여기신호의 상관관계 기반 joint coding을 이용한 MPEG-4 audio lossless coding 인코더 복잡도 감소 방법 (A Complexity Reduction Method of MPEG-4 Audio Lossless Coding Encoder by Using the Joint Coding Based on Cross Correlation of Residual)

  • 오디오 신호를 무손실 압축하여 휴대용 멀티미디어 기기에서 최고의 오디오 품질을 제공하는 기기들이 등장하고 있으며, 무손실 오디오 압축을 위한 기술에서는 2006년 MPEG-4 audio lessless coding(ALS)와 MPEG-4 scalable lossless coding(SLS)가 국제 표준으로 채택 되었다. 2009년에는 MPEG에서 최대 스테레오 음원까지만 지원하는 MPEG-4 ALS simple profile을 정의하였다. 표준화된 무손실 오디오 코덱이 휴대용 멀티미디어 기기에서 널리 이용되기 위해서는 휴대용 멀티미디어 기기에서 가장 널리 쓰이는 스테레오 조건에서 낮은 복잡도를 보여야 한다. 하지만 기존 연구에서는 MPEG-4 ALS의 압축률을 향상시키거나, 혹은 다채널 환경에서의 복잡도를 개선하기 위한 연구들이 주로 이루어졌다. 본 논문에서는 MPEG-4 ALS 인코더의 복잡도와 압축률을 분석하고, 이를 바탕으로 MPEG-4 ALS simple profile 조건에서 MPEG-4 ALS 인코더의 복잡도를 개선하기 위한 방법을 제안한다. 분석 결과 MPEG-4 ALS 인코더의 analysis 블록에서 전체 복잡도의 75% 발생하므로, 기존의 연구에서 개발된 저 복잡도 필터를 인코더에 적용하여 복잡도를 감소시키며, joint coding의 압축 효율과 여기 신호의 상호상관계수의 관계를 기반으로 joint coding 결정 방법을 제안한다. 제안된 방법과 저 복잡도 필터가 포함된 MPEG-4 ALS 인코더의 성능은 MPEG-4 conformance test 파일과 일반 음악 파일을 이용하여 복잡도 및 압축률로써 평가된다. 실험 결과 제안된 방법이 적용되었을 경우 압축률은 유사하면서 인코더 복잡도가 24% 감소한다. 이를 통해 본 논문에서 제안된 방법이 MPEG-4 ALS 인코더의 복잡도 감소에 탁월한 성능을 가짐을 보였다.

Chlorsulfuron 및 Imazaquin에 의한 옥수수 Acetolactate Synthase 활성의 저해특성 (Inhibition Characteristics of Chlorsulfuron and Imazaquin on Acetolactate Synthase Activity of Corn Plants)

  • 옥수수 잎으로부터 추출한 ALS를 대상으로 하여 CHL과 IMA의 상호작용, ALS의 기질인 pyruvate 및 2-ketobutyrate에 대한 친화도, 저해양식의 차이 등을 조사한 결과는 다음과 같다. 1. ALS 에 대한 CHL의 $I_{50}$값은 100nM이었으며 IMA의 값은 $5{\mu}M$로 나타나 CHL의 ALS 저해활성은 IMA보다 약 50배 정도 높았다. 2. ALS에 대한 시험관내에서의 실험에서 두 제초제의 동시처리시 상가적인 ALS의 저해효과가 나타났으나, 상호 순차적 체계처리의 경우에서는 먼저 처리한 제초제에 의한 저해효과가 지배적으로 나타났다. 3. 제초제에 의한 ALS 활성의 저해는 시험관내에서 10분 이내에 대부분 이루어졌다. 4. ALS의 활성은 분지아미노산에 의해 저해되었는데 Val과 Leu에 의한 저해가 Ile에 의한 저해보다 크게 나타났다. 또한 이들을 동시에 혼합처리하면 0.1mM의 농도에서도 ALS의 활성이 65% 정도 저하되었다. 5. 2-Ketobutyrate를 ALS의 기질로 첨가하면 pyruvate만을 기질로 사용하였을 때보다 측정가능한 ALS 활성이 2-ketobutyrate에 의해 농도의존적으로 감소되는 경향이었다. 6. ALS의 기질인 pyruvate에 대해 CH은 비경쟁적 저해제로 IMA는 반경쟁적 저해제로 작용하였다.

  • Acetolactate synthase (ALS, EC 4.1.3.18) is the first common enzyme in the biosynthesis of leucine, isoleucine, and valine. It is the target enzyme for several classes of herbicides, including the sulfonylureas, the imidazolinones, the mazolopyrimidines, the pyrimidyl-oxy-benzoates, the pyrimidyl-thio-benzens, and the 4,6-dimethoxypyrimidines. An amino-terminal fragment of the sulfonylurea-resistant ALS gene (SurB) from Nicotiana tabaccum was cloned into the bacterial expression vector pGEX-2T. The resulting recombinant plasmid pGEX-ALS1 was used to transform Escherichia coli strain BL21, and the tobacco ALS was expressed in the bacteria as a protein fused with glutathione S-transferase (GST). Polyclonal antibodies against the fusion product (GST-ALS) were produced, and the sensitivity of GST-ALS with the rabbit anti-GST-ALS IgG was up to 50 ng. This antibody was used for Western blot analysis of the partially purified ALS from barley shoots. The results suggest that the polyclonal antibody produced in this study can be used to detect plant ALS.

The Interaction of Barley Acetolactate Synthase with 4,6-Dimethoxypyrimidine Inhibitors

  • Acetolactate synthase (ALS) catalyzes the first common step in the biosynthesis of valine, leucine, and isoleucine. ALS is the target enzyme for several classes of structually diverse herbicides. We have synthesized 4,6-dimethoxypyrimidine derivatives as ALS inhibitors, and their inhibitory activities on barley ALS were determined. $IC_{50}$ values for the derivatives are 0.2~200 ${\mu}m$. K11570, the most potent ALS inhibitor with $IC_{50}$ of 0.2 ${\mu}m$, showed mixed-type inhibition with respect to substrate pyruvate, and the progress curves for ALS inhibition by K11570 indicated that the amount of inhibition increased with time. Inhibition-competition experiments were carried out and indicated that three different classes of inhibitors, K11570, a sulfonylurea Ally, and leucine, bind to ALS in a mutually exclusive manner. Chemical modification of tryptophanyl and tyrosyl residues of ALS decreased the sensitivity of ALS to K11570, while cysteine modification did not affect the sensitivity. These results suggest that tryptophanyl and tyrosynyl residues are probably located at or near the inhibitor binding site.

The Effects of Sera from Amyotrophic Lateral Sclerosis Patients on Neuromuscular Transmission and Calcium Channels in Mice

  • Amyotrophic lateral sclerosis (ALS) is a degenerative neuromuscular disease of unknown etiology in which the upper and lower motor neurons are progressively destroyed. Recent evidences support the role of autoimmune mechanisms in the pathogenesis of ALS. This study investigated the effects of sera from ALS patients on neuromuscular transmission in phrenic nerve-hemidiaphragm preparations and on calcium currents of single isolated dorsal root ganglion (DRG) cells in mice. Mice were injected with either control sera from healthy adults or ALS sera from 18 patients with ALS of sporadic form, for three days. Miniature end plate potential (MEPP) and nerve-evoked end plate potential (EPP) were measured using intracellular recording technique and the quantal content was determined. Single isolated DRG cells were voltage-clamped with the whole-cell configuration and membrane currents were recorded. Sera from 14 of 18 ALS patients caused a significant increase in MEPP frequency in normal Ringer's solution $(4.62{\pm}0.14\;Hz)$ compared with the control $(2.18{\pm}0.15\;Hz).$ In a high $Mg^{2+}/low\;Ca^{2+}$ solution, sera from 13 of 18 ALS patients caused a significant increase in MEPP frequency, from $2.18{\pm}0.31$ Hz to $6.09{\pm}0.38$ Hz. Sera from 11 of 18 patients produced a significant increase of nerve-evoked EPP amplitude, from $0.92{\pm}0.05$ mV to $1.30{\pm}0.04$ mV, while the other seven ALS sera did not alter EPP amplitude. In the ALS group, EPP quantal content was also elevated by the sera of 14 patients (from $1.49{\pm}0.07$ to $2.35{\pm}0.07).$ MEPP frequency and amplitude in wobbler mouse were $4.03{\pm}0.53$ Hz and $1.37{\pm}0.18$ mV, respectively, which were significantly higher than those of wobbler controls (wobblers without the symptoms of wobbler). Sera from ALS patients significantly reduced HVA calcium currents of DRG cells to 42.7% at -10 mV. Furthermore, the inactivation curve shifted to more negative potentials with its half-inactivation potential changed by 6.98 mV. There were, however, significant changes neither in the reversal potential of $I_{Ca}$ nor in the I-V curve. From these results it was concluded that: 1) The serum factors of sporadic ALS patients increase neuromuscular transmission and can alter motor nerve terminal presynaptic function. This suggests that ALS serum factors may play an important role in the early stage of ALS, and 2) Calcium currents in DRG cells were reduced and rapidly inactivated by ALS sera, suggesting that in these cells, ALS serum factors may exert interaction with the calcium channel.

Effect of Pyrimidylsalicylate on the Valine Sensitive Acetolactate Synthase Purified from Serroatia marcescens

  • The inhibitory effect of herbicides such as sulfonylurea derivatives, imidazolinones and pyrimidylsalicylate has been examined on the purified valine sensitive acetolactate synthase (ALS) from Serratia marcescens. The concentration of sulfometuron methyl which inhibits 50% of the ALS activity was 2.5 mM. The required concentrations of triasulfuron, primisulfuron methyl and imazaquin for the 50% inhibition of the ALS activity were 1 mM. The resistance of Serratia ALS to sulfometuron methyl, imazapyr and imazaquin is similar to that of E. coli ALS 1. However, pyrimidylsalicylate showed a potent inhibitory effect on the Serratia ALS almost 13 times more potent than on E. coli ALS II, which is known as herbicide-sensitive isozyme. The inhibitory mode was competitive against pyruvate. 150 value was determined to be $17{\mu}M$ in an assay mixture containing 20 mM pyruvate, and the $K_1$, value was calculated to be $0.4{\mu}m$ from the modified double reciprocal plot of 1/V versus $1/S^2$.

RNA-Binding Proteins in Amyotrophic Lateral Sclerosis

  • Significant research efforts are ongoing to elucidate the complex molecular mechanisms underlying amyotrophic lateral sclerosis (ALS), which may in turn pinpoint potential therapeutic targets for treatment. The ALS research field has evolved with recent discoveries of numerous genetic mutations in ALS patients, many of which are in genes encoding RNA binding proteins (RBPs), including TDP-43, FUS, ATXN2, TAF15, EWSR1, hnRNPA1, hnRNPA2/B1, MATR3 and TIA1. Accumulating evidence from studies on these ALS-linked RBPs suggests that dysregulation of RNA metabolism, cytoplasmic mislocalization of RBPs, dysfunction in stress granule dynamics of RBPs and increased propensity of mutant RBPs to aggregate may lead to ALS pathogenesis. Here, we review current knowledge of the biological function of these RBPs and the contributions of ALS-linked mutations to disease pathogenesis.

근위축성 측삭경화증 환자 1례에 대한 임상증례 (Case of Amyotrophic Lateral Sclerosis(ALS) with Oriental Medical Treatment Evaluated by K-ALSFRS-R)

  • Objects : This study is a report on treatment effect of the case that treated by using of oriental medical treatment to Amyotrophic Lateral Sclerosis(ALS) patient. Methods : The patient diagnosed with ALS was treated by using of acupuncture, physical therapy, Su-gi therapy, thermotherapy and herbal medicine during 180 days. K-ALSFRS-R score was used as the tool of treatment effect. Results : Despite the K-ALSFRS-R score reduction, the oriental medical treatment maintain the respiratory function and a speech. Conclusions : The oriental medical treatment is effective on local symptoms of ALS, but there were no functional improvement of ALS in this case study. It is necessary to have more examination about ALS.

  • 본 연구는 한국에서 발생하고 있는 설포닐우레아계 제초제 저항성 및 감수성 물달개비의 ALS 염기서열 분석에 의한 ALS 유전자의 특성과 제초제 저항성 메카니즘을 구명하기 위하여 실시하였다. 감수성 및 저항성 두 계통의 biotype에서 815개의 염기서열을 분석하였다. 분석된 염기서열에서 감수성 biotype으로부터 4개 clone, 저항성 계통으로부터 15개 clone, 총 19개의 clone들을 크게 4 group으로 분류할 수 있었다. 첫 번째 group은 저항성 및 감수성 biotype의 ALS 유전자 염기서열이 차이가 없었으며, 두 번째 그룹은 아미노산 197번째의 proline이 DNA의 점돌연변이(point mutation)에 의해 serine으로 변화된 부분이다. 세 번째 group은 6곳에서 점돌연변이에 의한 아미노산치환이 발생하였으며, 네 번째 group는 첫 번째 group과 세 번째 group 중간적 특성으로 3곳에서 염기서열 점돌연변이에 의해 아미노산 치환이 있었다. 따라서 하나의 물달개비 식물체에서 여러 가지 형태의 ALS 유전자가 존재할 수 있으며, 물달개비 저항성 biotype의 ALS 유전자는 Domain A 부분에 있는 아미노산 197번째 proline이 염기서열 변화에 의해 serine으로 돌연변이 되었다.

근위축성 측삭경화증의 한방치료에 관한 문헌적 고찰연구 (A Literature Review of Korean Medical Studies of Amyotrophic Lateral Sclerosis (ALS))

  • Objectives : There have been many reports that show that Korean Medical Treatment of ALS patients can maintain or improve their major and minor symptoms, but this has not been reviewed in literature. The aim of this study is to review studies of ALS patients treated with Korean Medicine. Methods : A search was completed of studies containing word ALS in English and in Korean from KIOM OASIS. Results : There were no cases of ALS patients that were fully cured. However with Korean Medical treatment, there were articles that reported improvement or maintenance of major symptoms, recovery of minor symptoms like pain, chest discomfort, insomnia, cough, sputum, etc, and success in the long term care of ALS patients. Conclusions : Although the current study has limitations, our findings support the potential use and effectiveness of Korean Medical Treatment for ALS patients.
