Acanthopanax senticosus Reverses Fatty Liver Disease and Hyperglycemia in ob/ob Mice

  • Non-alcoholic fatty liver disease (NAFLD) is common in obesity. However, weight reduction alone does not prevent the progression of NAFLD to end-stage disease associated with the development of cirrhosis and liver disease. In a previous experiment, 50% ethanol extract of Acanthopanax senticosus stem bark (ASSB) was found to reduce body weight and insulin resistance in high fat diet-induced hyperglycemic and hyperlipidemic ICR mice. To evaluate the anti-steatosis action of ASSB, insulin-resistant ob/ob mice with fatty livers were treated with ASSB ethanol extract for an 8 week-period. ASSB ethanol extract reversed the hepatomegaly, as evident in reduction of % liver weight/body weight ratio. ASSB ethanol extract also specifically lowered circulating glucose and lipids, and enhanced insulin action in the liver. These changes culminated in inhibition of triglyceride synthesis in non-adipose tissues including liver and skeletal muscle. Gene expression studies confirmed reductions in glucose 6-phosphatase and lipogenic enzymes in the liver. These results demonstrate that ASSB ethanol extract is an effective treatment for insulin resistance and hepatic steatosis in ob/ob mice by decreasing hepatic lipid synthesis.

Effects of the Water Extract from the Stem Bark of Acanthopanax senticosus on Hyperlipidemia in Rats (가시오갈피 줄기의 물 추출물이 지질대사에 미치는 영향)

  • The effects of the water extract from the stem bark of Acanthopanax senticosus were evaluated on hyperlipidemic rats induced by lipid rich diet or poloxamer-407. The water extracts, when administered orally for 3 consecutive days in hyperlipidemic rats induced by poloxamer-407 (1 ml of 30%), was found to cause a significant decrease in plasma cholesterol and triglyceride concentrations. The water extracts, when treated orally for 5 consecutive days also showed a significant inhibition of serum total cholesterol and triglyceride level in rats treated with lipid rich diet (15% cholesterol, 1% sodium cholate and 84% com oil). HDL-cholesterol, however, was increased significantly.

  • We previously reported that the phytoprotein extracted from Acanthopanax senticosus (PA) had anti-carcinogenic anti-metastatic activity via increase of inhibition of gap junctional intercellular communication. In the present study investigated the immunomodulatory mechanism of phytoprotein isolated from the stem bark of Acanthopanax sentic (PA). PA was found to significantly stimulate macrophages producing TNF-α and IL-1β in vitro. It also showed tumori activity indicating that PA had the ability to stimulate macrophage directly. Moreover, PA induced the CDB/sup +/ CTL cy responses to recognize antigen on the B16-BL6 melanoma cells. Treatment of PA with B16-BL6 melanoma cells increased the proliferation of splenocytes compared with untreated control. These results demonstrate that PA immunomodulatory activity suggesting a useful anti-tumor agent applicable to treatment and prevention of cancer.

Separating of Falcarinol from Acanthopanax senticosus (가시오가피(Acanthopanax senticosus)에서 polyacetylene계 화합물인 falcarinol의 동정)

  • To detect falcarinol in Acanthopanax senticosus, its stem and bark were extracted with methanol fractionated with petroleum ether and diethyl ether, and separated by silica gel column chromatography, Resulting six crude samples were compared with standard falcarinol through TLC on silica gel plates. Fraction 6 showed $R_{f}$ value of 0.46 similar to that of standard falcarinol. Through analytical reverse phase HPLC/PDA, UV spectra of standard falcarinol and traction 6 recorded between 200 and 340 nm showed identical peaks and UV spectra patterns. GC/MS revealed standard falcarinol and fraction 6 have equal retention times of 7.4 and 8.5 min before and after TMS-derivatization, respectively. Standard falcarinol and fraction 6 showed more similar spectrum patterns after TMS-derivatization than before. These results confirm presence of falcarinol in A. senticosus.

Identification and Characterization of SOD Isoenzymes in Acanthopanax koreanum Plants (섬오갈피나무에서 SOD Isoenzyme의 식별 및 특성규명)

  • The isoenzyme patterns and activities of superoxide dismutase(SOD) were investigated from leaves of Araliaceae plants. Of the eight isoenzymes, two isoenzymes(SOD 4 and SOD 6) were prevalent to leaves of Araliaceae plants. The patterns of these two isoenzymes were most various in the leaves of Acanthopanax senticosus for. inermis, while their activity was highest in the leaves of A. koreanum. These two isoenzymes were respectively identified as Fe-SOD and CuZn-SOD, based on selective inhibition with KCN or$H_2O_2$. The SOD isoenzyme patterns did not differed among stem barks, root barks and leaves of A. koreanum. However, the activities of Fe-SOD and CuZn-SOD were higher in the root bark and in leaves, respectively. Both of Fe-SOD and CuZn-SOD were stable for 1 hr at 30-4$0^{\circ}C$, while unstable above 5$0^{\circ}C$.

  • 1. The Korean Acanthopanax genus includes 12 kinds consisting of 9 species and 3 forma. 2. The Korean Oga-pi which is on market sale has been used as bark for the medicinal purpose, and Oga-pi shall use Radicis Cortex. That is why it is basic rule that herbalogy shall use Radicis Cortex. 3. The origin of Oga-pi on sale is Acanthopanax sessiliflorum forma chungbunensis C.S. Yook. 4. $C_{HUNG}\;and\;N_AKAI'S}$ report on A. koreanum told us that there are brown hair on the mid-leaf junction, but in addition to it, our investigation was resulted in the fact that there are thorn along mid-rib sometimes. 5. 2 kinds of new forma are similar to A. sessiliflorum, but are different in the view-point of chemotaxonomy, compared with A. sessiliflorum. In its morphology, we can find some difference between 2 kinds of new forma and A. sessiliflorum. Our effort of examination on documents tell us that the all plants growing in the central part of our country is A. sessiliflorum forma chungbunensis C.S. Yook. The one which has thorn on both side among the plants collected in Mt. Dukyu, is called A. sessiliflorum forma nambunensis C.S. Yook. 6. A. sessiliflorum is growing in the southern part in Korea, and most Chungbu Oga-pi A. sessiliflorum forma chungbunensis in the central part of our country. For the convenience of our study, the key of Korean Acanthopanax plant is classified into, I-IV, as shown on the following items: I. No hair on both side of leaf A. Flower stalk is longer than petiole, and there are thorn under the petiole (5-7 stigma).${\cdots}A.\;sieboldianum$. B. Flower stalk is longer than petiole, or same length. The serration lie down, and the stem has short thorn (stigma is divided into 3 part).${\cdots\;\cdots}A.\;seoulense$ II. There are a lot of thorn or hair on back of leaf. A. Neuroprotective Effect of $NeuBo153^{\circledR}$ on Transient Focal Cerebral Ischemia in Rats ($NeuBo153^{\circledR}$의 중풍동물 모델에 대한 뇌신경 보호효과)

  • Objectives : The purpose of the present study is to observe the neuroprotective effect of the $NeuBo153^{\circledR}$ on transient focal cerebral ischemia in rats. Methods : $NeuBo153^{\circledR}$ was made by mixing the herbs, mainly the root of Panax ginseng, the root of Rehmannia glutinosa and Poria cocos, the stem bark of Acanthopanax senticosus, the root of Scutellaria baicalensis and Mel, and heating for 96 hours. Transient Focal cerebral ischemia (2 h of ischemia, 22 h of reperfusion) was induced by intraluminal suture method with SD rats. Sensory motor function was tested by rotarod test, prehensile traction test, beam balance test and foot fault test at 24 h after ischemia. The brain slices were stained by 2% 2, 3, 5-triphenyltetrazolium chloride and the infarct volume was measured by graphic analyzer at 24 h after ischemia. Results : $NeuBo153^{\circledR}$ treated group did not show significant differences compared with vehicle treated group in body temperature. Oral administration of $NeuBo153^{\circledR}$ reduced brain infarct volume by 29.7% compared with vehicle treated group. $NeuBo153^{\circledR}$ also showed protective effects on sensory motor functional deficits. Conclusion : $NeuBo153^{\circledR}$ treatment reduced brain damage and improved functional deficits induced by MCAo. It showed neuroprotective effects even when treatment was relayed 2 h after injury. Further research is required to evaluating long term functional recovery am accurate therapeutic range and mechanisms.
