인체암세포주에 대한 천연자원의 세포독성 검색 (Ⅱ) (Cytotoxic Activities of Herbal Drugs Against Human Cancer Cell Lines (Ⅱ))

  • In our continuing search for new antineoplastic agents from natural products, one hundred and thirty-five herbal drugs were extracted with petroleum ether/ether (1:1), ethyl acetate and methyl alcohol, successively and their cytotoxicities were evaluated against A549 (human lung carcinoma) and SK-OV-3(human ovary adenocarcinoma) cell lines. Among them, fifteen kinds of ether extracts, eighteen kinds of ethyl acetate extracts and seven kinds of methanol extracts showed significant cytotoxic activities (above 70% inhibition) against A549 cell lines at a concentration of $40\;{\mu}g/ml,$ while ten kinds of ether extracts, thirteen kinds of ethyl acetate extracts and six kinds of methanol extracts demonstrated significant cytotoxic activities against SK-OV-3 cell lines at the above same concentration.

인체암세포주에 대한 천연자원의 세포독성 검색 (I) (Cytotoxic Activities of Herbal Drugs against Human Cancer Cell Lines (I))

  • For the search of new antineoplastic agents from natural resources, two hudred and one kinds of oriental medicinal drugs were extracted with petroleum ether/ether(1:1), ethyl acetate and methyl alcohol, successively and their cytotoxicities were evaluated against A549 (human lung carcinoma) and SK-OV-3 (human ovary adenocarcinoma) cell lines. Among them, thirty kinds of ether extracts, forty-one kinds of ethyl acetate extracts and nine kinds of methanol extracts showed significant cytotoxic activities (above 70% inhibition) against A549 cell lines at a concentration of $40\;{\mu}g/ml$. And also, twenty-four kinds of ether extracts, thirty-one kinds of ethyl acetate extracts and six kinds of methanol extracts showed significant cytotoxic activities against SK-OV-3 cell lines at the same concentration.

발효콩 추출물의 항돌연변이원성 및 세포독성 효과 (Antimutagenic and Cytotoxicity Effects of Fermented Soybean Extract)

  • This study was carried out to determine the antimutagenic and anticancer effects of fermented soybean using Ames test and cytotoxicity, respectively. The ethyl acetate fraction (200 g/plate) of fermented soybean in the Salmonella typhimurium TA100 strain showed 86.6% of inhibition rate against the mutagenesis induced by N-methyl-N'-nitro-N-nitrosoguanidine(MNNG). In addition, the suppression of ethyl acetate fraction with same concentration of fermented soybean in the Salmonella typhimurium TA98 and TAI00 strains showed 82.4% and 90.8% inhibition against 3-amino-l,4-dimethyl-5H-pyrido-(4,3-b)indol (Trp-P-l), respectively. The cytotoxicity effects of fermented soybean against the cell lines with human lung carcinoma (A549), human gastric carcinoma (AGS) and human breast adenocarcinoma (MCF-7) were inhibited with the increase of the extract concentration. The treatment of 1.0 mg/mL ethyl acetate fraction of fermented soybean showed strong cytotoxicities of 71.6%, 91.5% and 80.7% against A549, AGS and MCF-7, respectively.

광천수의 항산화성, 항돌연변이원성 및 세포독성 효과 (Antioxidative, Antimutagenic and Cytotoxic Effects of the Mineral Water)

  • 지하 943 m(3호공)로부터 취수한 경산 광천수의 유$\cdot$무기성분을 분석한 결과 총 17종류의 성분 중 무기물은 11 종류가 검출되었으며 무기성분 중에서는 나트륨과 칼슘 성분이 많이 함유되어 있는 것이 특징이었다. 그리고 구리와 납과 같은 중금속과 탄산가스 및 유화수소는 검출되지 않았다. 그리고 항산화 활성, 항돌연변이원성 및 인간암세포 성장억제 실험 결과 항산화 활성에서는 비교적 약한 활성을 나타내었다. MNNG, 4NQO, Trp-P-1및 $B({\alpha})P$와 같은 돌연변이원을 이용한 미생물 수준에서의 항돌연변이원성 실험 결과 S. typhimurium TA98에 대해서 간접변이원인 Trp-P-1은 시료 농도 $200{\mu}g/plate$ 첨가 시 $54\%$의 억제 활성을 나타내었다. 또한 S. typhimurium TA100 균주에서 4NQO 및 MNNG는 각각 $65.8\%$$58.6\%$의 억제활성을 보였으며 $B({\alpha})P$ 및 Trp-P-1에 대해서도 각각 67%와 63%의 높은 억제활성을 나타내었다. 그리고 광천수의 항암활성을 규명하기 위한 암세포 성장억제 효과에서는 시료농도 $50{\mu}g/well$ 첨가 시 A549, Hela, AGS 및 MCF-7에 대해서 각각 $66\%,\;45.6\%,\;37.7\%$$47.6\%$의 억제효과를 나타내었다. 또한 인간 정상 신세포 293에 대한 온천수 농도에 따른 세포독성 효과는 $20\%$ 이하의 낮은 생육 억제율을 보였으며 이것은 광천수가 정상세포에 대해서는 비교적 낮은 독성효과를 나타냄을 알 수 있었다.

해양심층수염 및 다시마분말 첨가 고추장추출물의 항돌연변이성 및 암세포 성장억제효과 (Antimutagenic and Cytotoxic Effects of Kochujang Extracts Added Deep Sea Water Salt and Sea Tangle)

  • 본 연구는 Ames test와 SRB assay를 통해 SK와 SDK 메탄올 추출물을 이용하여 항돌연변이원성과 세포독성 효과에 대해 살펴보았다. S. Typhimurium TA98과 TA100 균주를 사용하여 돌연변이원성을 실시한 결과 SK와 SDK 메탄올 추출물 자체의 돌연변이원성은 없었으며, MNNG와 4NQO를 유도하여 높은 항돌연변이 효과를 나타내었다. S.Typhimurium TA98에서 4NQO를 유도하여 SDK($200{\mu}g$/plate) 메탄올 추출물 처리 시에 71.4%의 억제효과를 나타내었으며, 반면에 S. Typhimurium TA100에 4NQO와 MNNG의 돌연변이를 유도하였을 때 각각 56.1%와 83.6%의 억제율을 보였다. 암세포 성장 억제효과를 검토한 결과 샘플처리 시 농도 의존적 증가를 보였으며, SDK 메탄올 추출물 1 mg/mL 첨가 시 HeLa, Hep3B, MCF-7, AGS 및 A549에서 각각 61.5%, 61.3%, 51.4%, 57.9% 및 77.7%의 억제효과를 나타내었다. SK 메탄올 추출물 첨가 결과 $51{\sim}58%$ 정도의 억제효과를 보였다. 대조적으로 SDK 메탄올 추출물을 1mg/mL 첨가 시 293에서 $2{\sim}38%$의 세포독성을 나타내었다.

Radix Tetrastigma Hemsleyani Flavone Induces Apoptosis in Human Lung Carcinoma A549 Cells by Modulating the MAPK Pathway

  • Radix Tetrastigma Hemsleyani Flavone (RTHF) is widely used as a traditional herb for its detoxification and anti-inflammation activity. Recently, several studies have shown that RTHF can inhibit growth and induce apoptosis in human cancer cell lines. However, the mechanisms are not completely understood yet. In this study we investigated the potential effects of RTHF on growth and apoptosis in human lung adenocarcinoma A549 cells as well as its mechanisms. A549 cells were treated with RTHF at various concentrations for different times. In vitro the MTT assay showed that RTHF had obvious anti-proliferation effects on A549 cells in a dose- and time-dependent manner. Cell morphological changes observed by inverted microscope and Hoechst33258 methods were compared with apoptotic changes observed by fluorescence microscope. Cell apoptosis inspected by flow cytometry showed significant increase in the treatment group over the control group (P<0.01). Expression of apoptosis related Bax/Bcl-2, caspases and MAPK pathway proteins were detected by Western blotting. The results showed that RTHF up-regulated the Bax/Bcl-2 ratio and cle-caspase3/9, cle-PARP expression in a dose-dependent manner. Expression of p-p38 increased, p-ERK decreased significantly and that of p-JNK was little changed in the RTHF group when compared with the control group. These results suggest that RTHF might exert anti-growth and apoptosis activity against lung cancer A549 cells through activation of caspases and Bcl-2 family proteins and the MAPK pathway, therefore presenting as a promising therapeutic agent for the treatment of lung cancer.

꾸지뽕나무 메탄올 추출물의 세포독성 (Cytotoxicity of Methanol Extracts from Cudrania tricuspidata Bureau)

  • In order to develop as a natural source of anticancer materials of Cudrania tricuspidata, the cytotoxicity of methanol extracts by harvesting parts and times against 8 cell lines including 293 (normal kidney cells) and A-431 (epidermoid carcinoma cells) were investigated using MTT assay. All harvesting parts had hardly cytotoxicity against 293. And methanol extracts of stem bark and root bark showed very high cytotoxicities against 7 cancer cell lines. The cytotoxicity was the highest against HeLa (cervix adenocarcinoma cells) and followed by MCF-7 (breast adenocarcinoma cells), AGS (stomach adenocarcinoma cells), HT-29 (colon adenocarcinoma cells), HepG2 (hepatoblastoma cells), A549 (lung carcinoma cells) and A-431. By the way, leaf extract had a cytotoxicity against only AGS and ripe fruit extract had no cytotoxicity. Among harvesting times, the cytotoxicity of root bark were high from April to September but that of stem bark showed a little difference. These results showed that anticancer activities of Cudrania tricuspidata extracts were eventful changes by harvesting parts and times.

Comparative proteomics and global genome-wide expression data implicate role of ARMC8 in lung cancer

  • Background: Cancer loci comprise heterogeneous cell populations with diverse cellular secretions. Therefore, disseminating cancer-specific or cancer-associated protein antigens from tissue lysates could only be marginally correct, if otherwise not validated against precise standards. Materials and Methods: In this study, 2DE proteomic profiles were examined from lysates of 13 lung-adenocarcinoma tissue samples and matched against the A549 cell line proteome. A549 matched-cancer-specific hits were analyzed and characterized by MALDI-TOF/MS. Results: Comparative analysis identified a total of 13 protein spots with differential expression. These proteins were found to be involved in critical cellular functions regulating pyrimidine metabolism, pentose phosphate pathway and integrin signaling. Gene ontology based analysis classified majority of protein hits responsible for metabolic processes. Among these, only a single non-predictive protein spot was found to be a cancer cell specific hit, identified as Armadillo repeat-containing protein 8 (ARMC8). Pathway reconstruction studies showed that ARMC8 lies at the centre of cancer metabolic pathways. Conclusions: The findings in this report are suggestive of a regulatory role of ARMC8 in control of proliferation and differentiation in lung adenocarcinomas.

메밀 국수 추출물의 항돌연변이원성 및 세포독성 효과 (Antimutagenic and Cytotoxic Effects of Fagopyrum esculentum Moenech Noodles Extracts)

  • 메밀 국수의 주 재료인 메밀분의 함량을 60%와 70%로 다르게 하여 건면, 압출면 그리고, 생면의 추출물을 조제하여 항돌연변이원성과 세포독성 효과를 조사하였다. 세포독성 효과를 알아보기 위해서 A549, Hep3B, MCF-7, AGS와 HeLa를 사용하였다. 이들 시료의 추출물의 암세포 성장 억제효과를 조사한 결과, 메밀 생면은 자궁암세포에서 68.5%의 높은 억제 활성을 나타내었으며 압출면의 경우는 위암세포에서 75.7%의 억제율을 나타내었다. 건면의 경우는 폐암 세포에서 80%의 높은 억제효과를 나타내었다. 그리고 돌연 변이성 및 항돌연변이 활성은 S. Typhimurium TA98과 TA100 균주를 이용한 돌연변이성 실험 결과 메밀면 자체에는 돌연변이성이 없었으며, 항돌연변이원성 실험에서는 직접변이원인 MNNG$(0.4{\mu}g/place)$의 경우 TA100 균주에서 시료농도 $160{\mu}g/place$에서 메밀분 70% 혼합분 생면 추출물의 경우 41%의 억제효과를 나타내었으며 4NQO$(0.15{\mu}g/place)$에 대해서는 같은 시료농도에서 TA98 균주에 대해서 43%의 억제 효과를 나타내었다. 그리고 메밀분 70% 혼합분 압출면의 경우 28%의 낮은 효과를 보였으나 S. Typhimurium TA98 균주에서는 같은 시료농도에서 45%의 억제효과를 나타내었으며 공시 시료 모두 농도 의존적으로 억제하는 것으로 나타났다.

추출용매와 부위에 따른 고추 추출물의 세포독성 (Cytotoxicity of Extracts from Korean Pepper (Capsicum annuum L.) by Extraction Solvents and Plant Parts)

  • In order to find out anticancer activity of Korean pepper (Capsicum annuum L.), the cytotoxicity against 8 cell lines including 293 (normal kidney cells) and A-431 (epidermoid carcinoma cells) of extracts by extraction solvents and plant parts were investigated using MTT assay. Also the correlation between content of capsaicin known as anticancer ingredient and cytotoxicity of extracts from pepper were analyzed. The distilled water extracts from seed and germinated seed showed very high cytotoxicity against 6 cancer cell lines including A549 (lung carcinoma cells), AGS (stomach adenocarcinoma cells), HeLa (cervix adenocarcinoma cells), HepG2 (hepatoblastoma cells), HT-29 (colon adenocarcinoma cells), and MCF-7 (breast adenocarcinoma cells). But 80% ethanol and methanol extracts showed cytotoxicity against 293 and AGS. The $RC_{50}$, that was, the concentration of sample required for 50% reduction of cell viability, of seed and germinated seed extracts against AGS were $33.4{\sim}389.1{\mu}g/m{\ell}$ and $63.9{\sim}1,316.7{\mu}g/m{\ell}$, respectively, so anticancer activity was higher in seed than in germinated seed. In capsaicin contents, seed with high cytotoxicity and pericarp with a little cytotoxicity contained $47.4{\sim}1,260.0{\mu}g/g$ and $58.3{\sim}1,498.0{\mu}g/g$, respectively. As these results, the correlation was not between cytotoxicity and capsaicin content.
