Analysis of Effect of Environment on Growth and Yield of Autumn Kimchi Cabbage in Jeonnam Province using Big Data (빅데이터를 활용한 재배환경이 전라남도 지방 가을배추의 생육과 수량에 미치는 영향 분석)

  • This study was conducted to evaluate the effect of environment factors on the growth of autumn season cultivation of Kimchi cabbage using the big data in terms of public open data(weather, soil information, and growth of crop, etc.). The growth data and the environment data such as temperature, daylength, and rainfall from 2010 to 2019 were collected. As a result of composing the correlation matrix, the height and leaf number showed high correlation in growing degree days(GDDs) and daylength, and the yield showed negative correlation in growing degree days and the concentration of clay. GDDs and daylength explained about 89% and 84% of variation in height, respectively. These two environmental factors also explained about 85% and 79% of variation in leaf numbers, respectively. In contrast, the coefficient of determination was low for yield when GDDs and concentration of clay was used. The outcome of regional statistical analysis indicated that relationship between yield and sum of sand and silt were high in Haenam and Jindo areas. Hierarchical cluster analysis, which was performed to verify the association of yield, GDDs, and concentration of clay, showed that Haenam and Jindo were clustered together. Although GDDs and yield vary by year and region, and there are regions with similar concentration of clays, observation data are grouped as the result. These suggests that GDDs and soil texture are expected to be related to yield. The cluster analysis results can be used for further data analysis and agricultural policy establishment.

Direct Action of Relaxin on the Pig Mammary Glands to Promote Development of the Alveoli and Remodeling of the Extracellular Matrix (Relaxin Hormone의 직접젇인 작용에 의한 돼지 유선상피세포의 발달과 세포외간 기질의 분해개조 촉진)

  • Our previous studies demonstrated that relaxin in concert with estrogen promotes development of the mammary parenchyma during the last third of gestation in gilts, and the specific relaxin-binding sites were present in the mammary gland. This study was conducted to determine if relaxin-binding sites in the mammary gland were functional relaxin receptors. Three cycling cross-bred gilts were bilaterally ovariectomized on day 0 of the experiment. Beginning on day 15 and continuing through day 29 post-surgery, the gilt received an im. injection of estradiol benzoate at 12-hr intervals. Beginning on day 22 post-surgery, highly purified porcine relaxin was administered(lug/hr) into the left fourth mammary gland from the anterior end via miniature osmotic pump. Physiological saline was administered to the right fourth mammary gland. The gilt was sacrificed on day 29 post-surgery and histological characteristics of the mammary parenchyma were examined. The mammary glands treated locally with saline showed little, if any, lobulo-alveolar development, whereas the mammary glands treated with relaxin showed not only marked lobulo-alveolar development but also prominent secretions in the alveoli. The saline-treated glands were characterized by relatively dense and highly organized collagen fiber bundles. Whereas, in the relaxin-treated mammary glands, collagen fiber bundles were dispersed and loosely organized. In conclusion, relaxin-binding sites in the mammary gland are functional relaxin receptors and relaxin acts directly on the pig mammary gland to promote development of the alveoli and remodeling of the extracellular matrix.

Comparative Functional Analysis of the Malate Dehydrogenase(Mor2) during in vitro Maturation of the Mouse and Porcine Oocytes (체외성숙 과정 중 생쥐와 돼지 난자의 Malate Dehydrogenase(Mor2)의 기능에 대한 비교 분석)

  • Contrast to mouse where its in vitro maturation rates are high without specific supplements or presence of the cumulus cells, there are some species, such as porcine, where its in vitro oocyte maturation rates are still very low. This comparative study was conducted to investigate the role of malate dehydrogenase(Mor2) during oocyte maturation by RNAi in the mouse and porcine. The Mor2 double-stranded RNA(dsRNA) was prepared speciesspecifically and microinjected into the cytoplasm of denuded germinal vesicle(GV) oocytes. Oocytes were cultured for 48 h(porcine) and 16 h(mouse) in M199 with 10% porcine follicular fluid, pyruvate, p-FSH, EGF, cystein, and estradiol-$17{\beta}$. We measured changes in oocyte morphology, maturation rates and mRNA levels after Mor2 RNAi. We confirmed gene sequence-specific knock down of Mor2 mRNA in both species after Mor2 RNAi. In contrast to our previous finding that mMor2 RNAi resulted in GV arrest in the mouse, we found that pMor2 RNAi resulted in MI arrest in denuded porcine oocytes(58%), but developed to MII(84.4%) in COCs. To determine whether this difference between mouse and porcine RNAi is due to differences in culture media, we cultured mouse oocytes in the M199 media for 16 h after mMor2 RNAi. Mouse oocytes were developed to MII stage(62%) and there was no statistical difference compared to that of non-injected(76.8%) and buffer-injected(73.3%) control groups. Therefore, we concluded that the mouse and porcine oocytes are having different metabolic systems in relation to malate dehydrogenase for oocyte maturation. This could be a basis for differences in maturation rates in vitro in two species. Further scrutinized studies on the metabolic pathways would led us in finding better culture system to improve oocyte maturation rates in vitro, especially in more challenging species like the porcine.

Characterization Analysis for Cardiogenic Potential of Three Human Adult Stem Cells (세 종류 줄기세포의 특성 분석과 지방유래 줄기세포의 심근세포로의 분화)

  • In the present study, we isolated three human adult stem cells including adipose tissue-derived stem cells(HAD), umbilical cord-derived stem cells(HUC), and amnion-derived stem cells(HAM) and analysed their characteristics. And we examined whether HAD could be used as therapeutical cells for the heart diseases. Both HAM and HUC appeared very similar morphology but HAD was different. Doubling time of HUC was most fast, but total doubling numbers of HUC was same with HAM. Total doubling numbers of HAD was much more than others. Expression patterns of genes and proteins of three human adult stem cells were very similar. Also they were differentiated into adipocytes, osteocytes, and chondrocytes. In addition, they expressed many cardiomyocyte-related genes. But expression pattern of genes is a little different. When HAD were cultivated in the presence or absence of various combinations of BMP and FGF after 5-azacytidine expose for 24 h, expression of Cmlc-1, and ${\alpha}1c$ genes was significantly increased. However, expression of troponin T, troponin I and Kv4.3 genes was not changed. Based on these observations, it is suggested that HAD, HUC, and HAM might be used as potentially therapeutical cells for clinical application.

Diagnosis and management of the early defoliation of Korean flowering cherry in Gurye (구례 왕벚나무 가로수의 조기낙엽 진단 및 친환경적 관리 방법)

  • The Korean flowering cherry is a popular tree. However, the trees have started to defoliate early, including those in Gurye. Thus, it is necessary to identify the causes of the early defoliation and how to manage them. Therefore, the purpose of this study was threefold: 1) to investigate the early defoliation of flowering cherry trees, 2) to identify the differences in growth and flowering of the trees in response to treatment, and 3) to suggest an optimal treatment for the trees. The experiment was conducted in Gurye at a site 3km long with 102 flowering cherry trees along a street. There were three treatments: control, an environmentally friendly insecticide and a disinfectant(treatment 1), and an environmentally friendly insecticide, a disinfectant, and irrigation (treatment 2). The trees in Gurye were compared to trees on Jeju Island. The defoliation rates of the flowering cherry in Gurye were significantly higher than those on Jeju Island at each measurement time. Within Gurye, the defoliation rate was significantly higher in the trees of the control than in the two treatments and only the trees in the control from Gurye had shot-hole disease. Post-treatment, twigs collected from the control during April showed significantly poorer growth, lower numbers of flowering buds per twig, and lower numbers of foliar buds. The trees given treatment 1 showed significantly greater growth, number of total buds, flowering buds per twig, and density of buds than the trees given treatment 2. After analyzing the correlations, it was established that early defoliation led to poorer growth, affecting the flowering of the trees.

Contents of Sodium and Potassium for Restaurant Dishes in Seoul (서울지역 음식점 메뉴의 나트륨 및 칼륨 함량 비교)

  • To determine the contents of sodium (Na) and potassium (K) for restaurant dishes, 194 cases classified 26 kinds of restaurant dishes were collected from restaurants in Seoul and were analysed using the atomic absorption spectroscopy. Average contents of Na and K in Kimchi among restaurant dishes were highest as $554.9{\pm}155.5mg/100g$, $225.1{\pm}69.1mg/100g$, while average contents of Na and K were lowest in Seolleongtang as $24.2{\pm}5.8mg/100g$, $12.7{\pm}3.9mg/100g$ respectively. The contents between Na and K in restaurant dishes had a strong correlation of positive linear judging from 0.782 (p-value < 0.01) of the correlation coefficient. The Na/K ratio of Bulgogi was appropriate as $1.17{\pm}0.44$, while those of Udon, banquet noodles, Kalguksu were very high as shown more than 10. The contents of Na and K were examined 5 dish groups as Gook/Tang, Jjigae/Jeongol, stir-fried dish, noodle and Kimchi. Average contents of Na and K classified by dish groups were significantly difference (p-value < 0.001). Average content of Na among dish groups was highest in Kimchi as $554.9{\pm}155.5mg/100g$ followed by Jjigae/Jeongol, Gook/Tang, stir-fried dish and noodle. Similar to Na content, average content of K was highest in Kimchi as $225.1{\pm}69.1mg/100g$ followed by stir-fried dish, Jjigae/Jeongol, Gook/Tang and noodle. The ratio of Na/K by classified groups of dishes was shown as significantly difference (p-value < 0.001). Stir-fried dish was $1.18{\pm}0.44$ close to 1 followed by Kimchi, Jjigae/Jeongol, Gook/Tang, and noodle.

Development and Validation of an Analytical Method for Determination of Fungicide Tridemorph in Agricultural Commodities Using LC-MS/MS (LC-MS/MS를 이용한 농산물 중 살균제 tridemorph의 시험법 개발 및 검증)

  • The purpose of this study was developed for the determination of tridemorph in agricultural commodities samples. Tridemorph residues in samples were extracted with acetonitrile, partitioned with saline water, and then purified using and aminopropyl ($NH_2$) SPE catridge. The purified samples were quantified and confirmed via liquid chromatograph-tandem mass spectrometer (LC-MS/MS) in positive ion mode using multiple reaction monitoring (MRM). Matrix-matched calibration curves were linear over the calibration ranges (0.005~2.5 ng) into a blank extract with $r^2$ > 0.999. The limits of detection and quantification were 0.001 and 0.005 mg/kg, respectively. The average recovery ranged between 75.9% and 103.7% at different concentration levels (LOQ, 10 LOQ, 50 LOQ, n = 5) with relative standard deviations (RSDs) less than 9.0%. An interlaboratory study was conducted to validate the method by Korea Advanced Food Research Institute. The average recovery ranged between 87.0% and 109.2% at different concentration levels (LOQ, $10{\times}LOQ$, $50{\times}LOQ$, n = 5) with relative standard deviations (RSDs) less than 8.0%. All values were consistent with the criteria ranges requested in the Codex guidelines (CAC/GL40, 2003) and Food Safety Evaluation Department guidelines (2016). The results prove that the developed analytical methods is accurate, effective and sensitive for tridemorph determination.

Development and Validation of an Analytical Method for Determination of Fungicide Benzovindiflupyr in Agricultural Commodities Using LC-MS/MS (LC-MS/MS를 이용한 농산물 중 살균제 벤조빈디플루피르의 잔류시험법 개발 및 검증)

  • Benzovindiflupyr is a new pyrazole carboxamide fungicide that inhibits succinate dehydrogenase of mitochondrial respiratory chain. This study was carried out to develop an analytical method for the determination of benzovindiflupyr residues in agricultural commodities using LC-MS/MS. The benzovindiflupyr residues in samples were extracted by using acetonitrile, partitioned with dichloromethane, and then purified with silica solid phase extraction (SPE) cartridge. Correlation coefficient ($r^2$) of benzovindiflupyr standard solution was 0.99 over the calibration ranges ($0.001{\sim}0.5{\mu}g/mL$). Recovery tests were conducted on 5 representative agricultural commodities (mandarin, green pepper, potato, soybean, and hulled rice) to validate the analytical method. The recoveries ranged from 79.3% to 110.0% and then relative standard deviation (RSD) was less than 9.1%. Also the limit of detection (LOD) and limit of quantification (LOQ) were 0.0005 and 0.005 mg/kg, respectively. The recoveries of interlaboratory validation ranged from 83.4% to 117.3% and the coefficient of variation (CV) was 9.0%. All results were followed with Codex guideline (CAC/GL 40) and Ministry of Food and Safety guideline (MFDS, 2016). The proposed new analytical method proved to be accurate, effective, and sensitive for benzovindiflupyr determination and would be used as an official analytical method.

Effects of Storage Temperature and Grain Moisture Content on the Contaminaton of Fusarium and Fusariotoxin in Hulled Barley Grains (겉보리의 저장온도와 수분함량이 붉은곰팡이병균과 곰팡이독소 오염에 미치는 영향)

  • Fusarium graminearum is a mycotoxigenic plant pathogen, which could remain in harvested barley grains and produces mycotoxins at preferable conditions during storage. To elucidate the factors affecting contamination of Fusarium and Fusariotoxin in hulled barley during storage, three hulled barley grain samples were collected from Jeolla province. Moisture content of each sample was adjusted to 14% and 20%, respectively, then stored in two warehouses where temperature was controlled differently: one controlled below $12^{\circ}C$, and the other with no control. While monitoring temperature and relative humidity of warehouses hourly, grain moisture content, Fusarium occurrence, and mycotoxin level was analyzed at 1, 3, 6, and 12 month after storage. The average monthly temperature and relative humidity ranged $3{\sim}29^{\circ}C$, and 58~70% in warehouse without temperature control, whereas $3{\sim}13^{\circ}C$ and 62~74% in warehouse controlled below $12^{\circ}C$. Grain moisture content of the samples decreased in both warehouses except 14% samples which increased in the warehouse with temperature control. Fusarium frequency of the contaminated grains decreased continuously in the warehouse without temperature control. But in the warehouse below $12^{\circ}C$, Fusarium decreasing rate was slower because of high grain moisture content. In most samples, nivalenol was detected more in the warehouse without temperature control after 12 month but there was little difference after 1, 3, and 6 month. Therefore, it will be efficient to store hulled barley in the warehouse controlled below $12^{\circ}C$ to reduce Fusarium contamination when the barley is not dried properly. In addition, when storage period exceeds 12 month, it is recommended to store hulled barley in a warehouse controlled below $12^{\circ}C$ to reduce nivalenol contamination.

Frameless Fractionated Stereotactic Radiaton Therapy in Recurrent Head & Neck Cancers (국소재발된 두경부종양의 무고정틀 정위적 분할방사선치료)

  • Background & Objectives: Frameless fractionated stereotactic radiotherapy(FFSRT) is a modification of stereotactic radiosurgery(SRS) with radiobiologic advantage of fractionation without losing mechanical accuracy of SRS. Local recurrence of head and neck cancer at or near skull base benefit from reirradiation. Main barrier to successful palliation is dose limitation secondary to normal tissue tolerance. We try to evaluate the efficacy and safety of FFSRT as a new modality of reirradaton in these challenging patients. Materials & Methods: Seven patients with recurrent head & neck cancer involving at or near skull base received FFSRT from September 1995 to November 1997. Six patients with nasopharyngeal cancer had received induction chemotherapy and curative radiation therapy. One patient with maxillary sinus cancer had received total maxillectomy and postoperative radiation therapy as a initial treatment. Follow-up ranged from 11 to 32 months with median of 24 months. Three of 7 patients received hyperfractionated radiation therapy(1.1-1.2Gy/fraction, bid, total 19.8-24Gy) just before FFSRT. All patients received FFSRT(3-5Gy/fraction, total 15-30Gy/5-10fractions). Chemotherapy(cis-platin $100mg/m^2$) were given concurrently with FFSRT in four patients. Second course of FFSRT were given in 4 patients with progression or recurrence after initial FFSRT. Because IF(irregularity factor; ratio of surface area of target to the surface area of sphere with same volume as a target) is too big to use conventional stereotactic RT using multiple arc method for protection of radiation damage to critical normal tissue, all patients received FFSRT with conformal method using irregular static ports. Results: Five of 7 patients showed complete remission in follow-up CT &/or MRI. Three of these five patients who developed marginal, in-field, and out-field recurrences, respectively. Another one of complete responders has been dead of G-I bleeding without evidence of local recurrence. One partial responder who showed progressive disease 15 months after initial FFSRT has received additional FFSRT, and then he is well-being with symptomatic improvement. One minmal responder who showed progression of locoregional disease 9 months after $1^{st}$ FFSRT has received 2nd FFSRT, and then he is alive with stable disease. Five of 7 case had showed direct invasion to skull base and had complaint headache and various symptoms of cranial nerve involvement. Four of these five case showed improvement of neurologic symptoms after FFSRT. No significant neurologic complicaltion related to FFSRT was observed during follow-up periods. Tumor volumes were ranged from 3.9 to 50.7 cc and surface area ranged from 16.1 to $114.9cm^2$. IF ranged from 1.21 to 1.74. The average ratio of volume of prescription isodose shell to target volume was 1.02 that indicated the improvement of target coverage and dose distribution with FFSRT with conformal method compared to target coverage with FFSRT with multiple arc method. Conclusion: Our initial experience suggests that FFSRT with conformal method was relatively effective and safe modality in the treatment of recurrent head and neck cancer involving at or near skull base. Treatment benefit included good palliation of symptoms and reasonable radiographic response. However, more experience and additional follow-up are needed to better assess its ultimate role in treating these challenging patients.
