Micropropagation of Lillium Oriental Hybrid 'Casa Blanca' using Bulblet Sections with swollen Basal Plate in Bioreactor (생물반응기에서 저반부가 비대된 자구 절편체에 의한 오리엔탈 나리 'Casa Blanca' 의 대량증식)

  • A series of studies were carried out to establish micropropagation system, using airlift bioreactors (ebb $\varepsilon$ flood type, 5 L), of Lilium oriental hybrid 'Casa Blanca'. The bulblets with swollen basal plate were formed from bulb scales, then proliferated to bulblet clusters with swollen basal plate. Finally normal bulblets were formed from the sections. Bulblet formation and proliferation with swollen basal plate were not accomplished entirely in liquid culture of 5 L airlift bioreactors, but leafy bulb scales grew vigorously. Bulblet clusters with swollen basal plate were proliferated by periodic immersion culture. Bulblet proliferation was not affected by light, but scale leaves grew under light. MS medium containing 2.0 mg/L benzyl adenine (BA) and 0.3 mg/L indole acetic acid (IAA) was favorable to the bulblet proliferation with swollen basal plate. In liquid culture of 5 L bioreactors, bulblets from bulblet sections with swollen basal plate grew vigorously on MS medium with 70 g/L sucrose. It was effective for bulblet growth to replace the new medium after 8 weeks in culture during 16 weeks of cultural period. 15 g injection of bulblet sections as a cultural material was suitable for bulblet growth in 5 L bioreactors.

Advanced Wastewater Treatment of Low Concentration Ammonia Using the Immobilized Nitrifier Consortium (고정화 질화세균을 이용한 저농도 암모니아 폐수의 고도처리)

  • This study was performed in the airlift bioreactor using the nitrifier consortium entrapped in polyvinyl alcohol(PVA) for removing low concentration total ammonia nitrogen(TAN). At the superficial air velocity of 0.83 cm/sec, TAN removal rate and removal efficiency was $316.6{\pm}7.2g/m^3{\cdot}day$ and $92.8{\pm}2.2%$ respectively. Removal rate was continuously increased with decreasing hydraulic residence time(HRT) from 0.5 hr to 0.05 hr, whereas removal efficiency decreased with decreasing HRT. The optimum temperature for nitrification was $30^{\circ}C$ at which removal efficiency was $95.5{\pm}1.5%$. Nitrification was effectively performed at low temperature, $10^{\circ}C$. In the pH range from 7 to 9 in the bioreactor, removal rate and removal efficiency was $310{\pm}10g/m^3{\cdot}day$ and $94{\pm}3%$ respectively.

  • In microbial coal desulfurization process (MCDP) by using Acidithiobacillus ferrooxidans, the effect of process variables on pyritic sulfur removal efficiency has been investigated. The inhibitory effect of toxic materials contained in coal matrix on the activity of desulfurizing bacteria have been evaluated in coal extracts, and the results showed that the method was useful to evaluate the applicability of a coal which is to be desulfurization to MCDP. The removal efficiency increased with decreasing particle size and decreases with increasing pulp density, but has no significant influence of particle size and pup densities at high pulp densities over 20 wt%. The mass transfers of gaseous nutrients such as oxygen and carbon dioxide into coal slurry with various pulp densities and coal particle size has been studied in an airlift bioreactor. Mass transfer coefficient was independent of pulp density in coal slurry with fine particle below 175 $\mu\textrm{m}$, but significantly decreased with increasing pulp density over 225 $\mu\textrm{m}$. The coal particles over 575 $\mu\textrm{m}$ were significantly settled to the bottom of bioreactor resulting in poor mixing. Considering mass transfer, pulp density and coal mixing, an optimal size of coal particle for the microbial coal desulfurization process seems to be about 500 $\mu\textrm{m}$.

  • The production of laccase by Fomitella fraxinea was studied. The addition of minerals were necessary far laccase production by Fomitella fraxinea. Jar fermentor and Air-sparging fermentor performed high productivity In laccase activity by F. fraxinea. Laccase activity reached 3,540 in 8 days (Jar fermentor) and 3,100 in 6 days (Air-sparging fermentor) respectively.

  • A nitrifier consortium immobilized in polyvinyl alcohol was used to remove ammonia nitrogen from synthetic wastewater in an airlift bioreactor. The minimum aeration rates were 0.4, 0.6, 0.8 and 1.0 vvm for 5, 10, 15 and 20% immobilized bead packing volume fraction, respectively. The efficient packing fraction and the aeration rate for ammonia nitrogen removal were 15% and 2.4 vvm, respectively. With a hydraulic retention time of 0.5hr, the removal rate and the efficiency of ammonia nitrogen removal were 1685 g/㎥$.$day and 48% at an influent ammonia nitrogen concentration of 75 g/㎥.

Production of Itaconic Acid at Various Bioreactors (다양한 생물반응기에서 이타콘산의 생산)

  • A suitable culture method and bioreactor type for itaconic acid production were chosen by comparing the maximal concentration of itaconic acid produced in various systems. In batch culture, the maximal concentration of itaconic acid produced in a bubble column reactor was about 5% greater than that produced in stirred-tank or external-loop airlift reactor. These results were thought to be due to lower shear force and higher mass transfer efficiency in a bubble column reactor in comparison with other reactors. Moreover, the fed-batch mode in a bubble column was found to be a suitable one, producing about 25% higher concentration of itaconic acid compared to batch mode.

  • This study was performed by the airlift bioreactor using the nitrifier consortium entrapped in polyvinyl alcohol(PVA) for removing low concentration total ammonia nitrogen(TAN).   • The optimization of submerged culture conditions for mycelial growth and exopolysaccharide (EPS) production in an edible mushroom Tremella fuciformis were studied in shake flasks and bioreactors. The temperature of $28^{\circ}C$ and pH 8 in the beginning of fermentation in agitated flasks was the most efficient condition to obtain maximum mycelial biomass and EPS. The optimal medium constituents were as follows (g l-1): glucose 20, tryptone 2, $KH_2PO_4$ 0.46, $K_2HPO_4$ 1 and $MgSO_4H_2O$ 0.5. The fungus was cultivated under various agitation and aeration conditions in a 5L stirred-tank bioreactor. The maximum cell mass and EPS production were obtained at a relatively high agitation speed of 200 rpm and at an aeration rate of 2 vvm. The flow behavior of the fermentation broth was Newtonian and the maximum apparent viscosity (35 cP) was observed at a highly aerated condition (2 vvm). The EPS productivity in an airlift reactor was higher than that in the stirred-tank reactor. The EPS was protein-bound polysaccharides consisted of mainly mannose, xylose, and fructose. The molecular weights of EPS were determined to be $1.3{\sim}1.5{\times}10^6$.

  • This experiment was conducted to micropropagate bulblets via shoot cluster formation and massproduce normal bulblets from the sections of proliferated shoot clusters in Lilium asiatic hybrid 'Hae Hwa'. The induction of shoot clusters from the culture of bulblet sections was more effective than that of bulb scales on MS medium with 1.0 mg/L BA and 0.5 mg/L IAA. Proliferation of shoot clusters from the formed shoot cluster sections was the most favorable on medium containing 5.0 mg/L BA and 0.5 mg/L IAA. The formation and the growth of bulblets from shoot cluster sections were achieved effectively on medium with 60∼90 g/L sucrose. The leaves derived from shoot clusters grew vigorously but the bulblets from shoot clusters grew very poor in 5L air-lift bioreactor culture. By the addition of 30 mL fresh liquid medium containing doulble strength MS salts, 250 g/L sucrose and 5 g/L activated charcoal after 8 weeks in the shoot cluster culture on MS medium with 5.0 mg/L BA and 0.5 mg/L IAA, the number of bulblets was increased in light condition, but the growth of bulblets was not affected by light. Bulblet production was possible with the bulblet product at 53 to 68 mg in fresh weight by liquid medium addition after the proliferation of shoot cluster.
