Microleakage Assessment of a Pozzolan Cement-based Mineral Trioxide Aggregate Root Canal Sealer (포졸란 시멘트를 기반으로 하는 근관전색제의 치근단부 미세누출 평가)

  • This study aimed to assess microleakage of Endoseal MTA when it is used as a root canal sealer and a root canal filling material compared with conventional endodontic treatment materials Forty-two mature human permanent teeth with a single root canal were divided randomly into three experimental groups (n = 10) and two control groups (n = 6). Group A was obturated with AH $plus^{(R)}$ and gutta-percha (GP). Group E1 was obturated with Endoseal MTA and GP. Group E2 was obturated with Endoseal MTA only. The positive control group was obturated with GP only and the negative control group was obturated in the same way as the experimental groups. The samples were kept in saline solution for 24 hours and were immersed in 0.2% rhodamine B dye solution for 24 hours. Then the samples were split longitudinally and the micoleakage was assessed under a stereomicroscope. Complete microleakage was detected in all positive control group samples, whereas no microleakage was detected in the negative control group. There was no statistically significant difference between the experimental groups in the Kruskal-Wallis test. These results suggest that Endoseal MTA has potential use as a root canal sealer and a root canal filling material.

  • 2,3,7,8-Tetrachlorodibenzo-$\rho$-dioxin(TCDD) displays high toxicity in animals and has been implicated in human carcinogenesis. Although TCDD is recognized as potent carcinogens, relatively little is known about their role in the tumor promotion and carcinogenesis. It is known that TCDD can increase of cancer risk from various types of tissue by a mechanism possibly involving the aryl hydrocarbon receptor (AhR) activation. In this study, effects of TCDD on cellular proliferation of normal human skin and lung fibroblasts, Detroit551 and WI38 cells were investigated. In addition, to enhance our understanding of TCDD-mediated carcinogenesis, we have investigated process in which expression of Erk1/2, cyclinD1, oncogene such as Ha-ras and c-myc, and their cognate signaling pathway. TCDD that are potent activators of AhR-mediated activity was found to induce significant increase of cytochrome P4501A1 mRNA expression, suggesting a presence of functional AhR. These results support that CYP1A1 enzyme may be involved in the generation of TCDD-induced toxicity. Moreover mitogen-activated protein kinases (MARKs) phosphorylation and cyclin D1 overexpression are induced by TCDD, which corresponded with the progression of cellular proliferation. However, TCDD did not affected Ha-ras and c-myc mRNA expression. Taken together, it seems that TCDD are could be a part of cellular proliferation in non-tumorigenic normal human cells such as Detroit551 and WI38 cells through the upregulation of MAPKs signaling pathway regulating growth of cell population. Therefore, AhR-activating TCDD could potentially contribute to tumor promotion and Detroit551 and WI38 cells have been used as a detection system of tumorigenic effects of TCDD.

  • Let x : $M^{n}$ .rarw. $E^{m}$ be an isometric immersion of a manifold $M^{n}$ into the Euclidean space $E^{m}$ and .DELTA. the Laplacian of $M^{n}$ defined by -div.omicron.grad. The family of such immersions satisfying the condition .DELTA.x = .lambda.x, .lambda..mem.R, is characterized by a well known result ot Takahashi (8]): they are either minimal in $E^{m}$ or minimal in some Euclidean hypersphere. As a generalization of Takahashi's result, many authors ([3,6,7]) studied the hypersurfaces $M^{n}$ in $E^{n+1}$ satisfying .DELTA.x = Ax + b, where A is a square matrix and b is a vector in $E^{n+1}$, and they proved independently that such hypersurfaces are either minimal in $E^{n+1}$ or hyperspheres or spherical cylinders. Since .DELTA.x = -nH, the submanifolds mentioned above satisfy .DELTA.H = .lambda.H or .DELTA.H = AH, where H is the mean curvature vector field of M. And the family of hypersurfaces satisfying .DELTA.H = .lambda.H was explored for some cases in [4]. In this paper, we classify space curves x : R .rarw. $E^{3}$ satisfying .DELTA.x = Ax + b or .DELTA.H = AH, and find conditions for such curves to be equivalent.alent.alent.

  • Tin oxide thin films doped with silicon as anodes for lithium secondary battery were fabricated by R. F. magnetron sputtering technique. The electrochemical results for lithium secondary battery anodes showed that addition of silicon decreases the oxidic state of tin, and, hence, reduced the irreversible capacity during the first discharge/charge cycle. The (110),(101),(211) planes were grown with increasing substrate temperatures. The reversible capacity of thin films fabricated in conditions of $300^{\circ}C$ substrate temperature and 7:3 $Ar:O_2$ ratio was 700 mAh/g.

  • The effects of estrogen on apoptosis induced by 2,3,7,8-tetrachlorodibenzo-p-doxin (TCDD) were examined in cultured MCF-7 cells. TCDD stimulated apoptosis and inhibited the expression of bcl-2 gene in MCF-7 cells grown in the media supplemented with 10% fetal bovine serum. However, TCDD failed to induce apoptosis if cells were grown in the media deprived of all estrogen-like compounds. Removal of estrogen-like compounds from the growth media also led to the activation of bcl-2 gene expression in cells treated with TCDD. Combined treatment of estrogen with TCDD abrogated the binding of Aryl hydrocarbon Receptor (AhR)-TCDD complex to Dioxin response element (DRE) of bcl-2 gene leading to the inhibition of bcl-2 gene expression as well as stimulation of apoptosis. The present study suggests that the binding of estrogen receptor (ER)-estrogen complex to the estrogen responsive element (E) interferes with the binding of AhR- TCDD complex to the DRE and inhibits the bcl-2 expression.

Retreatability of two endodontic sealers, EndoSequence BC Sealer and AH Plus: a micro-computed tomographic comparison

  • Objectives: Recently, bioceramic sealers like EndoSequence BC Sealer (BC Sealer) have been introduced and are being used in endodontic practice. However, this sealer has limited research related to its retreatability. Hence, the aim of this study was to evaluate the retreatability of two sealers, BC Sealer as compared with AH Plus using micro-computed tomographic (micro-CT) analysis. Materials and Methods: Fifty-six extracted human maxillary incisors were instrumented and randomly divided into 4 groups of 14 teeth: 1A, gutta-percha, AH Plus retreated with chloroform; 1B, gutta-percha, AH Plus retreated without chloroform; 2A, gutta-percha, EndoSequence BC Sealer retreated with chloroform; 2B, gutta-percha, EndoSequence BC Sealer retreated without chloroform. Micro-CT scans were taken before and after obturation and retreatment and analyzed for the volume of residual material. The specimens were longitudinally sectioned and digitized images were taken with the dental operating microscope. Data was analyzed using an ANOVA and a post-hoc Tukey test. Fisher exact tests were performed to analyze the ability to regain patency. Results: There was significantly less residual root canal filling material in the AH Plus groups retreated with chloroform as compared to the others. The BC Sealer samples retreated with chloroform had better results than those retreated without chloroform. Furthermore, patency could be re-established in only 14% of teeth in the BC Sealer without chloroform group. Conclusion: The results of this study demonstrate that the BC Sealer group had significantly more residual filling material than the AH Plus group regardless of whether or not both sealers were retreated with chloroform.

  • There are growing concerns about human health effects of dioxin and dioxin like compounds such as polychlorinated dibenzo-p-dioxins (PCDDS), polychlorinated dibenzofurans (PCDFs), and polychlorinated biphenyls (PCBs), Earlier studies recognized that 2,3,7,8-tetrachloro dibenzo-p-dioxin (TCDD) and structually related dioxin like compounds invoke a number of common toxic responses that are mediated through a high-affinity cytosolic receptor protein, the AhR. Based on studies that Indicate th pivotal role of AhR in mediating most, if not all, of the dioxin toxicity, TCDD equivalency factor (TEF) approach was developed. This approach allowed the expression of toxic potential of a complex mixture as one integrated parameter, the toxic equivalency (TEQ) value.

In the present study, we have examined the effect of natural estrogen, phytoestrognes and environmental estrogens on the regulation of CYP1A1 gene expression in MCF-7 human breast cancer cell line. that ER and AhR are co-expressed. pCYP1A1 -luc reporter gene was transiently transfected into MCF-7 cells. These cells were treated with various chemicals and then luciferase assay was carried out. 17be1a-estradiol significantly inhibited TCDD stimulated luciferase activity dose dependently and this inhibition was partially recovered by concomitant treatment of tamoxifen. 17beta-estradiol metabolites, 2-hydroxyestradiol and 16alpha-estriol resulted in less potent inhibitory effect than estradiol and synthetic estrogen, diethylstilbestrol (DES) showed no effect on CYP1A1 gene expression. This study demonstrated that estrogen down-regulated TCDD stimulated CYP1A1 expression via ER mediation. And we have found out that several flavonoids such as genistein, kaempferol, daidzein, naringenin, and alkylphenols such as nonylphenol, 4-octylphenol and resveratrol also inhibited TCDD induced CYP1A1 expression like estrogen.

Initial Charge/Discharge of $LiCoO_2$ Composite Cathode with Various Content of Conductive Material for the Lithium ion Battery (리튬이온전지용 $LiCoO_2$정극의 도전재료에 따른 초기 충방전 특성)

  • Initial electrochemical characteristics of $LiCoO_2$ electrode for lithium ion battery with various content of super s black as conductive material were evaluated through the charge/discharge with the potential range of 4.3V to 2.0V versus $Li^+/Li^+$. The rate of C/4 and C/2 by the 3 electrode test cell composed with an electrolytic solution of 1 mol/l $LiPF_6/EC+DEC(1:3\;by\; weight)$. Lithium was used as reference electrode. High impedance charge behavior was observed at early stage of charge. In the case of $3\%w/w$ of super s black as conductive material, the specific resistance of the high impedance releasing was $3.82\;{\Omega}\;{\cdot}\;g-LiCoCo_2$ at the current density of $0.5 mA/cm^2$, which corresponds 7 times of the specific resistance of electrode $(0.728 g-LiCoO_2)$. At second charge, the specific resistance of the high impedance releasing was 63 mn · g-Lico02, which corresponds 12eio of the specific resistance of electrode and only $1.7\%$ of that of the first charge. The first charge and discharge specific capacities at C/4 rate were 160-161 and $153\~155mAh/g-LiCoO_2$, respectively, to lead $95.4\~96.4\%$ of coulombic efficiencies and ca. $6 mAh/g-LiCoO_2$ of initial irreversible specific capacity. Specific resistance at the end of charge and rest showed low value at content of super s black between 2 and $7\%w/w$, which agreed with characteristics of irreversible specific capacity. Capacity densities were reduced by the increasing the content of conductive material. They were 447 and 431mAh/ml when 2 and $2.9\%w/w$ of super s black were used, respectively, at the rate of C/4.
