다양한 스트레스에 유도되는 들잔디 ZjWRKY3, ZjWRKY7의 분석 (Analysis of ZjWRKY3, ZjWRKY7 induced by multiple stress in Zoysia japonica)

  • 식용작물, 사료, 잔디를 포함하는 모든 작물은 건조, 염, 저온, 고온 등의 여러 가지 환경스트레스의 영향을 빈번히 받기 때문에 작물의 생산성이 떨어지게 된다. 식물은 환경스트레스 상황에서 스스로 벗어날 수 없다. 따라서 식물은 환경 스트레스를 극복하는 방향으로 진화하였다. ARF, ABI3, NAC, HSF, WRKY 같은 환경 스트레스에 반응하는 유전자들이 식물에서 보고되었다. 이 유전자들은 환경스트레스에 반응하는 전사인자로, 식물의 스트레스반응 경로에 연관되어 있다. OsWRKY76의 경우에는 저온 및 병원균에 대한 내성을 증가시켰고, AtWRKY28 의 경우 여러 가지 환경스트레스에 관련이 있는 것으로 보고되었다. 들잔디는 정원이나 골프코스에서 가장 흔하게 사용되는 잔디이다. 하지만 들잔디에서는 아직 WRKY 유전자가 알려지지 않았다. 본 연구에서는 들잔디로부터 1개의 WRKY domain을 포함하는 ZjWRKY3, ZjWRKY7 를 분리하였다. ZjWRKY3과 ZjWRKY7은 저온, 건조, 염 스트레스에 발현이 증가하였다. 들잔디의 갈색퍼짐병을 일으키는 R. solani의 감염이 ZjWRKY3과 ZjWRKY7의 발현을 증가시켰다. 또한 ZjWRKY3, ZjWRKY7이 Zjchi 유전자 promoter의 W-box에 결합하여 전사를 조절한다는 사실을 확인 하였다. 따라서 ZjWRKY3, ZjWRKY7 유전자는 전사인자로서 환경스트레스 및 병원균 관련 하위 유전자들을 조절할 것으로 예상된다.

고온 및 건조 스트레스 조건 하에서 살리실산 경엽처리에 의한 고추의 광합성 특성 및 항산화효소 활성 증대 (Enhancement of Photosynthetic Characteristics and Antioxidant Enzyme Activities on Chili Pepper Plants by Salicylic Acid Foliar Application under High Temperature and Drought Stress Conditions)

  • 살리실산은 식물의 생장 및 발달, 항산화 방어기작, 광합성 작용 그리고 생물적 및 비생물적 스트레스 조건에서 다양한 생리적 기능을 조절하는 물질로 알려져 있다. 본 연구에서는 고온·건조 스트레스 조건에서 살리실산 경엽처리가 고추의 생육, 광합성 특성 및 항산화효소 활성에 미치는 영향을 구명하고자 하였다. 광합성 특성 측정결과 광합성 속도, 기공전도도 및 증산 속도가 증가하였고, 3회차 처리에서 가장 높았다. 세포내 MDA와 H2O2 함량은 살리실산 3회차 처리에서 현저하게 감소하는 경향을 보였다. APX, CAT, POD 및 SOD 활성이 현저하게 증가하였으며, 무처리 대비 최대 247, 318, 55 및 54% 증가하였다. 고추의 생육 특성은 무처리구와 유의한 차이를 보이지 않았으나, 상품 수량은 15% 정도 증가하였다. 이러한 결과들을 종합해 볼 때, 살리실산의 경엽처리는 고추의 광합성 특성과 항산화효소 활성을 증진시켜 고온·건조 스트레스에 의한 피해 경감에 긍정적 효과를 유발함을 확인하였다.

새싹보리 재배기간 중 수분스트레스 처리가 사포나린 함량에 미치는 영향 (Influence of Drought Stress Treatment on Saponarin Content during the Growing Period of Barley Sprouts)

  • BACKGROUND: Barley sprouts contain a large number of secondary metabolites such as polyphenols, saponarin, and policosanols. The synthesis of such secondary metabolites occurs as a defense mechanism against external environmental stresses. In particular, it has been widely known that drought stress (DS) increases the content of flavonoids in plants. The objective of this study was to investigate the effects of drought stress treatment on the saponarin content in barley sprouts during the growing period. METHODS AND RESULTS: In this study, changes in saponarin content with different DS exposure periods and times were evaluated under the hydroponic system. For establishing different DS treatment periods, water supply was stopped for 1, 2, and 3 days, once leaf length was at 10 cm. To control different DS treatment times, water supply was stopped for 2 days, once leaf lengths were 5, 10, and 15 cm. As a result, the water potential of barley sprouts decreased from -0.8 MPa (before DS treatment) to -1.2, -2.4, and -3.2 MPa (after DS treatment), and reversely recovered to -0.8 MPa after re-irrigation. When 10 cm leaves were subjected to DS for 1 and 2 days, the saponarin content increased by 12 and 10%, respectively, while it increased by 19% when DS was applied to the 5 cm leaves. CONCLUSION(S): The results of this study suggest that drought stress at the early stage of growth (5 cm) is most helpful to increase the saponarin content of barley sprouts.

규산비료 시용이 배추 묘의 생장과 환경내성에 미치는 영향 (Effect of Silicate Fertilizer on Growth, Physiology and Abiotic Stress Tolerance of Chinese Cabbage Seedlings)

  • 규산은 작물의 필수원소에는 포함되어있지 않으나, 화본과 작물을 중심으로 내도복성과 병충해 저항성의 향상, 군락구조 개선에 의한 광합성 능력의 향상 등에서 폭 넓게 그 유용성이 알려져 왔으며, 최근에는 원예작물에서도 규산질 비료의 시용이 수량이나 병충해저항성을 향상시키는 효과가 입증되고 있어 친환경농업 관점에서도 주목을 밭고 있다. 본 실험은 배추 육묘 중 규산질 비료의 시용이 묘소질과 저온, 고온, 건조 등 환경내성에 미치는 영향을 검토하기 위하여 수행하였다. 규산염 처리농도를 8, 16, 32, 64 및 128mM로 설계하여 주 2회 관주 처리 하고, 처리 3주 후에 생육조사 및 스트레스 내성에 대해 평가하였다. 생육조사 결과, 8, 16 및 32m의 농도에서는 대부분의 생육지표가 대조구에 비해 약간 증가하는 경향을 보였으나 8mM처리만 제외하고 통계적 유의차는 나타나지 않았다. 고농도인 128mM의 규산 처리구에서는 모든 생육 지표가 감소하였다. 총 뿌리 면적, 뿌리 길이 및 근단 수는 8, 16 및 32mM의 농도에서 증가했지만 64 및 128mM의 처리구에서는 감소하였다. 규산 처리 농도가 증가함에 따라 증산 속도는 감소한 반면 기공확산 저항은 증가하는 경향을 보였다. 상대적 이온 누출율도 대조구에 비해 규산염 처리구에서 감소되었으나, 처리 농도간 유의차는 나타나지 않았다. 규산처리에 의해 고온과 저온 장해 지표도 감소되었으며, 농도간에는16과 32mM이 가장 효과적이었다. 규산처리에 따라 건조내성도 증가하여 대조구는 단수 후 3일째부터 위조되기 시작하여 5일째는 전 개체가 위조하였으나, 규산처리구는 4일(8, 64, 128 mM) 또는 5일(16과 32mM) 부터 위조가 시작되어 6일(8mM)이나 7일(16, 32, 64및 128 mM)이 지나서야 모든 공시 개체가 위조되었다.

Assessment of Endophytic Fungal Diversity and Beyond

  • Endophytic fungi are microorganisms inhabiting living plant tissues without causing apparent harm to the host. They are drawing increasing attention due to their ability to produce various bioactive compounds as well as their effects on host growth and resistance to biotic and abiotic stresses. As a first step to assess biodiversity of plant associated fungi in Korea and the following evaluation on diverse biological activities, we are collecting endophytic fungi from plant in wild followed by systematic long-term storage in liquid nitrogen. Molecular identification using ITS sequences was also incorporated for pure culture by hyphal tip isolation. As of April 2015, about 1,400 fungal strains had been isolated from about 170 plant taxa. Fungal isolates belonging to Pleosporales, Diaporthales, Glomerellales, Hypocreales, and Xylariales were the most abundant. These collections are being used for several complementary researches, including screening of isolates with novel bioactive compounds or conferring drought stress resistance, phylogenetic and genomic study. Genome sequencing was performed for 3 isolates, one Xylaria sp. strain JS573 producing griseofulvin, an antifungal compound, and two Fusarium spp. strains JS626 and JS1030, which are assumed to be new species found in Korea. More detailed analysis on these genomes will be presented. These collections and genome informations will serve as invaluable resources for identifying novel bioactive materials in addition to expand our knowledge on fungal biodiversity.

Tyrosine phosphorylation as a signaling component for plant improvement

  • Plant genome analyses, including Arabidopsis thaliana showed a large gene family of plant receptor kinases with various extracellular ligand-binding domain. Now intensively studies to understand physiological and cellular functions for higher plant receptor kinases in diverse and complex biological processes including plant growth, development, ligands perception including steroid hormone and plant-microbe interactions. Brassinosteroids (BRs) as a one of well know steroid hormone are plant growth hormones that control biomass accumulation and also tolerance to many biotic and abiotic stress conditions and hence are of relevance to agriculture. BRI1 receptor kinase, which is localized in plasma membrane in the cell sense BRs and it bind to a receptor protein known as BRASSINOSTEROID INSENSITIVE 1 (BRI1). Recently, we reported that BRI1 and its co-receptor, BRI1-ASSOCIATED KINASE (BAK1) autophosphorylated on tyrosine residue (s) in vitro and in vivo and thus are dual-specificity kinases. Other plant receptor kinases are also phosphorylated on tyrosine residue (s). Post-translational modifications (PTMs) can be studied by altering the residue modified by directed mutagenesis to mimic the modified state or to prevent the modification. These approaches are useful to not only characterize the regulatory role of a given modification, but may also provide opportunities for plant improvement.

Developing genetic resources for pre-breeding in Brassica oleracea L.: an overview of the UK perspective

  • The vegetable brassicas are an important crop worldwide and are of significant commercial value. In order to ensure our targets for food security are met it is important that these crops are continually improved to increase sustainability of production, increase nutritional quality and reduce waste. Development of resistances against both biotic and abiotic stress are recognised as being key. Plant breeding plays a vital role in addressing these issues through the development of new and improved varieties. This continued improvement is becoming evermore dependent on our ability to identify and introgress beneficial alleles from 'exotic' germplasm into elite breeding material. Increasingly, more diverse germplasm such as those found in genebanks is being screened for benificial allelic variation, however, plant breeders often find it difficult to make use of such material due to the time required to remove undesirable characteristics from progeny due to linkage drag. This article describes how we have attempted to overcome this and develop resources that make the diversity available within the $Brassica$ $oleracea$ genepool more accessible.

작물의 생합성 칼슘 함량 증대 연구 현황 (Current status in calcium biofortification of crops)

  • Calcium is an essential nutrient for living organisms, with key structural and signaling roles. Its deficiency in plants can result in poor biotic and abiotic stress tolerance as well as reduced crop quality and yield. Calcium deficiency in humans causes various diseases such as osteoporosis and rickets. Biofortification of calcium in various food crops has been suggested as an economic and environmentally advantageous method to enhance human intake of calcium. Recent efforts to increase the levels of calcium in food crops have used calcium/proton antiporters ($CAXs$) and modified one to increase calcium transport into vacuoles through genetic engineering. It has been reported that overall calcium content of transgenic plants has been increased in their edible portions with some adverse effects. In conclusion, biofortification of calcium will add more value in crops as well as will be beneficial for animal and human. Therefore, more fundamental studies on the mechanisms of calcium ion storage and transporting are essential for more effective calcium biofortification.

Alternaria in Food: Ecophysiology, Mycotoxin Production and Toxicology

  • Alternaria species are common saprophytes or pathogens of a wide range of plants pre- and post-harvest. This review considers the relative importance of Alternaria species, their ecology, competitiveness, production of mycotoxins and the prevalence of the predominant mycotoxins in different food products. The available toxicity data on these toxins and the potential future impacts of Alternaria species and their toxicity in food products pre- and post-harvest are discussed. The growth of Alternaria species is influenced by interacting abiotic factors, especially water activity ($a_w$), temperature and pH. The boundary conditions which allow growth and toxin production have been identified in relation to different matrices including cereal grain, sorghum, cottonseed, tomato, and soya beans. The competitiveness of Alternaria species is related to their water stress tolerance, hydrolytic enzyme production and ability to produce mycotoxins. The relationship between A. tenuissima and other phyllosphere fungi has been examined and the relative competitiveness determined using both an Index of Dominance ($I_D$) and the Niche Overlap Index (NOI) based on carbon-utilisation patterns. The toxicology of some of the Alternaria mycotoxins have been studied; however, some data are still lacking. The isolation of Alternaria toxins in different food products including processed products is reviewed. The future implications of Alternaria colonization/infection and the role of their mycotoxins in food production chains pre- and post-harvest are discussed.

THE CLASSIFICATION SYSTEM OF RIVER HEALTH FOR THE ENVIRONMENTAL WATER QUALITY MANAGEMENT

  • South Africa has developed a policy and law that calls and provides for the equitable and sustainable use of water resources. Sustainable resource use is dependent on effective resource protection. Rivers are the most important freshwater resources in the country, and there is a focus on developing and applying methods to quantify what rivers need in terms of flow and water quality. These quantified and descriptive objectives are then related to specified levels of ecological health in a classification system. This paper provides an overview of an integrated and systematic methodology, where, fer each river, and each river reach, the natural condition and the present ecological condition are described, and a level/class of ecosystem health is selected. The class will define long term management goals. This procedure requires each ecosystem component to be quantified, starting with the abiotic template. A modified flow regime is modelled for each ecosystem health class, and the resultant fluvial geomorphology and hydraulic habitats are described. Then the water chemistry is described, and the water quality changes that are likely to occur as a consequence of altered flows are predicted. Finally, the responses to the stress imposed on the biota (fish, invertebrates and vegetation) by modified flow and water quality are predicted. All of the predicted responses are translated into descriptive and/or quantitative management objectives. The paper concludes with the recognition of active method development, and the enormous challenge of applying the methods, implementing the law, and achieving river protection and sustainable resource-use.
