Intersexuality of Crassostrea gigas and Ruditapes philippinarum in Southern Coastal Waters of Korea

  • Objectives : The aim of this study was to verify the intersexuality of Crassostrea gigas and Ruditapes philippinarum in southern coastal waters of Korea. Methods : Specimens of Crassostrea gigas (n=363) were collected from six areas of Tongyeong, Geoje and Yeosu. Ruditapes philippinarum (n=221) was collected from five areas of Yeosu. The sex ratio and intersexuality were determined after observing gonad preparations. Results : The rate of intersexuality in Crassostrea gigas was 16.25%, with females (24.79%; n=30/121) exhibiting a higher rate than males (11.98%; n=29/242). The rate of intersexuality in Ruditapes philippinarum was 24.43%, with females (37.76%; n=37/98) exhibiting a higher rate than males (13.82%; n=17/123). Conclusions : The results of this study suggest that intersexuality is induced by aquatic pollutants such as endocrine disrupting chemicals (EDCs).

당귀 추출물 정맥 주사가 Middle Cerebral Artery Occlusion 모델 흰쥐에서 Gliosis 억제에 미치는 영향 (The Effect of Intravenous Injection of the Water Extract of Angelica gigas Nakai on Gliosis in the Middle Cerebral Artery Occlusion Rats)

  • Objectives : Gliosis becomes physical and mechanical barrier to axonal regeneration. Reactive gliosis induced by middle cerebral artery occlusion is involved with up-regulation of CD81 and GFAP (Glial fibrillary acidic protein). The current study is to examine the effect of the Angelica gigas Nakai(intravenous injection. 100 mg/kg twice in a day) on CD81 and GFAP of the rat in the brain after middle cerebral artery occlusion. Methods : Cerebral infarction was induced by middle cerebral artery occlusion. And after intravenous injection of water extract of Angelica gigas Nakai, the size of cerebral infarction was measured. Examination of optical microscope were also used to detect the expression of CD81 and GFAP in the brain of the rat. Results : The following results were obtained : We found that size of cerebral infarcion induced by MCAO (Middle Cerebral Artery Occlusion) in rats were decreased after intravenous injection of Angelica gigas Nakai. We injected the extract of Angelica gigas Nakai to the MCAO in rats, and the optical microscope study showed that Angelica gigas Nakai had effect on protecting the cells of hippocampus. We found that GFAP, CD81 and ERK of the brain in rats with cerebral infarction after MCAO were meaningfully decreased after intravenous injecting Angelica gigas Nakai. We found that c-Fos expression of the brain in rats with cerebral infarction after MCAO were significantly increased after intravenous injecting Angelica gigas Nakai. Conclusions : These results indicate that Angelica gigas Nakai could suppress the reactive gliosis, which disturbs the astrocyte regeneration in the brain of the rat with cerebral infarction after MCAO by controlling the expression of CD81 and GFAP. And the effect may be modulated by the up-regulation of c-Fos and ERK.

Genome-Wide SSR 마커를 이용한 주요 산지별 참당귀의 유전다양성 분석 (Genetic Diversity of Angelica gigas Nakai Collected in Korea using Genome-Wide SSR Markers)

  • Background: Angelica gigas Nakai has been used as an herbal medicine in Eastern Asia for treating disorders in women for a long time. To date there are no studies on the genetic diversity of A. gigas. The present study aimed to study the genetic diversity of A. gigas variants using genome-wide simple sequence repeat (SSR) markers. Methods and Results: The genetic diversity of 199 variants of A. gigas cultivated in of different regions, was analyzed using 5 genome-wide SSR markers. The results revealed that the genetic variants were very diverse, and genetic analysis using the 5 SSR markers revealed high diversity among the variants. Conclusions: It is expected that the development of the true Angleical cultivar, by studying the system and group selection, can be achieved by genetic analysis using the developed markers, for generating a genetically fixed lineage and group selection.

일부 한약재의 수지상세포 활성화 효과 (Effect of Some Herbal Plant Extracts on the Activation of Dendritic Cells)

  • 본 실험에서는 일부 한약재를 이용하여 수지상세포의 활성화에 미치는 영향을 알아보았다. 그 결과, 실험에 사용한 여섯 가지 한약재 중 선별된 천궁과 당귀는 수지상세포의 항원 제시 능력에 영향을 미쳐, T 세포 증식 반응을 증가시켰고, IL-2와 IFN-r의 분비를 증가시키는 것으로 나타났다. 또한 이러한 T 세포의 활성은 수지상세포의 세포표면 단백질인 MHC classII와 CD86, 그리고 CD11c의 발현 증가에 의한 것임을 확인 할 수 있었다. 이상의 실험 결과, 본 실험에서 선별된 천궁과 당귀는 수지상세포를 활성화시키는 효과가 있는 것으로 생각된다.

당귀 추출물의 피부 흡수 증가를 위한 마이크로에멀젼 조성 (Microemulsion Fomulation for Enhanced Topical Absorption of Root Extract of Angelica gigas)

  • Angelica gigas is one of the most widely used herbal medicines in Asia. Root extract of Angelica gigas is known to have anti-oxidant activity and skin whitening effect. The aim of this study was to prepare microemulsion system of root extracts of Angelica gigas for topical delivery. Microemulsion was successfully prepared by using MCT (medium chain triglyceride) as an oil phase, Labrasol as a surfactant, and the mixture of propyleneglycol and phosphatidylcholine (4 : 1) as a cosurfactant. In vitro and in vivo skin permeation and deposition of decursin, as a marker, was determined using hairless mouse. Microemulsion significantly increased the in vitro skin permeation of decursin for up to 12 hours and was significantly higher than the control (water). Moreover, microemulsion formulation showed significantly higher skin deposition of decursin compared to the control in both in vitro and in vivo studies. Thus, microemulsion could be a useful vehicle for topical application of root extracts of Angelica gigas.

면역증강성 다당 생산을 위한 참당귀 세포배양의 최적조건 (Optimal Conditions for the Production of Immunostimulating Polysaccharides from the Suspension Culture of Angelica gigas Cells.)

  • 참당귀 세포(Angelica gigas H4)를 현탁배양하여 면역 증강성 다당을 생산하기 위한 최적배지 설계 및 생산조건 최적화 연구를 수행하였다. 우선 세포의 성장에 가장 좋은 배지로서 Schenk and Hildebrandt(SH) 배지를 선택하여 배양한 결과 $25^{\circ}C$, 암배양조건에서 각각 15.8g DCW/l와 0.85 g/l의 세포 및 다당을 얻을 수 있었다. 한편 다당 생산용 배지의 개발을 위하여 여러 가지 식물세포배양용 배지 중에서 다당 생산능이 가장 높은 배지로서 Gamborg B5 배지를 선정하였으며 B5 배지를 변형하여 면역증강성 다당생산을 위한 최적배지(PPM)를 개발하였다. 그리고 최적배지를 이용한 당귀 세포증식과 다당 생산을 위한 최적 pH는 6.0-6.5, 배양온도는 $20^{\circ}C$, 그리 고 광조건보다는 암조건이 좋은 것으로 밝혀졌다. 상기 최적조건에서의 최대 세포증식과 다당생산은 각각 14.8 g/l, 1.5 g/l 이었다.

참당귀(Angelica gigas Nakai) 개체군의 환경 특성 분석 (A Study on Environment Characteristics of the Angelica gigas Nakai Population)

  • This study was carried out to investigate vegetation structure and soil properties of Angelica gigas population distributed in Jeombongsan, Bangtaesan, Odaesan, Gyebangsan and Jirisan. From August 2007 until September 2008, 5m${\times}$5m quadrat was established in native area of Angelica gigas in order to record a dominants and coverage, and soil factors at 20 sites. It was found that the altitude in the distributed areas for Angelica gigas population was 710m or more. Angelica gigas population was classified into Parasencio auriculata var. kamtschatical dominant population, Pseudostellaria palibiniana dominant population, Isodon excisus dominant population and typical dominant population. In the site of study, soil pH, electrical conductivity, soil organic matter, available phosphorous, and exchangeable potassium, exchangeable calcium, exchangeable magnesium, exchangeable natrium concentration and total nitrogen were ranged from 5.1~6.2, 0.1~0.79dS/m, 2.21~22.11%, 9.56~37.97mg/kg, 1.85~23.88cmol+/kg, 0.21~1.18cmol+/kg, 1.07~5.09cmol+/kg, 0.04~0.14cmol+/kg and 0.09~1.04% respectively. The Parasencio auriculata var. kamtschatical dominant population was found in area of high altitude and had high slope degree. But Pseudostellaria palibiniana dominant population was found in area of low altitude and low slope degree. Isodon excisus dominant population was found to have higher soil pH and exchangeable potassium concentration than the other dominant population.

조미 자숙굴(Crassostrea gigas) 통조림 및 조미 구운굴(Crassostrea gigas) 통조림의 제조 및 품질특성 (Processing and Characteristics of Canned Seasoned Boiled Oyster Crassostrea gigas and Canned Seasoned Roasted Oyster Crassostrea gigas)

  • The purpose of this study was to collect basic data that could be used in the manufacture of two kinds of canned oyster Crassostrea gigas. The steamed oyster was prepared by shucking after boiling for 6 min at $105^{\circ}C$ and then washing and dewatering. The roasted oyster was prepared by baking steamed oyster at $140^{\circ}C$ for 20 min. The manufacturing methods of canned seasoned boiled oyster and canned seasoned roasted oyster were as follows. The boiled or roasted oyster (50 g) was added to a can (RR-90) along with a mixture of seasoning sauce 40 and then seamed using a vacuum seamer under 20 cm Hg after pre-exhausting at $90^{\circ}C$ for 20 min. The two kinds of canned oyster products produced under sterilization of Fo 12 min were tested for cultured bacteria, external appearance, proximate composition, pH, VBN (Volatile basic nitrogen), TBA (Thiobarbiuric aicd) value, amino-N, salinity, color value sensory evaluation, etc. Results showed that the canned seasoned roasted oyster had higher overall acceptability than the canned seasoned boiled oyster. The reason for this was judged to be that the process of roasting at $140^{\circ}C$ for 20 min influenced the sensory evaluation.

재배지 환경에 따른 참당귀 생육 및 지표성분 함량 비교 (Comparison of Growth and Contents of Active Ingredients of Angelica gigas Nakai under Different Cultivation Areas)

  • 참당귀(Angelica gigas Nakai)는 미나리과(Apiaceae) 당귀속(Angelica L.)에 속하는 다년생 식물로, 뿌리에 효능이 많아 약용 목적으로 재배된다. 본 연구에서는 주로 생산되는 참당귀 재배지의 특성에 대한 연구와 이에 따른 참당귀의 생장 및 지표성분 함량과의 상관관계를 구명하여 참당귀의 안정적인 한약재 수급을 위한 생산량 및 유용성분 함량 증가에 적합한 재배 환경에 대한 정보를 제공하는데 있다. 참당귀의 근직경은 평창 두일리가, 지근 개수는 평창 탑동리가 가장 많았다. 생중량은 평창 탑동리가 가장 높았으며, 건중량도 이와 같은 경향을 보였다. 재배지의 토성은 사질식양토(Sandy Clay Loam)였으며, 유기물 함량은 탑동리에서 3.63%로 가장 높은 함량을 보였다. 전 질소 함량은 봉화 남회룡리에서 0.19%로 가장 높았으며, 유효인산 함량은 고선리에서 높았다. 총 decursin 함량은 두일리에서 5.31%로 가장 높았다. 결과적으로 평창 두일리에서 참당귀의 생육이 가장 우수하였으며, decursin 함량 또한 높았다. 당귀의 수량이나 성분은 재배지역의 환경조건에 크게 영향을 받는데, 해발이 높은 지역에서 재배하는 것이 지표성분 함량과 생산량 증가에 유리하다고 사료되며, 환경조건과 생육 및 지표성분 함량과의 상관관계에 대한 기초자료가 될 것이라고 판단된다. 또한, 최적의 재배환경의 확립과 식물의 생리작용에 대한 연구의 기초가 될 수 있을 것이라고 판단된다.

Determination of Biosynthetic Pathway of Decursin in Hairy Root Culture of Angelica gigas

  • To establish the biosynthetic pathway of decursin in Angelica gigas Nakai, feeding experiment with stable isotope-labeled precursors were conducted. Umbelliferone and decursin were labeled with deuterium at C-3. The umbelliferone, the decursin, and other commercially available putative precursors, L-phenylalanine-ring-$d_5$ and trans-cinnamic acid-$d_7$, were fed to the hairy root culture of A. gigas. Each deuterated compound was incorporated into decursinol, decursinol angelate, and decursin as determined by mass spectrometric analysis. These findings confirmed the coumarin biosynthesis pathway sequence is composed of phenylalanine, cinnamic acid, umbelliferone, decursinol, and decursin.
