Studies on meat color, myoglobin content, enzyme activities, and genes associated with oxidative potential of pigs slaughtered at different growth stages

  • Objective: This experiment investigated meat color, myoglobin content, enzyme activities, and expression of genes associated with oxidative potential of pigs slaughtered at different growth stages. Methods: Sixty 4-week-old $Duroc{\times}Landrace{\times}Yorkshire$ pigs were assigned to 6 replicate groups, each containing 10 pigs. One pig from each group was sacrificed at day 35, 63, 98, and 161 to isolate longissimus dorsi and triceps muscles. Results: Meat color scores were higher in pigs at 35 d than those at 63 d and 98 d (p<0.05), and those at 98 d were lower than those at 161 d (p<0.05). The total myoglobin was higher on 161 d compared with those at 63 d and 98 d (p<0.05). Increase in the proportions of metmyoglobin and deoxymyoglobin and a decrease in oxymyoglobin were observed between days 35 and 161 (p<0.05). Meat color scores were correlated to the proportion of oxymyoglobin (r = 0.59, p<0.01), and negatively correlated with deoxymyoglobin and metmyoglobin content (r = -0.48 and -0.62, p<0.05). Malate dehydrogenase (MDH) activity at 35 d and 98 d was higher than that at 161 d (p<0.05). The highest lactate dehydrogenase/MDH ratio was achieved at 161 d (p<0.05). Calcineurin mRNA expression decreased at 35 d compared to that at 63 d and 98 d (p<0.05). Myocyte enhancer factor 2 mRNA results indicated a higher expression at 161 d than that at 63 d and 98 d (p<0.05). Conclusion: Porcine meat color, myoglobin content, enzyme activities, and genes associated with oxidative potential varied at different stages.

Isolation and Physiological Characteristics of Microorganisms Producing Extracellular Enzymes from Korean Traditional Soybean Sauce and Soybean Paste (전통 장류에서 세포외효소 분비능이 우수한 미생물의 분리 및 생리활성 특성)

  • We isolated microorganisms presenting high enzymatic activities for amylase, cellulase, protease, lipase or fibrinolysis from Korean traditional soybean sauce and paste. Then, the physiological properties and 16S rRNA sequences of isolated microorganisms were analyzed. All of the isolated 13 strains possessing high extra cellular enzyme activities have higher amylase and cellulase activities than Bacillus subtilis KACC 10114. All the selected strains have protease activities except for D2-14. Except D8-8 and K4-1, other strains have lipase activity. D2-7, D8-8 and K4-1 strains have higher fibrinolytic activities than others, while D8-2 strain has no activity. Most of the selected strains showed antibacterial activity even in gram positive and gram negative bacteria and yeast. Gene sequence analysis of 16S rRNA from isolated strains revealed that all the selected strains were member of Bacillus species.

  • A gene cluster for cyclohexanone oxidation was cloned from Rhodococcus sp. TK6, which is capable of growth on cyclohexanone as the sole carbon source. The 9,185-bp DNA sequence analysis revealed seven potential open reading frames (ORFs), designated as ssd-chnR-chnD-chnC-chnB-chnE-partial pcd. The chnBCDE genes encode enzymes for the four-step conversion of cyclohexanone to adipic acid, catalyzed by cyclohexanone monooxygenase (ChnB), $\varepsilon-caprolactone$ hydrolase (ChnC), 6-hydroxyhexanoate dehydrogenase (ChnD), and 6-oxohexanoate dehydrogenase (ChnE). Furthermore, the presence of a regulatory element in the downstream region of the chnD gene supports the notion that chnR is a putative regulatory gene. Among them, the activity of ChnB was confirmed and characterized, following their expression and purification in Escherichia coli harboring the modified chnB gene (chnB gene with 6 successive codons for His at the 3' terminus).

Analyses on the Status of Eggshell Breakage in the Step of Egg Conveyer System of the Laying Hen House (산란계사의 계란 이송단계에 따른 파란발생 실태분석)

  • Broken eggs not only cause the economic loss in the egg producing industry, but also may not be good for the consumer's health. In order to identify the causes of eggshell breakage in laying hen houses, analyses on the status of eggshell breakage in the step of egg conveyer system of the laying hen house were conducted. The survey of eggshell breakage rate was implemented by four different stages egg conveying system. In the results, the breakage rate showed 4.7% in the egg collection belt, 2.5% in the egg elevator, 1.7% in the egg grading system and 0.9% in the egg conveyor belt. In particular, to Hy-line Brown variety, as the hens' raising week old increased, the breakage rate of eggshell increased while the strength of eggshell decreased. In the regression analysis between the breakage rate of eggshell and the strength of eggshell, the coefficient of determination ($R^2$) by the test of significance was computed as 0.7, therefore the changes of the strength of eggshell affects the breakage rate of eggshell. However, between the strength of eggshell and thickness of egg shell, and between the strength of eggshell and strain of eggshell, there was no significant difference detected in the regression analysis.

Occurrence of Virus Diseases on Major Crops in 2008 (2008년 우리나라 주요 작물 바이러스병 발생 상황)

  • Viruses diagnosed on crops including rice plants from farmers or agricultural extension agencies cover the country were 11 species including Broad bean wilt virus 2 (BBWV2) in 2008. Tomato spotted wilt virus (TSWV) was the most important virus having the detection rate of 22.9%. Two viruses of Tomato yellow leaf curl virus (TYLCV) and Tobacco leaf curl virus (TLCV) inducing leaf yellow and curl diseases on tomatoes were occurred newly with the detection rate of 12.2% and 4.0%, respectively, in 2008. Rice stripe virus (RSV) was occurred on 869.5 ha mainly at Jindo and Haenam areas in Jeollanamdo province. At Jindo area, 12 plots were damaged severely with the infected hill rate of 83.8%. At the main production area of oriental melon at Seongju, almost all fruits from whole sale market at Seongju were infected with Cucumber green mottle mosaic virus (CGMMV) as the detection rate of 87%. The areas occurred TSWV in Korea were 25 totally from 2003 including 7 areas newly reported in 2008 including Naju in Jeoallanamdo. TSWV could be reduced as 0.1 % from 5.3% by covering insect proof net in vinyl house after chemical soil sterilization. Tomato yellow leaf curl disease was occurred on April in 2008 at Tongyoung area in Kyeongsangnamdo, and detected continuously at 13 areas, 7 in Kyeongsangnamdo, 4 in Jeollabukdo and 2 in Jejudo. Potato spindle tuber viroid (PSTVd) was occurred abruptly in a confined space of a civil breeding greenhouse and a cultivar evaluation field followed by disuse 17.4 M/T of potato tubers. No PSTVd was detected at 17 fields cultivated the related potatoes to the bred company by RT-PCR.

Neodothiora pruni sp. nov., a Biosurfactant-Producing Ascomycetous Yeast Species Isolated from Flower of Prunus mume

  • A yeast strain, designated as JAF-11T, was isolated from flower of Prunus mume Sieb. et Zucc. in Gwangyang, Republic of Korea. Phylogenetic analysis showed that strain JAF-11T was closely related to Neodothiora populina CPC 39399T with 2.07 % sequence divergence (12 nucleotide substitutions and three gaps in 581 nucleotides) in the D1/D2 domain of the large subunit (LSU) rRNA gene, and Rhizosphaera macrospora CBS 208.79T with 4.66 % sequence divergence (25 nucleotide substitutions and five gaps in 535 nucleotides) in the internal transcribed spacer (ITS) region. Further analysis based on the concatenated sequences of the D1/D2 domain of the LSU rRNA gene and the ITS region confirmed that strain JAF-11T was well-separated from Neodothiora populina CPC 39399T. In addition to the phylogenetic differences, strain JAF-11T was distinguished from its closest species, Neodothiora populina CPC 39399T and Rhizosphaera macrospora CBS 208.79T belonging to the family Dothioraceae by its phenotypic characteristics, such as assimilation of carbon sources. Hence, the name Neodothiora pruni sp. nov. is proposed with type strain JAF-11T (KACC 48808T; MB 850034).

Investigation of Optimal ionic Concentration of Nutrient Solution for the Water Culture of Young Welsh Onion (실파의 수경재배에 적합한 양액농도 구명)

  • The purpose of this experiment was to investigate optimal ionic concentration of nutrient solution for water culture of young welsh onion (Allium fistulosum). For the purpose of clarification of optimal nutrient concentration to maximize growth of young welsh onion, different nutrient concentrations of Yamazaki's solution for welsh onion seedling $(NO_3^--N\;9.0,\;NH_4^+-N\;3.0,\;PO_4^{3-}-P\;6.0,\;K^+7.0,\;Ca^{2+}\;2.0,\;Mg^{2+}\;2.0,\;and\;SO_4^{2-}-S\;4.4me{\cdot}L^{-1})$ which selected by prior experiment were treated as 0.6, 1.2, 1.8, and $2.4dS{\cdot}m^{-1}$. Increments of fresh weight, dry weight and top length were the highest in 1.2 and, in the next, were placed by the order of 1.8, 2.4, and $0.6dS{\cdot}m^{-1}$ The regression coefficients for the maximal growth of fresh weight of cv. 'Geurnjanguedaepa' and 'Tokyokuro' were $y=-42.091x^2+171.79x+11.047 (R^2=0.8946,\; R=0.9458^*)\;and\;y=-50.069x2+157.58x+15.414(R^2=0.9343,\;R=0.9692^{**})$, respectively, and optimal EC levels according to regression coefficients were 1.68 and $1.57dS{\cdot}m^{-1}$. As the conclusions, optimal nutrient levels far young welsh onion were $1.2dS{\cdot}m^{-1}$ EC in the early growth stage and $1.6\~l .7dS{\cdot}m^{-1}$ in the later growth stage.

Gear Rating and Contact Pattern Analysis for Rotavator Gearbox Using Actual Working Load (실 작업 부하를 이용한 로타베이터 기어박스의 강도 평가와 치면 접촉 패턴 해석)

  • The rotavator is attached to the three-point hitch at the rear of a tractor and uses the power take-off strength of the tractor to perform soil harrowing. During operation, the power transmitted to the gearbox of the rotavator varies with the soil characteristics and depth. These properties influence the reliability of the gearbox. In this study, actual load measurements and analyses were performed using a rotavator. In addition, the safety factor and fatigue life of the gearbox components were determined using the analysis results. Through analysis and tests, the contact pattern of the gear tooth surface was identified. The input power values of the gearbox were minimum and maximum at 54.5% and 84.5% of the tractor power, respectively. Based on the actual load analysis results, the strength and fatigue life of the gearbox components were satisfied. In addition, through the analysis and testing of the gear contact pattern, it was confirmed that a similar contact occurred. Through the analysis, the magnitude of the load acting on the tooth surface of the gear was confirmed.

Diversity of Butyrivibrio Group Bacteria in the Rumen of Goats and Its Response to the Supplementation of Garlic Oil

  • This study aimed to investigate the diversity of the Butyrivibrio group bacteria in goat rumen and its response to garlic oil (GO) supplementation as revealed by molecular analysis of cloned 16S rRNA genes. Six wethers fitted with ruminal fistulas were assigned to two groups for a cross-over design with 28-d experimental period and 14-d interval. Goats were fed a basal diet without (control) or with GO ruminal infusion (0.8 g/d). Ruminal contents were used for DNA extraction collected before morning feeding on d 28. A total bacterial clone library was firstly constructed by nearly full-length 16S rRNA gene cloned sequences using universal primers. The resulting plasmids selected by Butyrivibrio-specific primers were used to construct a Butyrivibrio group-specific bacterial clone library. Butyrivibrio group represented 12.98% and 10.95% of total bacteria in control and GO group, respectively. In libraries, clones were classified to the genus Pseudobutyrivibrio, Butyrivibrio and others within the family Lachnospiraceae. Additionally, some specific clones were observed in GO group, being classified to the genus Ruminococcus and others within the family Ruminococcaceae. Based on the criterion that the similarity was 97% or greater with database sequences, there were 29.73% and 18.42% of clones identified as known isolates (i.e. B. proteoclasticus and Ps. ruminis) in control and GO groups, respectively. Further clones identified as B. fibrisolvens (5.41%) and R. flavefaciens (7.89%) were specifically found in control and GO groups, respectively. The majority of clones resembled Ps. ruminis (98% to 99% similarity), except for Lachnospiraceae bacteria (87% to 92% similarity) in the two libraries. The two clone libraries also appeared different in Shannon diversity index (control 2.47 and GO group 2.91). Our results indicated that the Butyrivibrio group bacteria had a complex community with considerable unknown species in the goat rumen.
