A Plant Growth-Promoting Pseudomonas fluorescens GL20: Mechanism for Disease Suppression, Outer Membrane Receptors for Ferric Siderophore, and Genetic Improvement for Increased Biocontrol Efficacy

  • Pseudomonas fluorescens GL20 is a plant growth-promoting rhizobacterium that produces a large amount of hydroxamate siderophore under iron-limited conditions. The strain GL20 considerably inhibited the spore germination and hyphal growth of a plant pathogenic fungus, Fusarium solani, when iron was limited, significantly suppressed the root-rot disease on beans caused by F. solani, and enhanced the plant growth. The mechanism for the beneficial effect of strain GL20 on the disease suppression was due to the siderophore production, evidenced by mutant strains derived from the strain. Analysis of the outer membrane protein profile revealed that the growth of strain GL20 induced the synthesis of specific iron-regulated outer membrane proteins with molecular masses of 85- and 90 kDa as the high-affinity receptors for the ferric siderophore. In addition, a cross-feeding assay revealed the presence of multiple inducible receptors for heterologous siderophores in the strain. In order to induce increased efficacy and potential in biological control of plant disease, a siderophore-overproducing mutant, GL20-S207, was prepared by NTG mutagenesis. The mutant GL20-S207 produced nearly 2.3 times more siderophore than the parent strain. In pot trials of beans with F. solani, the mutant increased plant growth up to 1.5 times compared with that of the parent strain. These results suggest that the plant growth-promoting P. fluorescens GL20 and the genetically bred P. fluorescens GL20-S207 can play an important role in the biological control of soil-borne plant diseases in the rhizosphere.

  • Electrical characteristics of two types of 20 GHz Push-Push GaAs MESFET dielectric resonator oscillators having Wilkinson and T-junction power combiners for the output stage have been investigated. The Push-Push oscillator for suppressing fundamental frequency 10 GHz and enhancing 20 GHz has been designed and realized in microstrip configuration on 20 mil thick RT-Duroid(${\varepsilon}_r$=2.52) teflon substrate. Two different types of power combiners, T-junction and Wilkinson, have been considered. Whenever one type of the combiners has been adopted for the output circuit, output power, phase noise and fundamental frequency suppression characteristics of the oscillator have been measured. When the Wilkinson power combiner was used, a maximum output power of 5.67 dBm, a phase noise of -105.5 dBc/Hz at an offset frequency of 100 kHz and a fundamental frequency suppression of -29.33 dBc have been measured. When the T-junction power combiner was used, a maximum output power of -1.17 dBm, a phase noise of -102.2 dBc/Hz at an offset frequency of 100 kHz and a fundamental frequency suppression of -17.84 dBc have been measured.

A Study on the Performance Characteristics of a Chirp RTLS over Wireless Channel with Gaussian Noise (가우시안 잡음이 존재하는 무선채널에서 Chirp RTLS 시스템의 성능 특성에 관한 연구)

  • The chirp signaling has been mainly used in radar systems due to its good correlation characteristics, and nowadays it is applied to real time locating system(RTLS). The RTLS with chirp signaling was chosen as a standard such as ISO/IEC 24730-5 and IEEE 802.15.4a. In this paper, the performance of a real time locating system with chirp signaling was evaluated and simulated with relative distance error rates. We considered three cases of S/I = -30[dB], -20[dB], and -10[dB] with Rician factor K=10 and K=20. The performance was enhanced with K factor improvement by 25%, 27% and 50% for respective three cases of S/I. As results, in case of S/I < -20[dB], the minimum signal power is required for performance improvement even though the line of sight component is helpful. And also, in case of S/I ${\geq}$ -20[dB], as the line of sight component is stronger the better performance is obtained.

Effects of Storage Conditions on Qualities of Buttercup Squash (Kabocha) (밤호박의 저장 온도와 습도가 품질에 미치는 효과)

  • To establish the optimum conditions for storing buttercup squash, we examined the effectiveness of several storage methods and the quality of the squash under various storage conditions, including temperature (12 and $20^{\circ}C$) and relative humidity (20, 40, 60, and 80%). The spoiling rate of the squash was affected more by the storage temperature than the relative humidity, and the squash stored at $20^{\circ}C$ started to be deteriorated after 20 days of storage. At $20^{\circ}C$, soluble solid content gradually increased until 20 days of storage, and then it tended to decrease. The L-value had a tendency to increase with days of storage, and the a- and b-value also increased after 40 days. In addition, the color changes were great when the squash was stored at high temperature and high relative humidity. The total pectin content increased until 20 days at $20^{\circ}C$, and then it decreased, but less change was observed in the squash stored at $12^{\circ}C$. Overall, the results showed that storage at $20^{\circ}C$ after field curing resulted in excessive weight loss, color loss and poor eating quality, as well as a high level of decay (approximately 70%) after 40 days. However, the squash stored at $12^{\circ}C$ and 60% RH (relative humidity) showed less degreening and had a reduced level of decay, below 10%.

Evolutionary Theory and Twentieth Century Music -Focused on 4'33" by John Cage- (진화론으로 바라본 20세기 현대 음악 -존 케이지 4분 33초를 중심으로-)

  • 20th century music has been recognized as disconnected. Some criticized that it is not music. This controversy has been continued over decades in music history. This study applied Darwin's evolutionary theory to John Cage's 4'33", the most popular music in 20th century and tried to prove musical continuity embedded in it. 4'33" has always been a subject of controversy because it has no sound. However, the study found out that 4'33" has the common musical factor, the rest. 4'33" can be interpreted as the variation of the rest in evolutionary view. The rest has undergone a variety of variations in form and meaning since the beginning of the musical notation and was suitable for a new musical environment changed in 20th century. In the evolutionary view, 4'33" is evolved music, not disconnected.

  • Bacillus subtilis K-20 chunggukjang is widely used in making soy sauces and bean pastes which are Korean traditional fermented foods. Bacillus subtilis K-20 chunggukjang was cultured, and fermented at $40^{\circ}C$ and 90% humidity for 96 hr after homogenizing with garlic, garlic and onion, and garlic, onion, and ginger. As a result, a product with pizza flavor and taste was obtained from Bacillus subtilis K-20. This product could be used as a functional food to promote immunity.

A Study on Discriminative Criteria of 6 Kinds of Achyranthis Radix Using HPLC/DAD;Isolation and Identification of 20-hydroxyecdysone from Aclryranthes japonica $N-{AKAI}$ and Comparison of Patterns of Achyranthis Radix from Different Locations by HPLC (HPLC/DAD를 이용한 6종(種) 우슬(牛膝)의 분류기준 연구;우슬(牛膝)(쇠무릎, Achyranthes japonica $N_{AKAI}$)로부터 20-hydroxyecdysone 분리.동정 및 산지별 우슬의 HPLC 패턴 비교)

  • Objectives : This study was performed to investigate the discriminative criteria of 6 kinds of Achyranthis Radix by HPLC/DAD. Methods : 20-hydroxyecdysone is isolated by silica gel column chromatography ($CHCl_3$:MeOH, 7:1-1:1 v/v) and identified by nuclear magnetic resonance, A high-performance liquid chromatographic method with diode array detection was used to identify 20-hydroxyecdysone in A. japonica. The analysis was performed using $C_{18}$ column with isocratic elution consisted of 18% acetonitrile and 82% water and the detection was carried out by DAD at 254 nm. 6 kinds of Achyranthis Radix from different locations were extracted in MeOH. Each extracts was analyzed by HPLC in same condition as used in analysis of 20-hydroxyecdysone. The identities of each extracts were determined by comparing the retention time and UV spectrum with that of reference compound. Results : 1. A. japonica and A. bidentata showed the similar patterns of HPLC chromatogram and 20-hydroxycedysone was present in both of them because the peaks having the same retention time and UV spectrum as 20-hydroxyecdysone were shown in the HPLC chromatograms of A. japonica and A. bidentata 2. Cyathula officinalis and C. capitata showed the similar patterns of HPLC chromatogram. The peak having the same retention time and UV spectrum as 20-hydroxyecdysone was shown in the HPLC chromatogram of C. capitata but not shown in the HPLC chromatogram of C. officinalis. 3. Two species of medicinal drugs from Sacheon province showed similar patterns of HPLC chromatogram. Achyranthis Radix from Sacheon(wild) did not have 20-hydroxycedysone but Achyranthis Radix from Sacheon(cultivated) showed the peak having the same retention time as 20-hydroxyecdysone but UV spectrum of the peak was different from that of 20-hydroxyecdysone. Conclusions : These results suggested that 20-hydroxyecdysone could be the discriminative criteria for Achyranthis Radix contain 20-hydroxyecdysone though they belong to different genus and species. And the patterns of HPLC chromatogram also could be the discriminative criteria as the different species of Achyranthis Radix belonging to the same genus showed similar patterns of HPLC chromatogram.

Synergistic Killing Effect of Synthetic Peptide P20 and Cefotaxime on Methicillin-Resistant Nosocomial Isolates of Staphylococcus aureus

  • The salt resistance of antibacterial activity and synergistic effect with clinically used antibiotic agents are critical factors in developing effective peptide antibiotic drugs. For this reason, we investigated the resistance of antibacterial activity to antagonism induced by NaCl and $MgCl_2$ and the synergistic effect of P20 with cefotaxime. P20 is a 20-residue synthetic peptide derived from a cecropin A (CA)-melittin(ME) hybrid peptide. In this study, P20 was found to have potent antibacterial activity against clinically isolated methicillin-resistant Staphylococcus aureus (MRSA) strains without hemolytic activity against human erythrocytes. The combination study revealed that P20 in combination with cefotaxime showed synergistic antibacterial activity in an energy-dependent manner. We also confirmed the synergism between P20 and cefotaxime by fluorescence-activated flow cytometric analysis by staining bacterial cells with propidium iodide (PI) and bis-(1,3-dibutylbarbituric acid) trimethine oxonol (BOX). This study suggests that P20 may be useful as a therapeutic antibiotic peptide with synergistic effect in combination with conventional antibiotic agents.

A Study on the Syrup using for Yackwa (약과(藥果)에 쓰이는 Syrup에 관(關)한 연구(硏究))

  • Among many honeys, the buckwheat honey was the best one using for Yackwa(a Korean cookie). We made a substitute, similary syrups of buckwheat honey, and then we got the following conclusions through the case of the similar syrups. 1. The best syrups are A, B and C syrup. A syrup's composition rate was sugar (40%), corn-syrup (20%) and water (40%) before boiling for 10 minutes. B syrup's composition rate was water (40%), glucose (20%), corn-syrup (20%) and sugar (20%) before boiling for 15 minutes. C syrup's composition rate like B syrup's but boiled for 10 minutes. 2. The best syrups used for dough are A and B syrup. C syrup was good for soaking. When A syrup used for dough, we have to immediate soaking in A syrup after frying or soaking in C syrup an hour later after frying. When B syrup used for dough, we must soaking in C syrup an hour later after frying. 3. The rate of fat in the composition of buckwheat honey's, A syrup's and B syrup's Yackwa was 27.22%, 23.05%, 23.05% and 30.35%.

A Study on Voice Analysis by Withdrawal Symptoms Identification of Smartphone Addiction : 20 Male College Students (스마트폰 중독의 금단 현상에 따른 음성 분석 연구 : 20대 남성 대학생을 대상으로)

  • In this paper, voice analysis is performed to propose a quantitative indication of withdrawal symptoms of smartphone addiction, applied to 20 males in their 20s. For this, their smartphones were confiscated during a certain period, and we collected samples of voice signals from them before and after that period. The pitch variation and intensity magnitude of the voice samples were used in the experiment to investigate their anxiety resulted from withdrawal symptoms. The experimental results showed an increase in pitch variation from 90% of samples, and a decrease in intensity magnitude from 95% of them. Finally, we carried out a study to find a statistical significance from the extracted experimental results through statistical analysis.
