Gentisyl Alcohol, an Antioxidant from Microbial Metabolite, Induces Angiogenesis In Vitro

  • Gentisyl alcohol isolated from Penicillium sp. has an antioxidative activity, protecting cells from oxidative stresses. From our in vitro angiogenesis assays with bovine aortic endothelial cells (BAECs), gentisyl alcohol was newly identified as a pro-angiogenic small molecule that induces new blood vessel formation of the cells. Gentisyl alcohol stimulated the proliferation of BAECs in a dose-dependent manner. Moreover, it induced in vitro angiogenesis of BAECs such as invasiveness, migration, and tube formation of the endothelial cells. Effects of gentisyl alcohol on invasion and tube formation were also dose-dependent. These results demonstrate that gentisyl alcohol could affect the angiogenic phenotypes of endothelial cells and be developed as a new small molecule with pro-angiogenic activity.

Enzymatic Formation of Guaiacylglycerol 8-O-4'-(Coniferyl Alcohol) Ether from Coniferyl Alcohol with Enzyme Preparations of Eucommia ulmoides

  • Lignans and neolignans are optically active plant secondary metabolites. Research on biosynthesis of lignans has already been advanced especially for the formation of (+) pinoresinol but information on the biosynthesis of 8-O-4'- neolignans is still limited. Moreover, the chemical structure(position of substituents on aromatic rings) and stereochemistry of 8-O-4' neolignans is not clear. Katayama and Kado discovered that incubation of cell-free extracts from E. ulmoides with coniferyl alcohol in the presence of hydrogen peroxide gave (+)-erythro- and (-)-threo- guaiacylglycerol 8-O-4'-(coniferyl alcohol) ether (GGCE)(diastereomeric ratio, 3:2) which is the first report on enzymatic formation of optically active -8-O-4' neolignans from an achiral monolignol. In this aspect, enzymatic formation of guaiacyl 8-O-4' neolignan is noteworthy to clarify its stereochemistry from incubation of coniferyl alcohol with enzyme prepared from Eucommia ulmoides. In this experiment, soluble and insoluble enzymes prepared from E. ulmoides were incubated with 30 mM coniferyl alcohol(CA) for 60 min. The enzyme catalyzed GGCE, dehydrodiconiferyl alcohol(DHCA), and pinoresinol identified by reversed phase HPLC. Consequently, diastereomeric compositions of GGCE were determined as erythro and threo isomer. Enantiomeric composition was determined by the chiral column HPLC. Both enzyme preparations enantioselectively formed (-)-erythro, (+)-erythro and (+)-threo, (-)-threo-GGCEs respectively.

Effect of Liuweidihuang-tang on Alcohol-induced Decrease in New Cell Formation in Rat Dentate Gyrus

  • Liuweidihuang-tang has been traditionally used for the treatment of delayed mental and physical development in children, complications of diabetes. and glomerulonephritis. In the present study, the effect of Liuweidihuang-tang on cell proliferation in the dentate gyrus of alcohol-intoxicated Sprague-Dawley rats was investigated via 5-bromo-2' -deoxyuridine (BrdU) immunohistochemistry. Alcohol administration was shown to decrease the number of BrdU-positive cells in the dentate gyrus, while Liuweidihuang-tang treatment increased new cell formation in the dentate gyrus under normal conditions and alcohol intoxication. It was thus demonstrated that aqueous extract of Liuweidihuang-tang exerts a protective effect against alcohol-induced decrease in new cell formation.

  • Effects of iron compounds in known biomimetic oxidation systems (Gif IV and GoAgg II) have been studied on activity and ketone/alcohol selectivity of cyclohexane oxidation. Both ketone/alcohol ratio and cyclohexane conversion were affected by counter-ion Z of iron compounds Z-Fe. When Z has a more electron withdrawing property, the reactivity is increased and the formation of ketone is favored. From these experimental results, a new mechanism is proposed for the biomimetic oxidation system.

  • 본 연구에서 육계 추출물로부터 cinnamyl alcohol을 분리·정제하였다. 연구 결과, H3 분획100 ㎍/mL에서 부정근 형성율이 286.6%였으며, 표준품 cinnamyl alcohol 1 mM (134.2 ㎍/mL)의 형성율이 290%로 유사한 결과를 나타내었다. 따라서 육계 methanol 추출물에 함유되어있는 cinnamyl alcohol이 녹두의 부정근 형성을 유도하는 주된 화합물일 것으로 판단된다. 육계에서 동정한 cinnamyl alcohol은 현재 식품첨가제 및 향료로써 사용되고 있으므로 안전성이 이미 보장되어 있으며, 한약재로써 사용된 육계 잔류물을 활용하여 부정근 형성 유도물질을 추출한다면비용적인 측면에서 또한 긍정적인 요인으로 작용할 것으로 판단된다. 따라서 육계는 유기농 농자재로써 충분히 사용 가능할 뿐만 아니라 신규 천연 식물생장조절제의 개발용 소재로서도 활용 가능할 것으로 사료된다.

Effect of ethyl alcohol aging on the apatite formation of a low-modulus Ti-7.5Mo alloy treated with aqueous NaOH

  • The purpose of this experiment was to evaluate the apatite-formation abilities of low-modulus Ti-7.5Mo substrates treated with NaOH aqueous solutions and subsequent ethyl alcohol aging before soaking them in simulated body fluid. Specimens of Ti-7.5Mo were initially treated with 5 M NaOH at $60^{\circ}C$ for 24 h, resulting in the formation of a porous network structure composed of sodium hydrogen titanate. Afterwards, the specimens were aged in ethyl alcohol at $60^{\circ}C$ for 5 or 10 min, and subsequently immersed in simulated body fluid at $37^{\circ}C$ for 3, 7 and 14 days. Ethyl alcohol aging significantly increased the apatite-forming abilities of Ti-7.5Mo. The amount of apatite deposited on the Ti-7.5Mo after NaOH treatment and subsequent ethyl alcohol aging was much greater, especially after the Ti-7.5Mo specimens were aged for 5 min. Due to its excellent combination of bioactivity, low elastic modulus and low processing costs, the Ti-7.5Mo treated with NaOH aqueous solutions and subsequently aged in ethyl alcohol has promising heavy load-bearing applications.

  • Capsaicin(8-methyl-N-vanillyl-6-nonenamide: CAP) known well as a major compound of not taste in hot pepper, was investigated for the inhibition effect on in vitro nitrosation. CAP(100$\mu$mol) inhibited the formation of N-nitrosoproline(NPRO) and N-nitrosothioproline(NTPRO) by 56% and 26%, respectively. Vanillyl alcohol inhibited the nitrosation of proline by a concentration-dependent manner, and vanillic acid and vanillin were less effective in blocking the nitrosation of proline compared to CAP and anillyl alcohol. The inhibitory effect of NPRO formation by CAP was evaluated to similar with alpha-tocopherol, and vanillyl alcohol was more effective than alpha-tocopherol in blocking the nitrosation of proline. Our results suggested that CAP and its metabolites such as vanillyl alcohol could inhibit endogenous nitrosation in hydrophobic biological environment.

  • The author has observed the tissue reaction of the absolute alcohol infection of rat oral mucosa. 0.5ml absolute alcohol was injected subcutaneously on the mucobuccal fold of rat. And the rats were sacrifieced at intervals of one day, 3rd, 1 week, 2 week and 4 week after alcohol injection. The microscopic tissue sections were made and stained with hematoxylin and eosin. The results were are as follows; 1. Degeneration and shrinkage of fibroblasts and coagulative necrosis were observed one day to and three day after alcohol injection. 2. Although coagulative necrosis and tissue degeneration occurred, the inflammatory infiltration was not prominent especially there were scarcely any polymorphonuclear leukocytes in that field. 3. Granulation tissue with moderate small round cell infiltration were replaced the necrotic area at one week after injection and the fibroblast proliferate into the granulation tissue at two week group. 4.
