  • In this study, the composition and characteristics of poly-$\beta$-hydroxybutyrate (PHB) biosynthesized by Actinobacillus sp. EL-9 are investigated. PHB produced by Actinobacillus sp. EL-9 was identified as the homopolymer of 3-hydroxy-butyric acid (PHB) by infrared spectroscopy and nuclear magnetic resonance spectroscopy analysis. The melting tem-perature (T$_{m}$), and crystallization temperature(T$_{c}$) of PHB was 169.7$^{\circ}C$ and 69.13$^{\circ}C$, respectively. The viscosity on he basis of Brookfield viscometer was 6.01 ㎗/g. The viscosity-average molecular weight estimated by Mark-Ho-wink-Sakurada equation was 1.08$\times$10$^{6}$ ($\pm$3,000).00).

  • Microorganisms growing alcoholic distillery wastes were isolated from soil. Of them, the selected strain EL-9 had a capability of accumulating poly-$\beta$-hydroxybutyrate (PHB) when grown in alcoholic distillery wastes. The strain EL-9 was identified as a genus Actinobacillus based on the morphological, cultural, and biochemical characteristics. The strain EL-9 was named temporarily as Actino- bacillus sp. EL-9. The optimal temperature and pH for cell growth of Actinobacillus sp. EL-9 were 30$\circ$C and 7.0, respectively. The optimal medium compositions for cell growth comprised glucose 2%, NH$_{4}$NO$_{3}$ 0.15%, Na$_{2}$HP0$_{4}$-12H$_{2}$O 0.25%, KH$_{2}$PO$_{4}$ 0.25%, MgSO$_{4}$4-7H$_{2}$O 0.15%, FeSO$_{4}$-7H$_{2}$O 0.005%, CaCl$_{2}$-2H$_{2}$O 0.003% and trace element solution 5m/l. To investigate the optimal conditions for PHB production, two-stage culture technique was used. The result showed that the optimal conditions for PHB production are identical those for cell growth. When propionic acid was added as a cosubstrate, PHB/HV copolymer was produced.

  • Alcoholic distillery wastes are utilized as dual purposes to produce PHB in lower production cost and to reduce the amount of waste to be treated. In this study, various attempts were made to increase PHB production under various conditions by Actinobacillus sp. EL-9 in a shaker culture. The addition of glucose, NH$_{4}$NO$_{3}$ to alcoholic distillery wastes slightly promoted cell mass and PHB production. Enzyme hydrolysis of alcoholic distillery wastes increased the production of PHB than that of untreated waste and acid hydrolysis treatment. The PHB weight in alcoholic distillery wastes was 1.91 g/l. Fermentation process of PHB production reduced the amount of COD value up to 54%, which reduced organic loading rate and capacity of activated sludge system.

Enhanced Production of Succinic Acid by Actinobacillus succinogenes using the Production Medium Supplemented with Recombinant Carbonic Anhydrases (재조합 탄산무수화 효소 첨가 생산배지를 이용한 Actinobacillus succinogenes 유래의 숙신산 생산성 향상)

  • Succinic acid, a representative biomass-derived platform chemical, is a major fermentation product of Actinobacillus succinogenes. It is well known that carbon dioxide is consumed during the succinate fermentation, but the biochemical mechanism behind this phenomenon is not yet understood well. In this study, it was found that the addition of carbonic anhydrase (CA)s into media significantly enhances the succinic acid production by A. succinogenes during the fermentation supplied with carbon dioxide. It is likely that the (bi) carbonate produced by the CA activity from gaseous carbon dioxide is favoured by A. succinogenes for consumption and utilization. Therefore, the $MgCO_3$ requirement could be significantly reduced without compromising the succinate productivity. Furthermore, because of too high price of the commercial carbonic anhydrase, it was undertaken to economically overproduce a cyanobacterial carbonic anhydrase by the use of a recombinant Pichia pastoris. An expression vector system was constructed with the carbonic anhydrase gene PCR-cloned from Cyanobacterium Synechocystis sp., and introduced into P. pastoris for fermentation studies. About 95.9 g/L of succinic acid was produced in the production medium with 30 ppm of carbonic anhydrase, approximately 2 fold higher productivity compared to the parallel process with no supplementation of the enzyme. It is expected that this method can provide a valuable way of overcoming inefficiencies inherent in gas supply during $CO_2$-based bioprocesses like succinic acid fermentation.

Continuous Production of Succinic Acid Using an External Membrane Cell Recycle System

  • Succinic acid was produced by continuous fermentation of Actinobacillus succinogenes sp. 130Z in an external membrane cell recycle reactor to improve viable cell concentration and productivity. Using this system, cell concentration increased to 16.4 g/l at the dilution rate $0.2\;h^{-1}$, up to 3 times higher than that of batch culture, and the volumetric productivity of succinic acid increased up to 6.63 g/l/h at the dilution rate $0.5\;h^{-1}$, 5 times higher than that of batch fermentation. However, in the continuous culture using a high dilution rate, operational problems including severe membrane fouling and contamination by lactic acid producer were observed. Another succinic acid producer, Mannheimia succiniciproducens MBEL55E, was also utilized in this system, and the cell concentration and productivity of succinic acid at the dilution rate of $0.3\;h^{-1}$ were found to be above 3 and 2.3 times higher, respectively, compared with those obtained at the dilution rate of $0.1\;h^{-1}$. These observations give a deep insight into the process design for a continuous succinic acid production by microorganisms.

Comparison of Culture-dependent and DGGE based Method for the Analysis of Marine Bacterial Community (배양법과 DGGE에 의한 해양세균 군집의 비교분석)

  • Seasonal variation of marine bacterial community was analyzed in the surface sea water collected from one of the stations locating at Tongyeoung coastal area, Korea. The results obtained by the culture method through identification with the VITEK Microbe ID system after pure culture in the selective medium were compared with those obtained by the DGGE based 16S rRNA PCR method. The composition of the marine bacterial community in the sea water samples harvested in September, 2004, November, 2004, January, 2005, May, 2005 and August, 2005 determined by the culture method showed 5, 5, 4, 6, and 10 strains respectively. Pseudomonas fluorescens and Acinetobacter lwoffii were detected in all seasons. The other strains were identified to be Pseudomonas stutzeri, Sphingomonas paucimobilis, Burkholderia mallei and Chryseobacterium indologenes. In contrast, the 16S rRNA PCR-DGGE method detected 10, 11, 6, 9 and 13 populations respectively in the same sea water samples and the strains were identified to be Acinetobacter lwoffii, Burkholderia mallei, Pseudomonas fluoresence, Actinobacillus ureae, Burkholderia sp., Pseudomonas stutzeri, Roseobacter sp., Vibrio parahaemolyticue, Sphingomonas paucimobilis and Rugeria algocolus. This results indicated that the DGGE based 16S rRNA PCR method was more efficient than the culture method for the grasp of the characteristics of the marine bacterial community.

Investigation of respiratory disorders in slaughtered pigs (돼지의 호흡기질병 감염상황 조사)

  • An abattoir survey of pneumonia and other lesions in slaughtered pigs from 5 selected herds located In the Western Kyongbuk was carried out during the period from March to December 1995. Pneumonic lungs was attempted bacteriological finding and antibiotic susceptibilities. From 583 slaughtered pigs, 445(76.3%) case was pneumonic lungs, seasonal patterns were Winter, Spring, Autumn, Summer in the order named. Among them, 127(21.8%) case was mycopla-sma pneumonia and 65(11.2%) case was pleuropneumonia. In snout lesion grade from 337 slaughtered pigs, above grade II score was 107(31.8%). In the white spot of liver, grade I was 544(93.3%), grade II32(5.5%) and grade III 7(1.2%). In the gastric ulcer, normal was 350(60.0%), grade I168(28.8%), grade II59(10.1%) and grade III 6(1.1%). Among the pigs(n=271) with pneumonic lesions above 20%, 162 strains werr isolated from 87(32.1%) pigs. The bacteria isolated from pneumonic lesions was Pasteurella sp 61(37.7%), Streptococcus sp 31(19.1%), Actinobacillus sp 3(1.9%), Coliform bacteria 19(11.7%) and the other bacteria 48(29.6%) These isolates were highly susceptible to the antibiotics including ENR 142(87.7%), Cft 138(85.2%) and Cf 126(77.8%).

Distribution of Hemolytic Vibrio sp. in Sea Water of the Beaches of Busan During Mid-summer (여름철 부산시 소재 해수욕장 해수의 용혈성 비브리오균 분포)

  • The distribution of hemolytic Vibrio sp. from sea water of three main beaches located in Busan (Gwangan(G), Haeundae(H) and Songjeong(S) beaches) was investigated from June to September 2006 ; this is mid-summer and the main season for bathing. The monthly detection ratio from each beach was 29.2% (7 of 24 samples, G), 33.3% (8 of 24 samples, H), and 16.7% (4 of 24 samples, S). The most probable number(MPN) of strains detected ranged from 1.8-36(G), 1.8-180(H) and 1.8-18(S) MPN/100mL. Of the isolated strains, 24 strains showed definite hemolytic activity. These 24 strains were identified as Vibrio fluvialis, Vibrio vulnificus, Aeromonas hydrophila, Actinobacillus ureae and Eikenella corrodens. Vibrio fluvialis was detected from all three beaches investigated. Vibrio vulnificus was detected from Haeundae and Gwangan beaches. Gwangan beach had a higher detection ratio of Vibrio sp. than Haeundae and Songjeong beaches. These results suggest that seafood harvested from the vicinity of theses beaches may cause food poisoning and risk management to prevent Vibrio septicemia is required, especially for Haeundae and Gwangan beaches.

An abattoir survey of incidence of pneumonia in slaughter pigs and an investigation of microbiology of affected lungs (도축돈의 폐렴병변 분포조사 및 폐렴병소로부터 호기성균의 분리동정)

  • The present study was conducted to investigate the incidence of pneumonic lesions with special regard to enzootic pneumonia and the microbiology of pneumoic lungs from 544 slaughter pigs during the period from October 1995 to September 1996. The incidence of enzootic pneumonic lesion was 76.3% (41s/s44) and pleurisy was detected from 7.9% of slaughter pigs. Seasonal prevalence of pneumonic lesions in slaughter pigs were in order of prevalence of 82.9% in spring, 76.8% in winter, 74.8% in autumn and 69.0% in summer, respectively. Pasteurella multocida, Streptococcus sp, Str suis, Corynebacterium sp, Actinobacillus pleuropneumoniae, Hemophilus parasuis, and Klebsiella pneumoniae were detected in order of prevalence from 16.9%, 15.9%, 7.5%, 6.0%, 1.4%, 1.0% and 0.5% of 415 pneumonic lungs, respectively. P multocida were susceptible to oxytetracycline, polymyxin-B, streptomycin, and vancomycin, while the majority of them were resistant to amoxicillin, ampicillin, cephalothin, kanamycin, and penicillin-G. Str suis were susceptible to amoxicillin, ampicillin, cephalothin, penicillin-G, although the majority of them were resistant to erythromycin, oxytetracycline, streptomycin, vancomycin. A pleuropneumoniae were susceptible to ampicillin, and cephalothin, but the majority of them were resistant to oxytetracycline.
