Characterization of Paraplantaricin C7, a Novel Bacteriocin Produced by Lactobacillus paraplantarum C7 Isolated from Kimchi

  • A Lactobacillus paraplantarum strain producing a bacteriocin was isolated from kimchi using the spot-on-the lawn method and named L. paraplantarum C7 [15]. The bacteriocin, paraplantaricin C7, was found to inhibit certain Lactobacillus strains, including L. plantarum, L. pentosus, and L. delbrueckii subsp. lactis. It also inhibited Enterococcus faecalis, yet did not inhibit most of the other LAB (lactic acid bacteria) tested. The maximum level of paraplantaricin C7 activity was observed under the culture conditions of $25^{\circ}C$ and a constant pH of 4.5. Paraplantaricin C7 retained 90% of its activity after 10 min of treatment at $100^{\circ}C$ and remained stable within a pH range of 2-8. Based on a culture supernatant, paraplantaricin C7 was purified by DEAE-Sephacel column chromatography and $C_{18}$ reverse-phase HPLC. SDS-PAGE and activity staining were then conducted using the purified paraplantaricin C7, and its molecular mass determined to be about 3,800 Da. The 28 N-terminal amino acids from the purified paraplantaricin C7 were determined, and the structural gene encoding paraplantaricin C7, ppnC7, was cloned by PCR using degenerate primers based on the N-terminal amino acid sequence. The nucleotide sequences for ppnC7 and other neighboring orfs exhibited a limited homology to the previously reported plantaricin operon genes. Paraplantaricin C7 is a novel type II bacteriocin containing a double glycine leader sequence.

Effect of Potassium Oxide on the Early Hydration of Tricalciumaluminate (Tricalciumaluminate의 초기 수화반응에 미치는 $K_2O$의 영향)

  • Two kinds of clinker liquid melts, one containing 2.0% of $K_2O$ and the other without $K_2O$, were prepared with the similar composition as those developed during the firing of portland cement clinker. One portion of melt was quenched and found to consist of glass together with traces of proto-$C_3A$, and the other portion was allowed to cool spontaneously to crystallize $C_3A$ and calciumferrite well. The product obtained by crystallization of the melt without $K_2O$showed cubic $C_3A$, while orthorhombic $C_3A$ was formed from the crystallized product containing $K_2O$. Studies on the hydration behavior of the samples made from the melts with 30% of gypsum were carried out for 24 hours by observation at regular intervals ettringite, monosulphate hydrates and gypsum which were formed or consumed during hydration. The samples without $K_2O$ in the melts were hydrated wth addition of proper amount of $K_2SO_4$ in the water for hydration. Hydration behavior of glassified $C_3A$ showed that it has low reactivity relative to crystallized $C_3A$, and also hydration reactivity of orthorhombic $C_3A$ was much lower than that of cubic $C_3A$ in 60 minutes. Potassium sulphate in the solution reduced the hydration reactivity of $C_3A$ . Evolution peaks of hydration heat examined by twin-type conduction micro-calorimeter showed that glassified $C_3A$ without $K_2O$ had secondary peak after 9 hours and $C_3A$ containing $K_2O$ after 12 hours. When crystallized $C_3A$ was hydrated, initial peaks of hydration heat were considerably high and there was no secondary peak for cubic $C_3A$ but the secondary peak of orthorhombic $C_3A$ appeared after 4 hours.

Digital Signature Schemes with Restriction on Signing Capability (서명 능력을 제한하는 전자 서명 스킴)

  • In some practical circumstances, the ability of a signer should be restricted. In group signature schemes, a group member of a group may be allowed to generate signatures up to a certain number of times according to his/her position in the group. In proxy signature schemes, an original signer may want to allow a proxy signer to generate a certain number of signatures on behalf of the original signer. In the paper, we present signature schemes, called c-times signature schemes, that restrict the signing ability of a signer up to c times for pre-defined value c at set-up. The notion of c-times signature schemes are formally defined, and generic transformation from a signature scheme to a c-times signature scheme is suggested. The proposed scheme has a self-enforcement property such that if a signer generates c+1 or more signatures, his/her signature is forged. As a specific example, we present a secure c-times signature scheme $^c$DSA based on the DSA (Digital Signature Algorithm) by using a threshold scheme. Our transformation can be applied to other ElGamal-like signature schemes as well.

Effects of C3A Content on Chloride Concentration in Pore Solution (C3A함유량이 세공용액 Cl 농도에 미치는 영향)

  • 본 연구는 시멘트 경화체 중의 Cl 고정화 메카니즘을 규명하는 연구의 일환으로 시멘트의 C3A 함유량에 따른 Cl 고정화 효과를 세공용액 분석방법에 의해 조사한 것으로 C3A함유량 0.46 9.65%의 4가지 시멘트와 C3A를 함유하지 않은 초속경 시멘트 페이스트를 밀봉용기내에서 양생시켜 재령 28일에 세공용액을 추출하여 세공용액 중의 Cl 과 OH 농도를 측정, C3A함유량이 Cl 고정화에 미치는 영향을 검토한 것이다. 연구결과 세공용액 중의 Cl 농도는 NaCl 혼입량에 관계없이 시멘트 중의 C3A량이 증가함에 따라 낮아져 시멘트 경화체 내에서 Cl 의 고정화에 C3A가 매우 효과적임을 알 수 있었다. 세공용액의 Cl /OH 는 Cl 혼입량이 시멘트 중량의 0.3%인 경우 강재부동태막을 파괴하는 0.3보다 낮았으며 X선회절분석에 의해 C3A에 의한 고정화 메카니즘은 프리델씨염(Friedel's salt)의 생성에 의함을 확인하였다.

  • The NDT using ultrasonic is largely divided into A-Scan and C-Scan methods. Since A-Scan method is subject to subjective judgement by trained personnel, C-Scan method has been introduced, which presents the weld area in two dimensions by placing the transducers two dimensionally used in the A-Scan method. Therefore, it is necessary to develop equipment that can provide weld quality without the help of a welding expert and the presentation of effective C-Scan images. Thus, in this paper, the algorithms that express a low resolution 2-dimensional gray image formed by C-Scan method as a high-resolution color C-Scan image and automatically determine the weld quality from the generated C-Scan color image. The high resolution color C-Scan images proposed in this paper allow the exact shape of the weld point to be expressed, and an objective algorithm to use this image to automatically determine weld quality.

  • In this study, the characteristics of 3C-SiC ohmic contact were investigated. Titanium-tungsten(TiW) films were used for contact metalization. The ohmic contact resistivity between 3C-SiC and TiW was measured by HP4155 and then calculated with the circular transmission line method(C-TLM). And also the physical properties of TiW and the interface between TiW and 3C-SiC were analyzed using XRD and AES. TiW films make a good role of a diffusion barrier and their contact properties with 3C-SiC are stable at high temperature.

A Theoretical Study of Electronic Structure and Properties of the Neutral and Multiply Charged $C_{60}$

  • The electronic structures and properties of the neutral and multiply charged C60n ions (n=2+ to 6-) with spin states have been investigated by semi-empirical MNDO calculations. In the ground state, C601- has the lowest total energy and the highest binding energy. The neutral C60 ion is supposed to have a high ionization potential and a high electron affinity. The HOMO and LUMO positions are lower in the cationic C60 than in the anionic C60. The LUMO energy becomes increasingly positive from C601- to C606- and the HOMO energy becomes increasingly negative from C602+ to C60. The HOMO-LUMO gap of the neutral C60 ion is higher than that of the multiply charged C60 ions. From the HOMO-LUMO gap, it seems reasonable to expect that electrons of the multiply charged C60 ions will be more polarizable than those of the neutral C60 ion. The HOMO and LUMO energies increase as the negative charge increases.

  • The fabrication methods and requirements of the fiber, interphase, and matrix of nuclear grade $SiC_f/SiC$ composites are briefly reviewed. A CVI-processed $SiC_f/SiC$ composite with a PyC or $(PyC-SiC)_n$ interphase utilizing Hi-Nicalon Type S or Tyranno SA3 fiber is currently the best combination in terms of the irradiation performance. We also describe important material issues for the application of SiC composites to LWR fuel cladding. The kinetics of the SiC corrosion under LWR conditions needs to be clarified to confirm the possibility of a burn-up extension and the cost-benefit effect of the SiC composite cladding. In addition, the development of end-plug joining technology and fission products retention capability of the ceramic composite tube would be key challenges for the successful application of SiC composite cladding.

Improvement of a Nitrogen Fertilizer Recommendation Model by introducing a Concept of the Mitscherlich-Baule-Spillman Equation (토양검정에 의한 질소시비량 결정에 Mitscherlich-Baule-Spillman 개념의 응용)

  • In order to improve a soil test-based fertilization model for nitrogen, the Mitscherlich-Baule-Spillman equation modified by Bray and his coworkers was introduced and tested for its possibility of application. The coefficients $C_1$ and $C_2$ in the equation A were calculated by applying 10 pot experimental results done by the National Institute of Agricultural Sciences and Technology. Then, a regression(B) between C_1 and x, the fertilizer level was estimated. log(Ymax-Y) = logYmax-$C_1x-C_2b$ (A) where Ymax : maximum yield. Y : yield without or with a limited amount of fertilizer applied, x : application level of fertilizer, b : content of a nutrient in soil, $C_1$ and $C_2$ : coefficients of x and b, respectively. log(0.05)=$-C_1x-C_2b$ (B) Fertilizer requirement calculated by the equation B was significantly correlated with the real level of fertilizer applied for 95 percentage of the maximum yield. A more extensive and precise study on $C_1$ and $C_2$ in the equation A in expected to improve significantly the fitness of soil analysis to the fertilizer recommendation.

  • Observing that for any $\beta_c$-Wallman functor $A$ and any Tychonoff space X, there is a cover $(C_1(A(X), X), c_1)$ of X such that X is $A$-disconnected if and only if $c_1 : C_1(A(X), X) \longrightarrow X$ is a homeomorphism, we show that every Tychonoff space has the minimal $A$-disconnected cover. We also show that if X is weakly Lindelof or locally compact zero-dimensional space, then the minimal G-disconnected (equivalently, cloz)-cover is given by the space $C_1(A(X), X)$ which is a dense subspace of $E_cc(\betaX)$.
