  • A new method for micro-quantification of abscisic acid from plant tissue is described. GLC-ECD method of abscisic acid pertafluorobenzyl ester synthesized by reaction of plant extract with pentafluorobenzyl bromide showed higher sensitivity than that of abscisic acid methyl ester, and the detection limit of abscisic acid by the described method was as low as 5 pg.

  • In the barly coleptile sections treated with either $1{\times}10^{-5}M$ abscisic acid (ABA) or $1{\times}10^{-5}M$ gibberellic acid (GA), the time course changes of ribonuclease (RNase) activity and ribonucleic acid (RNA) profiles were studied. The results may be summarized as follows: 1. While GA suppressed RNase activity, ABA activated it. 2. High level of s-RNA and low level of r-RNA compared with normal plant sections in hormone-untreated coleoptiles seemed to be the results of increased RNase activity in the incubation period. 3. While GA retarded the decomposition of r-RNA, ABA activated it and the results seemed to be related with RNase activity. 4. GA activated the synthesis of RNA-DNA component, and ABA suppressed it. 5. Increase in the amount of s-RNA with the treatment of ABA may be due to the decomposition of r-RNA.

Analysis of the Change of Amino Acids by Abscisic acid and Polyamine Treatment in Radish Young Cotyledons (무우 유식물의 자엽에서 Abscisic acid와 Polyamine 처리에 의한 아미노산 변화의 분석)

  • The changes of amino acids in the cotyledons of spring - and winter radishes were analysed during the abscisic acid (ABA). ABA plus cycloheximide (CH) and ABA plus polyamine (PA) treatment. The total contents of amino acids were increased in spring radishes, and decreased in winter radishes by ABA. The contents of hydrophilic amino acids, proline, glycine, serine and cysteine in spring radishes (expecially proline), and of cysteine, leucine and phenylalanine in winter radishes (expecially cysteine and leucine)were increased, CH treatment resulted in the accumulation of amino acids by the inhibition of new synthesized protein, which synthesized against ABA induced dehydration both of radishes. On the contrary, the contents of amino acids were decreased in spring radishes, and increased in winter radishes during PA treatment. These results indicated that the accumulated hydrophilic amino acids and new synthesized proteins induced the adaptation of dehydration against stress and the role of ABA in accelerating of stress adaptation was mediated by polyamines.

  • This research aims at developing a new plant growth inhibitors related to abscisic acid by means of esterification of (S)-(+)-ABA with p-hydroxy methyl cinnamate and umbelliferone, and testing its biological activity on growth of rice seedlings. The over-all yield of ABA-methyl cinnamate(AC) and ABA-umbelliferone(AC) ester compounds were 83% and 78%, respectively. The growth inhibition activity of these synthetic compounds were shown about 3 to 10 times(AC) and 10 to 30 times(AU) higher than (S)-(+)-ABA.

  • Evolution of adaptive mechanisms to abiotic stress is essential for plant growth and development. Plants adapt to stress conditions by activating the abscisic acid (ABA) signaling pathway. It has been suggested that the ABA receptor, clade A protein phosphatase, SnRK2 type kinase, and SLAC1 anion channel are important components of the ABA signaling pathway. In this study, we report that the shaker type potassium (K+) channel, GORK, modulates plant responses to ABA and abiotic stresses. Our results indicate that the full length of PP2CA is needed to interact with the GORK C-terminal region. We identified a loss of function allele in gork that displayed ABA-hyposensitive phenotype. gork and pp2ca mutants showed opposite responses to ABA in seed germination and seedling growth. Additionally, gork mutant was tolerant to the NaCl and mannitol treatments, whereas pp2ca mutant was sensitive to the NaCl and mannitol treatments. Thus, our results indicate that GORK enhances the sensitivity to ABA and negatively regulates the mechanisms involved in high salinity and osmotic stresses via PP2CA-mediated signals.

Growth and Abscisic Acid Changes of Creeping Thyme in the Exposure of NaCl and Drought (염 및 건조스트레스 하에서 포복형 백리향의 생육과 Abscisic Acid 농도변화)

  • Experimental purpose was to evaluate growth characteristic and abscisic acid (ABA) responses against salt/drought stresses. In the shoot biomass, creeping thyme was tolerated in mild NaCl stress, ranging 0 to 100 mM, while it was severely reduced in higher salinity. Under constant drought stress, the shoot biomass of creeping thyme showed a worse value compared to that of 100 mM NaCl treatment. Chlorophyll degradation was more severe in immature leaf than mature leaf under salt and drought stresses. In salt stress, immature leaf produced much amounts of ABA compared to mature leaf and also immature leaf showed faster increase of ABA than that of mature leaf. In drought stress, immature leaf responded to stress within 24 hours by the increase of ABA, while mature leaf responded to at 72 hours. Our results recommended that the optimal salinity level of creeping thyme was 50~100 mM NaCl.

  • A radioimmunoassay technique has been developed for the determination of abscisic acid (ABA) in crude extracts from germinating rice (Oryza sativa L.). By this method, the changes in ABA level of rice during germination was investigated. The ABA content in rice seeds was found to be 76.5ng/g dry weight in Dong-jin variety and 91.1ng/g dry weight in Sam-gang variety. A rapid decrease in ABA content of rice occurred during germination within 24 hours after seed imbition. The decreasing rate of ABA content during germination showed a significant direct proportion to the imbibition temperature and water-absorbing rate of rice. The decrease in ABA content during germination was found to be caused partly by an elution of ABA from the tissue to the imbibing fluid, and partly by a metabolic conversion of ABA to another compounds. The germination process of rice occurred only when the tissue ABA level decreased below a certain level, and the decreasing rate of ABA level during germination correlated with the ability for germination at low temperature of rice.

Effects of Abscisic Acid (ABA) and Fluridone on Red Coloration of 'Hongro' Apple Fruit Skins (Abscisic acid(ABA) 및 fluridone의 처리가 'Hongro' 사과의 과피 착색에 미치는 영향)

  • The objective of this study was to determine the effect of exogenous abscisic acid (ABA) on the red coloration and endogenous ABA contents of apple fruit skins. ABA and fluridone (an ABA synthetic inhibitor, FD) was sprayed on 'Hongro' apple fruit skins at 107 days after full bloom (DAFB). Visual coloration and hunter's color values were not affected by the ABA and FD treatments. Anthocyanin contents in fruit skins increased similarly to hunter $a^*$ values of fruit skins, but ABA and FD did not affect its accumulations. Liquid chromatography analysis revealed that endogenous ABA contents in control fruit increased at first and then decreased from 12 hours after the treatment. ABA treatment increased ABA contents in fruit skins from 2 hour after the treatment and it lasted until the end of the treatments. FD decreased ABA contents in fruit skins from 6 hours after the treatment. ABA treatment increased MdNCED2 (an ABA biosynthetic gene), MdACO1 (an ethylene biosynthetic gene), and MdCHS and MdDFR expressions. However, MdUFGT expressions were not affected by ABA treatment.

  • Wheat coleoptile sections were treated with either $1.5{\times}10^{-5}M$ ABA or $5×10^{-5}M$ IAA in vitro, the results may be summarized as follows, 1. The treatmert of IAA decreased the level of high molecular weight RNA F2 and F3 but that with ABA increased the F4 level. 2. IAA caused an increased activity of G2 isozyme, while ABA suppressed the activity of G3 isozyme. 3. The results may suggest that there may exist common effects of IAA and ABA on RNA and RNase. 4. The latent RNase activity caused by SH blocking reagent (p-hydroxymercury benzoate, Pb et al) was not observed.

Effects of Abscisic acid on Ozone Injury in Rice (벼의 오존피해 경감에 미치는 ABA의 효과)

  • This study was carried out to investigate the effects or abscisic acid (ABA), three concentrations, on growth, activities of antioxidant-related enzymes and grain yield of two rice plants (Oryza sativa L. cv. Hwamyungbyeo and cv. Namchunbyeo) exposed to ozone (0.150 ppm) for 6 hours a day for 30 days. The leaf chlorop0hyll contents, plant height, and tillering numbers were not shown significant difference. But ABA 10$^{-5}$ M treatment affects to growth slightly. In all concentrations of ABA, superoxide dismutase (SOB) activities were increased at tillering stage of Namchunbyeo which was exposed to ozone. It is considered that the optimum concentration of ABA is 10$^{-5}$ M for minimizing loss of grain yield loss.
