Acidification and Biochar Effect on Ammonia Emission and Nitrogen Use Efficiency of Pig Slurry in the Vegetative Growth of Maize (Zea mays L.)

  • The objective of this study was to verify the effect of pig slurry application with acidification and biochar on feed value, nitrogen use efficiency (NUE) of maize forage, and ammonia (NH3) emission. The four treatments were applied: 1) non-pig slurry (only water as a control, C), 2) only pig slurry application (P), 3) acidified pig slurry application (AP), 4) acidified pig slurry application with biochar (APB). The pig slurry and biochar were applied at a rate of 150 kg N ha-1 and 300 kg ha-1, respectively. The AP and APB treatments enhanced all feed values compared to C and P treatments. The NUE for plant N was significantly increased 92.1% by AP and APB treatment, respectively, compared to the P treatment. On the other hand, feed values were not significantly different between AP and APB treatments. The acidification treatment with/without biochar significantly mitigated NH3 emission compared to the P treatment. The cumulative NH3 emission throughout the period of measurement decreased by 71.4% and 74.8% in the AP and APB treatments. Also, APB treatment reduced ammonia emission by 11.9% compared to AP treatment. The present study clearly showed that acidification and biochar can reduce ammonia emission from pig slurry application, and pig slurry application with acidification and biochar exhibited potential effects in feed value, NUE, and reducing N losses from pig slurry application through reduction of NH3 emission.

Food Functionality and Bioactivity of Vacuum Freeze-dried Fish Roe Concentrates (동결건조 어류 알 농축물의 식품기능성 및 생리활성)

  • The purpose of this study was to evaluate the food functional properties and in vitro bioactivity of vacuum freeze-dried fish roe concentrates (FRCs) prepared from Alaska pollock Theragra chalcogramma (AP), bastard halibut Paralichthys olivaceus (BH) and skipjack tuna Katsuwonus pelamis (ST). All three species showed better buffering capacity on the alkaline side (pH 10-12) than on the acidic side. The water-holding capacities of the FRCs were 3.5, 8.5 and 4.2 g/g protein for AP, BH and ST, respectively, and were significantly higher than that of commercial egg white. The protein solubilities of the FRCs were 42.5% (AP), 50.0% (BH) and 13.9% (ST). The foaming capacities of the FRCs were not significantly different among the species (128.0% for AP, 128.3% for BH, and 143.3% for ST; P>0.05), and their foam stability was maintained at 53.0-74.2% for 60 minutes. The oil-in-water emulsifying activity indexes of AP and BH (19.5 and 20.2 ㎡/g protein, respectively) were significantly superior to that of ST (P<0.05). The 2,2-diphenyl-1-picrylhydrazyl and 2,2'-azino-bis-3-ethylbenzothia-zoline-6-sulfonic acid radical-scavenging activities (IC50, mg/mL) of the FRCs were in the ranges of 1.05-3.26 and 0.13-0.18 mg/mL, respectively, and the angiotensin I converting enzyme inhibitory activity was in the range of 0.97-1.89 mg/mL.

Monitoring of Pyrethroid Resistance Allele Frequency in the Common Bed Bug (Cimex lectularius) in the Republic of Korea

  • Two-point mutations (V419L and L925I) on the voltage-sensitive sodium channel of bed bugs (Cimex lectularius) are known to confer pyrethroid resistance. To determine the status of pyrethroid resistance in bed bugs in Korea, resistance allele frequencies of bed bug strains collected from several US military installations in Korea and Mokpo, Jeollanamdo, from 2009-2019 were monitored using a quantitative sequencing. Most bed bugs were determined to have both of the point mutations except a few specimens, collected in 2009, 2012 and 2014, having only a single point mutation (L925I). No susceptible allele was observed in any of the bed bugs examined, suggesting that pyrethroid resistance in bed bug populations in Korea has reached a serious level. Large scale monitoring is required to increase our knowledge on the distribution and prevalence of pyrethroid resistance in bed bug populations in Korea. Based on present study, it is urgent to restrict the use of pyrethroids and to introduce effective alternative insecticides. A nation-wide monitoring program to determine the pyrethroid resistance level in bed bugs and to select alternative insecticides should be implemented.

Study on Transcriptional Factors Activation Change of HL-60 cell Apoptosis by Hedyotis Diffusa's Methanol Extract (백화사설초 메탄올 추출물에 의한 HL-60 세포고사과정에 있어서의 transcriptional factors 활성변화 연구)

  • Objective : Hedyotis diffusa has been used as an anticancer agent for several decades in oriental medicine. We test whether the methanol extract of the herb affects transcriptional activation factors including $NF-{\kappa}B$ and AP-1. Methods : 1. HL-60 cells were treated with various concentrations(from 200 to $50{\mu}g/ml$) of methanol extract and $H_2O$ extract($200{\mu}g/ml$)of hedyotis diffusa, After 48h later, the cells were tested for viability by MTT assay. 2. The HL-60 cells were treated with $200{\mu}g/ml$ of methanol extract for the indicated periods. First. Nuclear extracts were isolated and incubated with oligonucleotide probe of $NF-{\kappa}B$ and AP-1. Second. Nuclear extracts were isolated and reacted with p50, p65. c-rel pan-Jun, c-Jun, JunB. JunD antibody on ice for 30min. Finally The cell lysates were prepared and analyzed by western blotting using anti-Fas, anti-FasL and anti-p53 antibody. Results : 1. The methanol extract decreases the viability of human lymphoid origin leukemia HL-60 cells in a dose-dependent manner. 2. $NF-{\kappa}B$ is rapidly activated by the addition of the methanol extract, reaches a peak at 30min and gradually returns to resting level. We confirm that $NF-{\kappa}B$ is a heterodimer mainly composed of p65 subunit with c-Rel. 3. Transcriptional activation of AP-1 is detected at 30min and reaches a maximum at 1hr after stimulation of the cells with the methanol extract. AP-1 is mainly composed with Jur-D and partially Jug-B proteins. 4. the methanol extract of Hedyotis diffusa induces the expression of Fas, Fas ligand and p53 proteins of HL-60 cells in a time dependent fashion. Conclusions : These results suggest that the methanol extract of Hedyotis diffusa exerts anticancer effects to induce the death of human leukomic HL-60 cells via activation of trascriptional factors such as $NF-{\kappa}B$ and AP-1, increase in expression of Fas mediated signalling proteins, and induction of tumor suppressor gene. p53.

  • Objectives: The purpose of this study was to compare the microshear bond strength (uSBS) of two totaletch and four self-etch adhesive systems and a flowable resin to enamel. Materials and Methods: Enamels of sixty human molars were used. They were divided into one of six equal groups (n = 10) by adhesives used; OS group (One-Step Plus), SB group (Single Bond), CE group (Clearfil SE Bond), TY group (Tyrian SPE/One-Step Plus), AP group (Adper Prompt L-Pop) and GB group (G-Bond). After enamel surfaces were treated with six adhesive systems, a flowable composite resin (Filek Z 350) was bonded to enamel surface using Tygon tubes. the bonded specimens were subjected to uSBS testing and the failure modes of each group were observed under FE-SEM. Results: 1. The uSBS of SB group was statistically higher than that of all other groups, and the uSBS of OS, SE and AP group was statistically higher than that of TY and GB group (p < 0.05). 2. The uSBS for TY group was statistically higher than that for GB group (p < 0.05). 3. Adhesive failures in TY and GB group and mixed failures in SB group and SE group were often analysed. One cohesive failure was observed in OS, SB, SE and AP group, respectively. Conclusions: Although adhesives using the same step were applied the enamel surface, the uSBS of a flowable resin to enamel was different.

Transcriptional Modulation of Metabolism-Related Genes in Brackish Water Flea Diaphanosoma celebensis Exposed to Mercury (수은 노출에 따른 기수산 물벼룩의 대사 관련 유전자의 발현 양상)

  • Mercury (Hg) is a major concern in marine environment because of their bioaccumulation and biomagnification properties, and adverse effects to aquatic organisms at even a trace amount. However, little information on the effects of Hg, compared to other heavy metals, is available in marine small crustaceans. Here, we investigated the transcriptional modulation of metabolism-related genes in the brackish water flea, Diaphanosoma celebensis after exposure to sublethal concentration (0.2, 0.4, 0.8 ㎍/l) of HgCl2 for 48 h. Relative mRNA expression levels of five detoxification enzyme-coding genes (cytochrome P450; cyp360A1, cyp361A1, cyp4AP3, cyp4C122, and cyp370C5) and six digestive enzyme-coding genes [alpha amylase (AMY), alpha amylase related protein (AMY-like), trypsin (TRYP), chymotrypsin-like protein (CHY), lipase (LIP), pancreatic lipase-related protein (PLRP)] were analyzed using quantitative real time reverse transcription polymerase chain reaction (qRT-PCR). As results, Hg increased the mRNA level of cyp370C5 (clan2) and cyp4AP3 (clan4) in a concentration dependent manner. A significant increase in TRYP mRNA was also concentration-dependently observed after exposure to Hg. These findings suggest that cyp370C5 and cyp4AP3 play a key role in Hg detoxification in D. celebensis, and Hg can affect energy metabolism by modulating the transcription of digestive enzyme. This study will provide better understanding the molecular effects of Hg in marine small crustacean.

Anti-wrinkling effects of "L-Skin Care" and molecular mechanisms on hairless mouse skin caused by chronic ultraviolet B irradiation.

  • Background: Naturally occurring antioxidants were used to regulate the skin damage caused by ultraviolet (UV) radiation because several antioxidants have demonstrated that they can inhibit wrinkle formation through prevention of matrix metalloproteinases (MMPs) and/or increase of collagen synthesis. We examined the effect of oral administration of the antioxidant mixture ("L-Skin Care") on UVB-induced wrinkle formation. In addition, we investigated the possible molecular mechanisms of photoprotection against UVB through inhibition of collagen-degrading MMP activity or through enhancing of pro collagen synthesis in mouse dorsal skin. Methods: Female SKH-l hairless mice were orally administrated "L-Skin Care" (test group) or vehicle (control group) for 10 weeks with UVB irradiation by three times a week. The intensity of irradiation was gradually increased from 30 to $180mJ/cm^2$. Microtopographic and histological assessments of the dorsal skins were carried out at the end of 10 weeks to evaluate wrinkle formation. Western blot analysis and EMSA were also carried out to investigate the changes in the balance of collagen synthesis and collagen degradation. Results: Our "L-Skin Care" significantly reduced UVB-induced wrinkle formation, accompanied by significant reduction of epidermal thickness, and UVB-induced hyperplasia, acanthosis and hyperkeratosis. Oral administration of "L-Skin Care" significantly prevented UVB-induced expressions of MMPs, mitogen-activated protein (MAP) kinases and activation of activator protein (AP)-1 transcriptional factor in addition to enhanced type I procollagen and transforming growth factor-$\beta$ (TGF-$\beta$) expression. Conclusion: Oral administration of "L-Skin Care" significantly inhibited wrinkle formation caused by chronic UVB irradiation through significant inhibition of UVB-induced MMP activity accompanied with enhancement of collagen synthesis.

Effects of Aeration Period and Temperature after Imbibition on Growth of Mungbean Sprouts (침종 이후의 Aeration 기간과 온도에 따른 숙주나물의 생장)

  • Lateral roots formed on mungbean sprouts should lower their quality. The study was carried out to clarify the effects of aeration periods (AP; 1, 2, 3, and 4 hours) and temperature (AT; 20, 30, and $40^{\circ}C$) after 5 hour seed imbibition into 50 ppm benzyladenopurine (BA) solution on growth and morphological characters of mungbean (cv. Keumseongnogdu, Owoolnogdu and Zhong Lu 1) sprouts. On the 6th day, the mungbean sprouts were sorted by 4 categories on the base of hypocotyl lengths; > 7cm, 4 to 7cm, < 4cm, and non-germination to calculate their composition rates, number of lateral roots, lengths of hypocotyl and root, diameters at middle and upper parts of hypocotyl, fraction fresh and dry weights were measured. AP more affected growth of the cultivars than AT showing little effect on them. In the composition rate of the above 4 categories, cv. Zhong Lu 1 had the highest rate in longer than 7cm but nearly the same rate in AP treatments. Rates of longer than 4cm hypocotyls in cv. Keumseongnogdu, Owoolnogdu were increased with longer AP but their rates of shorter than 4cm showed reverse response to the former. Formation rate and number of lateral roots per sprout were decreased with longer AP, showing more severe decrement when delayed 3 to 4 hour AP. Upper part of hypocotyls and roots were more thickened and shortened in longer AP, respectively. Total fresh weights had no significant difference between AP treatments while hypocotyl fresh weights were increased with longer AP. It was concluded that in mungbean sprout culture aeration from BA treatment to the first watering permitted at least 4 hours.

Formation of DNA-Protein Crosslink at Oxidized Abasic Site Mediated by Human DNA Polymerase Iota and Mitochondrial DNA Polymerase Gamma

  • Human genomic DNA is continuously attacked by oxygen radicals originated from cellular metabolic processes and numerous environmental carcinogens. 2-deoxyribonolactone (dL) is a major type of oxidized abasic (AP) lesion implicated in DNA strand scission, mutagenesis, and formation of covalent DNA-protein crosslink (DPC) with DNA polymerase (Pol) ${\beta}$. We show here that human DNA polymerase (Pol)${\iota}$ and mitochondrial $Pol{\gamma}$ give rise to stable DNA-protein crosslink (DPC) formation that is specifically mediated by dL lesion. $Pol{\gamma}$ mediates DPC formation at the incised dL residue by its 5'-deoxyribose-5-phosphate (dRP) lyase activity, while $Pol{\gamma}$ cross links with dL thorough its intrinsic dRP lyase and AP lyase activities. Reactivity in forming dL-mediated DPC was significantly higher with $Pol{\gamma}$ than with $Pol{\iota}$. DPC formation by $Pol{\gamma}$, however, can be reduced by an accessory factor of $Pol{\gamma}$ holoenzyme that may attenuate deleterious effects of crosslink adducts on mitochondrial DNA. Comparative kinetic analysis of DPC formation showed that the rate of DPC formation with either $Pol{\iota}$ or $Pol{\gamma}$ was lower than that with $Pol{\beta}$. These results revealed that the activity of catalytic lyase in DNA polymerases determine the efficiency of DPC formation with dL damages. Irreversible crosslink formation of such DNA polymerases by dL lesions may result in a prolonged strand scission and a suicide of DNA repair proteins, both of which could pose a threat to the genetic and structural integrity of DNA.

Quality Characteristics and Antioxidant Activities of Cookies Supplemented with Aronia Powder (아로니아 분말을 첨가한 쿠키의 품질 특성과 항산화 활성)

  • The feasibility of incorporating aronia powder (AP) as a value-added food ingredient into convenient food products was investigated using cookie as a model system. AP was incorporated into cookies at amounts of 0, 1, 2, 3, and 4% (w/w) based on total weight of wheat flour. The pH level of cookie dough decreased significantly with increasing levels of AP (P<0.05), whereas moisture content and density were not directly affected by levels of AP incorporation. The spread ratio increased significantly upon addition of AP; however, it was not significantly affected by level of AP incorporation. The loss rate of cookies decreased significantly with increasing levels of AP (P<0.05). In terms of color, lightness and yellowness decreased while redness increased significantly (P<0.05) with increasing levels of AP. Use of AP significantly decreased hardness of cookies (P<0.05), but no significant differences were found between control and 1% sample, 1% and 2% sample, 2% and 3% sample, and 3% and 4% sample (P>0.05). 2,2-Diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis-3-ethylbenzthiazoline-6-sulphonic acid (ABTS) radical scavenging activities were significantly elevated by AP addition, and they increased significantly as AP concentration increased in the formulation (P<0.05). Correlation analysis indicated that acceptances of color and chewiness were negatively correlated well with the level of AP incorporation, density and moisture content of cookie dough, and cookie redness, whereas they were positively correlated with pH of cookie dough, loss rate, and hardness (P<0.01 or P<0.05). Finally, consumer acceptance test indicated that the highest levels of AP incorporation (4%) had an adverse effect on general consumer preferences. In contrast, cookies with moderate levels of AP (2%) are recommended based on overall scores to take advantage of the antioxidant properties of AP without sacrificing consumer acceptability.
