오갈피(Acanthopanax sessiliflorum Seeman) 추출액이 유산균의 생육에 미치는 영향 (A Study on Effect of Acanthopanax sessiliflorum Seeman Water Extract on the Growth of Lactic Acid Bacteria)

  • This study examined the effect of medicinal herbs on the growth of 3 strains of lactic starter cultures in a MRS broth by adding a 0, 1, 3, 5, and 10% water extract. The pH, titratable acidity and OD of lactic acid bacteria were investigated in order to obtain fundamental knowledge for the development a new product, box thorn yogurt. In the case of adding the Acanthopanax sessiliflorum Seeman water extract, Streptococcus thermophilus was largely activated by the addition of the 5% extract. The growth of Lactobacillus acidophilus was activated by the extracts but it was depressed by the addition of the 3% Acanthopanax sessiliflorum Seeman water extract. Compared with the control, the growth of Bifidobacterium longum was largely activated by the addition of the 10% extract but it was depressed by the addition of the 5% Acanthopanax sessiliflorum Seeman water extract.

오갈피나무 종자(種子)의 성분(成分)에 관한 연구(硏究)(제1보)(第1報)-지방산(脂肪酸)과 Sterol 성분(成分)에 관하여- (Studies on the Constituents of Seeds of Acanthopanax sessiliflorum Seemann (I)-On Fatty Acids and Sterols-)

  • Fatty acids and sterols were isolated from petroleum ether extract of seeds of Acanthopanax sessiliflorum Seeman. Fatty acids were obtained from the saponified fraction and sterols from nonsaponified fraction of petroleum ether extract. Fatty acids were identified by gas liquid chromatography, and its composition was myristic acid, palmitic acid, stearic acid, oleic acid, linoleic acid and linolenic acid. The composition of sterols were determined by preparative TLC and gas liquid chromatography. It was confirmed to be a mixture of stigmasterol and ${\beta}-sitosterol$. These fatty acids and sterols were not previously reported from seeds of Acanthopanax sessiliflorum Seemann.

오가피속 식물 열매의 식물화학 성분 비교 (The Comparison of Phytochemical Components from the Berry of Acanthopanax Species)

  • This study was carried out to obtain the basic information that can be used to index Acanthopanax in eleven species of China and Korea. The phytochemical components from the berry of Acanthopanax species, were measured by the HPLC analysis. Protocatechuic acid, eleutheroside B, eleutheroside E, scopolin, rutin, hyperoside, chiisanoside, oleanolic acid were found in ethanol extracts from the berry of Acanthopanax species. Total phenolic compound of Acanthopanax sessiliflorum f. chungbuensis berry (0.682%) was about seven times higher than those of Acanthopanax divaricatus f. distigmatis berry (0.091%). As a result, the order of the eleutheroside E content was 1) Acanthopanax sessiliflorum f. chungbuensis (0.554%), 2) A. divaricatus var. albeofructus (0.501%), 3) A. divaricatus f. flavi-flos (0.452%). And also, the order of the chiisanoside content was 1) Acanthopanax senticosus var. subinermis (8.434%), 2) A. seoulense (0.94%), 3) A. divaricatus f. flavi-flos (0.798%).

  • The chemical constituents of root bark of Acanthopanax sessiliflorum Seemann(Araliaceae) which is distributed in southern region of Korea are studied. The five kinds of chemical constituents are isolated from MeOH extract of root bark by column fractionation and purified by recrystallization. Substance $I{\sim}III$ were identified as stigmasterol, ${\beta}-sitosterol$ and campesterol. Substance IV is suggested as a lignan substance based on chemical and spectral discussions. The molecular formula of substance $V\;show\;C_{20}H_{18}O_6$ and its melting point is $120.5{\sim}121.5^{\circ}$. Due to the spectral data, it is confirmed that substance V is a (+) sesamin of lignan.

한국산(韓國産 ) 오갈피나무속식물(屬植物)의 분포(分布)에 관하여 (Distribution of Acanthopanax Plants in Korea)

  • Acanthopanax sessiliflorum of family of Araliaceae, Korean wild plant was first described by T. $N_{AKAI}$ in 1909. Although Acanthopanax has been formerly classified by 2 genera, 8 species, but it could be rearranged by 1 genus, 7 species. A. sessiliflorum among 7 species was distributed almost all over the country and A. senticosus was available mainly in the northern part and distribution of A. senticosus was coming down southward alongside Mt. Taibaik and Mt. Sobaik from North. A. chiisanense was mainly distributed over the southern part in the middle area, and the other 4 species were rarely distributed in limited places.

  • 1. The Korean Acanthopanax genus includes 12 kinds consisting of 9 species and 3 forma. 2. The Korean Oga-pi which is on market sale has been used as bark for the medicinal purpose, and Oga-pi shall use Radicis Cortex. That is why it is basic rule that herbalogy shall use Radicis Cortex. 3. The origin of Oga-pi on sale is Acanthopanax sessiliflorum forma chungbunensis C.S. Yook. 4. $C_{HUNG}\;and\;N_AKAI'S}$ report on A. koreanum told us that there are brown hair on the mid-leaf junction, but in addition to it, our investigation was resulted in the fact that there are thorn along mid-rib sometimes. 5. 2 kinds of new forma are similar to A. sessiliflorum, but are different in the view-point of chemotaxonomy, compared with A. sessiliflorum. In its morphology, we can find some difference between 2 kinds of new forma and A. sessiliflorum. Our effort of examination on documents tell us that the all plants growing in the central part of our country is A. sessiliflorum forma chungbunensis C.S. Yook. The one which has thorn on both side among the plants collected in Mt. Dukyu, is called A. sessiliflorum forma nambunensis C.S. Yook. 6. A. sessiliflorum is growing in the southern part in Korea, and most Chungbu Oga-pi A. sessiliflorum forma chungbunensis in the central part of our country. For the convenience of our study, the key of Korean Acanthopanax plant is classified into, I-IV, as shown on the following items: I. No hair on both side of leaf A. Flower stalk is longer than petiole, and there are thorn under the petiole (5-7 stigma).${\cdots}A.\;sieboldianum$. B. Flower stalk is longer than petiole, or same length. Antimicrobial Effect of Acanthopanax sessiliflorum Fruit Extracts against Selected Oral Bacteria

  • This study aimed to evaluate the antimicrobial effects of Acanthopanax sessiliflorum fruit (ASF; Ogaza) extracts on Streptococcus mutans and Streptococcus sobrinus, which are agents that cause dental caries, and on Streptococcus mitis and Streptococcus salivarius, the microbial flora of the oral cavity. The ASF extracts obtained using 70% ethanol were fractionated in the order of ethyl acetate and n-Butanol, concentrated under reduced pressure, and lyophilized to give powdery solvent extracts. The antimicrobial activity of ASF extracts from each solvent was examined using the disk diffusion method. As a result, only those extracts obtained using an ethyl acetate solvent showed antimicrobial activity. These extracts were selected, and the minimum inhibitory concentration was measured by disk diffusion method at various extract concentrations. Results showed a minimum inhibitory concentration of 32 mg/ml. The viable cell count was measured to confirm the minimum bactericidal concentration. Results showed a minimum bactericidal concentration of 64 mg/ml. In the cytotoxicity test using normal human dermal fibroblast cells, the absorbance value of the test group was similar to that of the control group at 0.64, 1.28, and 6.4 mg/ml. The bacteria and their colonies were examined using a scanning electron microscope. Boundaries between the antimicrobial activity region and non-antimicrobial activity region were observed around the paper disk, which was immersed in the extract with 32 mg/ml concentration. Bacterial colonization was not observed in the area with antimicrobial activity. This finding suggests that ASF extracts can inhibit the growth of some microorganisms in the oral cavity, in addition to the effects of these extracts known to date. In particular, ASF extracts may be used as a preparation for preventing dental caries by adding the extract to the toothpaste or oral mouthwash.

오가피추출물의 멜라닌 생성 저해 효과 (The Inhibitory Effects of Acanthopanax sessiliflorum Seeman on Melanogenesis)

  • 본 연구에서는 천연 미백소재 개발을 위하여 오가피에서 추출한 페놀산 분획의 멜라닌 생성에 연관된 생리활성을 분석하였다. 오가피에서 추출한 페놀산 분획은 free radical 소거활성(DPPH)에서 $IC_{50}=3.43{\pm}0.35{\mu}g/mL$, superoxide radicals 소거활성 $IC_{50}=158.91{\pm}1.57{\mu}g/mL$을 나타내었다. B16 melanoma 세포에서의 멜라닌 생합성 저해효과는 농도 의존적으로 저해하여 $100{\mu}g/mL$에서 $27.2{\pm}2.65%$의 저해율을 보였다. 그리고 오가피에서 추출한 페놀산 분획은 $100{\mu}g/mL$에서 세포 내 tyrosinase의 활성을 $53.67{\pm}8.55%$ 저해하였으며, Western blot을 이용하여 tyrosinase와 tyrosinase 관련 단백질(TRP-2)의 발현 감소를 확인하였다. 이러한 결과를 볼 때, 오가피에서 추출한 페놀산 분획은 tyrosinase의 활성 및 발현을 억제함으로써 멜라닌 생성을 감소시키는 것으로 사료된다. 그러므로 오가피에서 추출한 페놀산 분획추출물은 미백용도의 기능성 원료로써 큰 가능성을 가지고 있는 것으로 판단된다.

  • Acanthopanax cortex (Acanthopanax sessiliflorum, Araliaceae, KP VIII), an important herbal drug, has been used as tonic, antistress and immuno-enhancing drugs in Korea. And Peiplocae cortex(Periploca sepium, Asclepiadaceae, CP 2000) has been used as cardiotonic, anti-inflammatory, and sedative effect in china. These are called “Namogapi” of Acanthopanax cortex and “Bukogapi” of Periplocae cortex in Chinese herbal market. These herbal medicines are sometimes circulated as same herbal medicine “Ogapi”.
