Antioxidant & Antibacterial effects of Artichoke (Cynara Scolymus L.) leaf by various extract solvents

  • Antioxidant is an important role to protect the human body against damage by reactive oxygen species. However, the excessive intake of such antioxidant is known to cause a serious poisonous influence on one's liver, lungs and circulating system. Therefore, it is necessary to develop a safe natural antioxidant. For the purpose of developing natural antioxidant and antibacterial, the antioxidant activity and antibacterial effects of various extract solvents from Artichoke (Cynara Scolymus L.) leaf were determined. In this study, the extracts of Artichoke leaf dried from solvent extraction were examined by means of DPPH free radical scavenging activity and ABTS free radical scavenging activity. The effect of free radical scavenging compared with $\alpha$-tocopherol and L-ascorbic acid. In Artichoke leaf extract, evaluated by using DPPH and ABTS showed that the highest antioxidant activities were found to be in methanol extracts from DPPH radical ($IC_{50}$: $20.06{\mu}g\;mL^{-1}$), ABTS radical ($IC_{50}$: $16.01{\mu}g\;mL^{-1}$) and followed by ethanol > methyl chloride > ethyl acetate > n-Hexane. By using disc diffusion method, the antibacterial activity showed that the Artichoke leaf extract was found to be most effective against all of the tested organisms and the methyl chloride extract showed the most significant antibacterial effect against all of tests among 5 solvents extract, followed by ethyl acetate > n-Hexane > ethanol > methanol. As a result, optimal in antioxidant activity for Artichoke (Cynara Scolymus L.) leaf is methanol extract and for antibacterial effect is Methyl Chloride extract.

버섯균사체 발효 뽕잎 추출물의 항산화 활성 (Antioxidant Activity of Mulberry Leaves Extract Fermented by Hericium erinaceum)

  • 본 연구에서는 뽕잎(Morus alba)을 노루궁뎅이 버섯균사체(Hericium erinaceum)로 발효시킨 열수 추출물(MA-HE)의 항산화 활성을 확인하고자 하였다. MA-HE의 항산화 활성을 DPPH radical, ABTS radical 소거활성 측정을 통해 알아보았다. 그 결과 DPPH radical 소거활성은 $500{\mu}g/mL$ 농도에서 61.73%, ABTS radical 소거활성은 $250{\mu}g/mL$ 농도에서 97.39%로 나타났다. MA-HE은 DNA의 산화적 손상을 효과적으로 억제하였다. 또한 생체고분자물질인 사람의 혈청단백질의 산화적 손상을 억제하였다. 세포에 $H_2O_2$를 처리하였을 때 세포생존율에 비하여 발효물을 $100{\mu}g/mL$ 농도로 전 처리한 세포생존율은 8% 증가했으며, 발효물을 $50{\mu}g/mL$ 농도로 처리했을 경우 세포 내 활성산소의 축적이 유의적으로 감소되었다. 본 연구 결과는 MA-HE이 항산화 활성을 가지고 있으며 산화적 스트레스에 의해 야기되는 세포 독성에 대한 보호 작용도 뛰어난 것으로 사료되었다.

동백나무 추출물을 향장소재로 사용하기 위한 항균, 항산화 활성 탐색 (Antimicrobial and Antioxidant Activity of Camellia japonica Extracts for Cosmetic Applications)

  • This study was carried out to investigate the antimicrobial and antioxidative activities of Camellia japonica extracts for cosmetic applications. Antimicrobial effects of C. japonica were determined against Bacillus cereus by methanol extract of new leaf, stem and stem-leaf; Malassezia pachydermatis, by methanol extract of new leaf and stem-leaf. A methanol extract of new leaf of C. japonica showed strong antimicrobial effect using paper disc method against most species especially in Staphylococcus aureus. Antioxidative activities of C. japonica seed oils were determined by DPPH and ABTS radical scavenging activities. The value of $EC_{50}$ of DPPH scavenging activity was 500 mg/mL and that of ABTS scavenging activity was 96.10 mg/mL. C. japonica oil extracts showed lower antioxidative activities than those of gallic acid and ${\alpha}$-tocopherol. Electron microscopic observation of damaged virgin hairs of different ages gave a stabilizing effects after C. japonica seed oil treatment. These results indicated that the extracts of stem, leaf and seed of C. japonica could be used as cosmetic ingredient combined with appropriate formula.

솔잎, 녹차 및 비타민나무 잎 혼합추출물의 항산화효과 (Antioxidant effect of complex extracts from pine needle, green tea, and sea buckthorn leaves)

  • 본 연구에서는 솔잎, 녹차 및 비타민나무 잎의 추출물과 그 혼합추출물의 항산화 활성을 조사하였다. 시료 7종의 총 폴리페놀 및 총 플라보노이드 함량은 SE가 가장 높게 나타났다. DPPH, ABTS+ radical 소거능 및 환원력은 SE가 가장 높은 활성을 보였고, superoxide radical 소거능과 아질산염은 GE의 활성이 가장 높았다. PE는 대부분의 실험에서 녹차 및 비타민나무 잎과의 혼합추출물이 단일추출물에 비해 높은 활성을 보였다. GE는 DPPH radical 소거능(300 ppm), ABTS+ radical 소거능(300 ppm) 및 환원력(100 및 500 ppm)에서 비타민나무 잎과 혼합추출하면 그 활성이 높아졌고, SE는 superoxide radical 소거능(100 ppm)과 아질산염 소거능(300 ppm)에서 녹차와 혼합추출하면 활성이 높아지는 경향을 보였다. 이상의 결과에서 PE는 단일 및 혼합추출물 7종 중에서 가장 낮은 활성을 나타낸 시료로 녹차 및 비타민나무 잎과 함께 혼합(PGSE)하여 사용한다면 솔잎의 낮은 항산화 활성을 개선시킬 수 있을 것으로 사료된다. 또한 본 연구 결과는 솔잎, 녹차 및 비타민나무 잎을 이용한 건강 기능성 식품 산업에서 기초자료로 활용할 수 있을 것으로 생각된다.

지역별 두중(杜仲)의 지표성분 및 항산화 활성 비교 (Comparison of Anti-oxidant Activity and Marker Compounds in Eucommiae Cortex Samples from Regional Groups)

  • Objectives : It is necessary to manage herbal medicines based on effectiveness by comparing the efficacy of herbal medicines by region. In this study, we compared anti-oxidative activity and marker compounds of Eucommiae Cortex by regional groups. Methods : Eucommiae Cortex grown and harvested in Gangjin, Sancheong, Yeongwol, Jangsu, and Jecheon were used. Eucommiae Cortex was extracted in distilled water at $100^{\circ}C$ for 3 hours, and filtered. filtered. Extract samples were freeze-dried at $-80^{\circ}C$. Comparison of anti-oxidant activity in Eucommiae Cortex samples from regional groups was measured in 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging, 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) free radical scavenging, and ferric reducing antioxidant power (FRAP) between regional groups of Eucommiae Cortex. In addition, high-performance liquid chromatography (HPLC) analysis was conducted to compare pinoresinol diglucoside concentration by regional groups. Results : HPLC analysis found that pinoresinol diglucoside concentration, widely known as the marker compound of Eucommiae Cortex, was the highest in Gangwon Yeongwol. There was a significant difference in anti-oxidative activity of Eucommiae Cortex between regional groups as discovered in DPPH, ABTS and FRAP assays. DPPH free radical scavenging was the highest in Jeonbuk Jangsu. ABTS free radical scavenging was the highest in Jeonbuk Jangsu and Chungbuck Jecheon. FRAP was the highest in Jeonbuk Jangsu. Conclusions : Although pinoresinol diglucoside concentration was high, anti-oxidative activity was not proportionately high. Pinoresinol diglucoside concentration was the highest in Gangwon Yeongwol. Anti-oxidative activity was the highest in Jeonbuk Jangsu.

고추장과 발효액이 첨가된 알래스카 산 프리미엄 명란의 ABTS, FRAP, total phenolic acid의 항산화 특성 분석 (Antioxidant Properties (ABTS, FRAP, Total Phenolic Content) of Alaska and Gochujang Pollock Roes and Fermented Pollock Roe Seasoning)

  • 알래스카 산 대구(Gadus chalcogrammus)는 북태평양에 분포하는 대구과 한류 어종이다. 알래스카 산 대구의 명란 원료, 프리미엄 고추장 명란, 프리미엄 발효 명란을 증류수와 에탄올로 추출하여 2,2'-azino-bis (3-ethylbenzthiazoline-6-sulphonic acid) (ABTS+), 철 환원 항산화능(Ferric Reducing Antioxidant Potential, FRAP), 총 페놀함량을 평가하였다. 명란 원료의 증류수와 에탄올 추출물의 ABTS+ 소거능은 1.0 mg/ml 농도에서 각각 50.1%, 53.1%였다. 고추장 명란의 증류수와 에탄올 추출물의 ABTS+ 소거능은 1.0 mg/ml 농도에서 각각 71.3%, 71.6%였다. 발효 명란의 증류수와 에탄올 추출물의 ABTS+ 소거능은 1.0 mg/ml 농도에서 각각 68.7%, 70.4%였다. 명란원료의 증류수와 에탄올 추출물의 ABTS+에 대한 최소 저해 농도 값($EC_{50}$)은 각각 12.49 ug/ml과 12.21 ug/ml이었다. 고추장 명란의 증류수와 에탄올 추출물의 FRAP에 대한 최소 저해 농도 값($EC_{50}$)은 각각 10.67 ug/ml과 10.56 ug/ml이었다. 발효 명란의 증류수와 에탄올 추출물의 총 페놀 함량에 대한 최소 저해 농도 값($EC_{50}$)은 각각 10.45 ug/ml과 10.31 ug/ml이었다. Gallic acid를 대조구로 할 때 총 페놀 함량에 대한 상대적 활성은 명란 원료가 52.0%, 고추장 명란이 61.1%, 발효 명란이 63.6%였다. 명란 원료보다 고추장 명란과 발효 명란에서 ABTS+, FRAP 소거능 및 총 페놀 함량이 우수하였다.

여주 품종별 폴리페놀, 플라보노이드 함량과 라디칼 소거활성 및 아질산염 소거능 (Different of Total Phenolics and Flavonoids, Radical Scavenging Activities and Nitrite Scavenging Effects of Momordica charantia L. According to Cultivars)

  • This study was carried out in order to determine the biological activities such as antioxidant activities in nine cultivars of Momordica charantia L. (Korea, China, Japan and Philippine native cultivars). Their antioxidant activities were measured using DPPH free radical scavenging, ABTS cation radical scavenging, and Nitrite scavenging ability. The highest total polyphenol content (16.82 ${\mu}g/m{\ell}$) measured in the native Korea cultivar, and this value was 4.0 ${\mu}g/m{\ell}$ higher than that of the 'Verde Beuhas' cultivar (12.82 ${\mu}g/m{\ell}$). The 'Peacock' cultivar had the highest total flavonoid amount which was 4.38 ${\mu}g/m{\ell}$. The free radical scavenging activity using DPPH method was the highest in the native China cultivar ($RC_{50}$ = 102.6 ${\mu}g/m{\ell}$). ABTS cation radical scavenging activity according to cultivars was significantly higher in 'Peacock'. Nitrite scavenging ability showed the most remarkable effect at the pH 1.2, exhibited to 81.5${\sim}$86.9% by addition of ethanol extract 1${\mu}g/m{\ell}$ from Momordica charantia L. These results suggest that Momordica charantia L. had the potent biological activities, and that their activities exhibited differently depending on cultivars.

오리나무 가지 유래 Oregonin의 인체 모유두 세포 Apoptosis 조절 효능 (Modulative Effect of Human Hair Dermal Papilla Cell Apoptosis by Oregonin from the Braches of Alnus japonica)

  • 최근 식품의약품안전처에서 그동안 의약외품으로 분류되었던 '탈모 증상 완화' 관련 제품이 기능성 화장품의 범주에 포함됨에 따라서 관련 산업계에 큰 관심 중 하나라고 할 수 있다. 본 연구에서는 기존 의약품들의 한계점인 부작용의 위험률이 적으면서 탈모를 과학적으로 조절시켜줄 수 있는 객관적인 근거를 제시할 수 있는 천연물 유래 신소재를 발굴하고자, 오리나무 가지로부터 분리한 oregonin은 강력한 활성산소종 소거능을 가지며 탈모와 연관성이 높은 모유두 세포의 사멸을 명확하게 조절함을 확인하였다. 결론적으로 oregonin을 포함한 오리나무 추출물은 두피에서 발생한 활성산소를 강력하게 제거하는 효능을 기반으로 하여, 산화적 스트레스에 의한 모유두세포의 사멸 억제 및 조절을 통하여 탈모방지 효과를 기대할 수 있는 천연 탈모 방지 및 양모 신소재로서의 개발가능성이 있는 것으로 확인하였다.

비자 추출물의 식품부패균에 대한 항균효과 및 항산화활성 (Antimicrobial Effects on Food-Borne Pathogens and the Antioxidant Activity of Torreya Nucifera Extract)

  • This study investigates antimicrobial effects of food-borne pathogens and the antioxidant activity of Torreya nucifera extract. The growth of food-borne pathogens including Staphylococcus aureus, Pseudomonas aeruginosa, and Candida albicans was inhibited by the extract. The antimicrobial activity of the extract was highest for Staphylococcus aureus among seven gram-positive bacteria and for Pseudomonas aeruginosa among six gram-negative bacteria. The extract exhibited slightly lower DPPH radical-scavenging activity, but its ABTS radical-scavenging activity was higher than that of ${\alpha}$-tocopherol. The results demonstrate the extract's antimicrobial effects on food-borne pathogens as well as potent antioxidant capacity and suggest that Torreya nucifera may be used as a natural antibacterial agent and an effective antioxidant in food.

Quality Evaluation of Cheongpomuk Added with Chlorella Powder

  • This study examined the quality characteristics to evaluate the optimal mixing ratio of a chlorella powder in Cheongpomuk. The total polyphenol and total flavonoids contents of Cheongpomuk and chlorella powder was 279.90 ㎍/mL, 489.50 ㎍/mL and 48.79 ㎍/mL, 687.85 ㎍/mL respectively. The DPPH free radical scavenging activities and ABTS radical scavenging activities of chlorella powder was 65.39%, 91.28% respectively. The L values (58.85~26.23), a values (-0.44~-5.84) of the Hunter's color values decreased and b value (-10.98 ~4.12) increased significantly in proportion to the amount of chlorella powder. The total polyphenol contents (55.00~734.20 ㎍/mL), total flavonoids contents (41.00~88.29 ㎍/mL), DPPH free radical scavenging activities (35.10~65.50%) and ABTS radical scavenging activities (50.29~92.86%) of Cheongpomuk increased significantly in groups of chlorella powder. In the physical properties, the hardness (135.93~103.93 g/㎠), chewiness (6.76~3.76 g), springiness (4.92~4.36 %), and gumminess (147.70~94.23) of Cheongpomuk decreased significantly in proportion to the amount of chlorella powder. The Cheongpomuk containing 15% chlorella powder showed a high score with regard to color, flavor, moistness, chewiness and overall acceptance. The appropriate addition amount of chlorella powder for making Cheongpomuk is 15%. This study indicate that Cheongpomuk treated with chlorella powder had the highest functional component and antioxidant activity.
