Amino Acid Contents of the Roots of Cynanchum Wilfori)

  • The aim of the presents study is to investigate and compare the antioxidative effects and qualities of the cultivating root of Cynanchum Wilfori, which is increasingly used in recent days, with those of the wilding root, which has mainly been used in the past in oriental medicine for a tonic and also for prevention and treatment of various geriatric diseases including aging progress. For comparison of their antioxidative effects, the activities of the total extracts on lipid peroxidation and the activity of aldehyde oxidase(EC 1.2.3.1) as well as xanthine oxidase(EC 1.2.3.2) were investigated in vitro. In addition, their inhibitory effects on the activity of 5-lipoxygenase, which is known to induce inflammation and concerned with free radicals, were also determined in vitro. Furthermore, the amino acid contents of both roots were analyzed in order to compare their qualities. The results are as follows: 1. The wilding root inhibited significantly the activity of 5-lipoxygenase, showing five times more portent than the cultivating root. 2. Both of the wilding root and the cultivating root inhibited aldehyde oxidase activity in a dose-dependant manner. The wilding root was more effective than the other. 3. Both of the wilding root and the cultivating root dose-dependently suppressed lipid peroxidation in rat brain, kidney, and liver. 4. The anti-peroxidative effects of both roots appeared to be most strong in brain and least in liver. In particular, the cultivating root exhibited a significant inhibition on brain lipid peroxidation. 5. The cultivating root contained 15 amino acids including five essential amino acids in contrast with the less contents in the wilding root.

Structure-Activity Relationships of Gagaminine and Its Derivatives on the Inhibition of Hepatic Aldehyde Oxidase Activity and Lipid Peroxidation

  • In order to determine the structure-activity relationships for antioxidative effects of gagaminine, a steroidal alkaloid isolated from the roots of Cynanchum wilfordi (Asclepiadaceae), two derivatives identified as sarcostin and penupogenin were prepared from gagaminine by hydrolysis and reduction. These compounds were evaluated for the inhibitory effects on the aidehyde oxidase activity and on lipid perbxidation in vitro. Furthermore, their effects were compared with those of gagaminine and the related compounds, cinnamic acid and nicotinic acid. The results of this study prove that the cinnamoyl group in the structure of gagaminine is critical in inhibition of the aldehyde oxidase activity while the nicotinoyl group may be necessary for anti-lipid peroxidation of the compound.

옥수수 (Zea mays) 뿌리에서 에탄올에 의한 알데히드 산화효소의 활성 증가 (Promotion of Aldehyde Oxidase Activities by Ethanol in Maize (Zea mays) Roots)

  • 옥수수의 원뿌리(primary root)를 재료로 외부에서 처리한 에탄올이 알데히드 산화효소의 활성을 변화시키는 현상을 관찰하였다. 에탄올의 촉진 효과는 처리된 에탄올의 농도에 따라 다르게 나타나 알데히드 산화효소는 0.2-0.4% 에탄올 처리 구간에서 대조구에 비하여 낮은 활성을 나타내었으며 0.8-1.0% 에탄올 처리 구간에서는 대조구에 비하여 높은 활성을 나타내었다. 에탄올에 의하여 알데히드 산화효소의 활성이 증가하는 조건에서도 두 개의 알데히드 산화효소 유전자 AO1과 AO2 의 전사 수준에는 변화가 없었다. 그러나 에탄올 처리는 알데히드 산화효소의 단백질 함량을 현저히 증가시켰으며, 이는 에탄올이 알데히드 산화효소 활성을 증가시키는 조절작용이 번역(translation) 단계에서 이루어진다는 사실을 보여주었다. 에탄올, 메탄올 그리고 이소프로판올을 처리한 실험 결과 에탄올에 의해서만 알데히드 산화효소의 활성 증가가 유도되었다. 에탄올은 식물체의 뿌리가 저산소 조건에서 에너지 확보를 위하여 발효를 진행하는 경우 자연적으로 식물체내 농도가 증가하는 물질이다. 따라서 에탄올 처리시에 알데히드 산화효소의 활성이 증가하는 현상은 알코올에 대한 일반적인 반응이 아니라 식물체의 뿌리가 생리적으로 경험할 수 있는 에탄올에 대하여 특이적으로 나타내는 반응이라는 결론을 얻었다.

The enzyme activities and the ratio of type conversion of xanthine oxidase were decreased only in Jwagyuyeum. But, We can't see special changes in Woogyuyeum. The enzyme activities of aldehyde oxidase was lowered in both herbs, especially Jwagyuyeum was much more done. These results suggest that Jwagyuyeum and Woogyuyeum decrease the activities of free radical generating enzymes such as xanthine oxidase and aldehyde oxidase which form lipid peroxide.

  • Jwagyuyeum and Woogyuyeum, being known to reinforce Kidney-yin and -yang, were tested for the effects of on free-radical generating enzyme and lipid peroxidation. In vitro, levels of lipid peroxide in tissues of liver were proportionally decreased to concentration of extracts prepared from Jwagyuyeum and Woogyuyeum. They were much more decreased, when lipid peroxidation was induced with ferrous iron (Fe+2). In vivo, after both herbs were administered to the rat, levels of lipid peroxide in liver were decreased only in Jwagyuyeum. And, enzyme activities of xanthine oxidase and aldehyde oxidase in liver were not changed. It was guessed that Jwagyuyeum and Woogyuyeum inhibited lipid peroxidation directly, or acted on free radical resolving enzymes which decrease lipid peroxide. Consequently both herbs, particularly in Jwagyuyeum might delay aging.

석창포 정유성분의 항산화활성 (Antioxdative Effect of the Essential Oil form the Rhizomes of Acorus gramineus)

  • 한방에서는 경련억제의 목적으로 처방되고 있는 석창포의 정유액을 분리하여 활성산소종 생성계 효소인 xanthine oxi-dase와 alehyde osidase에 대한 억제활성을 in vitro에서 측정하고 지질과산화 억제효과를 in vitro 와 in vivo에 각각 조사하였으며 DPPH radical 소거 효과를 검토하여 다음과 같은결과를 얻었다. 석창포 정유는 xanthine oxidase에 대해서는 0.01 mg/ml에서 13.3%, aldehyde oxidase에 대해선.s 0.001mg/ml에 서 5.0% 각각 효소 활성을 억제 하였으나, 괴량에서는 오히려 효소활성을 증가시키는것으로 나타났다. 지질과산화 억제효과는 1.0mg/ml에서 대조군에 비해 49.4% 억제하여 우수한 효과를 보였으며 향기를 7일간 흡입시킨 in vivo test에서는 인위적으로 지질과산화를 유발시킨 대조군에 비해 과산화지질이 16.7% 감소되었다. 그러나 DPPH radical에 대한 소거효과는 2.0 mg/ml에서약 4%의 미미한 효과를 나타내었다.

에탄올 전처치한 흰쥐에 Xylene 투여가 간조직 중 Xanthine Oxidase 활성 변동에 미치는 영향 (Effect of Ethanol-pretreatment on the Liver Xanthine Oxidase Activity in Xylene-treated Rats)

  • To evaluate an effect of ethanol pretreatment on the liver xanthine oxidase(XO) activity, 0.25ml of xylene(50% in olive oil) per 100g body weight was daily given four days to the rats at 2hrs after aministration of ethanol each day, while each control group(ethanol, xylene, olive oli) was treated as the same dose described as above. The animals were sacrificed at 24hrs after last injection. Xylene-treated rats showed the more decreased activity of liver XO compared to the control. But the pretreatment of ethanol to the xylene-treated rats enhanced the liver XO activity. Furthermore, the xylene-treated rats led to more increased Vmax value in liver XO compared to the only xylene-treated rats. On the other hadn, hepatic aldehyde dehydrogenase activity was more decreased in xylene-treated rats pretreated with ethanol than in xylene-treated rats. And the intermediated xylene metabolites, methyl benzylalcohol or aldehyde inhibited the XO activity "in vitro". In conclusion, the phenomenon that pretreatment of ethanol to the xylene-treated rats led to the enhancement of liver XO activity, may be due to an influence of acetaldehyde.taldehyde.

Testosterone에 의한 승홍의 신독성 유발에 의한 연구 (The role of testosterone on nephrotoxicity of mercuric chloride)

  • Mercuric chloride, inorganic compound, is one of the most important drugs that has been used in the field of argriculture, antisyphilitica and anticeptics, but it is not used clinically at present. We have studied the effect of testosterone on the mercuric chloride-induced nephrotoxicity. Renal lipid peroxide concentration of male rat treated with mercuric chloride was significantly increased in comparison with that of the female rat, it showed similar effects on testosterone pretreatment. Changes in renal catalase and gluta- thione peroxidase activities were not siginificantly different in testosterone-treated groups. But, renal xanthine oxidase and aldehyde oxidase activities of testrosterone-treated group given mercuric chloride significantly increased in comparison with that of the testoste- rone-treated alone. Animals treated with testosterone prior to mercuric chloride showed more severe damage on histological observations than those treated with testosterone only. Consequently, we suggest that the mercuric chloride-induced nephrotoxicity might be renal lipid peroxide generating enzyme system by testosterone.
