Studies on the Shelf-life Extension of Jeotkal, Salted and Fermented Seafood (젓갈류의 유통기한 연장을 위한 연구)

  • To develop natural food preservatives for extending the shelf-life of jeotkal (salted and fermented seafood), antimicrobial substances were extracted from 32 types of medicinal herbs and edible plants using 95% ethanol. Among the extracts, Glycyrrhizae radix, Curcumae domestica, Galla rhois, and Resina pini showed relatively high inhibitory effects on the growth of the microorganisms isolated from the deteriorated jeotkal. We selected and tested the extract from Recina pini as a natural jeotkal preservative. This ethanol extract was purified partially by adding equal quantity of water, through which 77% of insoluble materials were removed as impurities. In manufacturing modified jeotkal using squid, sucrose and starch syrup were substituted with sorbitol, $glucono-{\delta}-lactone$ was added instead of vitamin C and lactic acid, and sterilized hot pepper was used instead of natural one. The shelf-life of modified jeotkal was prolonged by 4 days compared with the control jeotkal when stored at $20^{\circ}C$, while that of modified jeotkal containing 1.0% partially purified Recina pini extract was prolonged by 6 days compared to the control. The same tests were conducted for the changran (stomach and intestine of Alaska pollack) jeotkal preservation. The shelf-life of the control jeotkal was 24 days, whereas the modified jeotkal and the Resina pini extract-containing modified jeotkal maintained their qualities without changes in microbial and chemical characteristics for 90 days at $20^{\circ}C$ storage.

The Influence of AH-26 and Zinc Oxide-Eugenol Root Canal Sealer on the Shear Bond Strength of Composite Resin to Dentin (AH-26 및 산화아연유지놀 근관실러가 상아질에 대한 복합레진의 전단결합강도에 미치는 영향)

  • The purpose of this study was to evaluate the influence of the AH-26 root canal sealer on the shear bond strength of composite resin to dentin. One hundred and forty four (144) extracted, sound human molars were used. After embedding in a cylindrical mold, the occlusal part of the anatomical crown was cut away and trimmed in order to create a flat dentin surface. The teeth were randomly divided into three groups; the AH-26 sealer was applied to the AH-26 group, and zinc-oxide eugenol (ZOE) paste was applied to the ZOE group. The dentin surface of the control group did not receive any sealer. A mount jig was placed against the surface of the teeth and the One-step dentin bonding agent was applied after acid etching. Charisma composite resin was packed into the mold and light cured. After polymerization, the alignment tube and mold were removed and the specimens were placed in distilled water at $37^{\circ}C$ for twenty four hours. The shear bond strength was measured by an Instron testing machine. The data for each group were subjected to one-way ANOVA and Tukey's studentized rank test so as to make comparisons between the groups. The AH-26 group and the control group showed significantly higher shear bond strength than the ZOE group (p<0.05). There were no significant differences between the AH-26 group and the control one (p>0.05). Under the conditions of this study, the AH-26 root canal sealer did not seem to affect the shear bond strength of the composite resin to dentin while the ZOE sealer did. Therefore, there may be no decrease in bond strength when the composite resin core is built up immediately after a canal filling with AH-26 as a root canal sealer.

Protective Effects of Sea Buckthorn (Hippophae rhamnoides L.) Leaves Fermented with Hericium erinaceum Mycelium against Oxidative Modification of Biological Macromolecules and Cell Death (노루궁뎅이 버섯균사체를 이용한 비타민나무 발효물이 생체고분자의 산화적 변형과 세포사멸에 미치는 보호 영향)

  • In this study, hot water extract from sea buckthorn (Hippophae rhamnoides L.) leaves fermented with Hericium erinaceum mycelium (SBT-HE) was assessed for protection against oxidative modification of biological macromolecules and cell death. Antioxidant activity of SBT-HE was evaluated based on 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical, 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical, and peroxyl radical scavenging assays. SBT-HE showed 65.06% DPPH radical scavenging activity at $500{\mu}g/mL$, 98.83% ABTS radical scavenging activity at $50{\mu}g/mL$, and 44.03% peroxyl radical scavenging activity at $100{\mu}g/mL$. SBT-HE significantly inhibited DNA strand breakage induced by peroxyl radical. SBT-HE also prevented peroxyl radical-mediated human serum albumin modification. SBT-HE effectively inhibited $H_2O_2$-induced cell death and significantly increased cell survival by 21.59% at $100{\mu}g/mL$. SBT-HE also reduced intracellular reactive oxygen species levels in $H_2O_2$-treated cells. The results suggest that SBT-HE can contribute to antioxidant activity and protect cells from oxidative stress-induced cell injury.

Temperature Effect on Nitrification and Interrelationship between Nitrifiable NO3-N and Tobacco Productivity in Some Tobacco Tillage Soils with Different Soil pH (토양(土壤)pH가 상이(相異)한 몇가지 연초경작지(煙草耕作地) 토양(土壤)에서 질산화작용(窒酸化作用)에 대한 온도효과(溫度效果) 및 NO3-N와 잎담배 생산성(生産性)과의 상호관계(相互關係))

  • An incubation study was conducted to examine the effect of soil pH and temperature on nitrification potential of 8 different soil series applied with no-N and 200 ugN/g soil as a compound fertilizer at 60 % moisture content of maximum water holding capacity for 8 weeks, whose series were ranged from acid to mild alkali as Gopyeong(Jincheon, pH 4.51), Yesan(Jincheon, pH 4.54), Jigog(Eumseong, pH 4.71), Songsan(Goesan, pH 5.01), Angye(Seongju, pH 5.34), Banho(Seongju, pH 5.73), Weongog(Jincheon, pH 5.93), and Banho(Seongju, pH 7.70), respectively. Interrelationship between the nitrifiable and the net $NO_3-N$(N added plot-no-N plot) accumulated in the soil and tobacco yield in the no fertilizer plot were investigated as well. 1. Nitrification response was various according to soil characteristics at each temperature condition showing that nitrifiable $NO_3-N$ values of the soils were much higher at $25^{\circ}C$ than $15^{\circ}C$. And difference of nitrification potential affected by temperature was markedly distinguishable from 2 weeks after incubation and was showing a tendency to reduce with increasing of soil pH. 2. At each temperature condition, net $NO_3-N$ accumulated at 2 and 4 weeks after incubation was positively correlated with soil pH. 3. Tobacco yield in the no fertilizer plot was more highly correlated with the values of nitrifiable and net $NO_3-N$ accumulated at $15^{\circ}C$ similar to soil temperature in rhizosphere of early stage of tobacco growth than those at optimum temperature($25^{\circ}C$).

Production of yuzu granules using enzyme treated yuzu pulp powder and evaluation of its physiochemical and functional characterization (유자박 식이섬유를 이용한 유자과립 제조 및 이화학적 특성조사)

  • In this study, solubilized yuzu pulp powder (EYP) was produced using enzyme treated yuzu pulp powder (YP) and used to manufacture yuzu granules (0-20% EYP content). The physicochemical, product stability, and functional properties of Yuzu granules were compared among five enzyme treatments. Among the five treatments, CL had the highest YP solubilization yield (48.68%). Microstructural observation of EYP using FE-SEM revealed that its surface became irregular and porous after enzymatic treatment. Compared to YP, EYP had 2 times lower insoluble dietary fibers and 3 times lower hemicellulose and cellulose content. Among the yuzu granules, IV (yuzu granules with 15% EYP) had an excellent water and oil holding capacity and flowability. IV granule had the highest narirutin and hesperidin content of 3.4 mg and 2.2 mg/g DW, respectively and the highest antioxidant (68.4%) and tyrosinase inhibitory activities (82.5%). Therefore, EYP or granule with EYP can be used as a functional component in food industry or pharmaceutical field.

Comparison of Quality and Bioactive Compounds in Chicken Thigh Meat from Conventional and Animal Welfare Farm in Korea (국내 일반 및 동물복지 육계 다리육의 냉장 저장 기간 중 품질과 생리활성기능 성분 비교)

  • This study was conducted to evaluate the difference in the quality of chicken thigh meat from conventional and animal welfare farms during refrigeration storage over 9 days. Chicken thigh meat from conventional (CTC, n = 30) and animal welfare farms (CTW, n = 30) was tested. The pH value was significantly lower in CTW (6.28) than in CTC (6.37) on day 1; however, no significant differences were found on subsequent days. The yellowness of CTW was higher than that of CTC on day 1, but CTW showed lower yellowness than did CTC on day 7 and 9. The cooking loss, water holding capacity, lightness, redness, and coliform levels of CTC did not show any significant difference when compared with CTW during storage. The shear force of CTW was significantly higher than that of CTC on day 1, 3, 7, and 9. Total microorganism and coliform in CTC and CTW increased with increasing storage days. On day 7 and 9, the total microorganism level of CTW was lower than that of CTC. The thiobarbituric acid value of CTW was lower than that of CTC on day 9. The volatile basic nitrogen (VBN) of CTW was lower than that of CTC during storage. Anserine content and 1,1-diphenyl-2-picryl-hydrazyl(DPPH) scavenging activity of CTW was significantly higher than CTC on day 1. These results suggest that CTW stayed fresher for longer than did CTC because of low total microorganism level on day 7 and 9, and VBN during refrigerated storage.

Protective effect of Gabjubaekmok (Diospyros kaki) extract against amyloid beta (Aβ)-induced cognitive impairment in a mouse model (아밀로이드 베타(amyloid beta)로 유도된 인지장애 마우스 모델에서 갑주백목(Diospyros kaki) 추출물의 인지기능 및 뇌 신경세포 보호 효과)

  • The current study investigated the effect of Gabjubaekmok (Diospyros kaki) ethanolic extract (GEE) on $H_2O_2$-induced human neuroblastoma MC-IXC cells and amyloid beta $(A{\beta})_{1-42}$-induced ICR (Institute of Cancer Research) mice. GEE showed significant antioxidant activity that was evaluated based on ABTS, DPPH scavenging activity, and inhibition of malondialdehyde (MDA) and acetylcholinesterase activity. Further, GEE inhibited ROS production and increased cell viability in $H_2O_2$-induced MC-IXC cells. Administration of GEE ameliorated the cognitive dysfunction on $A{\beta}$-induced ICR mice as evaluated using Y-maze, passive avoidance, and Morris water maze tests. Results of ex vivo test using brain tissues showed that, GEE protected the cholinergic system and mitochondrial functions by increasing the levels of antioxidants such as ROS, mitochondrial membrane potential (MMP), and adenosine triphosphate (ATP) against $A{\beta}$-induced cognitive dysfunction. Moreover, GEE decreasd the expression levels of apoptosis-related proteins such as $TNF-{\alpha}$, p-JNK, p-tau, BAX and caspase 3. While, expression levels of p-Akt and $p-GSK3{\beta}$ increased than $A{\beta}$ group. Finally, gallic acid was identified as the main compound of GEE using high performance liquid chromatography.

Biological Activity of Viola mandshurica Fermented with Bacillus methylotrophicus CBMB205 and Leuconostoc pseudomesenteroides NRIC1777 (미생물 분리주를 이용한 제비꽃 발효액의 생리활성 변화)

  • The aim of this study was to improve the efficacy and functionality of Viola mandshurica (VM). A water suspension of VM power was fermented for 72 hr with Bacillus methylotrophicus CBMB205 (BM) and Leuconostoc pseudomesenteroides NRIC1777 (LP) isolated from kimchi. The antioxidant activity and reducing power of fermented VM, its total phenolic and flavonoid compounds, as well its inhibitory activity on ${\alpha}$-amylase, ${\alpha}$-glucosidase, and pancreatic lipase were determined and compared to those of non-fermented VM (NVM), a negative control. The total phenolic and flavonoid compounds of VM fermented with BM and LP were higher than those of NVM by 1.4, 1.17, and about 3 times. There was no difference in 2, 2'-diphenyl-1-picryl hydrazyl (DPPH) radical scavenging activity between fermented VM and NVM. However, there was a 2.1- and 1.6-fold increase in 2, 2'-azino-bis-(3-ethylbenzothzoline-6-sulfonic acid) (ABTS) radical scavenging activity in VM fermented with BM and LP, respectively. The reducing power of BM was 1.6 times as high as NVM, but no significant difference was found between LP and NVM. Fermented VM's inhibitory activity on ${\alpha}$-amylase, ${\alpha}$-glucosidase, and pancreatic lipase was much higher than that of NVM. Fermenting VM with BM was superior to fermenting it with LP, except flavonoid content. Taken together, VM fermented with BM could be used as a functional food and as an additive to cosmetics.

Development and Validation of an Analytical Method for Quinoxyfen in Agricultural Products using QuEChERS and LC-MS/MS (QuEChERS법 및 LC-MS/MS를 이용한 농산물 중 살균제 Quinoxyfen의 잔류시험법 개발 및 검증)

  • An analytical method was developed for the determination of quinoxyfen in agricultural products using the QuEChERS (Quick, Easy, Cheap, Effective, Rugged and Safe) method by liquid chromatography-tandem mass spectrometry (LC-MS/MS). The samples were extracted with 1% acetic acid in acetonitrile and water was removed by liquid-liquid partitioning with $MgSO_4$ (anhydrous magnesium sulfate) and sodium acetate. Dispersive solid-phase extraction (d-SPE) cleanup was carried out using $MgSO_4$, PSA (primary secondary amine), $C_{18}$ (octadecyl) and GCB (graphitized carbon black). The analytes were quantified and confirmed by using LC-MS/MS in positive mode with MRM (multiple reaction monitoring). The matrix-matched calibration curves were constructed using six levels ($0.001-0.25{\mu}g/mL$) and the coefficient of determination ($R^2$) was above 0.99. Recovery results at three concentrations (LOQ, 10 LOQ, and 50 LOQ, n=5) were in the range of 73.5-86.7% with RSDs (relative standard deviations) of less than 8.9%. For inter-laboratory validation, the average recovery was 77.2-95.4% and the CV (coefficient of variation) was below 14.5%. All results were consistent with the criteria ranges requested in the Codex guidelines (CAC/GL 40-1993, 2003) and Food Safety Evaluation Department guidelines (2016). The proposed analytical method was accurate, effective and sensitive for quinoxyfen determination in agricultural commodities. This study could be useful for the safe management of quinoxyfen residues in agricultural products.

Effects of Dietary Supplementation of Enzyme complex on Growth Performance, Carcass Characteristics and Meat storability in Broiler Chickens (사료 내 복합효소제의 첨가가 육계 생산성 및 도체특성 및 저장성에 미치는 영향)

  • This study was conducted to investigate the effects of dietary supplementation of enzyme complex (metalloprotease and xylanase) on growth performance, carcass characteristics and meat storability of broiler chickens. The experiment utilized a 2 (basal and low spec diets) ${\times}$ 3 (0, 0.5 or 1 g/kg of enzyme supplementation) factorial arrangement. A total of 360 male Ross broilers were randomly assigned into the following six groups: BD-T0 (basal diet + enzyme complex 0 g/kg), BD-T1 (basal diet + enzyme complex 0.5 g/kg), BD-T2 (basal diet + enzyme complex 1 g/kg), LD-T0 (low spec diet + enzyme complex 0 g/kg), LD-T1 (low spec diet + enzyme complex 0.5 g/kg), LD-T2 (low spec diet + enzyme complex 1 g/kg). Feed and water were provided ad libitum for 42 days, during which time feed intake and body weight were measured at one-week intervals. After the feeding trial, chickens were slaughtered by exsanguination and samples were collected. Feed intake was lower in the enzyme supplemented groups than the non-supplemented groups (p<0.05), as were body weight and gain (p<0.05). Thus, the feed conversion ratio improved in the enzyme supplemented groups relative to the non-supplemented groups (p<0.05). The carcass rate and production index were also significantly improved by supplementation with enzyme complex (p<0.05). Finally, the thiobarbituric acid reactive substances (TBARS) values were lower in the enzyme supplementation groups after 6 storage days (p<0.05). In conclusion, dietary supplementation with enzyme complex including metalloprotease and xylanase improved growth performance, carcass characteristics, and storability of broiler chicken meat.
