Components of human breast milk: from macronutrient to microbiome and microRNA

  • Human breast milk (HBM) is essential for the infant's growth and development right after birth and is an irreplaceable source of nutrition for early human survival. Various infant formulas have many similarities to HBM in many components, but there is no perfect substitute for HBM. Recently, various breast milk components and their roles have been studied according to the development of various analysis techniques. As is already well known, HBM contains about 87%-88% water, and 124-g/L solid components as macronutrients, including about 7% (60-70 g/L) carbohydrates, 1% (8-10 g/L) protein, and 3.8% (35-40 g/L) fat. The composition may vary depending on the environmental factors, including maternal diet. Colostrum is low in fat but high in protein and relatively rich in immuneprotective components. Although HBM contains enough vitamins to ensure normal growth of the infant, vitamins D and K may be insufficient, and the infant may require their supplementation. Growth factors in HBM also serve as various bioactive proteins and peptides on the intestinal tract, vasculature, nervous system, and endocrine system. In the past, HBM of a healthy mother was thought to be sterile. However, several subsequent studies have confirmed the presence of rich and diverse microbial communities in HBM. Some studies suggested that the genera Staphylococcus and Streptococcus may be universally predominant in HBM, but the origin of microbiota still remains controversial. Lastly, milk is the one of most abundant body fluid of microRNAs, which are known to play a role in various functions, such as immunoprotection and developmental programming, through delivering from HBM and absorption by intestinal epithelial cells. In conclusion, HBM is the most important source of nutrition for infants and includes microbiomes and miRNAs for growth, development, and immunity.

Coix lacryma-jobi var. mayuen Stapf Sprout Extract Ameliorates High-Fat Diet-Induced Obesity by Upregulating LKB1/AMPK Signaling (LKB1/AMPK 신호 전달 경로의 활성화로 인한 새싹율무 열수 추출물의 항비만 효과)

  • Objectives : AMP-activated protein kinase (AMPK) is a key metabolic regulator that reduces lipogenesis. AMPK is mainly activated via phosphorylation of liver kinase B (LKB) 1 under energy stress. Here, we highlighted the anti-obesity effect and underlying mechanism of Coix lacryma-jobi var. mayuen Stapf sprout water extract (CSW) sprout extract in connection with the LKB1/AMPK signaling pathway. Methods : C57BL/6 mice (20~25 g) fed HFD to induce obesity and at the same time administered CSW 100 mg/kg (CSWL; (CSWL; CSW low concentration) or CSW 200 mg/kg (CSWH; CSW high concentration) or Garcinia extract (Garcinia) 200 mg/kg orally for 6 weeks. Body weight and food intake were measured at the same time each day. After 6 weeks of CSW administration, liver tissue and serum were obtained through an autopsy. After the end of the experiment, biochemical analysis (triglycerides (TG), total cholesterol (TC), HDL-cholesterol, and LDL-cholesterol) was performed on the serum. And then, protein levels related to TG and TC synthesis were measured through western blot analysis in liver tissue. Results : As a result, serum TG, TC, and LDL-cholesterol levels were significantly increased in the control group and significantly decreased in the CSW administration group. On the other hand, the HDL-cholesterol level was increased in the CSW-administered group. And as a result of Western blot analysis, CSW significantly increased the phosphorylation of LKB1 & AMPK, and remarkably decreased the expression of factors related to TG and TC synthesis. Conclusions : Our findings suggest that CSW influences the TG and TC synthesis to positively affect HFD-induced obesity in C57BL/6 mice.

Effects of Dietary Cholesterol on Male Reproductive Tracts by Regulating PCSK9 Gene (콜레스테롤 식이가 Pcsk9 유전자 조절을 통해 남성 생식기관에 미치는 영향)

  • Proprotein convertase subtilisin/kexin type 9 (PCSK9), is a protein mainly secreted by a liver. The PCSK9 plays an important role in low density lipoprotein (LDL) metabolism acting as a repressor of LDL receptor through transportation of the LDLR to the lysosome for degradation. Thus, the PCSK9 inhibitor suppresses PCSK9-regulated degradation of the LDL receptor as a LDL-lowering medicine. However, little is known about the role of PCSK9 in the reproductive system. Therefore, in the present study, we investigated Pcsk9 expression in male reproductive tracts including penises, prostates and testes using rats in response to their diets between a normal diet and a high-fat diet with cholesterol. Based on our previous study, the high-fat diet elevates concentration of total cholesterol and LDL in serum whereas it reduces the concentration of plasma high density lipoprotein (HDL). In addition, it dramatically affects to morphological changes of the male reproductive organs. Consistent with these results, the expression of Pcsk9 was substantially decreased in the penile tissues (P < 0.001) from rats fed a high fat diet as compared to a normal diet. Moreover, it slightly reduced in the prostate and testes (P < 0.05) of rats in response to a high fat diet. Localization of Pcsk9 was predominantly detected in urethral epithelium of penises, cylinder-shaped cells of prostate glands, and spermatogonia, spermatocytes and spermatid of testes of rats. Collectively, results of current study provide invaluable insights into the Pcsk9 gene with respect to its tissue- and cell-specific expression by a high fat diet with cholesterol.

PARTIAL REPLACEMENT OF GRASS SILAGE WITH WHOLE-CROP CEREAL SILAGE FOR GROWING BEEF CATTLE

  • A study was conducted to investigate the effect of different inclusion levels of urea treated whole-crop wheat silage (UWCWS) in grass silage based rations on the performance of growing beef cattle. The winter wheat (variety, Riband) was harvested (in the summer of 1991) at a dry matter proportion of 520 g/kg and treated with feed grade urea at the rate of 37 kg/tonne crop dry matter and preserved in a heavy duty plastic bag using a silo press. The urea treated whole crop wheat silage (UWCWS) was mixed with grass silage to replace 0.00 (S100), 0.33 (S33) and 0.67 (S67) parts of the forage dry matter and fed ad libitum in a cross over design to 18 Simmental X Holstein Friesian growing beef animals. Two energy sources {one high in starch, rolled barley (RB) and one high in digestible fibre, sugar beet pulp (SBP)} were fed to supply sufficient energy for the efficient use of nitrogen by the rumen micro-organisms. The data on DMIF (dry matter intake of forage), TDMI (total dry matter intake), DLWG (daily live weight gain), FCR (feed conversion ratio) were recorded and faecal samples were collected to determine the digestibility coefficients. Results revealed that with the inclusion of UWCW in the animals' diets the DMI of the forage was significantly increased (p < 0.05). The highest DMIF was found in the treatment "S33" ($6.28{\pm}0.25kg$) where 67% of the silage dry matter was replaced with the UWCW and the lowest value for DMIF was observed in the control treatment ($5.03{\pm}0.23kg$). The DLWG did not differ significantly between the treatments. However, treatment "S100" showed a trend towards a superior DLWG. Feed conversion ratio in the control treatment differed significantly from "S67" and "S33". The addition of the UWCW in the animals' diet resulted in the lower FCR There was no effect of type of energy supplement on any aspect of performance either overall or in interaction with grass silage: UWCWS ratio. The regression and correlation coefficients for DMIF (r = 5.22 + 0.0184x*), DLWG (r = $1.04-0.00086x^{NS}$) and FCR (r = 4.78 = 0.022x*) on the inclusion of UWCW in the diet were calculated. The effect of the inclusion of UWCW on the overall digestibility coefficients was significant (p < 0.05). The addition of the UWCWS in the diet decreased the digestibility of the DM, OM, ADF and NFE but effect on the protein digestibility was non significant. The results of present study suggests that a DLWG slightly over 1 kg can be achieved with UWCW during the store period (period in which animal performance targets are low especially during winter) and the prediction of ME was overestimated as the high intake of DM did not reflect in improved animal performance.

Total Dietary Fiber and Mineral Absorption

  • The consumption of foods rich in TDF should not be associated with impaired mineral absorp-tion and long-term mineral status. In surveys of populations consuming high amounts of TDF e.g Third World populations and vegetarinas gross deficiencies in mineral nutrition have not been noted. If mineral status is low among these groups it is most likely caused by the inadequacy or imbalance of the diet and not by the TDF. The key word is interaction which should be inte-rpreted in dietary imbalances that produce nut-rient deficiencies. There are no strong data to support the concept that TDF inhibits mineral absorption through a binding chelation mechanism. Limited data sug-gest that positively charged groups on polymers such as chitosan and cholestyramine will decrease iron absorption in humans and animals. Because TDF does not contain positively charged groups future research should be directed at the possible role of protein consumed along with TDF and the combination of effects on mineral nutrition Phytic acid is acknowledged as a potent chela-tor of zinc. However its association with zinc and its propensity to lower Zn bioavaiability may enhance the absorption of other elements notably copper and iron. The importance of interactions among nutrients including TDF will gain addi-tional attention in the scientific community. Soluble and insoluble dietary fiber function di-fferently in the intestine. Insoluble fibers accele-rate movement through the intestine. Soluble die-tary fibers appear to regulated blood concentra-tions of glucose and cholesterol albeit by some unknown mechanism. In creased viscosity produ-ced by the SDF in the intestine may provide an explanation of how this class of polymers affects plasma glucose cholesterol and other nutrients. Employing a double-perfusion technique in the rat we demonstrated that viscosity produced by SDF will delay transfer of zinc into the circulatory system. This delayed absorption should not be interpreted as decreased utilization. A great deal of additional research is required to prove the importance of luminaly viscosity produced by SDF on slowing nutrient absorption or regulating bllod nutrient homeostasis. Increased intake of TDF in the total human diet appears desirable. A dietary intake of 35g/day should not be considered to have a negative effect on mineral absorption. It is important to educate people that an intake of more than 35g TDF/day may cause an imbalance in the diet that can adve-rsely affect mineral utilization. Acknowledgments. Customer's perception and preference for Japanese Chain Restaurants in Seoul & Kyunggi Province (서울.경기지역 일식 체인전문점에 대한 인지도와 선호도)

  • The objectives of this study were, 1) to investigate the perception and preference for Japanese chain restaurants due to demographics and consumption behaviors, and 2) to research improvements for Japanese chain restaurants according to the customers' dissatisfaction. Frequency analysis and one-way ANOVA were used to analyze the data. Three hundred questionnaires were distributed and 254 were returned(84.66%). The customers' perception and preference about Japanese chain restaurants were significantly related with each other. The differences of perception and preference due to demographics and consumption behaviors were significant. The most dissatisfied selection attributes were price, number of Korean dishes, number of branch offices, and advertisements, in order. From examining the progressive circmnstances of Korean food-service industry and the social trends toward a preference for healthy, special ethnic food and dishes for diet control and high protein-low fat, it is apparent that food-service businesses related to Japanese food have the potential for success. The results of this study should provide valuable information for administrators and managers in the hospitality industry.

Diet-ralated Factors of Overweight Adolescent Girls (여중생의 비만과 관련된 식이요인에 관한 연구)

  • The purpose of this study was to search the obese related dietary factors of young adolescent girls for developing nutrition education program. In the cross-sectional study of 371 young adolescent girls, 25$\%$ were moderately obese, 13.2$\%$ obese, and a total of 39.1 were over moderately obese. The characteristics of obese girls were as follows. First, obese girls consumed more energy than is needed for physical activity. Their average daily energy intakes were not significantly higher and the daily energy expenditures were significantly lower than those of normal girls. Second, obese girls showed higher intakes of protein and fat, and ate more animal foods than normal girls. Fourth, obese girls skipped meals more often, especially breakfast. They ate less rice. For both normal and obese girls, their common dietary problems were low intake of calcium and iron, and high intake of snack foods. The result of this study shows that nutrition education programs for obese adolescent girls should be designed for eating habits, food balance and moderation. future research is to develop the effective nutrition education program for the obese adolescent girls.

The Positive Effect of LuoHanGuo as Sugar Substitute on Blood Glucose and Metabolism in Streptozotocin-Induced Diabetic Mice

  • This study intended to compare the effects of the sugar-Omija extract tea (SO) and LuoHanGuo-Omija extract tea (LO) to blood glucose levels and metabolism among streptozotocin (STZ)-induced diabetic mice and to prove the positive effects of LuoHanGuo extract as a sugar substitute. According to the results, the rate of blood glucose increase and the blood glucose level was reduced, and the weight was increased. Aspartate aminotransferase (AST), alanine aminotransferase (ALT), triglyceride, low density lipoprotein cholesterol, and total cholesterol was decreased, and albumin, total protein, and high density lipoprotein cholesterol (HDL-C) was increased in the STZ/LO group as compared to the STZ group. Compared to the STZ/SO group, the rate of blood glucose increase and glucose tolerance was decreased. AST, ALT, and total cholesterol was decreased, and HDL-C level was high. In conclusion, the LO displayed hypoglycemic effect, prevented weight loss caused by diabetes, recovered liver function, and improved lipid metabolism. In addition, such positive effects were obviously shown during comparison with the experimental group treated with the SO. Therefore, the LO were considered as favorable food supplements that can be safely taken by persons with diabetes, obesity, hypertension, and heart disease; they have to control their sugar intake as well as their diet.

Patients' Evaluation on Foodservice in University and General Hospitals (대학병원 종합병원 입원환자의 급식에 대한 평가)

  • This study was conducted to investigate and to improve the actual condition of food service for patients in hospitals. For this purpose, questionnaires were distributed to 283 patients admitted to a university hospital and three general hospitals between January 27 and February 15, 1997. The department participated in the study included internal medicine, surgery, obstetrics, orthopedics and so forth. For meal time, 61.1$\%$ of patients wanted to eat breakfast at 8 am, 55.8$\%$ lunch at noon, and 73.5$\%$ dinner at 6 pm. The patients complained about unsatisfactory hospital food itself by 37.8$\%$, about insufficient food amount by 19.6$\%$, about menu with no choice by 41.2$\%$ and about low variety of the meals by 32.7$\%$. Sixty two point nine percent of the subjects enjoyed snacks between meals because of poor appetite at meal time(46.1$\%$), delayed food service(39.9$\%$) and others(11.2$\%$). The types of diet were mainly regular ones(58.6$\%$) with some high protein(12.4%) and diabetic sensitive ones(7.1$\%$). As eating place, the patient's prefered bed(51.9$\%$), room-table(27.2$\%$) and dining room(17.7$\%$). Fifty-five percent of them also wanted hospital foods available to their caring relatives. A Comparison of Herbage Utilization by Sheep on Native and Improved Pastures (면양에 의한 야초지와 개량초지의 초류이용성 비교)

  • The preference, chemical composition, intake, digestibility and chewing efficiency of collected herbage sample from sheep using esophageal fistula and fecal collection method were compared to those of offered herbage samples on the native and improved pastures at the 3rd flush stage(25 cm grass height). Experimental field was treated by one plot design and the begining of September 1987, at Chungnam National University, Daejon. The results obtained are summarized as follows: 1. Preference indices were not different among herbage species on improved pasture, while those on in native pasture were markedly different and this difference seemed to depend on diet selection. 2. Chemical compositions of collected herbage samples were different from those of offered herbage samples. Crude protein content was significantly high on native pasture, but NDF, cellulose and lignin content were significantly low on improved pasture (p < 0.01). 3. DM intake, digestibility and herbage utilization percent on improved pasture were significantly higher than those on native pasture (P < 0.01). 4. Eating rate, ruminating and chewing efficiency were influenced by the amounts of NDF intake. Therefore, eating rate, ruminating and chewing efficiency on improved pasture tended to be higher than those on native pasture (p <0.01).
