Characterization of Bacillus licheniformis KJ-9 Isolated from Soil (토양으로부터 분리한 Bacillus licheniformis KJ 9의 특성)

  • In order to produce high-quality fermenting composts, a microorganism was isolated from the natural world. The bacterium has not only in high enzyme activities but also had good antimicrobial activities against phytopathogenic microorganisms. Its cultivating characteristics were then investigated. Bacterium KJ-9, which contains high CMCase, protease and chitinase activities and excellent antimicrobial activities against phytopathogenic microorganisms, was separated from leaf mold and identified as Bacillus licheniformis by two methods: Bergey's Manual of Systematic Bacteriology and API 50 CHL Carbohydrate Test Kit (Bio Merieux, France) using an ATB (Automated Identification) computer system (Bio Merieux, France). Optimal medium for cultivation of B. licheniformis was 2% soluble starch as a carbon source, 0.5% yeast extract as a nitrogen source and 0.05% $MgSO_4{\cdot}7H_2O$. Optimal growth conditions of pH, temperature and shake speed were pH 7.0, $50^{\circ}C$ and 180 rpm, respectively. Culture broth of B. licheniformis KJ-9 cultured for 36~60 hr was effective in fungicidal activities against plant pathogens including Botrytis cinerea, Corynespora cassicola, Fusarium oxysporum, and Rhizoctonia solani.

  • Thermophilic anaerobic digestion (TAD) is characterized by higher biogas production rates as a result of assumedly faster microbial metabolic conversion rates compared to mesophilic AD. It was hypothesized that the thermophilic microbiome with its lower diversity than the mesophilic one is more susceptible to disturbances introduced by alterations in the operating factors, as an example, the supply of nitrogen-rich feedstock such as poultry manure (PM). Laboratory scaled TAD experiments using cattle slurry and increasing amounts of PM were carried out to investigate the (in-) stability of the process performance caused by the accumulation of ammonium and ammonia with special emphasis on the microbial community structure and its dynamic variation. The results revealed that the moderate PM addition, i.e., 25% (vol/vol based on volatile substances) PM, resulted in a reorganization of the microbial community structure which was still working sufficiently. With 50% PM application, the microbial community was further stepwise re-organized and was able to compensate for the high cytotoxic ammonia contents only for a short time resulting in consequent process disturbance and final process failure. This study demonstrated the ability of the acclimated thermophilic microbial community to tolerate a certain amount of nitrogen-rich substrate.

  • Ground Complex, which is located at Naro Space Center, consists of Assembly Complex(AC) and Launch Complex(LC) which is necessary for successful launch of KSLV-I(NARO). AC consists of Assembly/Testing Building(ATB), Payload Processing Building(PPB), Kick Motor Building(KMB). The purpose of AC is accepting of KSLV-I components, testing, checkout, assembly(disassembly) of the launch vehicle(LV), readiness for transferring LV to LC, accepting of integrated Launch Vehicle(ILV) in case of launch cancellation and short/long time storage, and so on. Qualification tests(QT) for the total system at AC are carried out to check hardware used for operations with first stage unit mockup, upper stage unit Mockup and integrated mockup(GTV). The qualification tests is carried out according to program and procedures of QT. By course of this process, AC is certificated that all the systems and facilities of AC are guaranteed by the fulfillment of technological operations envisioned in the program of qualification tests during the work with the mock-up.

  • A circular saw is an effective tool for cutting glass and carbon-fiber hybrid composites. This study investigated tool wear and cut quality when reusing saw blades. The carbide saws wear four times faster than the new ones, and polycrystalline diamond (PCD) is very resistant to tool wear, except at the end of its lifespan. The cut cross-section quality is affected by the blade type, tool wear, and spindle speed. Alternate top bevel (ATB)-type blades are suitable for cutting fiber-reinforced plastics, but triple-chip grind (TCG)-type blades are unsuitable because they cause fiber-pullout defects. Tool wear and low spindle speeds increase the occurrence of arc scratches, due to the rear saw blade. A microscopic examination showed that the burr, which is a mixture of fiber chips and epoxy matrix, was bonded on top, and glass-fiber delamination occurred on the bottom glass-fiber-reinforced polymer (GFRP) surface.

HEN Simulation of a Controlled Fluid Flow-Based Neural Cooling Probe Used for the Treatment of Focal and Spontaneous Epilepsy

  • Brain disorders such as epilepsy is a condition that affects an estimated 2.7 million Americans, 50,000,000 worldwide, approximately 200,000 new cases of epilepsy are diagnosed each year. Of the major chronic medical conditions, epilepsy is among the least understood. Scientists are conducting research to determine appropriate treatments, such as the use of drugs, vagus nerve stimulation, brain stimulation, and Peltier chip-based focal cooling. However, brain stimulation and Peltier chip-based stimulation processes cannot effectively stop seizures. This paper presents simulation of a novel heat enchanger network(HEN) technique designed to stop seizures by using a neural cooling probe to stop focal and spontaneous seizures by cooling the brain. The designed probe was composed of a U-shaped tube through which cold fluid flowed in order to reduce the temperature of the brain. The simulation results demonstrated that the neural probe could cool a 7 $mm^2$ area of the brain when the fluid was flowing atb a velocity of 0.55 m/s. It also showed that the neural cooling probe required 23 % less energy to produce cooling when compared to the Peltier chip-based cooling system.

The Number and Distribution of Bacteria falling in the Air of Dental Clinics (치과의원 공기중의 공중낙하세균수와 세균분포)

  • The purpose of this study was to investigate the distribution of microorganisms and the degree of contamination in the air of the dental clinics and to offer basic data as to the contamination of medical equipment and the prevention of the clinics. With this in mind, the researcher gathered air samples from the waiting rooms and medical offices of nine dental clinics in the city of J, South Korea with the use of a method of natural inattention and an air sampler and cultivated the samples on the plain table and drew from it bacteria falling and separated and sorted out the colony with the help of ATB and detected the distribution of the germs. The results are following, The number of bacteria falling in the air of the dental clinics was less than 10(CFU/Plate) with the exception of one dental clinic. This implies that they fit in with standards for hygiene. The number of bacteria falling in the air of the medical offices was less than 10(CFU/Plate) with the exception of one dental clinic. This implies that they fit in with standards for hygiene. The survey on the detection of staph. aureus reveals that all the dental clinics with the exception of B dental clinic proving to be positive had non-pathogenic staphylococci detected. The survey on the detection of pathogenic gram negative bacilli indicates that all the dental clinics but one were none detected. The survey on the distribution of germs shows that germs in 7 out of 9 dental clinics were none detected, and that they in four out of 9 waiting rooms were none detected. All the germs detected in the others were mostly non-pathogenic. The study shows that all the subject dental clinics but one were hygienically controlled and that there was a difference in accordance with cleaning and sterilization. This means that dental clinics should be equipped with systematic programs for cleaning and sterilization designed to prevent infection.

Fabrication and Characteristics of Resin-Type Neutron Shielding Materials for Spent Fuel Shipping Cask (사용후핵연료 수송용기에 사용될 수지계 중성자 차폐재 제조 및 특성)

  • Resin-type neutron shielding materials, KNS-115A, 115B and 115C have been fabricated to be used for spent fuel shipping cask. The base material is epoxy resin, and polypropylene, aluminium hydroxide and boron carbide are added. These shielding materials offer good fluidity at processing, which makes it possible to apply this resin shield to complicated geometric shapes such as shipping cask. Several measurements were made for the shielding materials to evaluate the shielding property, combustion characteristics, fire resistance, thermal and mechanical properties. The neutron shielding ability of the shielding materials is estimated to be better than that of foreign's shielding material, NS-4-FR, due to higher hydrogen atomic density. Other properties of the shielding materials are as follows: Onset temperatures; $267{\sim}270^{\circ}C$, thermal conductivities; $0.62{\sim}0.72W/m{\cdot}K$, combustion characteristics; <$800^{\circ}C$, ATB(average time of burning); <5sec, AEB(average extent of burning) ; <5mm, tensile strengths; $2.3{\sim}3.0kg/mm^2$, compressive strengths; $5.3{\sim}13.3kg/mm^2$, flexural strengths; $4.4{\sim}5.4kg/mm^2$.

Studies on Bacterial Characteristics of Bacillus cereus Group LS-1 Isolated from Suyeong Bay (수영만에서 분리된 Bacillus cereus Group LS-1 의 세균학적 특성에 관한 연구)

  • These studies were carried out to identify Bacillus cereus group 1..5-] strain isolated from 5uyeong Bay. This strain was differentiated from B. cereus group using conventional, API system and fatty acid composition analysis. Colony characteristics were opague. mucoid, entire margin. convex. circular and non hemolysis on sheep blood agar plates, and were observed with central spore forming positive bacilli in a Gram stained preparation. and had no motility. The carbohydrates tested; glucose.maltose, and sucrose were assimilated but neither trehalose nor salicin were assimilated. This strain ultilized gelatin and was also inhibited by 6.5% NaCI. The results of biochemical examination were differented from B. cereus group LS-1 compared with others B. cereus group. The fatty acid composition contained major amounts of branched chain acids. iso $C_{15}$ and iso $C_{13}$ and the range of chain length was $C_{12}$ to C"$C_{17}$ and n$C_{15}$, acid was not detected. Automated fatty acid computer profile indicated "B. mycoides GC subgroup B of 0.312 similarity index." The results agreed with other research cases. On the other hand. A TB computer prolile index of API system (API 50 CHB & API 20E) identified" Doubtful profile of 99.7% B. firmus" . These results were presented with considerable discrepancies between API system and fatty acid analysis. With 67 biochemical characters. the similarity matrix of B. mycaides (KCTC 1033). B. thuringiensis (KCTC 1033). B. cereus (5-3) and B. mycoides (S-12) showed 42%. 42%. 59%, and 52%. respectively. Through the key tests and fatty acid analyses. we could notice the appearance of B. mycoides of the B. cereus group and this leads us to suspect the existence of a new biotype B. mycoides.

Fabrication and Characteristics of Epoxy Resin-Type Based Neutron Shielding Materials (에폭시수지계 중성자 차폐재 제조 및 특성)

  • New neutron shielding materials, KNS-201, KNS-301 and KNS-601 have been fabricated to be used for radioactive material shipping and storage cask. The base materials are a modified and a hydrogenated bisphenol- A type and novolac type epoxy resin, and aluminium hydroxide and boron carbide are added. These shielding materials offer good fluidity at processing, which makes it possible to form this resin shield into complicated geometric shapes such as radioactive material shipping and storage cask. Several measurements were made for the shielding materials to evaluate the thermal and mechanical properties and radiation resistance. The properties of the shielding materials are as follows: onset temperatures 2S7~28$0^{\circ}C$, thermal conductivities 0.9S~1.14W/m. K, thermal expansion coefficients 0.77~1.26x$10_{-6}{\circ}C_{-1}$, combustion characteristics < 80$0^{\circ}C$, ATB(average time of burning) < 5sec, AEB(average extent of burning) < 5mm, tensile strengths 2.5~3.2kg/$\textrm{mm}^2$, compressive strengths 13.2~1S.2kg/$\textrm{mm}^2$, flexural strengths 5.2 -6.4kg/$\textrm{mm}^2$. In general, the concerned properties of KNS-201, KNS-301 and KNS-601 were revealed to be better than those of NS-4- FR. foreign neutron shielding material. It is also observed that the radiation resistance of KNS- 601 was better than those of KNS-201 and KNS-301.
