Analysis of Weathering Sensitivity by Swelling of Domestic Highway Sites (국내 고속도로현장의 스웰링에 의한 풍화민감도 분석)

  • This study aims to observe the swelling representative rocks in Korea and to suggest improvements in the use of test methods and prior analysis in relation to the weathering of rocks. The swelling test and analysis were performed on the drilling cores obtained for the ground investigation at the domestic highway construction site. For the method of determining the absorption expansion index of rocks, the method proposed in "Standard Methods for Sample Collection and Specimen Preparation" of ISRM and Korean Rock Engineers Standard Rock Test Method was used. The specimen for the measurement of the expansion displacement was cylindrical with a height of 10 cm and a diameter of 5 cm. The existing swelling analysis method evaluates the sensitivity to weathering by using the maximum expansion displacement, but since the classification by bedrock grade is unclear, it is reasonable to use the rate of change of the expansion displacement according to the immersion time. It is necessary to conduct an experiment to distinguish between weathering and fault deterioration. In addition, long-term weathering prediction technology for each cancer type is needed through the expansion displacement analysis of the chemical weathering stage.

Experimental Study of the Oven Dried Soil Weight Measurement Using Singularity Analysis (특이점 분석에 의한 노건조된 흙의 건조무게 측정에 관한 실험적 연구)

  • The dry unit weight of the soil can be obtained by measuring the weight of the oven dried soil. According to the Korean Industrial Standards (KS F), oven dried soil at 110±5℃ continuously increases in weight when exposed to air. However, there is no regulation on the weight measurement time of oven dried soil, making it difficult to accurately measure the dry weight. In this study, a method to easily determine the dry weight was presented through the analysis of the weight singularity that appears during weight measurement. The weight singularity represents the smallest value among the measured weights, and is the weight at which the effect of moisture absorption in the air of the sample is minimized. In the course of the experiment, a container was selected using a statistical analysis method, and a photograph of the soil samples were presented using an optical microscope. In addition, the temperature of the weight singularity was measured using a non-contact infrared thermometer. As a result of analyzing the weight singularity of six types of soil, including Jumunjin sand, Naeseong stream sand, Yecheon weathered granite soil, Jeju sand, Sabkha sand, and Ulleung sand, the weight singularity of oven dried sample appeared between 8 and 27 seconds after weight measurement, and the temperature ranged from 103.4 to 108.13℃. The weight decrease rate of the singularity was 0.0066 to 0.0085% depending on the soil samples.

Environmental Maintenance Technology for Concrete Manufacturing Industry by Using an Automatic Fugitive Dust Reduction System (비산먼지 자동 저감시스템을 이용한 콘크리트 제조업 환경 유지관리 기술)

  • Fine dust is a cause of serious ecological problems, and fugitive dust generated from construction sites is a major source of fine dust in Korea. However, at construction sites, including concrete manufacturing industry sites, measurements are rarely made at the fugitive dust generation stage, and passive removal methods are the majority. Therefore, in this study, a fugitive dust measurement method suitable for managing fugitive dust generated during aggregate unloading in the concrete manufacturing industry sites was selected. In addition, the purpose was to analyze the amount of fugitive dust reduction according to the operation of the reduction system by applying the automatic fugitive dust reduction system to the aggregate unloading site. As a result, the reliability of the light scattering method was secured through the comparative measurement of the beta-ray absorption method and the light scattering method, and the light scattering method correction coefficient was calculated and applied to the measured value of the fugitive dust particle mass concentration at the concrete manufacturing industry sites. In addition, the fugitive dust reduction rate according to the operation of the automatic fugitive dust reduction system was derived.

Application of Effective Earthquake Force by the Boundary Reaction Method and a PML for Nonlinear Time-Domain Soil-Structure Interaction Analysis of a Standard Nuclear Power Plant Structure (원전구조물의 비선형 시간영역 SSI 해석을 위한 경계반력법에 의한 유효지진하중과 PML의 적용)

  • Considering the non-linear behavior of structure and soil when evaluating a nuclear power plant's seismic safety under a beyond-design basis earthquake is essential. In order to obtain the nonlinear response of a nuclear power plant structure, a time-domain SSI analysis method that considers the nonlinearity of soil and structure and the nonlinear Soil-Structure Interaction (SSI) effect is necessary. The Boundary Reaction Method (BRM) is a time-domain SSI analysis method. The BRM can be applied effectively with a Perfectly Matched Layer (PML), which is an effective energy absorbing boundary condition. The BRM has a characteristic that the magnitude of the response in far-field soil increases as the boundary interface of the effective seismic load moves outward. In addition, the PML has poor absorption performance of low-frequency waves. For this reason, the accuracy of the low-frequency response may be degraded when analyzing the combination of the BRM and the PML. In this study, the accuracy of the analysis response was improved by adjusting the PML input parameters to improve this problem. The accuracy of the response was evaluated by using the analysis response using KIESSI-3D, a frequency domain SSI analysis program, as a reference solution. As a result of the analysis applying the optimal PML parameter, the average error rate of the acceleration response spectrum for 9 degrees of freedom of the structure was 3.40%, which was highly similar to the reference result. In addition, time-domain nonlinear SSI analysis was performed with the soil's nonlinearity to show this study's applicability. As a result of nonlinear SSI analysis, plastic deformation was concentrated in the soil around the foundation. The analysis results found that the analysis method combining BRM and PML can be effectively applied to the seismic response analysis of nuclear power plant structures.

Evaluation of porcine urine-derived cells as nuclei donor for somatic cell nuclear transfer

  • Background: Somatic cell nuclear transfer (SCNT) is used widely in cloning, stem cell research, and regenerative medicine. The type of donor cells is a key factor affecting the SCNT efficiency. Objectives: This study examined whether urine-derived somatic cells could be used as donors for SCNT in pigs. Methods: The viability of cells isolated from urine was assessed using trypan blue and propidium iodide staining. The H3K9me3/H3K27me3 level of the cells was analyzed by immunofluorescence. The in vitro developmental ability of SCNT embryos was evaluated by the blastocyst rate and the expression levels of the core pluripotency factor. Blastocyst cell apoptosis was examined using a terminal deoxynucleotidyl transferase dUTP nick end-labeling assay. The in vivo developmental ability of SCNT embryos was evaluated after embryo transfer. Results: Most sow urine-derived cells were viable and could be cultured and propagated easily. On the other hand, most of the somatic cells isolated from the boar urine exhibited poor cellular activity. The in vitro development efficiency between the embryos produced by SCNT using porcine embryonic fibroblasts (PEFs) and urine-derived cells were similar. Moreover, The H3K9me3 in SCNT embryos produced from sow urine-derived cells and PEFs at the four-cell stage showed similar intensity. The levels of Oct4, Nanog, and Sox2 expression in blastocysts were similar in the two groups. Furthermore, there is a similar apoptotic level of cloned embryos produced by the two types of cells. Finally, the full-term development ability of the cloned embryos was evaluated, and the cloned fetuses from the urine-derived cells showed absorption. Conclusions: Sow urine-derived cells could be used to produce SCNT embryos.

Physiological response of red macroalgae Pyropia yezoensis (Bangiales, Rhodophyta) to light quality: a short-term adaptation

  • Light quality is a common environmental factor which influences the metabolism of biochemical substances in algae and leads to the response of algal growth and development. Pyropia yezoensis is a kind of economic macroalgae that naturally grows in the intertidal zone where the light environment changes dramatically. In the present study, P. yezoensis thalli were treated under white light (control) and monochromatic lights with primary colors (blue, green, and red) for 14 days to explore their physiological response to light quality. During the first 3 days of treatment, P. yezoensis grew faster under blue light than other light qualities. In the next 11 days, it showed better adaptation to green light, with higher growth rate and photosynthetic capacity (reflected by a higher rETRmax = 61.58 and Ek = 237.78). A higher non-photochemical quenching was observed in the treatment of red light than others for 14 days. Furthermore, the response of P. yezoensis to light quality also results in the difference of photosynthetic pigment contents. The monochromatic light could reduce the synthesis of all pigments, but the reduction degree was different, which may relate to the spectral absorption characteristics of pigments. It was speculated that P. yezoensis adapted to a specific or changing light environments by regulating the synthesis of pigments to achieve the best use of light energy in photosynthesis and premium growth and metabolism.

  • Trimethylamine (TMA) is one of the difficult odorous compounds for the collection and analysis. Although sulfuric acid absorption and/or sulfuric acid impregnated filter method are commonly recommended for its sampling, these methods also suffer from difficulties involved in sample treatment and operational procedures. Hence, as an ancillary approach to measure TMA, we investigated the combination of bag sampling and SPME analysis for TMA measurements. For the purpose of our study, we investigated the following three subjects: 1) temporal variability of standard storage, 2) bag loss effect of TMS, and 3) TMA loss due to repetitive analysis of an identical bag sample. According to our storage test up to 7 or 20 dyas, TMA loss were found to occur up to 40 to 50% within relatively short period of up to 48 hrs depending on its concentration ranges. When the tests were made for bag loss by transferring TMA standards across different size bags, we were able to find that the extent of bag loss are not significant with 5 to 20% loss rate. Finally, the TMA sorptive loss via its exposure to SPME fiber was generally estimated to run from 2 to 3%.

Evaluation of engineering characteristics and field applicability of inorganic thixotropic-grout for backfilling of shield TBM tail voids (쉴드 TBM 뒤채움용 무기계 가소성 그라우트의 공학적 특성 및 현장적용성 평가)

  • The focus of this study is to evaluate the field applicability of the newly developed inorganic thixotropic-grout in various ways. In order to do this, the volume stability and the permeability of the inorganic thixotropic-grout have been measured and compared to the existing silica type grouts. In addition, the filling capability of the grout into the tail void has been evaluated through both an experiment with a miniature tail-void filling equipment and a test filling at the shield TBM construction site. The volume loss of the inorganic thixotropic-grout after a 14 day-curing under the atmosphere condition was appeared to be minimal. The excellent waterproofing ability of the inorganic thixotropic-grout was confirmed through a permeability test. The toxicity of the inorganic thixotropic-grout has been evaluated through a toxicity test of aquatic fishes (KS M 0111) and the pH value of the liquid eluviated from inorganic thixotropic-grout was measured as an average of 8.0 and a fatality rate of goldfish after 96 hours was 10% or so. The existence of harmful heavy metals in the liquid eluviated from the inorganic thixotropic-grout has been also examined through an atomic absorption spectroscopy (AAS) test. Any of harmful heavy metals were not detected and the detected level of $Cr^{6+}$ and Cd was far below the standard. Based on both an experiment with a miniature tail-void filling equipment and a test filling at the shield TBM construction site, the filling ability of inorganic thixotropic-grout into the tail void was proved to be excellent.

Calculation of the Theoretical Total Amount Ratio of Di(2-ethyl-hexyl) Phthalate in Indoor Air and Floor Dust in a Test House (모의 실험주택 모니터링 결과를 활용한 실내공기 및 바닥먼지 중 Di(2-ethyl-hexyl) phthalate (DEHP)의 이론적 총량 비율 산출)

  • Background: Human exposure to phthalates in indoor environments occurs via dermal absorption, oral ingestion of indoor dust, and inhalation of indoor air. However, systematic studies to investigate the exposure rate to phthalates among the three exposure routes in indoor environments are currently limited. Objectives: A theoretical exposure ratio between inhalation and oral exposure was calculated based on the total amount of di(2-ethyl-hexyl) phthalate (DEHP) emitted into indoor air and deposited into floor dust in a test house. Methods: Flooring and wallpaper containing DEHP were installed in a test house and the concentration of DEHP in both indoor air and floor dust were monitored for five months. Based on the measured DEHP concentrations, the theoretical total amount ratio of DEHP that could be exposed through inhalation and oral ingestion was calculated. Results: Considering the period of operation in the test house, the theoretical total amount of DEHP through inhalation and oral ingestion exposures in the entire test house space was calculated to be 0.014 mg and 5.5 mg, respectively. The exposure ratio of the two routes between inhalation and oral exposure corresponding to the total DEHP amount in flooring and wallpaper was 6.0×10-7% and 2.3×10-4%, indicating that theoretical oral exposure to DEHP is approximately 380 times higher than inhalation. Conclusions: Monitoring results from a test house has shown that oral exposure is the main exposure route for DEHP in indoor environments. The experimental design employed in this study and theoretical exposure ratio obtained can be applied to investigate actual exposure to DEHP and to determine the exposure characteristics of various types of semi-volatile organic compounds.

The Relationship between Image Parameters and SAR for Each Sequence of MRI (MRI 검사의 시퀀스 별 영상 변수와 SAR의 관계)

  • This study analyzed the relationship between image parameters and specific absorption rate (SAR) in various sequence environments to optimize SAR. For this purpose, image parameters were adjusted for T2, T1, STIR, T1 FLAIR, and T2 FLAIR sequences in a 3.0T MRI, and the whole body (WB) SAR and head SAR calculated by the device were measured. Then, the SAR was evaluated by adjusting the number of images and the flip angle (FA) of the refocusing RF. As a result, SAR increased as the number of image increased in all sequences. T1 and T1 FLAIR had correlation coefficients (r) of 0.876, 0.876 (WB SAR, head SAR), 0.867, 0.867 (WB SAR, head SAR), respectively, and STIR had the highest correlation with 0.898 and 0.899 (WB SAR, head SAR). showed (p<0.05). When applied by increasing the refocusing FA, WB SAR and head SAR increased overall in all sequences. The T1 and T2 sequences showed high correlation with correlation coefficients (r) of 0.897, 0.898 (WB SAR, head SAR) and 0.914, 0.915 (WB SAR, head SAR), respectively, while the sequences to which the inversion recovery technique was applied had relatively low FA, showed less sensitivity to increase. Therefore, in a sequence with a relatively low TR, minimizing the number of image and applying the fast spin echo to reduce the refocusing FA in a sequence with a high duty cycle are effective in reducing SAR.
