Modification of Papain with $IO_4-Oxidized$ Soluble Starch ($IO_4$-산화 가용성 전분에 의한 파파인의 변형)

  • Periodate-oxidized soluble starch was reacted with papain at pH 4.0, pH 7.0, and pH 9.7, and an oxidized soluble starch-papain conjugate was produced. When compared with native papain, the specific activity decreased to 60%, in both the modified papain reacted with 0.4% $NaBH_4$ and in the modified papain not reacted with $NaBH_4$. The specific activity decreased to 70% in the modified papains reacted with 1.5% $NaBH_4$ and 4.0% $NaBH_4$, respectively. The reduction by $NaBH_4$ did not have an effect in the thermal stability of either the modified or nonmodified papain. An activity of 54.7% remained in the papain modified at pH 4.0, which was incubated at $80^{\circ}C$ for 40 min. The papains modified at pH 7.0 and pH 9.7 and incubated for 40 min at two different temperatures, respectively, were stable to $60^{\circ}C$, and at $80^{\circ}C$ their activities at 56.3% and 44.1 %, respectively. The modified papain's thermal stability pattern was similar to that of native papain, with no increase in its statbility. In the range of pH $2.0{\sim}13.0$, the stability of the papain modified at pH 4.0 decreased greatly between pH $3.0{\sim}5.0$, but it was similar to the native papain at other pH values. The stability of the papain modified at pH 7.0 showed a similar pattern to the native papain at pH $2.0{\sim}6.0$, while its stability increased when moving into the alkali pH range. The papain modified at pH 9.7 also had increased stability, when moving into the alkali range. The results of Hammerstein milk casein, which was reacted with the papains modified at pH 4.0, pH 7.0, and pH 9.7, respectively, and analyzed by FPLC, showed different peaks according to the different modification pHs, and the greatest peak differences were observed with the modification at pH 9.7.

A Study on Nutrient Intake Status and Food Sources of Iron by Dietary Iron Density of High School Girls in Seoul (서울지역 여고생들의 식이 철분밀도에 따른 영양섭취상태 및 철분 급원식품에 관한 연구)

  • This study was conducted to examine nutrient intake status and iron food sources by dietary iron density of high school girls in Seoul. The subjects of 226 girls were divided into High group (${\geq}$ 6 mg/1,000 kcal, N=115) and Low group (< 6 mg/1,000 kcal, N=111) by dietary iron density. The nutrient intake data obtained by 24-hour recall method were analyzed by Can Pro 3.0 software. Mean age of all subjects was 16.4 years old, heights and weights of High group and Low group were 164.5 cm, 53.4 kg and 161.7 cm, 51.7 kg, respectively. The body mass index (BMI) of High group and Low group was 20.5 kg/m$^2$ and 19.8 kg/m$^2$, respectively. Most nutrient intakes except energy and lipid intakes of High group were higher than those of Low group. High group showed significantly higher intakes of total iron, vegetable iron and animal iron than Low group. Ca and folate intakes of High group were under 75% of the recommended intake (RI) and Ca, iron, folate and vitamin C intakes of Low group were under 65% of RI. The percentage of subjects who consumed iron less than estimated average requirements (EAR) were 40.0% in High group and 77.5% in Low group. Total food intakes of High group showed higher than that of Low group. Total animal food intakes were significantly higher and total vegetable food intakes were significantly lower in Low group than those of High group. Iron intake from meats, fishes, shell fishes and seasonings were significantly higher in High group than Low group. Iron intake from milk and dairy products were significantly lower in High group than Low group. Major food sources of iron were rice, bean curd, pork, and egg in order among both groups.

Effects of Concentrate Feeding on Rumen Papillae Development in Hanwoo Calves before Weaning (이유 전 농후사료 (무조사료) 급여가 한우 송아지의 반추위 유두 발달에 미치는 영향)

  • The present study was conducted to investigate the effects of concentrate feeding on rumen papillae development in suckling Hanwoo calves before weaning (60 days of age). Twenty-four Hanwoo calves (12 heifers and 12 bulls) at six days of age were randomly assigned to one of three dietary treatments [hay and two levels (low and high) of concentrate feeding] and given each diet with free access to their dams for suckling until 60 days of age. At 60 days of age, two calves from each treatment were sacrificed and used for post-mortem examination of rumen papillae development. Feed intake between 31 and 60 days of age was significantly higher (P<0.05) for the high concentrate treatment (380.2 g/d) than for other treatments (58.3 and 76.9 g/d for hay and low concentrate treatments, respectively). Although feed intake showed a large difference between the low and high concentrate treatments, body weights at birth and 60 days of age were similar between the two treatments, suggesting that calves in the low concentrate feeding suckled more milk from their dams than those in the high concentrate feeding. The ratio of reticulo-rumen weight to body weight at 60 days of age was significantly increased (P<0.05) in the high concentrate (1.39%), compared with those in the hay (0.85%) and low concentrate (1.06%) treatments. Furthermore, the high concentrate feeding significantly increased both rumen papillae length and width, compared with the hay and low concentrate feeding group. This was also detected clearly by visual observation. The blood concentration of ${\beta}$-hydroxybutyrate was significantly higher for the high concentrate (176.4 ${\mu}mol/L$) than for other treatments (58.9 and 59.2 ${\mu}mol/L$ for the hay and the low concentrate group, respectively). In conclusion, the results of the present study showed that, to achieve a large development of rumen papillae before weaning (60 days of age) in suckling Hanwoo calves, the amount of concentrate intake should be important. And also the results implied that hay might not be included in the diet for suckling Hanwoo calves before weaning (60 days of age).

A Survey of Food and Nutrient Intakes of the Eged People in Rural Area, Gyeongbuk Yecheon (경북 예천 농촌지역 거주노인의 연령대별 영양소 및 식품섭취량 조사)

  • This research has done for 262 people of the aged men and women that are more than 65 years old who are live in 9 areas of Yecheon as target; through twice off ace to face interview for 24 hours recall method, the result of food intake for 2 days is as following. In case of energy, the aged men (women) of sixties ingested 67.7 $(72.0)\%$ of Korean RDA by 1,369 (1264) kcal, for the ages of seventies and eighties, 68.9 $(66.9)\%$ of Korean RDA and 76.3 $(65.8)\%$ by each 1,309 (1104) kcal and 1,368 (1052) kcal. The aged men ingested protein $46.0\~49.6 g$ ($70.6\~82.9\%$ of RDA), and aged women ingested protein $32.7\~40.2 g$ ($59.4\~73.0\%$ of RDA). Calcium intake of aged men was 388.8 mg, 319.8 mg, 284.4 mg by age range, and aged women was 291.9 mg ($41.6\%$ of RDA), 246.5 mg ($35.3\%$ of RDA), 240.1 mg ($34.3\%$ of RDA). Iron intake of aged men was $8.6\~8.9 mg$ ($72\~74%$ of RDA), and aged women ingested 8.6 mg ($71.3\%$ of RDA), 7.5 mg (62.6 of RDA$\%$), 6.6 mg ($55.4\%$ of RDA) for iron by age range. Vitamin $B_1$ intake of aged men was $0.62\~0.71 mg$ ($62\~71\%$ of RDA), and aged women's intake was $0.50\~0.60 mg$ ($50\~60\%$ of RDA). Vitamin $B_2$ intake of aged men was $0.59\~0.60 mg$ ($49\%$ of RDA), and aged women's intake was $0.45\~0.50 mg$ ($37\~42\%$ of RDA). Vitamin C intake by age range, in case of aged men (women) in sixties was 53.1 (48.9) mg, in seventies was 49.9 (33.2) mg and more than eighties was 34.1 (33.4) mg. The average food intake by age range, in aged men (women) of sixties was 828.9 (670.8) g and seventies was 726.8 (568.8) g and more than eighties was 656.0 (525.3) g. Plant food intake of aged men was 490.8-569.5 g and aged women was 417.9-537.7 g. Aged men (women) of MAR by age range, sixties was 0.60 (0.58), seventies was 0.59 (0.50) and more than eighties was 0.56 (0.49), respectively. INQ for protein, phosphorus, iron, vitamin A, vitamin B, niacin, vitamin C was more than 1 in 60's and 70's aged men, but there was no nutrients in eighties of aged women. Aged men and women's KDDS points represent average 3.14 and 3.04 (out of 5 points), and while intake of the milk was the most lacking, but intake of the fruit was the most lacking in DDS.

A Study on the Children's Eating Habits and Food Preference according to Their Parents' Economic Status (I) - Seoul & Gyeonggi (Incheon) Area - (부모의 경제수준에 따른 자녀의 식습관과 식품기호도에 관한 연구 (I) -서울.경기 (인천)지역을 중심으로-)

  • This research was carried out a to investigate the food habit and preference of parents' social and economic level with 681 children (145 of Kindergarten, 300 of Children, 236 of Adolescences) in Seoul and Gyeonggi area using question naires. The parents' income level divided into 2 groups, less than 4 million won and equal or greater than 4 million won. Most of the middle years were over-weighing but kindergarten and adolescences had standard weights. The majority of parents had Bachelor degrees, most of father were office workers, majority of mothers of kindergarten with parents' income level less than 4 million won were professional women and mothers of the rest of the groups were mostly house wives. The middle years with parents income level less than 4 million son were skipping meals most frequently, but other groups didn't show much differences in their eating habits. All age groups showed that they like meats the most and dislike vegetables the worst. All age groups also showed that they mostly eat out 1-2 times a week and the next was 3-4 times a week. All age groups preferred ice creams, fruits, juices and snacks for their desserts. The middle years with parents' income equal or greater than 4 million won also showed high preference on strawberry and chocolate flavored milk, burgers and pizzas. The food s that preferred to eat when dining out were Chinese foods (Ja-jang-myeon and sweet and sour pork) for kindergarten group, Korean foods (kalbi and bulgogi) for the middle years with parents' income level less than 4 million won, family restaurant food (steak and rib) for the middle years with parents' income level equal or greater than 4 million won and Korean foods (kalbi and bulgogi) for the adolescences. The preferred cooking methods were roast (fish and sea weeds) for the kindergarten, roasted meat for the middle years, Kimchi and bean paste pot stew for the adolescences with parents' income level less than 4 million won and roasted meats for the adolescences with parents' income level equal or greater than 4 million won. The results showed that the adolescences with higher parental income lever preferred meats.

Quality Characteristics and Inhibition Activity against Helicobacter pylori KCCM 40449 of Liquorice Yogurts Manufactured by Exopolysaccharide Producing Lactic Acid Bacteria (Exopolysaccharide 생성 유산균을 이용한 감초 추출물 첨가 Yoghurt의 품질특성 및 Helicobacter pylori KCCM 40449 억제활성)

  • This study was carried out to fortify the antimicrobial activity of yoghurt by adding liquorice extract to it. The liquorice extracts (1 mg/mL) showed relatively high antibacterial activity against H. pylori KCCM 40449 (p < 0.05). The solvent liquorice extracts of minimal inhibitory concentrations (MIC) against H. pylori KCCM 40449 were 25- 100 ${\mu}g$/mL. Lactobacillus amylovorus DU-21 with high EPS production ability were inoulated to milk after the addition of different amounts of liquorice extracts (0.0%, 0.05%, 0.1% and 0.2%). The physico-chemical characteristics of yoghurts added with liquorice extracts were examined. The initial pH, titratable acidity, viscosity and viable cell counts of the yoghurt added liquorice extracts were 3.41-3.51, 1.021-1.091%, 1,686-1,930 cp and 9.41-9.38 Log CFU/mL, respectively. The viscosity and syneresis of yoghurt were better than that of the control. Antimicrobial activity against H. pylori KCCM 40449 increased with increasing addition of liquorice extract. However, the sensory score of yoghurt added with different amounts of liquorice extracts was lower than that of the control (p < 0.05). As a result of the sensory evaluations, the flavor, taste, texture, color and overall acceptability of the yoghurt with 0.05% liquorice extract were found to be much better than those of the other groups (p < 0.05). Overall, the optimal amount of liquorice extract added in the manufacture of yoghurt was 0.05% of the total weight. Further studies on increment of antimicrobial activity and palatability of liquorice extract added yoghurt are necessary.

Study on the Manufacturing Properties of Korean-type Koumiss (한국형 Koumiss제조 특성에 관한 연구)

  • For this study, Korean-type Koumiss was made by the fermentation of mixed cultures, in which yeast, Kuyveromyces, and microflora, Streptococcus thermophiles and Lactobacillus bulgaricus, were inoculated into 10% skimmed milk with added whey powder(control: A, 2%: B, 4%: C, 6%: D, and 8%: E). Fat, protein, lactose, titratable acidity, pH, the number of lactic acid bacteria, the number of yeast, alcohol content, volatile fatty acids, volatile free amino acids and minerals were measured in the products. The results were as follows: As the dosage of whey powder increased, fat increased from 0.74% in the control to 2.30% in sample E, protein increased from 2.95% in the control to 4.39% in sample E and lactose increased from 3.10% in the control to 7.43% in sample E. Titratable acidity and pH increased gradually. The number of lactic acid bacteria increased from $10^9\;cfu/m{\ell}$ in the control to $3.8{\times}10^9\;cfu/m{\ell}$ in sample E, and the number of yeast increased from $6.1{\times}10^7\;cfu/m{\ell}$ in the control to $1.65{\times}10^8\;cfu/m{\ell}$ in sample E, according to the increase of whey powder content. For alcohol content, the average values were 0.863%, 0.967%, 0.890%, 1.290%, and 1.313% for the control and samples B, C, D, and E, respectively. As the dosage of whey powder increased, alcohol content showed a tendency to gradually increase. The average alcohol content of E was 1.313 and this was higher than the alcohol content of Kazahstana-type Koumiss with 1.08%. Sixteen types of free amino acids were detected. Glycine was the lowest in the control at $0.38mg/m{\ell}$ and sample E contained $0.64mg/m{\ell}$. Histidine was also low in the control at $0.42mg/m{\ell}$ and sample E contained $0.65mg/m{\ell}$. On the other hand, glutamic acid was highest at $4.13mg/m{\ell}$ in the control whereas sample E had $6.96mg/m{\ell}$. Proline was also high in the control at $1.71mg/m{\ell}$ in control, but E contained $2.80mg/m{\ell}$. Aspartic acid and leucine were greater in sample E than in the control. For volatile free fatty acids, content generally had a tendency to increase in the control, and samples B, C, D, and E. Content of acetic acid gradually increased from $12,661{\mu}g/100m{\ell}$ in the control to $37,140{\mu}g/m{\ell}$ in sample E. Butyric acid was not detected in the control and was measured as $1,950{\mu}g/100m{\ell}$ in sample E. Caproic acid content was $177{\mu}g/100m{\ell}$ in the control and $812{\mu}g/100m{\ell}$ in sample E, and it increased according to the increase of whey powder content. Valeric acid was measured in a small amount in the control as $22{\mu}g/100m{\ell}$, but it was not detected in any other case. Mineral contents of Ca, P, and Mg increased from 1,042.38 ppm, 863.61 ppm, and 101.28 ppm in the control to 1,535.12 ppm, 1,336.71 ppm, and 162.44 ppm in sample E, respectively. Na content was increased from 447.19 ppm in the control to 1,001.57 ppm in sample E. The content of K was increased from 1,266.39 ppm in the control to 2,613.93 ppm in E. Mineral content also increased with whey powder content. In sensory evaluations, the scores increased as whey powder content increased. Flavor was lowest in the control with 6.3 points and highest in E with 8.2 points. Body and texture were highest at 4.2 points in the control, which did not have added whey powder. In the case of appearance, there were no great differences among the samples.

Effect of Lactic Acid Bacteria and Formic Acid on the Silage Quality of Whole Crop Rice at Different Maturity (유산균 및 개미산 첨가가 수확시기별 벼 사일리지의 발효 품질 및 사료성분에 미치는 영향)

  • Silage additives are needed to increase the quality of whole crop rice silage which seldom produce without the additives due to both high pH and butyric acid concentrations. Little information, however, is available about the silage fermentation of whole crop rice added with silage additives in Korea. This study was conducted to determine the optimum levels of silage additives by evaluating the effects of latic acid bacteria (LAB) and formic acid concentrations on the silage quality of whole crop rice harvested at different mature stages. Field study was established early in May until October 7th on a rice field at Yupori, Sinbuk-yeup, Chunchon, Kangwon-Do. "Ilpum" mutant rice was harvested at six different mature stages; booting stage (17 Aug.), milk-ripe stage (27 Aug.), dough stage (7 Sep.), yellow ripe stage (17 Sep.), dead ripe stage (27 Sep.) and full ripe stage (7 Oct.). Each sample was ensiled in three different ways; with 1) LAB (0.05, 0.1 and 0.2% of sample wt), 2) formic acids (0.2, 0.3 and 0.4% of sample wt.) and 3) no additive. The additive levels did not affect dry matter content, crude protein, fiber and total digestable neutriant concentrations at all stages. Addition of additives significantly decreased the silage pH and butyric acid concentrations which tended to be more decreased with higher levels of additives. Latic acid concentrations were higher with the use of additives, especially with LAB. The lower concentrations of ammonia-N were observed in additive treatments at all stages, but the concentrations of ammonia-N did not differ according to the additve levels after yellow ripe stage (0.69, 0.60 and 0.71% of DM in 0.05, 01 and 0.2% of LAB, respectively; 0.64 0.59 and 0.75% of DM in 0.2, 0.3 and 0.4% of formic acid, respectively). These results indicate that the optimum addition levels of LAB and formic acid are 0.5∼0.1% and 0.2∼0.3%, respectively, on which the high quality of rice whole crop silage was produced. produced.

The Therapeutic Effect of Lactobacillus reuteri in Acute Diarrhea in Infants and Toddlers (영유아의 급성 설사에서 Lactobacillus reuteri의 치료 효과)

  • Purpose : Certain strains of lactobacilli are known to accelerate recovery from acute diarrhea. Lactobacillus reuteri is isolated from human breast milk and a commonly occurring Lactobacillus species with therapeutic potential in acute diarrhea. The purpose of the present study was to investigate the therapeutic effect of L. reuteri in acute diarrhea in young children. Methods : Fifty patients between 6 and 36 months of age hospitalized with acute diarrhea (rotavirus in 40 percent) were randomized into two groups to receive either $10^8$ colony-forming units of L. reuteri or a matching placebo, twice a day for their length of hospitalization, or for up to 5 days. Antidiarrheal drugs were not prescribed to either group. The clinical outcome of diarrhea was evaluated. Results : The mean duration of watery diarrhea after initiation of treatment was 2.3 days for the L. leuteri group(n=25) vs. 2.9 days for the placebo group(n=25)(P=0.072). By the second day of treatment, watery diarrhea persisted in 64 percent of patients receiving L. reuteri, compared to 84 percent of those receiving placebo(P=0.006). On the second day, the mean frequency of watery diarrhea was 1.9 in the L. leuteri group and 3.4 in the placebo(P=0.046). Also, vomiting continued to the second day in 16 percent of patients receiving L. reuteri and 40 percent of those recieving placebo(P=0.031). Conclusion : L. reuteri is effective as a therapeutic agent in acute diarrhea in children.

'Dakyeong', Earley-heading, Resistance to Lodging and High-yielding Forage Oats Cultivar (조숙 내도복 다수성 추파용 귀리 품종 '다경')

  • 'Dakyeong' (Avena sativa L.), a winter oats for forage use, was developed by the breeding team at National Institute of Crop Science, RDA in 2016. It was derived from a cross between 'CI7505'(IT133304) and 'Swan'(IT197920). Subsequent generations followed by the cross were handled in bulk and pedigree selection programs at Iksan and Jeonju, respectively. After preliminary and advanced yield test for 2 years, 'SO2004009-B-B-10-8-3-9', designated as a line name of 'Gwiri91', were subsequently evaluated for earliness and forage yield during 3 years in four parts such as Jeju (upland), Yesan (upland), Iksan (upland), and Jeonju (paddy), from 2014 to 2016, and finally named as 'Dakyeong'. Cultivar 'Dakyong' has leaves of dark green color, thick diameter culm and long grain of brown color. Over 3 years, the heading date of 'Dakyeong' was about 5 days earlier than that of check cultivar 'Samhan' (April 30 and May 5, respectively), and their average forage dry matter yield harvested at milk-ripe stage was higher 12% ($15.7tone\;ha^{-1}$) than $14.0tone\;ha^{-1}$ of check cultivar. Cultivar 'Dakyeong' was lower than the check cultivar 'Samhan' in terms of the protein content (6.1% and 7.0%, respectively) and total digestible nutrients (62.1%, and 62.5%, respectively), while the TDN yield was more than the check ($7.79tone\;ha^{-1}$ and $7.64tone\;ha^{-1}$, respectively). Fall sowing cropping of 'Dakyeong' is recommended only in areas where average daily minimum mean temperatures in January are higher than $-6^{\circ}C$, and it should not be cultivated in mountain areas, where frost damage is likely to occur.
