A bacterial strain, KSl, possessing strong antifungal activity was isolated from soil samples of ginseng fields and identified as Bacillus subtilis. In greenhouse test, the culture filtrate of B. subtilis KS1 showed strong protective effect against several fungal diseases of agricultural plants such as cucumber gray mold and wheat leaf rust. In addition, the crude butanol fraction of the culture filtrate exhibited antagonistic effect against several fungi including plant or human pathogens, such as Botrytis maydis, Chytridium lagenarium and Candida albicans. The antifungal compound, SW1, produced by B. subtilis KS1 was purified through consecutive chromatographic separations on a pep-RPC column and a ${\mu}$ Bondapak $C_{18}$ reverse phase column. Temperature and pH showed little effect on the stability of the compound in the ranges $-20-121^{\circ}C$ and pH 4.0-10.0, respectively. The composition and structural characteristics of SW1 were analysed by HPLC and by $^1H-,\;^1H-^1H-COSY$, NOESY, COSY-NOESY and HOHAHA NMR spectroscopy, respectively, which revealed that the compound belongs to iturin A, a typical cyclic antifungal compound produced by B. subtilis. In contrast to the previously reported iturin A compounds which have one or no $-CH_3$ side chain in the hydrophobic hydrocarbon chain of ${\beta}-amino$ acids, SW1 was shown to have a ${\beta}-amino$ acid containing 12-carbon skeleton with two $-CH_3$ side chains.
One of the most significant environmental issues in abandoned coal mine is acidic drainage which gives rise to the many environmental problems that acidifying streams water, sedimentation of iron/aluminium hydroxide, and pollution of water and soil. Water and precipitate samples for experiments were collected from stream and bottom in the pit mouth of Sambong mine. Mine water shows pH range from 7.24 to 7.94 in winter and 3.87 to 5.73 in summer season. The EC shows range from 432 to $897{\mu}S/cm$ at the stream receiving mine water. The highest concentrations of cations such as Mg, Al, Ca, and Mn are showing 15.50, 4.56, 85.30, 12.76 mg/L in the pit mouth, respectively. The reddish brown precipitates (Munsell color 10R-5YR in winter and 2.5YR-5Y in summer) consist mainly of 2-line ferrihydrite and schwertmannite. The precipitates are characterized by rod or cylindrical forms, and coccus or sphere of 0.1 to $0.5{\mu}m$ in diameter.
Sphingomonas sp. KS 301, which was isolated from oil contaminated soil, was shown to have five different SODs (SODI, II, III, IV, V) which can be separated by DEAE-Sepharose chromatography, and SOD III was finally purified in this study by ammonium sulfate precipitation, DEAE-Sepharose chromatography, Superose 12 gel filtration and Uno-Q1 ion exchange chromatography. The molecular weight of SOD III was 23 kDa as determined by SDS-PAGE and the apparent molecular weight of the native enzyme was estimated to be approximately 71 kDa by Superose-12 gel filtration chromatography. These data suggest that the purified SOD consists of at least two subunits. The specific activity of the SOD III was higher than Mn type or Fe type SOD of Escherichia coli by 5 fold. To determine the type of SOD III, inhibitory effects of $NaN_{3},\;H_{2}O_{2},\;KCN$ were examined. 10 mM $NaN_{3}$ was able to inhibit 56% of the SOD III activity, which indicates that this SOD is Mn type. The optimum pH of the SOD III was 7.0 and the optimum temperature was $20^{\circ}C$. N-terminal amino acid sequence of purified SOD III was most similar to those of Psudomonase ovalis and Vibrio cholerae among bacteria.
Issues of declining growth and vigor in forests are major concerns in many areas around the world, especially in response to predictions in the 1980s of widespread forest declines. This paper summarizes the current state of knowledge for forests in North America, Europe, and East Asia. Forest declines near point-sources of pollution(such as metal smelters) have been well recognized for a century, but evidence of widespread impacts away from point-sources remains relatively uncommon. In North America, significant forest decline has resulted from high concentrations of ozone near Los Angeles, California, and around Mexico City. Some high-elevation forests of red spruce in the eastern U.S. have declined in the past 20 years ; evidence indicates a role of low-pH fog in reducing they cold-tolerance of spruce. In Europe. most attention has focused on Norway spruce stands that developed yellow foliage, needle loss, and in some cases mortality. This syndrome appears to be related generally to an inadequate supply of magnesium, perhaps coupled with a very high supply of nitrogen. Despite localized areas that show declining trees, overall stand growth and standing biomass in Europe increased from. 1970 to 1990. Much less information is available for East Asia. Many industrialized regions in China have a pH of rain <4.5, and some connections between pollution and forest decline have been suggested. Pollution impacts on forests near cities in Korea include needle chlorosis, reduced needle retention, and declining species diversity. Overall, temperate forests show no widespread declines, and no evidence of substantial effects of pollutants on forest growth or vigor. Small areas showing declining forests may indeed demonstrate pollution impacts, and may provide cause for concerns about future impacts on larger areas.
For obtaining the basic data for establishing plan on the conservation of the Gameunsa 3-story and Naweonri 5-story Pagodas located at the Kyeongju city, the characteristics of the rock and weathering phenomena have been investigated. The former consists of quartz-rlch granite containing small amount of biotite, and the latter of alkali granite with abundant perthite, These rock phases are nearly identical to the marginal phase of medium-grained hornblende-biotite granodiorite and alkali granite respectively, which are distributed around the Kyeongju city. The rock weathering may be governed mainly by chemical weathering of feldspar following physical segregation of quartz grains and pervasive moss. The feldspar easily dissolve In the solution with pH<7 to precipitate clay mineral such as a kaolinite as a secondary phase on the feldspar surface. However, the chemical weathering of feldspar may continue when the surface is washed by the rain according to removal of the reprecipitated phase. On forwarding, the weathering may be greatly Influenced by the acid rain. Exfoliation and weathering along igneous lineation resulting in exfoliating along the structural line are the characteristic weathering phenomena. Also the secondary small cracks are irregularly developed on the rocks due to different strain on places by the overall structural unbalance of the pagodas. Along these cracks, the rain water intrudes deeply into the rocks and weathering occurs intensively compared to other parts. Weathering may be artificially promoted by the grinding or sculpturing when the pagodas were made. Because it may influence on the physical properties of the rocks as well as destruct the surface of the feldspar crystals, the major constituents of the rocks, it results in providing the environment of easy chemical weathering along time. For conservation, the pagodas must be structurally balanced by compacting the soil basement and supplementing rocks on the destroyed part. On the exfoliated part it is better not to be artifically treated as using cementing material. But the cracks may be filled up by cementing material to avoid the intrusion of acidic water. To supplement the rocks on the destroyed part, it may be better to use similar rock phases from identical biotite granite and alkali granite masses around the Kyeongju city.
This study is on the corrosion of iron objects caused by sulfides in undersea environment. The corrosion state of objects in seawater and their damage state after underwater and left in highly humid air were studied. The samples of this study were four iron objects which had been taken out from undersea mud layer located in Taean Mado, Chungcheongnamdo. SEM-EDS and XRD analyse on the objects to check whether they have sulfides or not. The result of analysis suggested that the major component of corrosion product generated in undersea deposit soil is sulfur(S) and iron sulfide(FeS) is formed as sulfide. However, there was no clear corrosion on the surface of objects which was exposed to sea water because of the impact of concretion which covered the surface. In order to check the damage status of iron objects after they had been taken out of sea water, exposure tests in high humidity environment and dehumidified environment were done on the corrosion products. The result of the test suggested that the oxidization of iron sulfide corrosion product makes iron sulfate ($FeSO_4$) and sulfuric acid ($H_2SO_4$) and they can cause secondary corrosion of iron objects. Therefore, it is believed that the iron sulfide corrosion product of iron objects taken out from underwater environment should be removed by all means and the keeping environment of the iron objects should also maintain dehumidified state.
In this study, we introduced experience participated in ELPAT (Environmental Lead for Proficiency Analytical Testing) program that is administered by the American Industrial Hygiene Association and EPA. The 126 sample results of total 128 samples met reference value, as a result accepted 'proficient'. The concentration of Pb in air, paint chips, soils and dust wipes are $0.0089{\sim}0.3956mg/m^3$, 0.0500~8.9149%, 29.0${\sim}$1697 mg/kg and $18.00{\sim}900.3{\mu}g/sample$, respectively. The lead in environmental matrix prepared adequate techniques and analysed by flame atomic absorption spectrometry. The lead in air was extracted hydrochloric acid, in paint chips and soils were digested with microwave and hotplate and in dust wipes were prepared microwave digestion. No differences of each preparation techniques was found and all methods were to be proper.
To investigate optimal conditions for plant regeneration in Platycodon grandiflorum (Jacq. A. DC.).Both leaf and hypocotyl explants were cultured on Murashige& Skoog's (MS) medium supplemented with combinations of 0.1, 0.5, 1.0, or 2.0 mg/L cytokinins (BA and kinetin) and 1.0 mg/L 2,4-D for 6 weeks, respectively. According to the type of explant, the total shoot organogenesis (56.38%) in leaf explants was higher than in hypocotyls (28.20%). In comparison with kinetin and BA for the plant regeneration, the frequency (70.38%) of leaf explants was higher in combination with kinetin and 2,4-D than of BA with 2,4-D (42.38%), whereas the frequency (35.56%) of hypocotyls explants was higher in BA combination than kinetin combination (20.83%). Thehighest frequency (94.20%) was observed from the cultures of leaf explants on the MS medium supplemented with 1.0 mg/L kinetin and 1.0 mg/L 2,4-D. Upon transfer onto 1/2 MS basal medium containing 3% sucrose, shoots developed into plantlets with roots, and were well grown in soil in the greenhouse. These results lead us to speculate that the optimization of culture conditions was responsible for the mass propagation from in vitro cultures of Platycodon grandiflorum (Jacq. A. DC.).
To elucidate a method of preventing dental caries, strains producing lytic enzymes were isolated and their characteristics were investigated. Among 5,00 alkalophilic strains isolated from soil, 22 strains showed lytic activity against Streptococcus mutans. Strain No. 4830, with the highest lytic activity, was selected for further study. Strain 4830 showed 94% sequence homology with the 16S rDNA sequence of Bacillus alcalophilus, but it was concluded to be different from Bacillus alcalophilus because of its biochemical characteristics. The strain was named Bacillus sp. 4830. The lytic enzyme from Bacillus sp. 4830 was purified by ethanol precipitation and CM-agarose column chromatography. The molecular weight of the lytic enzyme was determined to be 28 kDa by SDS-PAGE. The lytic enzyme was stable between pH 5.0 and pH 11 and up to $40^{\circ}C$. The optimal pH and temperature for the lytic activity was 9.0 and $50^{\circ}C$, respectively.

The growth characteristics of Candida tropicalis KIST 359, isolated from soil samples collected at an oil depot in Korea, have been studied by cultivating batches under varying conditions. The conclusions of the study were: 1. The yeast easily assimilates hydrocarbons in a range of $C_{14}-C_{17}$, and the optimum cultivation temperature and pH are $30^{\circ}C$ and 5.5, respectively. 2. Using this strain of micro-organism, gas oil gives a higher cell yield than kerosine and with gas oil except urea all other nitrogen sources $(NH_4NO_3,\;NH_4Cl,\;(NH_4)_2SO_4\;and\;(NH_4)_2HPO_4)$ similarly support a satisfactory growth of the yeast. 3. The highest yield is obtained with a gas oil level of 10%(v/v), and concentrations of nitrogen source and $MgSO_4{\cdot}7H_2O$ of 0.5 and 0.05%(w/v), respectively. 4. The protein content of dried yeast cells is 59.8%. Its amino acid composition can be compared well with that of FAO provisional patterns, but with a low methionine and a high lysine content.
