Effects of Korean Mountain Ginseng Leaves and Its Active Constituents on Scopolamine-induced Amnesia in Mouse (장뇌삼 잎 추출물의 분획물이 Scopolamine으로 유도된 Mouse의 기억력 개선에 미치는 영향)

  • Objectives: The purpose of this study was to characterize the effect of the fraction of Korean mountain ginseng folium (FKG) on the learning and memory impairments induced by scopolamine. Methods: The memory ameliorating effect of FKG was investigated using a passive avoidance test, the Y-maze test, and the Morris water maze test in mice. Drug-induced amnesia was induced by treating animals with scopolamine(1mg/kg, i.p.). Results: FKG (2 or 4mg/kg, p.o.) administration significantly reversed scopolamine-induced cognitive impairments in mice by the passive avoidance test and the Y-maze test(P<0.05), and also improved escape latency in the Morris water maze test at 2 or 4mg/kg(P<0.05). Although FKG has little inhibitory activity for AChE (IC50 value; 1847 ${\mu}g/ml$) in an invitro study, phosphorylated extracellular signal-regulated kinase(pERK) was increased by the administration of FKG inhippocampus on immunohistochemistry. Conclusions: These results suggest that FKG may be a useful cognitive impairment treatment, and its beneficial effects are mediated, in part, via activation of ERK pathway.

Influence of FCCP on Catecholamine Release in the Rat Adrenal Medulla

  • The aim of the present study was to investigate the effect of FCCP (carbonyl cyanide p-trifluoromethoxyphenyIhydrazone), which is a potent mitochondrial uncoupler, on secretion of catecholamines (CA) from the perfused model of the rat adrenal gland and to establish the mechanism of its action. The perfusion of FCCP (3 ${\times}$ $10^{-5}$ M) into an adrenal vein of for 90 min resulted in great increases in CA secretions. Tachyphylaxis to CA-releasing effect of FCCP was not observed by repeated perfusion of it. The CA-releasing effects of FCCP were depressed by pre-treatment with pirenzepine, chlorisondamine, nicardipine, TMB-8, and the perfusion of EGTA plus $Ca^{2+}$-free medium. In the presence of FCCP (3 ${\times}$ $10^{-5}$ M), the CA secretory responses induced by Ach (5.32 ${\times}$ $10^{-3}$ M), and DMPP ($10^{-4}$ M) were significantly enhanced. Furthermore, the perfusion of CCCP (3 ${\times}$ $10^{-5}$ M), a similar mitochondrial uncoupler, into an adrenal vein for 90 min also caused an increased response in CA secretion. Taken together these experimental results indicate that FCCP causes the CA secretion the perfused rat adrenal medulla in a calcium-dependent fashion. It is suggested that this facilitatory effects of FCCP may be mediated by cholinergic receptor stimulation, which is relevant to both stimulation of the $Ca^{2+}$ influx and $Ca^{2+}$ release from cytoplasmic $Ca^{2+}$ stores.

Identification of Octopine Type Ti Plasmid in Agrobacterium tumefaciens KU12 (Agrobacterium tumefaciens KU12내에 존재하는 Octopine Type Ti Plasmid의 확인)

  • Agrobacterium tumefaciens KU12 isolated from Korea is able to induce tumors on various plants and catabolize octopine as a sole carbon and nitrogen source. A, tumefaciens KU12 contains three plasmids. Their sizes are 45.5 kb. 240 kb. and > 240 kb. respectively. For the purpose of identification of octopine type Ti plasmid, avirulent A, tumefacients A136 is transformed with plasmids isolated from KU12 by direct transformation. Transformants containing Ti plasmid were grown on AB medium containing octopine as a sole nitrogen source. The isolated strain, named KU911, contains only 240 kb plasmid. As a result of induction of crown gall and Southern hybridization with other octopine Ti plasmid pTiAch5, 240 kb plasmid named pTiKU12 was Ti plasmid.

  • The application of Internet technology has created enormous impact on banking sector with the implementation of many techno-oriented services like Internet banking, EFT, branchless banking, Automated Clearing House (ACH) transactions etc. Study of customer's attitude in terms of trust, perceived risk and ease of use of a particular technology is as an important parameter for acceptance or rejection of a technology. To explore the customers'attitude for Internet banking this research is undertaken. The research is carried out in Pokhara valley which is the second largest city and tourism capital of Nepal. The study employs descriptive research design with stratified sampling procedure for eight top commercial banks. A set of 25 customers is taken from each selected 8 banks making a sample size of 200 respondents. A fixed set of question related to demographic factors is provided personally or by visiting the location of the customers of Internet banking service and collected accordingly. Reliability test is performed using Cronbach's alpha and data is analyzed using inferential statistics to present the results of the study. This study provides knowledge on the current scenario of Internet banking and helps banks in cost saving, mass customization, product innovation, improved marketing and communication. This study is very important for financial institutions like banks, government agencies and business houses to understand the perception of customers towards Internet banking and technology as a whole. The study also supplements the gap in literature on technology and banking in Nepal and serves as an important knowledge base.

  • To investigate the effect of pine needle extract (PNE) on membrane fluidirt and neurotransmiter-related enzymes in brain of Spragu-Dawley(SD), male SD rats were fed basic diets (control group), and experimantal diets (PNE group)with 0.5% and 0.1% fo PNE for 6 weeks. pine (pinus tabulaeformis C$_{ARR}$ is one of the popular plant drugs which has used as a medicine in Asia. Cholesterol levels in brain mitochondria of 0.5%-PNE and 0.1%-PNDE groups were significantly decreased in 15% and 25%, respectively, compared with control group, but cholesterol levels in brain microsomes of these PNE groups howed almost no change compared with control group. Lipofuscin accumulations in brain membranes of 0.5%-PNE and 0.1%-PNE groups were sgnificantly inhibited in 18% and 21%, respectively, compared with control group. Brain memberance fluidity was also activated in 50% and 100% by the administration of 0.5%-PNE and 0.1%-PNE. higher acetylcholinesterase(15% and25%) and lower monoamine oxidase B (25% and 15%0 activities were effectively modulated by the administration of 0.5%-PNE and 0.1%-PNDE. These results suggest that more beneficial effects such as inhibition of cholesterol and lipofuscin, increase of membrane fluidity, higher acetylcholinesterase and lower monoamone oxidase activities in brain membranes of SD rats may be effectively modulated by administration of pine needle extract (PNE).

  • Power consumption in the building thermal load could be the sum of the building fabric conduction load, building ventilation convection load and other such as radiation loss load. Dynamic Breathing Building (DBB) is the state-of-the-art to improve the wall insulation and indoor air quality(IAQ) performance as making air flow through the wall. This heat recovery type DBB contributes the power consumption saving due to the improved dynamic U-value. KIER twin test cell with static insulation(SI) and dynamic insulation(DI) at KIER was developed to test building power consumption at the real outside conditions. Then, the actual results were compared with the theory to predict the power consumption at the KIER twin test cell and introduced the building new radiation loss factor $\alpha$ to explain the difference between the both the theory and the actual case. As the results, the power consumption at the breathing DI wall building could saved 10.8% at the 2ACH(Air change per hour) compared with conventional insulation. The building radiation loss factor $\alpha$ for this test condition to calibrate the actual test was 0.55 in the test condition.

The Effects of Endogenously and Exogenously Induced Nitric Oxides on the Nociperception of Rats (내.외인성으로 유도된 Nitric Oxide가 흰쥐의 통각전달에 미치는 효과)

  • Nitric oxide is a labile, gaseous, broad spectrum second messenger that used in various tissues and cells. If it is induced by endogenously and exogenously in the neuronal cells, it is able to mediate analgesia or hyperalgesia at the periphery and in the spinal level respectively. This dual role of nitric oxide in the sensory system is very intriguing but has not been fully understood yet. In this experiment, acetylcholine (300 $\mu$g/paw), sodium nitroprusside (600 $\mu$g/paw), and L-arginine (300 $\mu$g/paw) represented antinociceptive effect to noxious topical stimulus, but pronociceptive responses followed by spinally application (20$\mu$g/5$\mu$l, 10$\mu$g/3$\mu$l, 500$\mu$g/5$\mu$l respectively). Calcium ion is critical element which activates nitric oxide synthase, therefore verapamil (300 $\mu$g/paw) and NOS inhibitor (20 mg/kg, L-NAME or L-NOArg) are injected into right hind paw (i.pl.). When verapamil is combined with NOS inhibitors analgesic effects through NO-cGMP pathway are inhibited as compared with ACh alone. Diluted formalin (2.5%), when injected into rats'hind paw (0.05 ml), elicited a biphasic algesic responses and nitric oxide had an analgesic effect on both $A\delta$ and C sensory nerve fibers which manipulate the phases respective1y. Nitric oxides, which produced from constitutive nitric oxide synthase, activated cyclooxygenase-type I and then prostaglandins are produced from them. So, indomethacin and ibuprofen, inhibitors of COX$_1$enzyme, when pretreated intraperitoneally (100 mg/kg) could reduce the hyperalgesic state. From these results, it is possible to imagine that the intrathecally administered NO donors expressed hyperalgesia through both long-term potentiation mechanism and arachidonic acid-prostaglandin cascade.

Synthesis of Selective Butyrylcholinesterase Inhibitors Coupled between α-Lipoic Acid and Polyphenols by Using 2-(Piperazin-1-yl)ethanol Linker

  • In the previous paper (Bull. Korean Chem. Soc., 2011, 32, 2997), the hybrid molecules between ${\alpha}$-lipoic acid (ALA) and polyphenols (PPs) connected with neutral 2-(2-aminoethoxy)ethanol linker (linker-1) showed new biological activity such as butyrylcholinesterase (BuChE) inhibition. In order to increase the binding affinity of the hybrid compounds to cholinesterase (ChE), the neutral 2-(2-aminoethoxy)ethanol (linker 1) was switched to the cationic 2-(piperazin-1-yl)ethanol linker (linker 2). The $IC_{50}$ values of the linker-2 hybrid molecules for BuChE inhibition were lower than those of linker-1 hybrid molecules (except 9-2) and they also had the same great selectivity for BuChE over AChE (> 800 fold) as linker-1 hybrid molecules. ALA-acetyl caffeic acid (10-2, ALA-AcCA) was shown as an effective inhibitor of BuChE ($IC_{50}=0.44{\pm}0.24{\mu}M$). A kinetic study using 7-2 showed that it is the same mixed type inhibition as 7-1. Its inhibition constant (Ki) to BuChE is $4.3{\pm}0.09{\mu}M$.

New Species and New Records of Buellia (Lichenized Ascomycetes) from Jeju Province, South Korea

  • A new species and 2 new records of lichen genus Buellia were discovered from Chuja-do Island in Jeju Province during a recent floristic survey: B. chujana X. Y. Wang, S. Y. Kondr., L. $L\ddot{o}k\ddot{o}s$ & J.-S. Hur sp. nov., B. halonia (Ach.) Tuck., and B. mamillana (Tuck.) W. A. Weber. The new species is characterized by a brown, areolate thallus, the presence of perlatolic acid, and a saxicolous habitat. Together with previously recorded species, 10 Buellia species were confirmed from Jeju-do Island. Among these species, 3 growing in the exposed rocky area contained xanthone (yellowish lichen thallus, UV + orange), indicating that production of xanthone in this genus might be a defense strategy against the harm of UV light. Although the genus Buellia has been thoroughly studied in Korea before, novel species have been discovered continuously, and large species diversity has been found in this crustose genus, even from a small rocky island. This study indicates that the coastal area harbors a vast number of crustose lichen species, and there is great potential to discover unknown lichens in the coastal rocky area in Korea.

Effects of Seonbangpaedoktang on secretion of airway mucin and contractility of tracheal smooth muscle (선방패독탕(仙方敗毒湯)이 호흡기 뮤신 분비 및 기관 평활근 긴장도에 미치는 영향)

  • Objectives : The author intended to investigate Seonbangpaedoktang (SBPT) significantly affect in vivo and in vitro mucin secretion from airway epithelial cells. Methods : In vivo experiment, the author induced hypersecretion of airway mucin, hyperplasia of tracheal goblet cells and the increase in intraepithelial mucosubstances. Effects of orally-administered SBPT during 1 week on in vivo mucin secretion and hyperplasia of tracheal goblet cells were assessed. For in vitro experiment, confluent hamster tracheal surface epithelial (HTSE) cells were metabolically radiolabeled and chased in the presence of SBPT to assess the effect of the agent on 3H-mucin secretion. Total elution profiles of control spent media and treatment sample through Sepharose CL-4B column were analysed. Possible cytotoxicity of the agent was assessed by measuring LDH release. Also, the effect of SBPT on contractility of isolated tracheal smooth muscle was investigated. Results : SBPT inhibited hypersecretion of in vivo mucin and inhibited the increase of number of goblet cells ; SBPT did not affect in vitro mucin secretion and the secretion of the other releasable glycoproteins with less molecular weight than mucin from cultured HTSE cells, without significant effect on LDH release; SBPT did not affect Ach-induced contraction of isolated tracheal smooth muscle. Conclusions : SBPT can inihibit hypersecretion of in vivo mucin and the author suggest that the effect SBPT with their components should investigate further.
