하고초 추출물의 항혈전 효능 및 혈소판 응집 억제작용 (Antithrombotic and Antiplatelet Activity of Extract from Prunella vulgaris)

  • 한방생약제의 항혈전 및 혈소판 응집 억제 효능을 탐색하기 위하여 하고초의 물 추출물로 혈전 용해능 활성과 혈액 응고시간 지연효과 즉 PT (prothrombin time), APTT (activated partial thromboplastin time)와 혈소판 응집억제 활성 등에 대해 항혈전 효능을 평가하였다. 혈전용해도를 측정하는 fibrin plate가 용해되어 형성된 투명환의 넓이를 측정하는 실험을 진행한 결과 혈전용해도가 농도의존적으로 효능을 나타내었다. 혈액 응고 cascade에 미치는 영향을 알아보기 위해 혈액 응고 시간 지연 및 단축 효과를 확인하고자 APTT와 PT에 미치는 영향을 조사한 결과 PT의 경우 10 mg/ml, 5 mg/ml의 경우에는 대조군보다 우수한 지연효과를 보였다. APTT의 경우에는 10 mg/ml, 5 mg/ml는 대조군과 비교하여 매우 탁월한 지연효과를 보이고, 2.5 mg/ml, 1.25 mg/ml에서도 높은 지연효과를 나타냈다. 혈소판의 응집에 따라 형성되는 두 전극 사이에 형성된 전기적 저항의 변화로 나타나는 실험을 시행한 결과 하고초의 ADP와 collagen에서 뛰어난 응집억제 활성을 보였다. 따라서 위의 항혈전 효능평가 실험결과를 볼 때 하고초를 향후에 혈전 질환의 치료제 개발에 효과적으로 이용될 수 있을 것으로 사료된다.

해양미세조류 에탄올 추출물의 항균활성에 관한 연구 (Antibacterial activity of ethanol extracts from marine micro-algae)

  • Mixed A, Chlorella vulgaris, Nannochloropsis oculata, Chaetoceros calcitrans 등 4종류의 미세조류로부터 99.9% 에탄올로 추출한 성분의 항균활성을 검증하기 위해 그람음성세균 2종(E. coli, S. Typhimurium)과 그람양성세균 2종(S. aureus, B. cereus)에 대하여 paper disk diffusion assay, MIC, MBC를 측정하였다. 4종류의 미세조류 추출물은 공시균주에 대하여 0.62-1.66 mg/mL의 MIC를 나타내었으며, C. vulgaris 추출물이 4종의 공시균주에 대한 MIC가 0.62 mg/mL로 가장 강한 항균활성을 나타내었으며, MBC 또한 C. vulgaris 에탄올 추출물이 E. coli에 대해 2.50 mg/mL, S. aureus, B. cereus와 S. Typhimurium에 대해서는 4.16-5.00 mg/mL로 가장 강한 항균활성을 나타내었다. 항균활성이 가장 강한 C. vulgaris 에탄올 추출물의 성분을 분석한 결과, 함량이 가장 많은 성분은 9,12-octadecadienoic acid (linoelaidic acid, peak 5, 7)로 38.8%이었으며, 그 다음이 2-Hexadecen-1-o1, 3,7,11,15-tetramethyl (phytol, peak 2,6)로 30.0%이었다. Octadecadienoic acid는 일명 linoelaidic acid로 탄소수 18개의 고도불포화지방산이며, 두 번째로 함량이 많은 Phytol은 클로로필을 구성하는 불포화 제1급 알코올의 일종으로서(엽록소의 구성 성분) 병원성 대장균 및 황색포도상구균에 항균활성을 나타낸다는 보고로 볼 때, 이 2가지 성분이 항균활성의 주성분인 것으로 사료된다.

"하고초(夏枯草)"의 생약학적 연구 (Pharmacognostical Studies oh the 'Ha Go Cho')

  • 'Ha Go Cho (夏枯草)' is one of the Chinese crude drugs used mainly as a diuretic. With regard to the botanical origin of 'Ha Go Cho', it was reported by Su et al. that those from China were originated from the fruited spica of Prunella vulgaris L. of Labiatae. It was, however, for the herba or spica of Prunella vulgaris L. var. lilacina Nakai from Korea. According to survey of markets in Korea, most of the materials collected in the markets seemed to be originated from Prunella plant, while some seemed to be Thesium plant of Santalaceae. To clarify the botanical origin of 'Ha Go Cho', the anatomical characteristics of Prunella vulgaris L. var. lilacina Nakai and Thesium chinense Turcz. were studied. As a result, it was clarified that some 'Ha Go Cho' from Korea were the herba or spica of Prunella vulgaris var. lilacina, whereas some others were the herba of Thesium chinense.

지질 고 생산성 Chlorella vulgaris 변이주 분리 및 특성 분석 (Isolation of Chlorella vulgaris Mutants Producing High Lipid and their Characterization)

  • 미세조류인 Chlorella vulgaris (C. vulgaris)는 다른 미세조류에 비해 다량의 지질을 함유한다. 본 연구는 자외선 조사를 통해 변이주를 유도하고자 하였다. C. vulgaris에 자외선을 1, 2, 3, 4, 5 min간 조사하였으며 자외선 조사 시간에 비례하여 세포 생존율 및 색소 함량이 감소함을 확인하였다. 자외선 조사를 통해 C. vulgaris의 변이주를 유도하였으며, 지질 축적량을 기준으로 두 개의 균주를 선별하였다. 선별한 변이주는 UM10, 15로 명명하였으며, 야생균주와 동일한 환경에서 배양하였다. 변이주의 특성 분석을 위해 21일간 배양 후 세포 생장률, 건조 중량, 색소 함량, 지질 함량을 측정하였다. 그 결과, 두 종의 변이주는 야생균주에 비해 약 1.4배 높은 세포 생장률을 보였으며, 건조 중량은 약 1.5배 증가하였다. 광합성을 통한 미세조류 내 색소 함량을 알아보기 위해 클로로필과 카로티노이드 함량을 측정하였다. 변이주의 클로로필 및 카로티노이드 함량 모두 야생균주에 비해 약 10% 감소함을 확인하였다. 변이주의 지질 함량은 야생균주에 비해 약 1.2, 1.5배 증가하였다.

Parametric study of brewery wastewater effluent treatment using Chlorella vulgaris microalgae

  • The aim of this study was to evaluate the biomass and lipid production of Chlorella vulgaris and its nutrient removal capability for treatment of brewery wastewater effluent. The results indicate that the maximum biochemical oxygen demand (BOD) (91.43%) and chemical oxygen demand (COD) (83.11%) were removed by C. vulgaris with aeration in the absence of light. A maximum of 0.917 g/L of dry biomass was obtained with aeration in the dark conditions, which also demonstrated the highest amount of unsaturated fatty acids at 83.22%. However, the removal of total nitrogen (TN) and total phosphorus (TP) with these aeration and light conditions was 9.7% and 11.86% greater than that of other conditions. The removal of BOD and COD and the production of biomass and lipids with aeration in the dark and the TN and TP removal with aeration and light were more effective than other conditions in the brewery wastewater effluent in the presence of C. vulgaris.

  • Algal growth studies of Chlorella strains were conducted in a batch mode with bench type experiments. Carbon dioxide fixation rates of the following green microalgae were determined: Chlorella sp. H84, Chlorella sp. A2, Chlorella sorokiniana UTEX 1230, Chlorella vulgaris, and Chlorella pyrenoidosa. C. vulgaris, among other strains of microalgae, showed the highest growth rate (1.17 optical density/5 days). Cultivating conditions for C. vulgaris that produced the highest growth rate were at concentrations of 243 ${\mu}g\;CO_2$/mL, 10 mM ammonia, and 1 mM phosphate, with an initial pH range of 7-8.

트립토판과 리보플라빈을 이용한 광역동 치료의 여드름 치료 효과 (Topical Photodynamic Therapy with Triptophan and Riboflavin for the Treatment of Acne vulgaris)

  • Objective : Photodynamic therapy (PDT) using topical aminolevulinic acid (ALA) has increasingly been used for the treatment of acne vulgaris and several studies have shown its clinical efficacy. However, ALA-PDT needs a relatively long incubation period and is frequently associated with adverse effects. Triptophan and Riboflavin has been introduced as a new photosensitizer for the treatment of acne recently. Method : 11 patients with acne vulgaris were received PDT with tryptophan and riboflavin four to six times over two to three months. The photos of each patient were taken before and after treatment. Result and conclusion : PDT with tryptophan and riboflavin may be an effective and safe treatment for the Acne vulgaris patients.

The Pear Black Necrotic Leaf Spot Disease Virus Transmitted by Talaromyces flavus Displays Pathogenicity Similar to Apple stem grooving virus Strains

  • The pathogenicity to pear trees and other experimental hosts of the Apple stem grooving virus Korean isolate (ASGV-K) carried by a fungal vector, Talaromyces flavus was examined. ASGV-harboring T. flavus induced mild symptoms on virus-free pears. Symptom severity was intermediate between pears showing typical PBNLS and virus-free pears. Ten cultivars of Phaseolus vulgaris showed 35%-90% infectivity by direct infiltration into leaves and roots by ASGV-harboring T. flavus. Application of fungal cultures to soils showed 0%-70% infectivity depending on the P. vulgaris cultivar. Sap extracted from ASGV-infected Chenopodium quinoa induced similar symptoms on P. vulgaris at 25 days after inoculation. Similar symptoms were also detected on P. vulgaris which were inoculated with ASGV-harboring T.flavus. When healthy P. vulgaris leaves were challenged with sap extracted from P. vulgaris leaves infected with ASGV-harboring T. flavus, typical symptoms were observed. These data suggest that T. flavus mediates the transfer of ASGV to host plants.

하고초 약침이 LPS로 유발된 급성염증 백서 모델에 미치는 영향 (Effects of Prunella vulgaris Pharmacopuncture on Lipopolysaccharide-Induced Acute Inflammatory Rat Model)

  • Objectives : To investigate the anti-inflammatory effects of Prunella vulgaris pharmacopuncture in lipopolysaccharide (LPS)-induced inflammatory rat model. Methods : Sprague-Dawley rats were divided into 5 groups; normal control (n=8), LPS control (n=8), LPS+Prunella vulgaris pharmacopuncture at CV4 (CV4, n=8), LPS+Prunella vulgaris pharmacopuncture at ST36 (ST36, n=8), and LPS+Prunella vulgaris pharmacopuncture at CV12 (CV12, n=8). Pharmacopuncture was given every two days for 4 weeks followed by inflammation induction by peritoneal LPS injection (5mg/kg). Proinflammatory cytokines including interleukin-$1{\beta}$ (IL-$1{\beta}$), interleukin-6 (IL-6), tumor necrosis factor-$\alpha$ (TNF-$\alpha$), interleukin-10 (IL-10), thiobarbituric acid reactive substance (TBARS) from blood and liver tissue were compared before and 5 hrs after inflammation induction. Results : In CV4 and CV12 groups, plasma IL-$1{\beta}$, IL-6 and TNF-$\alpha$ levels increased by LPS injection, significantly decreased 5 hrs after injection (p<0.05). For CV12 group, plasma IL-10 concentration significantly increased (p<0.05). Liver IL-$1{\beta}$ and IL-6 levles significantly decreased in CV4 and CV12 groups (P<0.05), while normal and LPS control groups were not significantly different in TNF-$\alpha$ and IL-10 levels. Plasma TBARS concentration was significantly decreased in CV12 group, while there was no significant difference among LPS control and pharmacopuncture groups for liver TBARS concentration. Conclusions : Based on the present findings, Prunella vulgaris pharmacopuncture at CV12 may have a potentially preventive anti-inflammatory effect in an LPS-induced inflammatory rat model.
