Effects of Alanine and Glutamine Supplementation on Alcohol Metabolism in ICR Mice

  • This study was conducted to investigate the effects of oral supplementation of alanine and glutamine on alcohol metabolism. The subjects were 70 male ICR mice weighing 25-30 g. The animals were raised on standard rations artier weaning. After 24 hours of fasting, all the animals were given a peritoneal injection of 20% alcohol. Then, they were randomly divided into two groups: control and experimental. Fifteen minutes after the injection of alcohol, the mice in the experimental group wer given an oral solution of alanine(5 mM, 2 g/kg B. W) and glutamine (5 mM, 2g/kg B.W). The concentration of alcohol in the blood was measured in all the mice 20 minutes after they received the alochol, and the measurements continued every 20 minutes up to 140 minutes. The experimental group sustained lower blood alcohol levels at every 20 minute time interval compared to the control group, showing that oral supplementation of alanine and glutamine increases the rate of alcohol metabolism. Furthermore, the total amount of alcohol remaining in the blood, determined by using the Area Under the Curve (AUG) method, was lower in the group supplemented with alanine and glutamine, However, the effectiveness of alanine and glutamine in increasing the rate of alcohol metabolism, compared to the control group, diminished with time throughout the experiment. In conclusion, alanine and glutamine supplementation appears to promote alcohol metabolism shorthy after alcohol intake.

  • Aldehyde and active form of free oxygen produced in alcohol metabolism in liver are the cause of liver cell damage. The main system of alcohol metabolism is composed of alcohol dehydrogenase(ADH), aldehyde dehydrogenase(ALDH) and cytochrome P4502E1. Alcohol dehydrogenase is reversible in alcohol metabolism. To block the backward reaction and enhance alcohol oxidation, acetaldehyde trapping agents were assayed. The assay was carried out by measuring decreasing NADH at 340nm, using acetaldcehyde and NADH as substrate and coenzyme respectively. (omitted)

Comparison of silkworm powder from 3 Bombyx mori varieties on alcohol metabolism in rats

  • Increased alcohol consumption is a burden on the world because it is associated with various health problems. However, the effects of silkworms on alcohol metabolism have not been studied yet. The hard-to-eat mature silkworms have become easier to ingest recently due to the development of technology, steam-lyophilising mature silkworm larvae. In this study, we investigated and compared the effects of SMSPs from three silkworm varieties, Baekokjam, Golden-silk and Yeonnokjam weaving white, golden, and light green cocoons on alcohol metabolism in vivo. Sprague-Dawley rats pretreated with three SMSPs (0.1 g/kg or 1 g/kg body weight) or normal diet (AIN-76A) for 2 weeks were subjected to intragastric administration of absolute ethanol (3 g/kg body weight, 3 h). Three SMSPs did not affect the final body weight and liver weight. All 3 SMSPs were effective to reduce the enzymes in alcohol metabolism, alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH), and liver damage and enzymes involved in liver damage, aspartate aminotransferase (AST) and alanine aminotransferase (ALT). Among SMSP from 3 varieties of silkworm, preadministration of 1 g/kg Baekokjam SMSP showed the most effective suppressive effect on the activities of ADH, ALDH, AST and ALT. The Baekokjam SMSP contained higher amounts of beneficial amino acids than Golden-silk or Yeonnokjam SMSP. These results suggest that Baekokjam SMSP might be used as a new and promising candidate for improving alcohol metabolism and liver injury through promoting rapid alcohol metabolism.

Effects of Phellinus spp. Extract on Alcohol Metabolic Enzymes in Alcohol-treated Rats

  • Alcoholism is a significant health problem in the world. The liver is the first and primary target organ for alcohol metabolism. Alcohol dehydrogenase and aldehyde dehydrogenase play important roles in the metabolism of alcohol and aldehyde. In this study, I aimed to investigate the eliminatory effects of a Phellinus spp. extract on alcohol metabolism in drunken Sprague-Dawley (SD) rats. Male SD rats were given Phellinus spp. extract at 30 min after 40% (5 g/kg) alcohol ingestion. To assay the effect of Phellinus spp. extract on blood alcohol concentration, blood samples were taken from the tail vein at 1, 3 and 5 h after alcohol ingestion. The concentrations of alcohol, alcohol dehydrogenase, and aldehyde dehydrogenase in Phellinus spp. extract treated rat were significantly lower than that of the control with a time-dependent manner. In addition, the alanine aminotransferase and aspartate aminotransferase activities of Phellinus spp. extract-treated groups were altered compared to those of the control group. These results suggest that Phellinus spp. extract intake can have a positive effect on the reduction of alcohol, alcohol dehydrogenase, and aldehyde dehydrogenase concentrations in the blood and may alleviate acute alcohol-induced hepatotoxicity by altering alcohol metabolic enzyme activities. Phellinus spp. extract is thus a good nutraceutical candidate.

Effects of Dried Persimmon Snacks on Alcohol Metabolism in Men

  • Persimmon has been known to help alcohol intoxication in Korea for a long time and is prepared as a processed food. The effect of dried persimmon snacks on alcohol metabolism was determined in vivo. Eight Korean men (25~27 years old) were administered 3.5 mL/kg of alcohol (22.5%, w/v) with or without a dried persimmon snack (2g/kg). The levels of alcohol in the blood and of acetaldehyde in the blood and the urine were determined by alcoholmeter and assay using alcohol dehydrogenase and HPLC, respectively. All subjects showed decreased levels of alcohol in blood, and six subjects showed a decrease of alcohol in urine after consumption of ethanol with dried persimmon snacks. Concentration of acetaldehyde in the blood an urine decreased significantly in three and five of the eight subjects, respectively. Reduction was more significant for alcohol than for alcohol than for acetaldehyde with administration of ethanol and dried persimmon snacks. This in vivo result suggests that dried persimmon snacks are effective in decreasing th concentration of alcohol after alcohol intake.

오미자 열매의 물추출물이 알콜대사에 미치는 효과 (Effects of Water Extracts in fruits of Omija (Schizandra chinensis Baillon) on Alcohol Metabolism)

  • To assess the effects of water extracts in fruits of Omija (Schizandra chinensis Baillon) on alcohol metabolism, rats were orally administrated with alcohol (25% alcohol, 0.75g/200g B.W., 40% alcohol, 0.8g/200g B.W.). The level of metabolites and enzyme activities of the serum and liver were unchanged by the 25% ethanol or 40% ethanol treatment with acute orally administration. Blood alcohol level was markdely decreased by the treatment with water extracts in fruits of Omija. The serum level of Urea nitrogen, Free fatty acid, GPT and LDH were tended to decreased, level of GOT was unchanged. Contents of hepatic microsomal protein, glycogen, pyruvate in the liver were increased by water extracts in fruits of Omija. In conclusion, the present study clearly demonstrates that water extract in fruits of Omija promotes the overall metabolism and detoxication of alcohol.

  • This study was designed to elucidate the effect of fermented blackberry drinks (BD) on alcohol metabolism and hangover in alcohol-treated rats. We showed that the administration of BD increased the activity of alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) in alcohol-treated rats. Moreover, the administration of BD reduced the serum alcohol and acetaldehyde concentrations in alcohol-treated rats. Furthermore, the administration of BD attenuated the levels of serum aspartate aminotransferase and alanine aminotransferase in alcohol-treated rats. Taken together, these results suggest that BD plays an important role in alcohol metabolism and liver function by reducing blood alcohol and acetaldehyde through the activation of ADH and ALDH in alcohol-treated rats and could be used as a functional anti-hangover drinks.

  • This study was undertaken to investigate the effect of the levels of dietary Zn and alcohol consumption on lipid metabolism in Sprague-Dawley male rats weighing 80 to 90g for eight weeks. Ninety rats were divided into nine groups according to Zn levels and alcohol consumption such as no alcohol group[low Zn diet group(<1ppm. LZ) control Zn diet group(30ppm. CZ) high Zn diet group(60ppm, HZ)] 10% alcohol consumption group[the same Zn levels as no alcohol group LZLA, CZLA, HZLA] 20% alcohol consumption group[the same Zn levels as no alcohol group, LZHA, CZHA, HZHA] The results obtained were summarized as following : 1) In the serum total cholesterol increased with increasing dietary Zn levels but decreased with alcohol consumption. HDL-cholesterol decreased with alcohol consumption. Triglyce-ride in alcohol group was higher than no alcohol group. In alcohol group triglyceride increased with decreasing dietary Zn levels. 2) In the liver total lipid in alcohol group was higher than no alcohol group HA group showed a significant increase. Triglyceride increased with alcohol consumption. 3) In the serum and liver Zn content increased with increasing dietary Zn levels but decreased with alcohol consumption.

  • Purpose: This study was conducted to investigate the effect of freeze-dried Gastrodiae rhizoma powder (GR) on the liver function and alcohol metabolism in alcohol treated rats. Methods: The rats were administered various concentrations of GR (100 mg, 200 mg, 300 mg/kg B.W.) for 3 days 1 hour before 50% (v/v) ethanol (3 g/kg B.W.) administration. Two tests focusing on liver function and alcohol metabolism in acute alcohol treatment were carried out. Results: Glutamic oxaloacetic transaminase activity was significantly increased by alcohol treatment, and was decreased by 100 mg GR administration. Acute ethanol treatment led to significant increase in alcohol and acetaldehyde levels of serum and liver. However, 100 mg GR administration led to a significant reduction in increased alcohol level in the serum with decreased alcohol dehydrogenase (ADH) activity and increased acetaldehyde level in liver was significantly reduced by three levels. Conclusion: These results suggest that GR can be effective in enhancing liver function and alcohol metabolism in the alcohol-treated rats. Studies on the appropriate dosage of GR should further be developed to treat alcohol detoxification and stimulate liver function.

Evaluation of the role of Lactobacillus casei on alcohol metabolism and liver functions of rats

  • Alcohol consumption causes numerous consequences on the health of the human body. Heavy drinking on a daily base has caused liver diseases. Furthermore, some products such as acetaldehyde produced from alcohol metabolism are more toxic than alcohol itself. This study was carried out to evaluate the role of Lactobacillus casei on alcohol metabolism, especially, the removal of the toxic effect of alcohol. The maximum alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) activities from L. casei were observed at 4 hr of culture. L. casei was confirmed to produce the ADH and ALDH by the SDS-PAGE. From in vivo test using SD rats with 22% alcoholic drink, blood alcohol concentration (BAC), glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GPT) of the rats feeding the medium containing L. casei were lower than those of the rats feeding the medium containing an alcoholic drink only This demonstrates that the ADH and ALDH produced by L. casei have virtual functions to detoxicate the alcohol in vivo and the fermentation broth of L. casei can be used as an alcohol detoxification drink.
