Antioxidant Properties of 7 Domestic Essential Oils and Identification of Physiologically Active Components of Essential Oils against Candida albicans (식물정유 7종의 항산화능 분석 및 Candida albicans 생장 억제 정유의 생리활성 성분 구명)

  • In this study, we selected two essential oils, Citrus unshiu and Cinnamomum cassia with superior antioxidant effects from the essential oils of 7 wild plants in South Korea and examined their antimicrobial activity against Candida albicans, which causes dermatitis to identify the antimicrobial components in the essential oils. As a result of measuring DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity, SC50 value of the Citrus unshiu essential oil was 0.010 mg/mL, while for the Cinnamomum cassia essential oil, SC50 value was 0.09 mg/mL. In addition, when ABTS (2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activity was measured, SC50 value of the Citrus unshiu essential oil was 0.09 mg/mL, while for the Cinnamomum cassia essential oil, it was 0.06 mg/mL, exhibiting high antioxidant activity. For the minimum inhibitory concentration (MIC), the essential oil of Cinnamomum cassia was 1.25 mg/mL and that of Citrus unshiu was 5 mg/mL, demonstrating a high antimicrobial activity of the Cinnamomum cassia essential oil. Through the thin layer chromatography (TLC) bioassay, we assessed the antimicrobial activity against C. albicans according to the fraction components of the two essential oils. Also, by using preparative TLC (prep. TLC), we obtained the active fractions, and by performing GC/MS analysis of the components with the same Rf value, we identified the antimicrobial-active components. As a result, the main components having antioxidant and antimicrobial activities were cinnamyl acetate, eucalyptol, linalool, and citral of the Cinnamomum cassia essential oil and linalool from the Citrus unshiu essential oil. Also, based on the analysis of the fractional components that showed antioxidant and antimicrobial activities in both of the two essential oils, it was found that linalool has antioxidant activity, while cinnamyl acetate, eucalyptol, citral, and geranyl acetate have antioxidant and antimicrobial activities.

A Study on the Cosmetic Materials of Abies nephrolepis MAX. Extracts with Whitening Activity (미백활성을 가진 분비나무(Abies nephrolepis MAX.) 추출물의 화장품 소재로써 연구)

  • We conducted a study on the whitening activity of Abies nephrolepis MAX. extracts. The electron-donating ability of 70% ethanol extracts from A. nephrolepis MAX. Stem (AN-S) and A. nephrolepis MAX. Leaf (AN-L) were found to be 89.4% and 90.9% at 1,000 ㎍/ml concentration, respectively. Their ABTS+ radical scavenging abilities were found to be 88.8% and 96% 500 ㎍/ml concentration, respectively. Their tyrosinase inhibitory effects were found to be 48.5% and 31.1% at 500 ㎍/ml concentration, respectively. Cell survival rates measured in B16F10 were examined with the AN-S and AN-L extracts. Results showed cell viabilities of 98.3% and 94.4% at 500 ㎍/ml concentration, respectively. The protein and mRNA expression inhibitory effects of AN-S and AN-L extracts were measured by Western blotting and RT-PCR, respectively, at 25, 50 and 100 ㎍/ml concentrations. The results of western blotting showed that the AN-S extract at 100 ㎍/ml concentration decreased the TRP-1 protein expression rate by 46%. The results of Western blotting also showed that the AN-L extract at 100 ㎍/ml concentration decreased the MITF protein expression rate was by 54.6%. Consequently, in the case of MITF, TRP-1, TRP-2, and tyrosinase, the amount of mRNA expressed decreased as the concentration of the extracts increased. Our results suggested that A. nephrolepis MAX. extracts can be used as a functional cosmetic material by confirming that they can be used as a whitening material.

Antioxidant and Antiwrinkle Effects of Persimmon Leaves extract (시엽(Persimmon Leaves) 에탄올 추출물의 항산화와 항주름 효과)

  • In this study, we investigated the antioxidant and anti-winkle activity in human fibroblast cell (CCD-986sk) of Persimmon Leaves (PL) as a cosmetic ingredient. As a result of investigating antioxidant activity through electron-donating ability and ABTS+ radical scavenging assay, the PL showed concentration-dependent antioxidant activity similar to ascorbic acid, a control group, at a concentration of 1,000 ㎍/ml. As a result of investigating the anti-wrinkle effect through elastase inhibition and collagenase inhibition assay, the PL showed concentration-dependent antioxidant activity similar to epigallocatechin gallate, a control group, at a concentration of 1,000 ㎍/ml. As a result of measuring the synthesis rate of pro-collagen type I and the inhibition rate of MMP-1 in UVB-induced CCD-986sk cells, the control group EGCG showed a 90.2% pro-collagen synthesis rate at 20 ㎍/ml and PL showed an 88.5% synthesis rate at 30 ㎍/ml. In addition, the inhibition rate of MMP-1 of 33.0% and 40.8% were confirmed in EGCG 20 ㎍/ml and PL 30 ㎍/ml, respectively. As a result of measuring the protein expression of pro-collagen type I and MMP-1 in the PL through western blot, it was confirmed that the protein expression of pro-collagen type I increased, and MMP-1 decreased when the PL was treated together compared to the UVB alone group. According to the above experimental results, it is expected to be used as a natural product material for cosmetics by confirming that the PL prevent photoaging caused by UVB stimulation and have antioxidant and anti-wrinkle effects.

Anti-inflammatory Effects of Rumohra adiantiformis Extracts Fermented with Bovista plumbea Mycelium in LPS-stimulated RAW 264.7 Cells (LPS로 자극된 RAW 264.7 세포에서 찹쌀떡버섯 균사체로 생물전환된 루모라고사리 추출물의 항염증 효과)

  • This study was designed to evaluate the anti-inflammatory effects of Rumohra adiantiformis extracts fermented with Bovista plumbea mycelium (B-RAE) in LPS-stimulated RAW 264.7 cells. The total polyphenol and total flavonoid content of B-RAE were 379.26±7.77 mg/g and 50.85±3.08 mg/g, respectively. The results of measuring the antioxidant activity of B-RAE showed that it scavenges 2, 2-diphenyl-1-picrylhydrazyl (DPPH), 2, 2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS), and superoxide anion radical in a dose-dependent manner. B-RAE inhibited nitric oxide (NO) production in a dose-dependent manner without affecting cell viability. The gene expression of pro-inflammatory cytokines such as tumor necrosis factor-α (TNF-α), interleukin-lβ (IL-1β), and IL-6 was measured using real time quantitative reverse transcription PCR (qRT-PCR). We found that, compared to the LPS-treated group, B-RAE significantly reduced the mRNA levels of the pro-inflammatory cytokines in a concentration-dependent manner. The expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2), the phosphorylation of transcription factors such as nuclear factor-κB (NF-κB), and the mitogen-activated protein kinase (MAPK) signaling pathway proteins were assessed using Western blot analysis. We found that B-RAE significantly suppressed the expression of iNOS and COX-2, but their expression was increased by LPS treatment. In addition, the phosphorylation of NF-κB and IκB, which was increased by LPS treatment, was reduced with B-RAE treatment. The effect of B-RAE on the phosphorylation of the MAPK signaling pathway proteins was measured, and the phosphorylation of extracellular signal-regulated kinase (ERK) and the p38 MAPK proteins decreased in a dose-dependent manner, while the phosphorylation of c-Jun N-terminal kinase (JNK) increased. These anti-inflammatory effects of B-RAE may thus have been achieved through the high antioxidant activity, the inhibition of NO production through the suppression of iNOS and COX-2 expression, the inhibition of the NF-κB pathway, and the suppression of pro-inflammatory cytokine expression.

Functional Components and Antioxidant Activities of Perilla Leaf Genetic Resource (잎들깨 유전자원의 기능성분 및 항산화 활성)

  • The objective of this study was to investigate the distribution of functional compounds in perilla leaves of various genetic resources and their antioxidant activities. A comprehensive analysis of functional compounds was conducted for 90 genetic resources, focusing on total polyphenol content (TPC), total flavonoid content (TFC), individual phenolic content (IPC), and lutein. Their antioxidant activities were then analyzed based on their radical scavenging capacity using ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) and DPPH (2,2-diphenyl-1-picrylhydrazyl). The TPC content exhibited a range of 13.19 to 35.85 mg gallic acid equivalent/g, whereas the TFC content varied from 11.74 to 46.51 mg catechin equivalent/g. Total IPC was detected in a range of 6,310.98 to 40,491.82 ㎍/g. Lutein was detected at levels between 70.97 and 597.97 ㎍/g. ABTS and DPPH radical scavenging activities of perilla leaves ranged from 30.39 to 58.58 mg trolox equivalent (TE)/g and from 7.74 to 46.56 mg TE/g, respectively. Furthermore, correlation analysis demonstrated that rosmarinic acid, a phenolic acid, exhibited a significantly positive correlation with antioxidant activity. These findings suggest that various genetic resource of perilla leaves could effectively mediate antioxidant capacity. Results of this study provide valuable information for use of perilla leaves in Korea as functional food materials.

Correlation between phytochemical contents in peel and flesh of cold-stored Fuji apple (저온 저장 후지 사과의 과피와 과육 중 파이토케미컬 함량의 상관관계)

  • This study determined the relationships between the total anthocyanin content in apple skin and the total polyphenols, flavonoids, proanthocyanidins, and soluble solids contents in the flesh of cold-stored Fuji apples. Total anthocyanin content in apple skin ranged from 0.130±0.005 mg CE/g fw to 0.262±0.028 mg CE/g fw, and the overall average was 0.200±0.008 mg CE/g fw. The total polyphenolic compounds in the flesh was ranged from a 4.283±0.141 mM GAE/g fw to 8.207±0.234 mM GAE/g fw, and the average content was 6.275±0.177 mM GAE/g fw. The total flavonoid content ranged from 4.510±0.080 mM QE/g fw to 2.467±0.458 mM QE/g fw, and the average total flavonoid content was about 3.586 mM QE/g fw. The total proanthocyanidin content was relatively high, ranging from 3.475±0.577 mM EE/g fw to 6.816±0.277 mM EE/g fw, and the soluble solid in the flesh was about 12 °Brix to 14 °Brix. The DPPH radical scavenging activity of extracts from apple flesh ranged from 66.36% to 94.99%, and the ascorbate equivalent concentration was 0.482 mM. The ABTS radical scavenging activity was 99.12% to 99.9%, indicating a higher inhibitory activity than the DPPH inhibitory activity, and the ascorbate equivalent concentration was 0.486 mM. The correlation between the total anthocyanin and total polyphenolic compounds was y = 15.192x + 3.2169 (R2=0.2748), but the concentration of total polyphenolic compounds increased when the total anthocyanin content was increased. The correlation equation of total anthocyanin with total flavonoids was y = 15.18x + 0.5555 (R2=0.6226), with total proanthocyanin was y = 14.918x + 2.3422 (R2=0.3372), and with soluble solid was y = 10.558x + 11.126 (R2=0.1925), indicating that the correlation of total anthocyanin with total flavonoid was higher than that with soluble solid.

Anti-wrinkle effects of solvent fractions from Jubak on CCD-986sk (CCD-986sk 세포 내 주박 분획물의 항주름 효능)

  • In this study, in order to evaluate the possibility of using Jubak as a functional cosmetic material, evaluation of antioxidant activity according to fractions and anti-wrinkle efficacy in CCD-986sk cells, a human fibroblast, were conducted. As a result of confirming the antioxidant activity by measuring ABTS+ radical scavenging ability, Jubak's Ethyl Acetate fractions was found to be 75.5% at a concentration of 1,000 ㎍/ml, showing the highest antioxidant activity among the extraction solvents. The wrinkle improvement effect was confirmed by measuring the inhibitory activity of elastase and collagenase, and in both test results, Jubak's Ethyl Acetate fractions showed the highest efficacy at a concentration of 1,000 ㎍/ml. As a result of measuring the synthesis rate of pro-collagen type I in CCD-986sk cells induced by UVB, Jubak showed the highest efficacy in the order of Ethyl Acetate, Water, Acetonitrile, and Hexan fractions at the same concentration of 20 ㎍/ml. As a result of measuring the inhibition rate of MMP-1, a collagen degrading enzyme, all four solvent fractions showed an efficacy of more than 70% at 20 ㎍/ml. As a result of measuring the mRNA expression levels of pro-collagen type I, MMP-1, and MMP-3 in a real-time PCR experiment, the protein expression level of pro-collagen type I increased when treated with Jubak fractions compared to the UVB group alone. The mRNA expression levels of MMP-1 and MMP-3 were confirmed to be decreased, and Ethyl Acetate fractions was the most effective in improving wrinkles after the control group (EGCG). As a result, it was confirmed that the Ethyl Acetate fractions among Jubak's solvent fractions has an anti-wrinkle effect against photoaging caused by UVB stimulation, and is expected to be used as a natural material for cosmetics.

Quality characteristics and antioxidant activity of rice muffins added with Orostachys japonicus powder (와송 분말을 첨가한 쌀머핀의 품질 특성 및 항산화 활성)

  • This study aimed to suggest the usability and optimal amount of Orostachys japonicas in foods. Orostachys japonicus powder was added at 0% (Con), 3% (O3), 5% (O5), 7% (O7), and 9% (O9) to rice muffins and the quality and antioxidant characteristics of the muffins were determined. The moisture content and pH of the muffins decreased as the amount of Orostachys japonicus powder added increased. The weight increased as the amount of Orostachys japonicas powder added increased, but the volume, specific volume, and baking loss rate decreased. The L-and b-values of the muffins decreased as the a-value increased. The texture, hardness, gumminess, and chewiness of muffins increased while adhesiveness, springiness, and cohesiveness decreased. Polyphenol, flavonoid, DPPH, and ABTS+ radical scavenging activity, as well as reducing power, increased as the amount of Orostachys japonicus powder added increased. In the sensory test, the 5-7% addition group showed high scores in appearance, flavor, taste, texture, and overall preference. The degree of flavor and astringency of muffins, which are evaluation items for characteristic strength, increased as the amount of Orostachys japonicus powder added increased, and the level of moistness decreased. Therefore, the addition of 5-7% of Orostachys japonicus powder is thought to have a positive effect on the muffins. The purpose of this study was to suggest the usability and optimal addition amount of Orostachys japonicus powder and to provide basic data on foods with Orostachys japonicus added.

Antioxidant Compounds and Activities of Foxtail Millet, Proso Millet and Sorghum with Different Pulverizing Methods (분쇄방법에 따른 조, 기장, 수수의 항산화성분 및 항산화활성)

  • We evaluated the antioxidant compounds and activity of the methanolic extracts of foxtail millet (FM), proso millet (PM), and sorghum (SG) using different pulverizing methods (pin mill and ultra fine). The particle size of the FM, PM, and SG were 102.12, 89.52, and $102.25\;{\mu}m$, respectively, using the pin mill pulverizer. The sizes were 9.43, 9.52, and $10.18\;{\mu}m$, respectively, using the ultra fine pulverizer. There was no difference in moisture, crude fat, ash, or protein content between the two different pulverizing methods. The total ${\gamma}$-oryzanol content of the FM using the pin mill and ultra fine pulverizers was 116.07 and $145.30\;{\mu}g$/g, respectively. The total polyphenol content of the SG using the pin mill and ultra fine pulverizers was 14.58 and 15.03 mg/g extract residue, respectively. There was no difference in total flavonoid or tannin content of the methanol extracts between the two different pulverizing methods. The major phenolic compounds in FM were pyrogallol, gallic acid, (+)-catechin, salicylic acid, ferulic acid, and rutin; in PM, they were pyrogallol, rutin, gallic acid, kaempfrol, and salicylic acid; in SG, they were (+)-catechin, salicylic acid, pyrogallol, myricetin, hesperidin and chlorogenic acid. SG had a higher radical scavenging activity than FM or PM extracts. The DPPH and ABTS radical scavenging activities of the SG extracts using the ultra fine pulverizer were 178.10 and 251.56 mg TE/g extract residue, respectively. We noted a significant correlation between free radical scavenging activity and polyphenolic compound.

Inhibitory activities on biological enzymes of extracts from Oplismenus undulatifolius (주름조개풀(Oplismenus undulatifolius) 추출물의 생리활성 효소 억제 효과)

  • The phenolic contents which were extracted with water and 70% ethanol from O. undulatifolius were 7.7, 10.1 mg/g, respectively. The 1,1-diphenyl-2-picrylhydrazyl free radical scavenging activity of water and ethanol extracts were 78, 82% at $50{\mu}g/mL$ phenolics, respectively. The 2,2'-Azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) radical cation decolorization activity were 92, 76% at $100{\mu}g/mL$ phenolics. Antioxidant protection factor in water and ethanol extracts at $200{\mu}g/mL$ phenolics were 1.51 and 2.08 PF, respectively. Thiobarbituric acid reactive substance were 84% in water extracts and 99% in ethanol extracts at $50{\mu}g/mL$ phenolics, respectively. The inhibition activity on ${\alpha}-Glucosidase$ was 44% in ethanol extracts at $200{\mu}g/mL$ phenolics. The inhibition activity on ${\alpha}-amylase$ was 37-88% in water extracts at $50-200{\mu}g/mL$ phenolics. The tyrosinase inhibition activity as whitening effect were 82% in ethanol extracts. The elastase inhibition activity were 4, 61% in water and ethanol extracts, respectively. The collagenase inhibition activity of antiwrinkle effect showed an excellent wrinkle improvement effect as 39% in water extracts and 67% in ethanol extracts at $200{\mu}g/mL$ phenolics, respectively. The hyaluronidase inhibition activity as anti-inflammation effect of ethanol extracts was confirmed to 46% of inhibition at $200{\mu}g/mL$ phenolic. The astringent effect of water and ethanol extracts was confirmed to 13, 32% of effect at $200{\mu}g/mL$ phenolic, respectively.
