Effects of Seaweed Extracts and Corn Starch on the Characteristics of Acorn Mooks (해조류와 옥수수 전분의 첨가가 도토리묵의 물성에 미치는 영향)

  • The physical and processing properties of acorn jelly were investigated to see the effect of polysaccharides in seaweed extract and corn starch. The yield of acorn jelly added starch concentration was slightly increased when the concentration was added more. However, moisture content and color had no significant changes with concentration. In the case of acorn jelly added sea tangle, color of acorn jelly was darker, but color of the jelly with carrageenan was lighter. According to the concentration of seaweed extract, the hardness was increased as concentration was added. The texture of acorn Jelly added agar had the highest binding. In the case of acorn jelly added corn starch, there were no significant changes, but this binding was stronger than the acorn jelly added extract of marine algae. In the physical and sensory properties of acorn jelly with corn starch, the ideal mixture ratio between the acorn jelly and the corn starch was 6:4.

  • The objective of this study is to investigate the effect of Codonopsis lanceolata Bentk et Hook on the growth of lactic acid bacteria and some harmful bacteria. Extract of C. lanceolata was obtained by hydrothermal extraction method and used intactly without concentration. To investigate the inhibitory effect on the growth of harmful bacteria, we used agar dilution method. On agar plate containing more than 25% extract, they showed decreased number of colonies.   • This study examied ned physical properties and surface characteristics of papers post-processed by Jumchi technique and stainability of extract from Amur Cork-Tree to use Dakji as clothing material. Then, it found the following results. After-treatment by Amorphophalus konjac K. Koch and persimmon juice showed stronger tensile strength and tearing strength than untreated samples and the drape stiffness was substantially increased. After-treatment by persimmon juice generated dyeing effects as well as excellent tensile strength and tearing strength all samples were generally dyed well by Amur Cork-Tree, While the untreated samples did not show any significant effects in dyeing for more than 5 minutes. It was found that after-treatment substantially contributed on concentration of dyeing as dyeing amounts of post-processed samples were gradually increased as time increased. After-treated sample with agar did not show any significant differences from untreated sample. Accordingly, it has to be studied further.

  • To enhance the shoot induction efficiency from nodal stem of yooza, the culture conditions such as basal medium, carbohydrate source, solidifying agent and the optimum concentration of plant growth regulators for shoot induction were investigated. The nodal explants were cultured better on MS medium than on MT, SH, B5 or W media in considering of shoot induction rate and mean shoot length. Solidifying agent in medium was better with 0.8% agar than with 0.3% agar, 1.2% agarose or 0.2% gelrite. Carbohydrate source in shoot induction medium was efficient with 30 g/L sucrose. The optimum concentrations of plant growth regulators were determined that 0.1 mg/L NAA as auxin was effective on the shoot induction, and 1.0 mg/L BAP as cytokinin induced multiple shoots efficiently. Shoot induction was the most effective on MS medium supplemented with 4 mg/L $GA_3$ in yooza.

Bacterial Color Response to Hexavalent Chromium, $Cr^{6+}$

  • A blue pigment-producing bacterium, Vogesella indigofera, was isolated and quantified for the relationship between its synthesis of a blue pigment and exposure concentrations of $Cr^{6+}$. The concentration of $Cr^{6+}$ and the percentage of blue colonies on agar plates was negatively correlated ($r^{2}$ =-0.8683). Critical concentrations inhibiting bacterial pigment production were found to be between 100-150 $\mu\textrm{g}$ $Cr^{6+}$/ml on agar plates and 200-300 $\mu\textrm{g}$ $Cr^{6+}$/ml in liquid culture. As the blue color is characteristic and easily observable, the bacterium Vogesella indigofera may have potential applications in the detection and monitoring of environmental pollution.

  • The fungistatic effects of extract and crystal (A) of Berberis Koreans Palibin, a common shrub in Korea, were obeserved and compared with undecylenic acid and vegetable oil. In vitro studies, the spores of fungi were inoculated on Sabouraud's glucose agar media which contained compounds of various concentration, and the growth of the fungi was observed for 3 weeks. The species of the fungi used in these experiments were Epidermophyton floccosum, Microsporum gypseum, Microsporum audouini, Microsporum canis, Microsporum nanum, Microsporum cookei, Trichophyton rubrum, Trichophyton mentagrophytes, Trichophyton tonsurans and Trichophyton verrucosum. The results of the studies were as follows: 1. The growth of E. floccosum, M. audouni, M. canis, M. nanum, M. cookei, was inhibited in Sabouraud's glucose agar media containing extract and crystal (A) of Berberis Koreana Palibin 1 mg/ml.

Study on Antimicrobial Activity of Extracts from Fritillaria unibracteata Hsiao et K.C. Hsia and F. ussuriensis Maxim.

  • Antimicrobial activities of methanol, ethanol, water, and $CH_2Cl_2$ extracts from Fritillaria unibracteata Hsiao et K.C. Hsia and F. ussuriensis Maxim. were investigated by disk-agar diffusion method. The result showed comparatively strong antimicrobial activity against several microorganisms. The extracts from F. unibracteata and F. ussuriensis dosedependently increased the activity. However, water and $CH_2Cl_2$ extracts showed no antimicrobial activity against 7 microorganisms. Especially, against the most sensitive microorganism Staphylococcus epidermidis, methanol extracts at highest concentration of 20 mg/mL exhibited the largest clear zone on plate by 6-12 mm and ethanol extracts on plate by 6-10 mm.

RELATIONSHIP OF THE BACTERIA INHIBITING PLAQUE FORMATION AND THE NUMBER OF ORAL BACTERIA (치태형성 억제세균과 구강내 세균수와의 관계)

  • The inhibition degree of the isolated bacteria on plaque formation of Streptococcus mutans, and the effect of these bacterial genus on the concentration of total bacteria in saliva were assessed with the following. The effectiveness of the isolated bacteria on the inhibition of plaque formation was assessed culturing Streptococcus mutans in the beaker with orthodontic wires. The mean weight of plaque produced on a wire was 152mg in the culture of Streptococcus mutans only, whereas being reduced to 4mg, 78mg, or 72mg in the combined culture of Streptococcus mutans and Enterococcus durans, Lactobacillus acidophilus, or Streptococcus oralis. The colony forming units (CFU) of Streptococcus mutans were $3.6{\times}10^8$ per ml in the culture of Streptococcus mutans, only, wheras being $1.4{\times}10^6,\;5.6{\times}10^6,\;or\;3.8{\times}10^6$ per ml in the combined culture of Streptococcus mutans and Enterococcus durans, Lactobacillus acidophilus, or Streptococcus oralis. When saliva from children was inoculated on brain heart infusion agar, the colony forming units of bacteria were $4.8{\times}10^6\;to\;1.3{\times}10^9$ per ml of saliva. The concentration of Enterococcus, Lactobacillus, or Streptococcus inhibiting Streptococcus mutans in saliva was not proportioned to that of total bacteria replicated on brain heart infusion agar. These results indicate that the isolated bacteria inhibited the replication of Streptococcus mutans, resulting into inhibiting the formation of plaque, but the concentration of Enterococcus, Lactobacillus, or Streptococcus inhibiting Streptococcus mutans, in saliva might not affect the total bacterial concentration of saliva.

Occurrence of Syzygites Megalocarpus on Wild Mushrooms and Its Cultural Characteristics (야생(野生)버섯의 기생균(寄生菌)인 이분지털곰팡이의 출현(出現)과 배양(培養) 특성(特性))

  • Syzygites megalocarpus occurred on the fruit bodies of Tricholoma matsutake, Leccinum rugosiceps, Pulveroboletus ravenelii, Russula emetica, and Amanita pseudoporphyria in the field. We could select Malt Extract Agar, Mueller Hinton Medium and Potato Dextrose Agar as optimal media among eight media. Syzygites megalocarpus showed an optimal temperature around $23^{\circ}C$ with optimal pH 6.0. Growth of S. megalocarpus on PDA was inhibited by 36.5% at 5% NaCl compared with without NaCl and did not grow at more than 8% NaCl in 2 days after inoculation. However, it grew 1.1 cm in 10% NaCl in 5 days after inoculation. Growth of Tricholoma matsutake on PDA was inhibited by increasing contration of NaCl and did not nearly grow at 2.5% NaCl in 60 days after inoculation. Because S. megalocarpus grew at high concentration of NaCl, we concluded that NaCl should not use for controlling S. megalocarpus on the fruit body of T. matsutake.

Effect of Medium Component on Plant Regeneration via Adventitious Bud Formation from Leaf Explant Cultures of Strawberry (Fragaria ananassa Duch.) (딸기 (Fragaria ananassa Duch.) 잎 절편체 배양으로부터 부정아 형성을 통한 식물체 재생)

  • This study was investigated to establish a regeneration system of plant via adventitious bud formation from leaf explant cultures of strawberry (Fragaria ananassa Duch). Effects of plant growth regulators (2,4-D, BAP), agar sucrose and myo-inositol on adventitious bud formation were investigated. When the leaf explants were cultured on MS medium supplemented with 0.1 mg/L 2,4-D and 3 mg/L BAP, the adventitious bud formation was most promoted. The adventitious bud formation was not induced from leaf explants cultured on MS medium containing 2,4-D alone. Adventitious bud formation was enhanced to almost 3 times on medium with low level of agar concentration (0.4%) in comparison with those on the medium with high level of agar (1%), but almost of shoot was vitrificated on the medium. Therefore, the normal adventitious bud formation from leaf explants was most effective on the medium containing 0.05 mg/L 2,4-D,1 mg/L BAP, 0.8% agar, 30 g/L sucrose and 100 mg/L myo-inositol. Thus, the mass propagation of healthy strawberry could be established using leaf explants.
