Inhibitory Activity on Angiotensin Converting Enzyme and Antioxidant Activity of Hovenia dulcis Thunb. Cortex Extract (헛개나무의 Angiotensin 전환 효소 저해 및 항산화 활성)

  • To develop a new functional materials, angiotensin converting enzyme (ACE) inhibitory activity, antioxidant effect and total phenolic content of Hovenia dulcis Thunb. cortex were evaluated. Methanol and water extract of H. dulcis inhibited ACE by 81% and 76%, respectively, at the concentration of $4,000\;{\mu}g\;m{\ell}^{-1}$ which were similar level with that (85%) of commercial peptide-type ACE inhibitor. Superoxide radical scavenging activity of two extracts $(99.5%{\sim}99.9%)$ were stronger than that (69%) of ascorbic acid at the final concentration of $200\;{\mu}g\;m{\ell}^{-1}$. Among the solvent fractions, ether and ethylacetate fraction showed also potent scavenging activities (91% and 85%) for superoxide radical. Inhibitory activities of two extracts on oxidation of human low density lipoprotein (LDL) which were similar with that of ${\alpha}-tocopherol$, were higher than 80% at the concentration of $50\;{\mu}g\;m{\ell}^{-1}$. Total phenol contents of methanol and water extracts were 7.2% and 3.6%, respectively, and that of ethylacetate showed the highest value as 60.8% among the solvent fractions. Therefore, it has been suggested that H. dulcis cortex could be a effective anti-hypertention and antioxidant resource to develope a new functional material.

  • Angiotensive I-converting enzyme inhibitory activity of various extracts from some rice brans were investigated with its optimal extraction conditions. Water extracts of Ilpumbyeo rice bran showed the highest ACE inhibitory activities of 7730%. ACE inhibitor from Ilpumbyeo rice bran was maximally extracted when its powder was treated with 10 times of distilled water for 12h at $30^{\circ}C$.

  • A halotolerant and extracellular protease-producing yeast was isolated from traditional Meju and identified as a strain of Hansenular polymorpha by investigating its microbiological characteristics. The optimum pH, temperature and NaCl concentration reauired for the growth of Hansenular polymorpha S-9 were found to be pH 6.0, 30$^{\circ}C$ and 0.5 M, respectively. Extracellular protease was produced maximally at 10 U ml(sup)-1 when Hansenular polymorpha S-9 was grown on the medium containing 1.0% beef extract and 0.1 M NaCl for 12 hr at 30$^{\circ}C$. About 13% of the angiotensin-converting enzyme (ACE) inhibitory activity was shown in the hydrolysates which were obtained from the digestion of soybean protein (6 mg) for 6 hr at 30$^{\circ}C$ by the crude enzyme (1 U).

Comparison of Physiological Activity in Different Parts of Dolsan Leaf Mustard (돌산 갓의 부위별 생리활성 작용의 비교)

  • Physiological activity in different parts of Dolsan leaf mustard (DLM) was investigated. To determine the physiological activity, DLM was crushed filtered centrifuged and then the supernatant was used as a sample. of 1%, 3% and 6%. By adding of improved root juice(20th day) with the concentration of 6% the cytotoxicity against HepG2 was the highest about 78.2%. And antioxidative effects in different parts of DLM was measured by using the DPPH method. Antioxidative effects was higher in all leaves than other parts. In particular antioxidative effects was the highest in leaves of traditional DLM at the 20th day of growth about 80.4%. In leaves of improved DLM at the 60th growth angiotensin-I converting enzyme inhibitory effect was the highest about 94.0%. Consequently there was not significant difference of physiological activity between improved and traditional DLM. However the cytotoxicity against HepG2 was the highest in roots of DLM. And the antioxidative and the ACE inhibitory effect in leaves of DLM were higher than those of other parts.

  • Hydrolyzates which inhibit the angiotensin-I converting enzyme(ACE) were prepared from defatted anchovy meal by pepsin. These were tested for inhibitory activity against ACE, which is one of the hypertension inducing factors. The ACE inhibitory activity of the hydrolyzates increased until 20hrs of hydrolysis had elapsed but slightly decreased after that time. And presence of $50\%$ ethanol soluble peptide-nitrogen increased slowly up to 12hrs of hydrolysis, and then mainly increased until 20hrs of hydrolysis was completed. From the profiles of gel permeation chromatography on a Bio-gel P-2 of $50\%$ ethanol soluble fraction obtained from hydrolyzate for 20hrs, the higher active fractions were 2'($IC_{50}=45\;{\mu}g\;protein/ml$) and 4'($IC_{50}=76\;{\mu}g\;protein/ml$). Amino acid analysis showed major quantities of glutamic acid, leucine, lysine for 2'and aspartic acid, threonine for 4' respectively.

  • Antibacterial, antifungal, anticancer, and ACE inhibitory activities of methanol, ethyl acetate, chloroform, and hexane extracts of garlic were investigated. Methanol extract showed highest yield of 7.9-8.8 brix. Ethyl acetate and chloroform extracts showed strong antibacterial activities as determined by inhibition zone (8-16mm) against B. subtilis and P. aeruginosa, and good antifungal activities as determined by inhibition zone (8-30mm) against A. niger, M. miehei, and T. reesei. Wando, Banyans, Chinese, and Seosan garlic methanol extracts showed 65.6, 60.4, 70.1, and 55.3% ACE inhibitory activities, 42.3, 53.8, 50.4, and 54.5% anticancer activities against SNU-1, and 11.3, 20.2, 36.6, and 30.5% anticancer activities against HeLa, respectively.

In-silico Studies of Boerhavia diffusa (Purnarnava) Phytoconstituents as ACE II Inhibitor: Strategies to Combat COVID-19 and Associated Diseases

  • COVID-19 caused a catastrophe in human health. People infected with COVID-19 also suffer from various clinical illnesses during and after the infection. The Boerhavia diffusa plant is well known for its antihypertensive activity. ACE-II inhibitors and calcium channel blockers are reported as mechanisms for the antihypertensive activity of B. diffusa phytoconstituents. Various studies have said ACE-II is the virus's binding site to attack host cells. COVID-19 treatment commonly employs a variety of synthetic antiviral and steroidal drugs. As a result, other clinical illnesses, such as hypertension and hyperglycemia, emerge as serious complications. Safe and effective drug delivery is a prime objective of the drug development process. COVID-19 is treated with various herbal treatments; however, they are not widely used due to their low potency. Many herbal plants and formulations are used to treat COVID-19 infection, in which B. diffusa is the most widely used plant. The current study relies on discovering active phytoconstituents with ACE-II inhibitory activity in the B. diffusa plant. As a result, it can be used as a treatment option for patients with COVID-19 and related diseases. Different phytoconstituents of the B. diffusa plant were selected from the reported literature. The activity of phytoconstituents against ACE-II proteins has been studied. Molecular docking and ligand-protein interaction computation tools are used in the in-silico experiment. Physicochemical, drug-likeness, water solubility, lipophilicity, and pharmacokinetic parameters are used to evaluate phytoconstituents. Liriodenine has the best drug-likeness, bioactivity, and binding score characteristics among the selected ligands. The in-silico study aims to find the therapeutic potential of B. diffusa phytoconstituents against ACE-II. Targeting ACE-II also shows an effect against SARS-CoV-2. It can serve as a rationale for designing a drug for patient infected with COVID-19 and associated diseases.

Comparison of Biological Activity Between Soybean Pastes Adding Sword Bean and General Soybean Pastes (작두콩 첨가 된장과 일반 된장의 생리활성 특성 비교)

  • This study was carried out to analyse the biological activity in soybean pastes with the sword beans and general soybean pastes. The electron donating activity of soybean pastes with the sword beans(J2, J4) was higher than other soybean pastes with value of $81.5{\pm}2.2%$ and $88.3{\pm}0.7%$, respectively. And the content of total phenol compounds was higher in J2 and J4 compared to the others with value of $1773.8{\pm}2.9\;mg/kg$ and $1785.7{\pm}3.9\;mg/kg$ respectively. ACE inhibitory effect was higher in J2 and J4 than other soybean pastes with value of $63.1{\pm}1.0%$, $62.1{\pm}1.4%$, respectively. And aglycone type(daidzein, genistein) isoflavone content was higher than glucoside type(daidzin, genistin). But tyrosinase inhibitory effect was not different among test samples. From the results, soybean pastes with the sword beans(J2, J4) have higher anti-oxidant activity and ACE inhibitory effect than other soybean pastes, therefore, it may have potential to prevent hypertension.

Functional Properties of Nutmeg (육두구의 생리활성에 관한 연구)

  • This study investigated the antioxidative and antimicrobial activities of nutmeg (water, ethanol extract and essential oil). The total phenol contents of water, ethanol extract and essential oil were 3.4%, 16.9%, and 3.2%, respectively. Hydrogen donating abilities of water, ethanol extract and essential oil at 1,000 ppm were 4.9%, 41.8% and 6.8%, respectively. The antioxidative activities in linoleic acid substrates were in the order of BHT > ethanol > extract > ${\alpha}$-tocopherol > essential oil > water extract. The antioxidative activities in linoleic acid emulsion substrates were in the order of BHT > water extract > essential oil > ethanol extract > ${\alpha}$-tocopherol. In antimicrobial activity, ethanol extract showed growth inhibition effect against Micrococcus luteus, Bacillus cereus and Salmonella enteritidis, and the essential oil showed growth inhibition effect against Micrococcus luteus. However, no antimicrobial activity of water extract was observed. The nitrite-scavenging abilities of water, ethanol extract and essential oil at 1,000 ppm were 4.5%, 28.8% and 98.8%, respectively, and the ACE inhibitory activities were 0.2%, 11.0% and 10.0%, respectively.

Isolation of Angiotensin Converting enzyme inhibitors from Ripe Cucurbita moschata Duch

  • Angiotensin converting enzyme (ACE) inhibitor acts on the inhibition of ACE and causes a decrease in blood pressure. There have been several reports on screening of ACE inhibitors from natural food products and protein hydrolysates of various food sources. Ripe Cucurbita moschata Duch has been used as an oriental medicine in Korea. To isolate ACE inhibitors, crude water extracts of the edible portion of ripe Cucurbita moschata Duch were obtained after heating in water at 95$^{\circ}C$ for 2 h. Crude extracts were then filtered using PM-10 and YM-1 membranes. The membrane-filtered solution was loaded onto Sephadex G-15 column equlibrated with a phosphate buffer. Among the four major fractions of gel permeation chromatography, the second fraction had the highest inhibitory activity of 65%. Further purification of the fraction using reversed-phase HPLC with a $C_{18}$ column produced ACE inhibitors, which were identified as a mixture having molecular mass of 222 and 273 by Tandem mass spectrometry.
